RESUMO

Spatial cognition research requires behavioral paradigms that can distinguish between different navigational elements, such as allocentric (map-like) navigation and egocentric (e.g., body centered) navigation. To fill this need, we developed a flexible experimental platform that can be quickly modified without the need for significant changes to software and hardware. In this paper, we present this inexpensive and flexible behavioral platform paired with software which we are making freely available. Our behavioral platform serves as the foundation for a range of experiments, and although developed for assessing spatial cognition, it also has applications in the nonspatial domain of behavioral testing. There are two components of the software platform, "Maze" and "Stim Trigger." While intended as a general platform, presently both programs can work in conjunction with Neuralynx and Open Ephys electrophysiology acquisition systems, allowing for precise time stamping of neural events. The Maze program includes functionality for automatic reward delivery based on user defined zones. "Stim Trigger" permits control of brain stimulation via any equipment that can be paired with an Arduino board. We seek to share our software and leverage the potential by expanding functionality in the future to meet the needs of a larger community of researchers.

Healthcare systems worldwide have been stressed to provide sufficient resources to serve the increasing and aging population in our society. The situation became more challenging at the time of pandemic. Technology advancement, especially the adoption of wearable health monitoring devices, has provided an important supplement to current clinical equipment. Most health monitoring devices are rigid, however, human tissues are soft. Such a difference has prohibited intimate contact between the two and jeopardized wearing comfortableness, which hurdles measurement accuracy especially during longtime usage. Here, we report a soft and stretchable photodiode that can conformally adhere onto the human body without any pressure and measure cardiovascular variables for an extended period with higher reliability than commercial devices. The photodiode used a composite light absorber consisting of an organic bulk heterojunction embedded into an elastic polymer matrix. It is discovered that the elastic polymer matrix not only improves the morphology of the bulk heterojunction for obtaining the desired mechanical properties but also alters its electronic band structure and improves the electrical properties that lead to a reduced dark current and enhanced photovoltage in the stretchable photodiode. The work has demonstrated high fidelity measurements and longtime monitoring of heat rate variability and oxygen saturation, potentially enabling next-generation wearable photoplethysmography devices for point-of-care diagnosis of cardiovascular diseases in a more accessible and affordable way.

Premise: The entire life cycle of ferns has been documented, yet their life histories are still poorly understood. In particular, the phenology of fern gametophytes remains largely unknown. To address this issue, we demonstrated a new ecological approach to explore the phenological link between spore release and gametophyte maturation within the life history of a tree fern species. Methods: We conducted a serial survey of Alsophila podophylla gametophyte abundance in the field, and recorded the time of its spore release. Every two months for one year, all terrestrial fern gametophytes in an unsampled subplot were collected and identified using tissue-direct PCR. Results: We found temporal differences in gametophyte abundances, with a sevenfold difference between the highest and lowest months. The number of spores released was linked to the gametophyte abundance two months later. The switch from gametophyte to juvenile sporophyte was found to be most correlated with precipitation. Discussion: The observed fluctuation in gametophyte abundance and population structure was likely associated with the phenology of spore release and environmental factors. Importantly, these findings provide the first evidence of phenological links between different developmental stages in a fern's life history.

The measurement of the size and timing of meals provides critical insight into the processes underlying food intake. While most work has been conducted with a single food or fluid, the availability of food choices can also influence eating and interact with these processes. The 5-Item Food Choice Monitor (FCM), a device that continuously measures eating and drinking behaviors of rats provided up to 5 foods and 2 fluids simultaneously, was designed to allow study of food choices simultaneously with meal patterns. To validate this device, adult male and female (n = 8 each) Sprague-Dawley rats were housed in the FCM. Food and fluid intake were measured continuously (22-h/day) while rats were presented water and powdered chow. Then a cafeteria diet of 5 foods varying in macronutrient content, texture, and flavors were offered along with water. Lastly, the 5 foods were offered along with 0.3 M sucrose and water. Analyses were conducted to find optimal criteria for parceling ingestive behavior into meals, and then meal patterns were quantified. Total intake, as assessed by FCM software, was in good concordance with that measured by an independent scale. A minimum meal size of 1 kcal and a meal termination criterion of 15-min accounted for >90% of total intake and produced meal dynamics that were in register with the literature. Use of the cafeteria diet allowed comparisons between meal patterns with a single food versus a multi-food diet, as well as analyses of macronutrient-related food choices across subsets of meals. The FCM proved to accurately measure food intake over a 22-h period and was able to detect differences and similarities in the meal patterns of rats as a function of sex and food choice availability. Combined with any number of experimental manipulations, the FCM holds great promise in the investigation of the physiological and neural controls of ingestive behavior in a dietary environment that allows food choices, more closely emulating human eating conditions.

Organelle genomes of land plants are predominately inherited maternally but in some cases can also be transmitted paternally or biparentally. Compared to seed plants (>83% genera of angiosperms and >12% genera of gymnosperms), plastid genome (plastome) inheritance has only been investigated in fewer than 2% of fern genera, and mitochondrial genome (mitogenome) from only one fern genus. We developed a new and efficient method to examine plastome and mitogenome inheritance in a fern species-Deparia lancea (Athyriaceae, Aspleniineae, Polypodiales), and found that plastid and mitochondrial DNAs were transmitted from only the maternal parentage to a next generation. To further examine whether both organelle genomes have the same manner of inheritance in other Deparia ferns, we sequenced both plastid and mitochondrial DNA regions of inter-species hybrids, and performed phylogenetic analyses to identify the origins of organellar DNA. Evidence from our experiments and phylogenetic analyses support that both organelle genomes in Deparia are uniparentally and maternally inherited. Most importantly, our study provides the first report of mitogenome inheritance in eupolypod ferns, and the second one among all ferns.

