RESUMO

In this study, the genetic parameters of major visceral diseases were estimated using the postmortem inspection records of 9057 fattening Japanese Black cattle in Shimane Prefecture, Japan, and the genetic correlation between visceral diseases and carcass traits was analyzed. There were six visceral diseases with a prevalence of 5% or higher, namely, pleurisy, pneumonia, bovine abdominal fat necrosis (BFN), rumenitis, hemorrhagic hepatitis, and perihepatitis. Variance components were estimated using the Gibbs sampling method, and the heritability of the visceral disease ranged from 0.07 to 0.49 for perihepatitis and BFN, respectively. Significant negative genetic correlations were identified between pleurisy and rib thickness (-0.32), BFN and carcass weight (-0.29), and BFN and rib eye area (-0.22). No significant genetic correlation was observed among the visceral diseases. The least squares analysis of variance suggested that some visceral diseases decrease the value of carcass traits. In particular, carcass weight and rib eye area in individuals with BFN were 11.7 kg and 1.87 cm2 lower than those of healthy cattle, respectively. Thus, it was inferred that genetic factors were involved in the visceral diseases of fattening Japanese Black cattle in Shimane Prefecture.

RESUMO

We analyzed the results of a nationwide questionnaire administered to 699 stimulant offenders and examined sex differences in the association between various psychosocial problems and treatment history for substance use disorder. Based on their attributes, we predominantly evaluated the treatment and support provided to women with substance use disorder. The rates of childhood (before the age of 18) traumatic experiences (physical, psychological, and sexual abuse and neglect) and lifetime intimate partner violence were significantly higher in women than in men. The history of treatment for substance use disorder was also significantly higher in women than in men, at 15.8% for men and 42.4% for women [χ2 (1) = 41.223, p < 0.001]. Logistic regression analysis was performed using the treatment history of substance use disorder as the dependent variable. The results showed that treatment history was significantly associated with the total drug abuse screening test-20 score and suicidal ideation in men and with survivors of child abuse and eating disorders in women. A comprehensive assessment is required for several issues, such as child abuse, domestic violence, trauma symptoms, eating disorders, and drug problems. Moreover, an integrated treatment for substance use disorder, trauma, and eating disorders is required for female stimulant offenders.

RESUMO

In this study, we developed a simple method for preparing highly dispersed, stable, and streptavidin (SA)-functionalized carboxymethyl dextran (CMD)-coated melamine nanoparticles (MNPs) in an aqueous buffer at neutral pH. Dynamic light scattering (DLS) revealed the agglomeration of MNPs in an aqueous buffer at neutral pH. When CMD, N-hydroxysuccinimide (NHS), and 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide (EDC) were simultaneously mixed with the MNPs, CMD was bound to the MNPs, promoting their dispersibility. Preparation of SA-CMD-MNPs was accomplished simply by adding SA solution to the CMD-MNPs. The amount of SA bound to the CMD-MNPs was quantified by the bicinchoninic assay, and the amount of SA molecules bound to each CMD-MNP was 417 ± 4. SA-CMD-MNPs exhibited high dispersity (polydispersity index = 0.058) in a neutral phosphate buffer and maintained it for 182 days with dispersion using a probe sonicator (5 s) before DLS characterization. The performance of the SA-CMD-MNPs in biosensing was evaluated by immunohistochemistry, which revealed that the nanoparticles could specifically stain MCF-7 cells derived from breast cancer cells with low HER2 expression. This study provides an effective method for synthesizing highly dispersible nanoparticles for biosensing.

