RESUMO

Subchronic administration of semicarbazide in the experiment with the rats was used to reduce the formaldehyde level in the organism in order to reveal the interaction between formaldehyde metabolism and biochemical parameters, which define the oxidant-antioxidant system condition and NO metabolism. It has been found that under semicarbazide impact the generation of free radicals, ROS, nitrite and nitrate were enhanced while aldehydes level was reduced that resulted from not only semicarbazide effect like the aldehydes acceptor, but the formaldehyde synthesis slowdown and acceleration of its transformation into format as well. We suppose that formaldehyde plays certain role in the development of connective tissue pathology.

RESUMO

Acetic and succinate acids KoA acyl derivatives interacting with formate were displayed to produce alpha-ketoacids--pyruvate and alpha-ketoglutarate. These acids also interact with formate and make pyruvic and malate acids, while alpha-ketoglutarate, evidently, tricarboxy acids. Interaction of formate with acetic and succinate acids inspite of occurring out of the tricarbone cycle increases the latter metabolic functions.

RESUMO

Catechins and proanthocyanidins have been studied for their effect on initial stage of the protein biosynthesis. It has been found that (+)-catechin in all concentrations studied does not influence the intensity of t-RNA aminoacylation by 14C-glycine and 14C-arginine. Proanthocyanidins, namely, dimers B-3, B-6 and trimer C2 in the concentration of 10(-3) M completely inhibit the initial stage of the protein biosynthesis.

RESUMO

The intensity of biosynthesis processes in animal organism has been studied as affected by long-term administration of morphine. It was established that morphine administration to rats for five weeks intensified protein biosynthesis in the brain, kidneys, skeletal muscles: specific radioactivity of blood serum proteins also increased. Incorporation of 2-/14C/glycine label to the brain, cardiac and skeletal muscles increased as affected by morphine: the label incorporation to the liver lipids decreased and that to the kidney and spleen lipids did not change. Specific radioactivity of glycogen multiply increased in the rat liver as affected by morphine.

RESUMO

Administration of morphine into rats at a dose of 30 mg/kg within 6 days led to a decrease in total rate of tRNA aminoacylation in liver tissue. Content of lactate, pyruvate, malate and alpha-ketoglutarate was decreased within 6 days-long course of morphine administration, while content of lactate was only altered after 5 weeks of the intoxication. Adaptation reactions appear to be increased with time in long-term intoxication with morphine.

RESUMO

After administration of ethanol into rats at a dose of 1.63 g/kg of body mass within two weeks the rate of sRNA aminoacylation was decreased if 14C-leucine and 14C chlorella protein hydrolysate were used. Addition of the alcohol into incubation media caused the ethanol-dependent inhibition of sRNA aminoacylation by valine. leucine, proline, histidine as well as by amino acid mixture from the protein hydrolysate. At the same time, aminoacylation of sRNA by glycine and aspartic acid was unaltered. The rate of sRNA aminoacylation by aspartic acid was increased in presence of 85 mmole of ethanol.

RESUMO

The feeding of excessive leucine and phenylalanine doses to rats against a background of protein deficiency is shown to cause changes in the liver tRNA aminoacylation as well as in the synthesis of acid-soluble skin collagen differing from the normal level in the amino acid composition, elution profiles and content of disulphide groups.

RESUMO

Protein deficiency and tryptophane load against its background lead to the acid-soluble collagen synthesis in the rat skin. The amino acid composition of the collagen differs from the norm. This is accompanied by changes in the free amino acid pool of blood serum and liver, under tryptophane load the free amino acids pool of the liver increasing twice as high. At the same time protein deficiency increases and tryptophane load decreases the level of tRNA amino acylation with tryptophane in the animal liver. Thus, protein deficiency and tryptophane load against its background cause deep changes in the protein biosynthesis.

RESUMO

The valine and methionine loading against a background of protein-free diet evokes interrelated changes in amino acid composition of acid-soluble skin collagen preparations and free amino acids pool of blood serum in rats. The isolated collagen preparations also differ essentially from the normal preparations in the content of disulphide groups and elution profiles.

RESUMO

Optimal conditions are determined for the rat liver tRNA amino acylation with valine and methionine in norm, under protein deficiency and feeding of valine and methionine excess against a background of protein-free diet. It is shown that the level of the liver tRNA amino acylation with the protein deficiency is considerably higher than in the normal state. The valine and methionine excess against a background of protein-free diet inhibits the intensity of the valyl-tRNA and methionyl-tRNA formation. The cross experiments in a heterosystem show that this inhibition is due to changes in the activity or content of valyl- and methionyl-tRNA-synthetases in the rat liver.

RESUMO

It is established that at early stages of the burn disease (deep burn of III-B degree, 20% of body surface) in rats there appears metabolic acidosis, the content of urea increases in the liver and blood serum, a pronounced hydration of the liver and muscles is observed in the burn zone. Administration per os of carbostimulin just a day after burn removes symptoms of metabolic acidosis. When the preparation is fed to rats during 7 days after burn the intensity of radioactive label incorporation into proteins, lipid and glycogen of the liver as well as in proteins and lipids of kidneys and spleen increases.

The effect of the D,L-tryptophan excess in the rat organism was studied as applied to the level of amino acylation of tRNA by 14C-tryptophan. It is established that the introduction of tryptophan for three days in doses of 50 and 75 mg per 100 g of the living weight evokes a 24 and 40% decrease, respectively, in the level of amino acylation of the rat livel tRNA by 14C-tryptophan. The ability of accepting 14C-tryptophan in total preparations of the experimental rats liver tRNA considerably lower as compared to norm. A short-term heating in the presence of magnesium ions partially restores their decreased acceptor ability. This gives ground to suppose that the molecules of tRNAtr in the total preparation of the rat liver tRNA with a great tryptophan excess in the organism partially change their conformation and this is one of the reasons of the decrease in the level of tRNA amino acylation by 14C-tryptophan. The decrease in the protein synthesis at the first stage with the presence of a great tryptophan excess in the organism is evident to be also connected with the inhibition of tryptophanyl-tRNA-synthetase activity.
