RESUMO

Vancomycin-resistant enterococci (VRE) poses a formidable challenge to public health due to its inherent resistance to multiple antibiotics coupled with the ability to transfer genetic determinants to dangerous pathogens like Methicillin-resistant Staphylococcus aureus (MRSA). The purpose of this study was to investigate the incidence of vancomycin resistance in enterococci among clinical isolates at a tertiary care military hospital in the eastern region of Saudi Arabia and to detect van genes using multiplex-PCR. Overall, 246 isolates of enterococci were collected from various clinical specimens. The isolates were identified, and antimicrobial susceptibility testing was done using the Vitek 2 system. Multiplex PCR was performed on the VRE isolates, thus identified to determine the van genes harbored. A total of 15 VRE were identified, of which 14 (93.3%) were Enterococcus faecium, and 1(6.7%) was Enterococcus casseliflavus with intrinsic vanC resistance. Of the 14 vancomycin-resistant Enterococcus faecium, 8 (57.1%) harbored vanB genes, while 6 (42.8%) harbored vanA genes. All the VRE were susceptible to linezolid and tigecycline. Our study detected a low prevalence (6.1%) of VRE among clinical isolates of enterococci and that the vanB gene predominates in such strains. Susceptibility profiles indicated that linezolid and tigecycline are still effective against these multidrug-resistant pathogens. Pus specimens yielded the highest percentage (53.3%) of isolates from which VRE was obtained, and this finding is novel among studies done in Saudi Arabia.

Natural algal bloom consists of promising algal species which could be a feasible option for the source of bulk biomass and biodiesel production. It has been found in five natural fresh water algal blooms (Uttar Pradesh, India), containing high nitrogen (N) (4.6 ± 0.32 mg/L) and phosphorus (P) (4.12 ± 0.29 mg/L) concentration during spring (23.9-25.9 °C) and summer season (32.0-35.0 °C). Among the isolated algae from naturally occurring bloom, Chlorella sorokiniana MKP01 exhibited highest biomass (1.02 ± 0.02 g/L) and lipid content (174.1 ± 9.6 mg/L) in untreated tap water and urea/single super phosphate (SSP) in the ratio (2:1). The biodiesel quality was assessed and found to be with the Indian and international standards. Algal bloom was artificially developed in the open pond containing 10,000 l tap water supplemented with Urea/SSP (2:1) for a consistent supply of bulk biomass, yielded 8 kg of total biomass and lipid 1.3 kg.

The present study dealt with biomass, lipid concentration, fatty acid profile and biodiesel properties of microalga Scenedesmus abundans under different phosphate concentrations, pH and light intensities, one at a time. Among different phosphate concentrations, higher biomass (770.10±11.0mg/L) and lipid concentration (176.87±4.6mg/L) were at the concentration of 60mg/L. Light intensity at 6000lux yielded higher biomass and lipid concentration of 742.0±9.7 and 243.15±9.1mg/L, respectively. The biomass (769.0±12.3mg/L) and lipid (179.47±5.5mg/L) concentration were highest at pH 8 and pH 6, respectively. All the culture treatments showed marked effect on the fatty acid profile and biodiesel properties of the extracted oil. FAME derived biodiesel properties were compared with European biodiesel standards (EN 14214), Indian biodiesel standards (IS 15607) and American biodiesel standards (ASTM D 6751-08) to assess the suitability of algal oil as biodiesel feedstock.