RESUMO

BACKGROUND: Adverse childhood experiences (ACEs) are distressing and/or traumatic events that occur during childhood that increase the risk of negative health outcomes in adulthood. OBJECTIVE: This study estimated the prevalence of ACEs in a nationwide sample of Japanese methamphetamine users in prison and examined associations among ACEs, suicidal ideation, and non-suicidal self-injury. PARTICIPANTS AND SETTING: Participants were 636 inmates (418 male and 218 female) who were newly incarcerated in Japan for Stimulants Control Act violations. METHODS: First, 699 participants completed an anonymous self-report questionnaire. Of these, 636 participants who did not have any missing responses were included in the analysis. After calculating descriptive statistics, the associations between ACEs and suicidal behaviors were assessed using binary logistic regression analyses. RESULTS: Results showed that 76.1 % of the participants reported at least one ACE before the age of 18, and female participants reported a significantly higher number of adversities than their male counterparts. The most common ACEs were parental death or divorce, followed by psychological abuse. Logistic regression analyses revealed that ACE scores significantly increased the risk of suicidal ideation (SI; adjusted odd ratio [AOR] = 1.18, p < .001) and non-suicidal self-injury (NSSI; AOR = 1.18, p < .001) after controlling for possible confounding variables. CONCLUSIONS: Findings suggest the importance of early prevention and intervention for traumatic experiences, and have implications for the recommendation of gender-responsive, trauma-focused interventions, especially for female inmates in the criminal justice system, to break the intergenerational chain of abuse. Future research directions and treatment are discussed.

RESUMO

This study aimed to examine the relationship between methamphetamine (MA) use and high-risk sexual behavior among newly incarcerated prisoners in Japan. We conducted a secondary analysis using cross-sectional survey data, which constituted a nationwide prison sample surveyed between July and November 2017. In total, 699 participants (462 male and 237 female prisoners), who completed the self-administered questionnaire during imprisonment were recruited for the study. The prevalence of high-risk sexual behaviors was as follows: inconsistent condom use (78.4% male, 81.7% female), multiple sexual partners (61.3% male, 41.3% female), history of sexually transmitted diseases (14.1% male, 23.6% female), and trading sex for drugs (15.6% male, 17.7% female). A logistic regression analysis showed that MA use during sex increased the likelihood of high-risk sexual behavior in both male (adjusted odds ratio [AOR] = 5.86; 95% confidence interval [CI] = 3.41-10.07) and female prisoners (AOR = 2.58; 95% CI = 1.33-5.00). Additionally, in female prisoners, a history of sharing MA injection (AOR = 1.60; 95% CI = 1.06-2.42) increased high-risk sexual behavior. Our results indicate that harm reduction programs tailored according to the specific necessities of each gender to reduce high-risk sexual behaviors among MA users should be included in Japanese drug policies.

RESUMO

BACKGROUND: There are a few studies on gender differences in terms of reasons and motives for methamphetamine use. Purpose/Objectives: To examine gender differences in triggers of methamphetamine use Methods: We provided a questionnaire survey to 699 inmates who were incarcerated because of violation of the Stimulants Control Act. Results: The results of DAST-20 (the severity of drug dependence) moderately correlated with the applicable numbers of the 37 situational triggers and 34 emotional triggers for stimulant use, respectively. Regarding gender differences, items including "when having sex" [46.3% (men) vs. 25.3% (women)], "when I have some money" (39.4% vs. 25.3%), and "after getting paid from work" (22.1% vs. 11.4%) were applicable in a significantly higher proportion in males (p < 0.001 and p = 0.008, respectively), whereas items including "worried about the body shape" (3.5% vs. 31.2%) and "after quarreling with someone" (11.7% vs. 29.1%) were applicable in a significantly higher proportion in females (p < 0.001 for both comparisons). Further, most trigger (emotions) items that exhibited gender differences were significantly higher in females. Many females answered that they had used drugs to relieve discomfort symptoms associated with menstrual periods. Conclusions/Importance: Thus, the awareness of important factors including learning how to deal with negative emotions (e.g. depression), repairing self-image, and recognizing physical and emotional changes associated with the menstrual cycle is helpful in providing proper care and treatment of drug addiction in females.

