In recent years, the free-range system for laying hens has increased, driven by societal sensitivity to animal welfare. This study aimed to comparatively analyze the total lipid, cholesterol, and fatty acid composition of egg yolks of late-age laying hens reared in enriched cages (C) and the free-range system (FR). Eggs were collected from Lohmann Brown Classic hens at the 68th, 70th, 72nd, and 74th week of age. The concentrations of total lipids and cholesterol were not affected (p > 0.05) by either factor. Egg yolk from the FR group showed lower (p < 0.01) monounsaturated fatty acids and higher (p < 0.01) polyunsaturated fatty acid (PUFA) compared with that of the C group. From a nutritional point of view, the PUFA n-6/n-3 and the PUFA/SFA ratios of egg yolk from the FR group were favorably lower and higher (p < 0.01) compared with the C one. Conversely, hen age did not affect (p > 0.05) the fatty acid composition of yolks. Interactions between factors were found for total n-3 and n-6 PUFA and the n-6/n-3 ratio (p < 0.01), as well as the thrombogenic index (p < 0.05). In conclusion, the results confirmed that the free-range system may improve the nutritional yolk fatty acid profile and its positive impact on human health.

The present study aimed to test the synbiotic PoultryStar® solUS delivered in ovo to evaluate its effect on hatchability, productive performance and meat quality, compared to its post-hatch administration in water. On the twelfth day of embryonic incubation, 1200 fertile eggs were divided into synbiotic groups injected with 2 mg/embryo (T1) and 3 mg/embryo (T2), a saline group injected with physiological saline and an uninjected control group (C). After hatching, 120 male chicks/group were reared and chicks from the saline group were supplemented with the synbiotic via drinking water (T3). Hatchability was low in both T1 and T2 groups. Growth performance was not affected by the treatments. However, in the second rearing phase (15-36 days), birds from the C and T3 groups were heavier than T1 birds, due to a higher feed intake and daily weight gain. Neither route of synbiotic administration influenced final body weight (at 56 days), weight and yield of the carcass or commercial cuts. Physico-chemical properties, total lipid, cholesterol and fatty acid composition of breast muscle were not affected by the treatments. Considering its exploratory nature, this study has raised many questions that need further investigation, such as the bioactive combination and the effect on embryonic development.

A study was carried out to evaluate the effect of the total replacement of flaked soybean (Glycine max L., SOY) with raw pea (Pisum sativum L., PEA) on the carcass and meat quality traits of two medium-growing broiler strains (Kabir Rosso Plus, KB; New Red, NR). Birds were housed in 20 pens (five replications/groups, six birds each). At 83 days of age, 40 birds (2/replication) were slaughtered and the pectoral muscle (PM) was removed for analyses. Diet did not affect slaughter weight, carcass traits and meat quality. A pea diet determined a significant increase of MUFA and a decrease of PUFA, n-3 and n-6 PUFA; hence, the pea-fed group had a lower PUFA/SFA and a higher n-6/n-3 ratios compared to the soy-fed. NR chickens were heavier, with higher carcass and cut weights (p < 0.01) compared to KB chickens. Interactions (p < 0.05) between factors were found for PM weight and yield. Meat from NR had a higher (p < 0.05) pH. Fatty acids were slightly affected by genotype. Replacing soybean with pea adversely affects meat fatty acid composition in terms of nutritional profile.

The effects of Glycine max L. (SOY) vs. Pisum sativum L. (PEA) in the diet were investigated on in vivo performance of two medium-growing broiler genotypes and on environmental impact. Sixty Kabir Red Plus and sixty New Red chickens were randomly allocated in 20 pens (n = 6 birds per pen). Each pen, i.e., experimental unit, received 1.18 kg dry matter (DM) including soybean (3.39%) in SOY, or pea bean (6.78%) in PEA groups. DM intake, DM refusals and bodyweight (BW) were recorded on pen basis, and average daily gain (ADG) and feed conversion rate (FCR) were calculated. Data on in vivo performance were processed by ANOVA General Linear Model followed by Student−Newman−Keuls post hoc test. Greenhouse gas (GHG) emissions were evaluated on a year basis by GLEAM-i software. The diet did not affect (p > 0.05) DM intake and DM refusals, BW, ADG and FCR. Diet x genotypes affected (p < 0.05) DM intake and DM refusals. PEA diet, cheaper than SOY diet, allowed the reduction in total GHG and CO2, and of meat emission intensity. However, an increase in total CH4 and N2O emissions was observed. The replacement of soybean with pea bean can represent a possible management strategy to reach trade-off between good farming practices and environmental protection on small-scale poultry farms of inner Mediterranean areas.