Intraventricular hemorrhage (IVH) is a common occurrence in the days immediately after premature birth. It has been correlated with outcomes such as periventricular leukomalacia (PVL), cerebral palsy and developmental delay. The causes and evolution of IVH are unclear; it has been associated with fluctuations in blood pressure, damage to the subventricular zone and seizures. At present, ultrasound is the most commonly used method for detection of IVH, but is used retrospectively. Without the presence of adequate therapies to avert IVH, the use of a continuous monitoring technique may be somewhat moot. While treatments to mitigate the damage caused by IVH are still under development, the principal benefit of a continuous monitoring technique will be in investigations into the etiology of IVH, and its associations with periventricular injury and blood pressure fluctuations. Electrical impedance tomography (EIT) is potentially of use in this context as accumulating blood displaces higher conductivity cerebrospinal fluid (CSF) in the ventricles. We devised an electrode array and EIT measurement strategy that performed well in detection of simulated ventricular blood in computer models and phantom studies. In this study we describe results of pilot in vivo experiments on neonatal piglets, and show that EIT has high sensitivity and specificity to small quantities of blood (<1 ml) introduced into the ventricle. EIT images were processed to an index representing the quantity of accumulated blood (the 'quantity index', QI). We found that QI values were linearly related to fluid quantity, and that the slope of the curve was consistent between measurements on different subjects. Linear discriminant analysis showed a false positive rate of 0%, and receiver operator characteristic analysis found area under curve values greater than 0.98 to administered volumes between 0.5, and 2.0 ml. We believe our study indicates that this method may be well suited to quantitative monitoring of IVH in newborns, simultaneously or interleaved with electroencephalograph assessments.

In recent years, to circumvent the interpretive limitations associated with intake tests commonly used to assess taste function in rodents, investigators have developed devices called gustometers to deliver small volumes of taste samples and measure immediate responses, thereby increasing confidence that the behavior of the animal is under orosensory control. Most of these gustometers can be used to measure unconditioned licking behavior to stimuli presented for short durations and/or can be used to train the animal to respond to various fluid stimuli differentially so as to obtain a reward and/or avoid punishment. Psychometric sensitivity and discrimination functions can thus be derived. Here, we describe a new gustometer design, successfully used in behavioral experiments, that was guided by our experience with an older version used for over 2 decades. The new computer-controlled gustometer features no dead space in stimulus delivery lines, effective cleaning of the licking substrate, and the ability to measure licking without passing electrical current through the animal. The parts and dimensions are detailed, and the benefits and limitations of certain design features are discussed. Schematics for key circuits are provided as supplemental information. Accordingly, it should be possible to fabricate this device in a fashion customized for one's needs.

We have developed a sensitive EIT protocol for detection of intraventricular bleeding. A common model of human neonates is the neonatal piglet. We used our method to test the sensitivity of our method and device to small amounts of blood-like fluid injected near the left and right ventricles of a piglet cadaver. Comparing blood-like fluid detection in open an closed piglet skulls, we found that we could detect amounts of blood less than 0.5 ml, which is smaller than that required for our target of detecting grade II intraventricular hemorrhages in human neonates.

The electrical impedance tomography (EIT) image reconstruction problem is ill posed and spatially variant. Because of the problem's ill-posed nature, small amounts of measurement noise can corrupt reconstructed images. The problem must be regularized to reduce image artifacts. In this paper, we focus on the spatially variant characteristics of the problem. Correcting errors due to spatial variance should improve reconstruction accuracy. In this paper, we present methods to normalize the spatially variant image reconstruction problem by equalizing the point spread function (PSF). In order to equalize the PSF, we used the reconstruction blurring properties obtained from the sensitivity matrix. We compared three mathematical normalization schemes: pixel-wise scaling (PWS), weighted pseudo-inversion (WPI) and weighted minimum norm method (WMNM) to equalize images. The quantity index (QI), defined as the integral of pixel values of an EIT conductivity image, was considered in investigating spatial variance. The QI values along with reconstructed images are presented for cases of two-dimensional full array and hemiarray electrode topologies. We found that a spatially invariant QI could be obtained by applying normalization methods based on equalization of the PSF using conventional regularized reconstruction methods such as truncated singular value decomposition (TSVD) and WMNM. We found that WMNM normalization applied to WMNM regularized reconstruction was the best of the methods tested overall, for both hemiarray and full array electrode topologies.

Intraventricular haemorrhage is a common cause of death in premature human infants. As preventative measures and treatments become available, a method for monitoring and detection is required. Electrical impedance tomography (EIT) is a viable monitoring method compared to modalities such as ultrasound, MRI or CT because of its low cost and contrast sensitivity to blood. However, its sensitivity to blood may be obscured by the low conductivity skull, high conductivity cerebrospinal fluid (CSF) and shape changes in the head and body. We estimated the sensitivity of three 16-electrode and impedance measurement configurations to bleeding using both idealized spherical and realistic geometry three-dimensional finite element models of the neonatal head. Sensitivity distribution responses to alterations in skull composition as well as introduction of conductivity anomalies were determined. Of the three patterns tested, a measurement scheme that employed electrodes at locations based on the 10-20 EEG layout, and impedance measurements involving current return over the anterior fontanelle produced superior distinguishabilities in regions near the lateral ventricles. This configuration also showed strongly improved sensitivities and selectivities when skull composition was varied to include the anterior fontanelle. A pattern using electrodes placed in a ring about the equator of the model had similar sensitivities but performed worse than the EEG layout in terms of selectivity. The third pattern performed worse than either the Ring or EEG-based patterns in terms of sensitivity. The overall performance of the EEG-based pattern on a spherical homogeneous model was maintained in a sensitivity matrix calculated using a homogeneous realistic geometry model.
