The aim of the study was to investigate if the application of a synthetic olfactory agonist (SOA) would reduce indicators of stress in sows, in response to a stressor prior to parturition, and if it would improve farrowing house performance of sows and their piglets. Two studies were conducted: an intensive study with 47 sows, either having their first or second litter (Control n = 24; SOA n = 23); and a commercial validation study with 418 sows, either having their first litter or have had multiple litters (Control n = 210; SOA n = 208). Within the intensive study, sows were housed with or without a synthetic olfactory agonist suspended in the creep area of the farrowing crate, whereas within the commercial validation study, sows were housed with or without a synthetic olfactory agonist suspended over the adjoining creep area of two farrowing crates. Within the intensive study, despite a discernible increase in cortisol concentration in response to a stressor (snout rope test), cortisol response was not different between treatments (p > 0.05). Farrowing duration in first-litter sows exposed to the SOA was decreased (p < 0.001) whilst there was no impact on farrowing duration in second litter sows. Piglets were not attracted by the SOA to increase their utilisation of the creep area and spent more time in proximity to the sow (p < 0.05). Within the commercial validation study, no impacts were seen on piglet production measures (p > 0.05). Largely the use of an SOA within the farrowing house did not impact the sow or her piglets in either the intensive study or commercial validation study. Based on these current results, the use of SOA within the farrowing house is not supported.

The ability to assess the welfare of animals is dependent on our ability to accurately determine their emotional (affective) state, with particular emphasis being placed on the identification of positive emotions. The challenge remains that current physiological and behavioral indices are either unable to distinguish between positive and negative emotional states, or they are simply not suitable for a production environment. Therefore, the development of novel measures of animal emotion is a necessity. Here we investigated the efficacy of microRNA (miRNA) in the brain and blood as biomarkers of emotional state in the pig. Female Large White × Landrace pigs (n = 24) were selected at weaning and trained to perform a judgment bias test (JBT), before being exposed for 5 weeks to either enriched (n = 12) or barren housing (n = 12) conditions. Pigs were tested on the JBT once prior to treatment, and immediately following treatment. MiRNA and neurotransmitters were analyzed in blood and brain tissue after euthanasia. Treatment had no effect on the outcomes of the JBT. There was also no effect of treatment on miRNA expression in blood or the brain (FDR p > 0.05). However, pigs exposed to enriched housing had elevated dopamine within the striatum compared to pigs in barren housing (p = 0.02). The results imply that either (a) miRNAs are not likely to be valid biomarkers of a positive affective state, at least under the type of conditions employed in this study, or (b) that the study design used to modify affective state was not able to create differential affective states, and therefore establish the validity of miRNA as biomarkers.

Animal welfare assessment requires measures for positive affective state. Pharmacological agents that manipulate affective state can be used to evaluate novel biomarkers for affective state assessment. However, to validate that an agent has modified brain function, a reliable indicator is required. Circulating cortisol has been used as a reporter for effective delivery of the antidepressant selective serotonin reuptake inhibitor (SSRI) fluoxetine hydrochloride to the brain in humans and sheep. Here, we tested cortisol as a reporter for effective delivery of fluoxetine hydrochloride to the pig brain. We hypothesized that following administration of SSRI, innervation of the serotonergic reward pathway would result in activation of the hypothalamic-pituitary-adrenal (HPA) axis, leading to increased circulating cortisol levels. Large White-Landrace gilts received either a single intravenous dose of 100 mg fluoxetine hydrochloride suspended in 10 mL saline (n = 4), or 10 mL saline alone (n = 4). Blood samples were collected every 15 min for one hour prior to, and six hours post-treatment. The interaction of treatment x time on mean plasma cortisol levels between 15-165 min post-treatment was significant (p = 0.048) with highest cortisol concentrations of SSRI treated pigs versus controls (+ 98%) at 135 min post-treatment. However, individual cortisol profiles after SSRI treatment revealed high inter-individual variation in response. We conclude that, while combined data imply that plasma cortisol may be a readout for SSRI efficacy, inter-individual variation in SSRI response may preclude application of this approach in the pig. Given the current limited sample size, further research to confirm these findings is needed.