RESUMO

One obstacle in diagnostic pathology is the harmonization of one drug-one diagnostic tests for programmed death ligand-1 (PD-L1). There are many challenges in accurate comparisons of diagnostic tests, such as differences in the titer of each antibody, detection system and dynamic range of visualization. Our previously developed digital immunostaining technique is highly sensitive and quantitative with the ability to quantify particles that bind in a one-to-one fashion with antibody in each cell. Determining the differences in the titer of each antibody with digital immunostaining may be beneficial for future harmonized analysis. To demonstrate the accuracy of digital immunostaining, the present study compared the number of particles with ELISA and nCounter data from five cell lines. NCI-H460 exhib-ited the highest level of PD-L1 protein, followed by A549, PC-3, NCI-H1299, and NCI-H446 cells. In addition, the PD-L1 mRNA values determined by nCounter corresponded with the order of the protein levels determined by ELISA. The present study revealed that digital immunostaining for PD-L1 was highly associated with ELISA and nCounter data. Among the four antibodies tested, the titer of all but SP142 coincided with ELISA and nCounter data. These results indicated that our digital immunostaining technique may be beneficial for future harmonized analysis.

RESUMO

The quantitative sensitivity and dynamic range of conventional immunohistochemistry (IHC) with 3,3'-diaminobenzidine (IHC-DAB) used in pathological diagnosis in hospitals are poor, because enzyme activity can affect the IHC-DAB chromogenic reaction. Although fluorescent IHC can effectively increase the quantitative sensitivity of conventional IHC, tissue autofluorescence interferes with the sensitivity. Here, we created new fluorescent nanoparticles called phosphor-integrated dots (PIDs). PIDs have 100-fold greater brightness and a more than 300-fold greater dynamic range than those of commercially available fluorescent nanoparticles, quantum dots, whose fluorescence intensity is comparable to tissue autofluorescence. Additionally, a newly developed image-processing method enabled the calculation of the PID particle number in the obtained image. To quantify the sensitivity of IHC using PIDs (IHC-PIDs), the IHC-PIDs method was compared with fluorescence-activated cell sorting (FACS), a method well suited for evaluating total protein amount, and the two values exhibited strong correlation (R = 0.94). We next applied IHC-PIDs to categorize the response to molecular target-based drug therapy in breast cancer patients. The results suggested that the PID particle number estimated by IHC-PIDs of breast cancer tissues obtained from biopsy before chemotherapy can provide a score for predicting the therapeutic effect of the human epidermal growth factor receptor 2-targeted drug trastuzumab.

RESUMO

BACKGROUND/AIM: Phagocytes recognize pathogens that enter the body as well as other abnormal and foreign materials that may exist within an organism (such as dead cells, oxidized lipids, and denatured proteins), and phagocytose and eliminate them to maintain a healthy state. In a previous study a simple prototype device was used, under development by Hamamatsu Photonics (Prototype), that detects fluorescence to determine the phagocytic activity of the murine macrophage cell line J774.1. The present study aimed to determine whether it was possible to detect phagocytic activity in a slight amount of human peripheral blood without using hemolysis. MATERIALS AND METHODS: Three microliters of human peripheral blood was drawn from the fingertip and mixed with 30 µg of pH-sensitive fluorescent particles. The fluorescence intensity of the human peripheral blood sample was then measured using the Prototype in development, cultured for 2 h at 37°C, and then re-measured. The phagocytes were observed under fluorescence microscopy and the phagocytosis rate of CD11b-positive cells was verified with a flow cytometer. RESULT: The phagocytic activity of non-hemolyzed human peripheral blood was measured using the Prototype under development; fluorescence after phagocytosis was detected. Furthermore, this was confirmed by both fluorescence microscopy and flow cytometry. The precision of the measurements of human peripheral blood phagocytic activity was verified with the Prototype using samples from three healthy individuals. The relationship between blood sugar levels and phagocytic activity before and after meal times was determined. Concerning exercise, phagocytic activity tended to decrease, although salivary amylase level increased in the healthy individual examined after exercise. CONCLUSION: The simple Prototype can measure phagocytic activity in a small amount of peripheral blood without hemolysis. The device allows for rapid and minimally-invasive detection of changes in phagocytic activity, which has conventionally been difficult. These findings provide promising evidence that assessment of individual phagocytic capacity can be made easier using this novel device.