The freshwater alga Spirogyra porticalis (Muell.) Cleve, a filamentous charophyte, collected from the Indian trans-Himalayan cold desert, was identified on the basis of morpho-anatomical characters. Extracts of this alga were made using solvents of varying polarity viz. n-hexane, acetonitrile, methanol and water. The antioxidant capacities and phenolic profile of the extracts were estimated. The methanol extract showing highest antioxidant capacity and rich phenolic attributes was further investigated and phytochemical profiling was conducted by gas chromatography-mass spectrometry (GC/MS) hyphenated technique. The cytotoxic activity of methanol extract was evaluated on human hepatocellular carcinoma HepG2 and colon carcinoma RKO cell lines. The anti-hypoxic effect of methanol extract of the alga was tested on in vivo animal system to confirm its potential to ameliorate oxidative stress. The antioxidant assays viz. ferric reducing antioxidant power (FRAP), 2,2'-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS), 1,1-diphenyl-2-picrylhydrazyl (DPPH) and nitric oxide (NO) radical scavenging capacities, ß-carotene-linoleic acid bleaching property and lipid peroxidation exhibited analogous results, wherein the algal extracts showed significantly high antioxidant potential. The extracts were also found to possess high content of total proanthocyanidin, flavonoid and polyphenol. GC/MS analysis revealed the presence of thirteen chemotypes in the methanol extract representing different phytochemical groups like fatty acid esters, sterols, unsaturated alcohols, alkynes etc. with substantial phyto-pharmaceutical importance. The methanol extract was observed to possess anticancer activity as revealed from studies on HepG2 and RKO cell lines. In the present study, S. porticalis methanol extract also provided protection from hypoxia-induced oxidative stress and accelerated the onset of adaptative changes in rats during exposure to hypobaric hypoxia. The bioactive phytochemicals present in this trans-Himalayan alga are of enormous interest and can be utilized sustainably for discovery of novel drugs against oxidative stress.

Present investigation studied the potential of fresh water green microalga Scenedesmus abundans as a feedstock for biodiesel production. To study the biomass and lipid yield, the culture was grown in BBM, Modified CHU-13 and BG-11 medium. Among the tested nitrogen concentration using Modified CHU-13 medium, the highest biomass and lipid yield of 1.113±0.05g/L and 489±23mg/L respectively was found in the culture medium with 0.32g/L of nitrogen (KNO3). Different lipid extraction as well as transesterification methods were also tested. Fatty acid profile of alga grown in large scale indigenous made photobioreactor has shown abundance of fatty acids with carbon chain length of C16 and C18. Various biodiesel properties such as cetane number, iodine value and saponification value were found to be in accordance with Brazilian National Petroleum Agency (ANP255) and European biodiesel standard EN14214 which makes S. abundans as a potential feedstock for biodiesel production.

RESUMO

Cyanobacteria are a rich source of vast array of bioactive molecules including toxins with wide pharmaceutical importance. They show varied bioactivities like antitumor, antiviral, antibacterial, antifungal, antimalarial, antimycotics, antiproliferative, cytotoxicity, immunosuppressive agents and multi-drug resistance reversers. A number of techniques are now developed and standardized for the extraction, isolation, detection and purification of cyanobacterial bioactive molecules. Some of the compounds are showing interesting results and have successfully reached to phase II and phase III of clinical trials. These compounds also serve as lead compounds for the development of synthetic analogues with improved bioactivity. Cyanobacterial bioactive molecules hold a bright and promising future in scientific research and great opportunity for drug discovery. This review mainly focuses on anticancerous, antiviral and antibacterial compounds from cyanobacteria; their clinical status; extraction and detection techniques.

In the course of systematic and ecological studies on algal flora of fresh water environment of three different agroclimatic zone of Uttar Pradesh revealed one hundred eighty two species represented by fifty-two genera inhabiting fresh water bodies having different physico-chemical properties. In both the regions members of order Conjugales were dominant and represented by ninety nine species belonging to fourteen genera. This is followed by Chlorococcales having fifty two species represented by nineteen genera and Chaetophorales with nine species of four genera only. In the central Uttar Pradesh a positive correlation was found between hydrogen ions concentration with temperature and species diversity, while in western Uttar Pradesh a highly positive correlation was found in electrical conductivity and total dissolved solids.

The paper deals with 45 species of 21 genera of fresh water blue green algae (BGA) from three different agro-climatic zones of Uttar Pradesh. Samples were collected from different habitats varying in physico-chemical properties. Out of 45 species, 13 species belonged to order Chroococcales, 31 to order Nostocales, while only 1 species belonged to order Stigonimatales i.e. Fischerella mucicola. The physico-chemical parameters like pH, temperature, dissolved oxygen, electrical conductivity, nitrate, nitrite and rainfall play an important role in the periodicity of BGA. A positive correlation was found between dissolved oxygen (DO) of different ponds and species diversity, except in the case of western region of Uttar Pradesh (Farukhabad and Mahoba districts) where a positive correlation was found in electrical conductivity and total dissolved solids.

Present study was carried out to examine the effect of Butachlor on growth and nitrogen fixation by Anabaena sphaerica. The increased concentration of the pesticide did not have any adverse effect on the alga. Rather it accelerated the algal contribution in terms of biomass and nitrogen fixation.