Galactooligosaccharides (GOS) that are delivered in ovo improve intestinal microbiota composition and mitigate the negative effects of heat stress in broiler chickens. Hubbard hybrids are slow-growing chickens with a high resistance to heat. In this paper, we determined the impact of GOS delivered in ovo on slow-growing chickens that are challenged with heat. The experiment was a 2 × 2 × 2 factorial design. On day 12 of incubation, GOS (3.5 mg/egg) was delivered into the egg (n = 300). Controls (C) were mock-injected with physiological saline (n = 300). After hatching, the GOS and C groups were split into thermal groups: thermoneutral (TN) and heat stress (HS). HS (30 °C) lasted for 14 days (days 36-50 post-hatching). The spleen (n = 8) was sampled after acute (8.5 h) and chronic (14 days) HS. The gene expression of immune-related (IL-2, IL-4, IL-6, IL-10, IL-12p40, and IL-17) and stress-related genes (HSP25, HSP90AA1, BAG3, CAT, and SOD) was detected with RT-qPCR. Chronic HS up-regulated the expression of the genes: IL-10, IL-12p40, SOD (p < 0.05), and CAT (p < 0.01). GOS delivered in ovo down-regulated IL-4 (acute p < 0.001; chronic p < 0.01), IL-12p40, CAT and SOD (chronic p < 0.05). The obtained results suggest that slow-growing hybrids are resistant to acute heat and tolerant to chronic heat, which can be supported with in ovo GOS administration.

This study investigated the effects of a galactooligosaccharide (GOS) prebiotic in ovo injected on intestinal transcriptome and plasma immune parameters of broiler chickens kept under thermoneutral (TN) or heat stress (HS) conditions. Fertilized Ross 308 eggs were injected in ovo with 0.2 mL physiological saline without (control, CON) or with 3.5 mg of GOS (GOS). Three-hundred male chicks/injection treatment (25 birds/pen) were kept in TN or HS (30 °C) conditions during the last growing phase, in a 2 × 2 factorial design. At slaughter, from 20 birds/injection group (half from TN and half from HS), jejunum and cecum were collected for transcriptome analysis, and plasma was collected. No differences in plasma parameters (IgA and IgG, serum amyloid) and no interaction between injection treatment and environment condition were found. GOS-enriched gene sets related to energetic metabolism in jejunum, and to lipid metabolism in cecum, were involved in gut barrier maintenance. A homogeneous reaction to heat stress was determined along the gut, which showed downregulation of the genes related to energy and immunity, irrespective of in ovo treatment. GOS efficacy in counteracting heat stress was scarce after ten days of environmental treatment, but the in ovo supplementation modulates group of genes in jejunum and cecum of broiler chickens.

A study was carried out to assess the efficacy of prebiotic delivered in ovo on performance, carcass traits, meat quality and gut health in the face of a natural coccidiosis infection in Kuroiler chickens. On d 12 of incubation, 150 fertile eggs were divided into a prebiotic group injected with trans-galactooligosaccharides (Bi2tos) and a control group uninjected. Hatched chicks from each group were further divided: One group received antibiotic chick formula while the other was left untreated, giving rise to 4 groups-Control (C), Antibiotic (A), Bi2tos (B), and Bi2tos + Antibiotic (AB). Prebiotic improved growth performance at six weeks of age, AB birds were the heaviest at the end of the rearing period. The highest intestinal lesion scores and oocyst counts were recorded in C birds. B group had a slightly higher carcass weight and cuts yields tended to be higher in treated groups compared to C. Meat from B group displayed a higher amount of polyunsaturated fatty acids compared to C and a positively lower n-6/n-3 ratio compared to C and A. In conclusion, prebiotics with or without antibiotics reduced severity of intestinal lesions and oocyst excretion induced by natural infection with Eimeria, with positive effects on Kuroiler chicken productive traits.

Intestinal mucosa is the interface between the microbial content of the gut and the host's milieu. The goal of this study was to modulate chicken intestinal microflora by in ovo stimulation with galactooligosaccharides (GOS) prebiotic and to demonstrate the molecular responses of the host. The animal trial was performed on meat-type chickens (Ross 308). GOS was delivered by in ovo injection performed into the air cell on day 12 of egg incubation. Analysis of microbial communities and mucosal gene expression was performed at slaughter (day 42 post-hatching). Chyme (for DNA isolation) and intestinal mucosa (for RNA isolation) from four distinct intestinal segments (duodenum, jejunum, ileum, and caecum) was sampled. The relative abundance of Bifidobacterium spp. and Lactobacillus spp. in DNA isolated from chyme samples was determined using qPCR. On the host side, the mRNA expression of 13 genes grouped into two panels was analysed with RT-qPCR. Panel (1) included genes related to intestinal innate immune responses (IL-1ß, IL-10 and IL-12p40, AvBD1 and CATHL2). Panel (2) contained genes involved in intestinal barrier function (MUC6, CLDN1 and TJAP1) and nutrients sensing (FFAR2 and FFAR4, GLUT1, GLUT2 and GLUT5). GOS increased the relative abundance of Bifidobacterium in caecum (from 1.3% to 3.9%). Distinct effects of GOS on gene expression were manifested in jejunum and caecum. Cytokine genes (IL-1ß, IL-10 and IL-12p40) were up-regulated in the jejunum and caecum of the GOS-treated group. Host defence peptides (AvBD1 and CATHL2) were up-regulated in the caecum of the GOS-treated group. Free fatty acid receptors (FFAR2 and FFAR4) were up-regulated in all three compartments of the intestine (except the duodenum). Glucose transporters were down-regulated in duodenum (GLUT2 and GLUT5) but up-regulated in the hindgut (GLUT1 and GLUT2). In conclusion, GOS delivered in ovo had a bifidogenic effect in adult chickens. It also modulated gene expression related to intestinal immune responses, gut barrier function, and nutrient sensing.