RESUMO

Research has consistently shown that actuarial measures are superior to unstructured clinical judgments in predicting recidivism of offenders. However, in a non-Western context, clinical judgment may capture contextually relevant risk-related factors. The purpose of the current research is to investigate the incremental value of clinical risk judgment in a sample of Japanese youths. With a sample of 299 released youths, the relative accuracy of the actuarial predictor (Youth Level of Service/Case Management Inventory [YLS/CMI]) and unstructured clinical judgment (recommendation of placement by psychologists) was examined. Results show that an unstructured clinical judgment failed to add incremental variance to the actuarial measure in the prediction of future offenses. The current study results are similar to other studies from North America. Possible reasons why unstructured risk judgment did not add incrementally to the actuarial assessment of risk are discussed. RESUMO

Research on correctional treatment based on the risk principle (Andrews & Bonta, 2010) has found that the intensity of intervention matched with the risk level of offenders' recidivism is successful to reduce recidivism. However, there is no research dealing with this issue in a non-Western context. The purpose of this study was to examine the validity of the risk principle in rehabilitation for Japanese juvenile delinquents. The sample was 321 Japanese male inmates of the juvenile detention and assessment centers. They were followed for an average of 573 days after discharge to assess recidivism. Among high-risk juvenile delinquents, those who were treated in the juvenile training school showed significantly lower rates of recidivism than those who were placed under community supervision, while among low risk juveniles, there was no significant difference in recidivism between the two treatment conditions. The results indicate that the risk principle is valid for the correctional treatment of Japanese juvenile delinquents, suggesting its cross-cultural universality. Implications for juvenile justice policies and directions for future research are discussed.

RESUMO

Treatment completion by violent offenders results in fewer victims and less violence in society. As researchers and members of society, we have a compelling interest in finding ways to keep violent male offenders in effective treatment programs. This study examines file-rated predictors of treatment attrition from an institutionally based program for persistently violent offenders. Each of the three prediction models of institutionally based treatment attrition included the predictors of motivation for assistance and prior treatment dosage: (a) the past criminal behavior model, (b) the recent antisocial behavior model, and (c) the non-antisocial instability model. Recent antisocial behavior did not improve the prediction of treatment attrition over the past criminal behavior model. Motivation for assistance did not make a contribution in the recent antisocial behavior or the non-antisocial instability models while prior treatment dosage consistently contributed to the prediction of attrition across the models. Recent non-antisocial behavior is important to offender treatment attrition.

RESUMO

On X-ray diagnostic technology, it is important to grasp a change of the X-ray high-voltage equipment and radiographic technique factors. The 1st questionnaire was performed in 1974, and it carried out after that every five years, and conducted 8th investigation. As a result, we requested 656 institutions and got a reply from 103 institutions. The response rate was 15.7%. For X-ray high-voltage equipment, the inverter-type device shifts to 83.6% from 73.4% of last time. X-ray high-voltage equipment will be shifted to inverter-type device the near future. For X-ray tube device, the target angle becomes more smaller than usual, and maximum anode heat content is increasing tendency. The spread of digital devices is being advanced, and especially the flat panel detector (FPD) increases in the devices. The spread of soft copy diagnoses is 73.8% for chest image diagnoses, and 49.5% for breast image diagnosis. For the device management, the ratio of institutions measured at purchase time was 91.2%. But, the ratio of institutions performed an invariability examination was 58.4%. It is required to grasp the performance of an X-ray equipment and peripheral equipment, and to perform accuracy control in order to obtain proper radiographic technique factors and imaging. In order to use it for the improvement in photography technology, we would like to continue to conduct this investigation periodically.

RESUMO

The main purpose of the current research is to examine the applicability of the Youth Level of Service/Case Management Inventory (YLS/CMI) in a Japanese juvenile offender population. Three hundred eighty-nine youths who were released from the five Juvenile Classification Homes were followed for approximately one and half years. Results show that the YLS/CMI total score significantly predict recidivism. Survival time analysis demonstrates that the YLS/CMI total score also significantly predicted faster time to recidivism. The overall findings support adequate predictive validity of the YLS/CMI total score, but subscales lacked content representativeness and predictive validity in this sample. Canadian and Japanese cultural differences in criminal history and substance abuse are contributors to the lack of content representativeness.

