Effect of six-month treatment with Ukrain on early osteoporosis induced by ovariectomy in rats. Part II: Preliminary studies of peripheral blood parameters.

Studies on Wistar rats have demonstrated that six months after ovariectomy no changes were observed in peripheral blood morphology other than a decreased leukocyte count. A similar effect was noted in the sham-operated group. The present results show that Ukrain treatment started on the second postoperative day and continued every other day for six months significantly increased leukocyte count in comparison with the untreated ovariectomized and sham-operated groups. The activities of alanine aminotransferase (ALT) and aspartate aminotransferase (AST) were also estimated in this study. In none of the groups studied was any change of ALT activity noted. The activity of AST was significantly increased in the sham-operated and untreated ovariectomized rats. After six-month treatment with Ukrain, the activity of this enzyme was significantly decreased in the serum of untreated ovariectomized rats. In rats pretreated with Ukrain some of the studied parameters of peripheral blood were similar to those of the control group.

[Comparison of prolactin levels in first, second and multiple physiological pregnancy]. / Porównanie zawartosci prolaktyny w ciazy fizjologicznej pierwszej, drugiej i wielokrotnej.

Serum prolactin level was estimated by radioimmunological method (RIA) in women during the first, the second and the multiple normal pregnancy. The value of prolactin was compared in the successive pregnancies and in the successive weeks of pregnancy. The dependence between concentration of prolactin and successive pregnancy was observed. A significant increase of prolactin was noted in the 4 week of multiple pregnancy. The increased level of prolactin was observed in the 10th, 12th, 14th, 40th and 42nd weeks of the first normal pregnancy.

[Alpha-fetoprotein in acute viral hepatitis type A]. / Alfa-fetoproteina w ostrym wirusowym zapaleniu watroby typu A.

Serum AFP levels were estimated by radioimmunoassay (RIA) at the first and the last but one day of hospitalization of 25 patients with acute viral hepatitis of type A. The observed group consisted of 11 women and 9 men. A significant increase in the serum AFP levels was found in the group of patients on the first day of hospitalization. Besides, a rise in the aminotransferases (ALT and AST) activity and in the serum total bilirubin level were observed. On the last but one day of hospitalization the mean value of serum AFP level and the activity of the investigated enzymes were diminished. Mean value of serum AFP level the and activity of aminotransferases were higher in patients than those in the control group. Only serum total bilirubin level was similar to the mean value of control group.

The study was aimed at the determination of serum levels of the investigated hormones (prolactin, gonadotrophins, testosterone and alpha-fetoprotein) in blood donors with antibodies to hepatitis C virus. The observed group consisted of 50 men of age between 22 and 47 years. The blood serum concentrations of hormones were determined by RIA method using the ready made reagent kits. No characteristic changes were observed in the serum hormones levels except for prolactin. Mean value of serum prolactin concentration was higher in blood donors than those in the control group.

Effect of three months treatment with Ukrain on peripheral blood morphology in rodents.

Studies on Albino Swiss mice and Wistar rats have demonstrated that Ukrain administered intraperitoneally (i.p.) for three months produces the following effects in the haematologic parameters: increased leucocytes and decreased thrombocytes. The haemoglobin and erythrocyte levels, as well as haematocrit value, were unchanged. In the leucogram changes were observed; i.e., a fall in the number of neutrophil segments and an increase in the lymphocyte count. The erythrocyte indices, P.C.V., M.C.V., M.C.H. and M.C.H.C. were not changed by a period of three months i.p. Ukrain administration. The observed changes were more marked in female than in male animals and were greater in mice than in rats.

Effect of single and three months treatment with Ukrain on aminotransferases (ALT and AST) and on the serum protein level in rodents.

The influence of Ukrain on the activity of aminotransferases (ALT and AST) and on the serum total protein content was estimated in mice and rats of both sexes receiving single or repeated doses of the drug. It was found that one hour after intraperitoneal (i.p.) administration of Ukrain no characteristic changes were recorded in the activity of the investigated enzymes, or in the serum protein content of animals of either sex. Similar effects were observed after three months treatment with Ukrain in rats of either sex. Only in mice receiving Ukrain for three months was a rise in ALT and AST activity found. No particular changes were observed in the total serum protein level, except for a small decreases in the sera of male mice.

Effect of single and prolonged administration of Ukrain on prolactin concentration in rats.

The effect of single and prolonged administration of Ukrain on the serum prolactin concentrations in rats of both sexes was investigated. One hour after intraperitoneal (i.p.) administration of Ukrain in rats, in doses of 7, 14 and 28 mg/kg, the drug did not affect their serum prolactin concentration. Only in a dose of 28 mg/kg did this drug decrease serum prolactin in a group of female rats. Repeated i.p. treatment (once daily for three months) with 7, 14 and 28 mg/kg of Ukrain significantly increased the serum prolactin concentration in rats of both sexes. The most marked effect was observed in the group of female rats.

Effects of serotonin-active drugs on prolactin secretion in male rats.

The effects of several serotonin acting drugs on serum prolactin levels were studied in male rats. Administration of the serotonin precursor, 5-hydroxytryptophan (5-HTP; 250 mg/kg ip) or pargyline (5 mg/kg ip) with 5-HTP (100 mg/kg ip) significantly increased serum prolactin concentration. Pretreatment of rats with the serotonin agonist, quipazine (10 mg/kg ip) caused increase in serum prolactin levels. Depletion of serotonin stores by pretreatment with p-chloro-phenylalanine methyl-ester (PCPA; 3 x 300 mg/kg ip) significantly prevent the effect of quipazine on prolactin levels. Serotonin uptake blocker, fluoxetine (10 mg/kg ip) produces an increase in the prolactin concentrations of the serum. Administration of the reserpine (2.5 mg/kg sc) did not affect serum prolactin levels. Both serotonin receptor blockers, cyproheptadine (0.5 mg/kg ip) and pizotifen (1 mg/kg ip) caused a week decrease in the serum prolactin levels. Significant changes in concentrations of 5-hydroxytryptamine (5-HT) and 5-hydroxyindoleacetic acid (5-HIAA) in brain of rats pretreated with above serotonin-active drugs were found. These results indicate that serotonergic system can modify secretion of prolactin.