Over the past several decades, as a result of the increasing growth rate and body size of modern hybrid birds, poultry industry has been facing up the occurrence of many breast meat abnormalities, such as White Striping (WS), Wooden Breast (WB) and Spaghetti Meat (SM), whose incidence has recently reached alarming levels. Thus, the present study aimed at simultaneously investigating the implications of WS, WB and SM abnormalities on meat colour, pH, proximate composition, fatty acid profile, collagen, thermal properties as well as texture and water mobility, assessed on both superficial and deep section of Pectoralis major muscle. Overall, the occurrence of breast abnormalities is associated with a higher ultimate pH and a significant increase in moisture and fat level, coupled with a decrease in protein and ash content with WB showing the most detrimental effect. However, fatty acid profile resulted to be mildly modified only by WS. Moreover, WB fillets showed significantly higher (P < .001) collagen content and stromal protein denaturation enthalpy associated with an altered water distribution and mobility within the muscle tissue. On the contrary, SM samples displayed a lower (P < .001) collagen cross-linking and a softer texture after the cooking. Overall, these findings reveal that the occurrence of WB abnormality exerts a more profound and prominent effect on meat quality traits rather than the presence not only of WS, but also of SM.

The purpose of this study was to examine the effect of in ovo injection of 2 different prebiotics, DiNovo (DN; Laminaria spp., extract containing laminarin and fucoidan) and Bi2tos (BI; non-digestive trans-galactooligosaccharides from milk lactose digested with Bifidobacterium bifidum NCIMB 41171), on growth, slaughter traits, intramuscular fat percentage (IF) and muscle fiber diameter, and lipid oxidation of meat in chickens reared under commercial conditions, following an in ovo trial protocol. On d 12 of embryonic incubation, 350,560 Ross 308 eggs were randomly divided into 3 experimental groups and automatically injected in ovo with: physiological saline (control group), BI at dose of 3.5 mg/embryo and DN at dose of 0.88 mg/embryo. Hatched chicks (males and females) were allocated dependent on treatment group into 3 poultry houses on each farm (3 farms in total) with a stocking density of 21.2 to 21.5 chicks/m2 At 42 d of age, 14 randomly chosen birds (7 males and 7 females), per each treatment from each farm, were individually weighed and slaughtered. The results showed no significant differences of final number of chickens/chicken house, mortality, BW per treatment, stocking density (kg/m2), feed intake, feed conversion rate (FCR), and European Broiler Index among 3 experimental groups. Treatments with BI and DN were associated with slight increases (P > 0.05) in average BW and a minor improvement (P > 0.05) of FCR in BI group. Slaughtered chickens from DN and BI treated groups had significantly increase of BW, carcass weight, carcass yield, and breast muscle weight compared with the control group. IF and muscle fiber diameter were similar among groups. Males had significantly higher slaughter traits compared to females, except for breast muscle yield. The prebiotic treatments led to a higher lipid oxidation in meat, even if the detected TBA reactive substances were below the critical value recognized for meat acceptability. In conclusion, in ovo administration of prebiotics was associated with improvements in a number of parameters of relevance to commercial poultry production.

The effects of dietary supplementation in pigs with plant extract (PE) from Lippia spp., titrated in verbascoside (5mg/kg feed), from weaning to slaughter (166days), on carcass characteristics, meat quality, collagen characteristics, oxidative stability and sensory attributes of Longissimus dorsi (LD) muscle were examined. Ten pigs per treatment were slaughter at a live weight of 109.5±1.4kg. No influence on carcass characteristics, LD meat quality parameters and collagen characteristics were observed. Dietary PE increased (P<0.001) α-tocopherol levels in LD muscle. Raw LD of pig fed PE showed lower (P<0.001) lipid oxidation levels than controls. A reduction (P=0.05) of fat odor and rancid flavor intensity in cooked LD muscle stored at 4°C for 24h was observed in the treated group. This study shows that PE is an effective antioxidant in pork meat, enhancing oxidative status and sensory attributes, without affecting other meat quality parameters.