RESUMO

Picture naming was investigated primarily to determine its dependence on certain imagery-related variables, with a secondary aim of developing a new set of Japanese norms for 360 pictures. Pictures refined from the original Nishimoto, Miyawaki, Ueda, Une, and Takahashi (Behavior Research Methods 37:398-416, 2005) set were used. Naming behaviors were measured using four imagery-related measures (imageability, vividness, image agreement, and image variability) and four conventional measures (naming time, name agreement, familiarity, and age of acquisition), as well as a number of other measures (17 total). A simultaneous multiple regression analysis performed on naming times showed that the most reliable predictor was H, a measure of name diversity; two image-related measures (image agreement and vividness) and age of acquisition also contributed substantially to the prediction of naming times. The accuracy of picture naming (measured as name agreement) was predicted by vividness, age of acquisition, familiarity, and image agreement. This suggests that certain processes involving mental imagery play a role in picture naming. The full set of norms and pictures may be downloaded from http://www.psychonomic.org/archive/ or along with the article from http://www.springerlink.com .

RESUMO

Increased activity of Ser/Thr protein phosphatases types 1 (PP1) and 2A (PP2A) during maladaptive cardiac hypertrophy contributes to cardiac dysfunction and eventual failure, partly through effects on calcium metabolism. A second maladaptive feature of pressure overload cardiac hypertrophy that instead leads to heart failure by interfering with cardiac contraction and intracellular transport is a dense microtubule network stabilized by decoration with microtubule-associated protein 4 (MAP4). In an earlier study we showed that the major determinant of MAP4-microtubule affinity, and thus microtubule network density and stability, is site-specific MAP4 dephosphorylation at Ser-924 and to a lesser extent at Ser-1056; this was found to be prominent in hypertrophied myocardium. Therefore, in seeking the etiology of this MAP4 dephosphorylation, we looked here at PP2A and PP1, as well as the upstream p21-activated kinase 1, in maladaptive pressure overload cardiac hypertrophy. The activity of each was increased persistently during maladaptive hypertrophy, and overexpression of PP2A or PP1 in normal hearts reproduced both the microtubule network phenotype and the dephosphorylation of MAP4 Ser-924 and Ser-1056 seen in hypertrophy. Given the major microtubule-based abnormalities of contractile and transport function in maladaptive hypertrophy, these findings constitute a second important mechanism for phosphatase-dependent pathology in the hypertrophied and failing heart.

RESUMO

Our purpose in the present study is to provide a normative set of nonsensical pictures known as droodles and to demonstrate the role of semantic comprehension in facilitating recall of pictorial stimuli. The set consists of 98 pairs of droodles. Experiment 1 standardized these pictorial stimuli with respect to several variables, such as appropriateness of verbal labels, relationship between two droodles, and correct recall. Appropriateness of verbal labels was rated higher for pictures presented in pairs than for pictures presented singly. Experiment 2 used the standardized set of droodles in a recall experiment similar to those of Bower, Karlin, and Dueck (1975) and others. As we expected, semantic interpretation can strongly facilitate recall. Multiple regression analysis showed that several measures had significant power of explanation for recall performance. The full set of norms and pictures from this article may be downloaded from http://brm.psychonomic-journals.org/content/supplemental.

RESUMO

Contrast enhanced magnetic resonance angiography (CE-MRA) is a common method for imaging the Adamkiewicz Artery (AKA). Test-bolus imaging methods have long been the mainstream, and recently, a time resolved technique (4D-MRA) is also reported. However, the current rates of detection for both are mixed, and the best method has not been established yet. For AKA, the present rate is 72% left, Th8~L1 91% that exists between, with a vessel diameter of 0.5 to 1.5 mm. Additionally, the AKA branch is known to have large individual differences, and thus, a wide range of high spatial resolution imaging. At our institution from January 2009, a 3 T system is installed, and 4D-MRA (time-resolved angiography with interleaved stochastic trajectories: TWIST) imaging became possible. TWIST is a sequence to balance the temporal resolution and spatial resolution, and is considered optimal for the imaging of AKA. In accordance with this idea, the first group of seven patients was imaged by TWIST, but in all cases, the results were not depicting AKA. In subsequent cases, the technique was changed to a 3D-gradient echo sequence with a Bolus-tracking method (Care bolus), and good results were obtained. In conclusion, AKA TWIST was not thought to depict the relationship between specific parameters. When using TWIST for imaging AKA, we will need to find out why it was not visualized, by examining AKA with simulated vessel phantoms, TWIST and Care Bolus. Furthermore, we report on our visual evaluation of the Retrospective clinical pictures.