Lactation anoestrus limits the flexibility of modern pig production systems such that any increase in lactation length reduces farrowing frequency, and thus profit. This review focuses on post-partum development of the sow's reproductive system, the physiology of lactation anoestrus and how it can be overcome, as well as the fertility of sows mated while lactating. The propensity for sows to ovulate spontaneously while lactating is high (24-31%), and a high proportion of sows will ovulate rapidly and synchronously in response to combinations of altered suckling (split weaning, interrupted suckling), daily boar contact, exogenous gonadotrophins, and group housing. The apparent ease with which lactation anoestrus can be overcome represents an opportunity to uncouple sow mating from weaning, thus reducing the impact of lactation length on productivity. This is especially true when considering the benefits of the described stimulation methods on the reproductive performance (i.e., shorter weaning to oestrus intervals and higher litter sizes) of the low proportion of sows that maintain lactation anoestrus.

The ability to stimulate a fertile oestrus during early lactation provides an opportunity to increase piglet weaning age whilst maintaining 2.4 litters per sow per year. This study evaluated the effects of boar contact beginning on day 7 of lactation and lactation length on lactation oestrus induction and subsequent reproductive output in multiparous sows (parity 2.9 ± 0.16; mean ± SEM; range 2-6). Large White × Landrace multiparous sows were allocated to one of four treatment groups; weaning at 26 days post-partum and no boar contact (control); weaning at 26 days and boar contact (BC); weaning at 7 days and boar contact (BC+short lactation (SL)); and weaning at 7 days and no boar contact (SL). Control and SL sows were taken for a short walk daily during which a backpressure test was performed in the absence of a boar. Oestrus expression within 26 days of farrowing was significantly higher in BC (67%) and BC+SL (93%) sows than control (11%) and SL (13%) sows (P<0.05). Of sows mated within 26 days of farrowing, there was no difference in the number of piglets born alive or stillborn. Therefore, boar contact is an effective method to stimulate oestrus within 26 days of farrowing in both lactating and weaned sows.

This study evaluated the effect of split weaning and fence-line boar exposure during lactation on the incidence of lactation oestrus. Large White and Large White × Landrace sows (parity 2.9 ± 0.17; mean ± SEM) were housed in conventional farrowing crates from day -4 to 30 post-parturition. Four treatments (n = 18) were used: control (SPW0): continuous lactation of 10 piglets with all piglets weaned on day 30 of lactation; and three split wean (SPW) treatments with 3 (SPW3), 5 (SPW5) or 7 (SPW7) of the heaviest piglets removed from the sow on day 18 lactation. From day 18 lactation all sows received 15 min daily, fence-line boar exposure in a detection mating area. Fewer sows in the SPW0 treatment (56% (10/18)) expressed a lactation oestrus compared to the SPW3, SPW5, and SPW7 treatments (83%; 89%; 94%, respectively). SPW0 sows had a lower subsequent total born compared to SPW5 or SPW7 sows (8.9 ± 1.1 vs. 12.5 ± 1.0 and 13.1 ± 1.1, respectively). Between day 18 and 30 of lactation, sows in SPW5 and SPW7 gained weight (4.5 ± 1.4 and 1.9 ± 1.4 kg, respectively) whereas SPW0 and SPW3 sows lost weight (4.9 ± 1.4 and 2.9 ± 1.4 kg, respectively) (P<0.05). Split weaned piglets were heavier at day 17 of age by 1.0 kg however by day 40 of age no weight differences were observed between piglets weaned on day 18 compared to day 30 (P<0.05). In conclusion, split weaning coupled with fence-line boar exposure in late lactation induced lactation oestrus in a higher proportion of sows compared to those suckling a normal litter size.

The sucrose breath test (SBT) is a simple noninvasive technique used currently to determine intestinal absorptive function in humans and rodents. However, to date, the test has not been adapted for use in swine. During weaning, intestinal sucrase activity in piglets temporarily declines in response to stressors and is commonly used as a marker of the intestinal response to weaning. Here we assessed the sucrose dose needed for using the SBT in piglets. Six randomly allocated piglets were orogastrically gavaged with (13)C-labeled sucrose at a dose of 2 g/kg; breath samples were collected for measurement of (13)CO2 on days 0 (approximately 17 h after weaning), 5, and 10 after weaning. The resultant SBT value (cumulative dose at 90 min) was decreased by 46% on day 5 after weaning relative to baseline levels, consistent with temporal changes in gastrointestinal sucrase activity associated with weaning. We conclude that a sucrose dose of 2 g/kg is satisfactory to conduct SBT studies in piglets. With further development, the SBT may provide a new tool to noninvasively monitor digestive function in weaned piglets, to assess the effects of nutritional strategies on intestinal health, and as an indicator of gut integrity and function in swine models of human gastrointestinal disease.