RESUMO

The gamma-Ga(2)O(3)-Al(2)O(3) mixed oxides with a spinel structure were prepared by the solvothermal reaction of gallium acetylacetonate and aluminum isopropoxide in diethylenetriamine. In the crystal structures of the catalysts obtained by the calcination of these mixed oxides, Ga(3+) and Al(3+) ions preferentially occupied tetrahedral and octahedral sites, respectively. The catalysts with low Ga contents had a unique structure with high surface areas and a concentration gradient of decreasing Ga content from the surface to the bulk. In methane-selective catalytic reduction (SCR) of NO, higher NO conversion to N(2) was attained on the catalyst with high occupation of Ga(3+) ions at tetrahedral sites and Al(3+) ions at octahedral sites. For the gamma-Ga(2)O(3)-Al(2)O(3) mixed oxide with a charged Ga molar content of 0.3 (ST(0.3)), tetrahedral and octahedral sites were solely occupied by Ga(3+) and Al(3+) ions, respectively, and the catalyst exhibited the highest NO conversion to N(2). Therefore, it was concluded that the active site for methane-SCR of NO is tetrahedral Ga(3+) ion and octahedral Al(3+) ion, which are linked to each other. Nitrogen monoxide is adsorbed on the isolated hydroxyl group attached to Al(3+) ions and then oxidized by O(2) yielding surface nitrate species. Tetrahedral Ga(3+) ions work as Lewis acid sites for the activation of methane because of their coordinative unsaturation. The Ga(3+) ions in the gamma-Ga(2)O(3)-Al(2)O(3) catalyst have a redox property, which plays important roles in both the oxidation of NO to surface nitrate species and the activation of methane. The most important factor for this catalyst is that the sites for the formation of surface nitrate species reside next to the methane activation sites, which facilitates the reaction between surface nitrate species and the activated species derived from methane, thus mitigating the consumption of methane by simple combustion with O(2). Therefore, ST(0.3), which has the largest number of ensembles of the tetrahedral Ga(3+) ions and octahedral Al(3+) ions, shows the highest activity for methane-SCR of NO.

RESUMO

Postrelease studies can provide data with which to evaluate expectations based on prerelease tests of biological control insects. In 2004, we observed Trichosirocalus horridus Panzer (Coleoptera: Curculionidae), the rosette weevil introduced into North America against Eurasian thistles, feeding on native tall thistle, Cirsium altissimum L. Spreng., in tallgrass prairie. In this study, we examined the rosette weevil's use of tall thistle, compared with its use of the co-occurring exotic bull thistle, C. vulgare (Savi) Tenore. For both thistle species, we quantified weevil frequency, abundance, and seasonal variation in incidence, using both timed observations at two sites over two growing seasons (2004, 2005) and dissections of thistle flowering shoots from 13 sites (2005). Based on prerelease information, we expected the Eurasian thistle to be the quantitatively preferred host plant for this Eurasian weevil. Instead, we found that both the frequency of infestation and the mean number of adult rosette weevils per plant were at least as high, and sometimes higher, on the native thistle as on the exotic thistle. Furthermore, adult weevil phenology coincided on the two host species. This study provides new quantitative evidence of nontarget feeding by another weevil released for thistle biological control; and it raises important questions for further research. We conclude that continued new releases, as well as augmentation of existing populations, of T. horridus should wait until more research is done on the impact of the nontarget occurrence now reported for this biological control insect.
