To clarify the impact of human activities on the natural environment, as well as the current ecological risks to the environment surrounding Zhushan Bay in Taihu Lake, the characteristics of deposited organic materials, including elements and 16 polycyclic aromatic hydrocarbons (∑16PAHs), in a sediment core from Taihu Lake were determined. The nitrogen (N), carbon (C), hydrogen (H), and sulfur (S) contents ranged from 0.08 to 0.3%, 0.83 to 3.6%, 0.63 to 1.12%, and 0.02 to 0.24%, respectively. The most abundant element in the core was C followed by H, S, and N, while elemental C and the C/H ratio displayed a decreasing trend with depth. The ∑16PAH concentration was in the range of 1807.48-4674.83 ng g-1, showing a downward trend with depth, with some fluctuations. Three-ring PAHs dominated in surface sediment, while 5-ring PAHs dominated at a depth of 55-93 cm. Six-ring PAHs appeared in the 1830s and gradually increased over time before slowly decreasing from 2005 onward due to the establishment of environmental protection measures. The ratio of PAH monomers indicated that PAHs in samples from a depth of 0 to 55 cm were mainly derived from the combustion of liquid fossil fuels, while the PAHs in the deeper samples mainly originated from petroleum. The results of a principal component analysis (PCA) showed that the PAHs in the sediment core of Taihu Lake were mainly derived from the combustion of fossil fuels, such as diesel, petroleum, gasoline, and coal. The contributions of biomass combustion, liquid fossil fuel combustion, coal combustion, and unknown source were 8.99%, 52.68%, 1.65%, and 36.68%, respectively. The results of a toxicity analysis indicated that most of the PAH monomers had little impact on the ecology, and the annual increase of a small number of monomers might have toxic effects on the biological community, resulting in a serious ecological risks, that requires the imposition of control measures.

Objective To establish a prognosis model using immune-related genes in hepatocellular carcinoma (HCC) patients, which could provide a theoretical foundation for HCC immunotherapy. Methods Immune-related genes were identified by differential expression analysis, and risk prognosis prediction models were established using univariate, multivariate Cox and Least absolute shrinkage and selection operator (LASSO) regression analysis. The predictive value of the prognostic model was evaluated using the concordance index (C-index), receiver operating characteristic curve (ROC curve), and calibration curve and decision curve. In addition, risk score was used to stratify patients to assess prognostic differences in patients at different risk levels. Results We identified 1403 immune-related genes, mainly involved in biological processes such as immune response, adaptive immune response, and immunoglobulin production, as well as pathways such as cytokine interactions, chemokine signaling pathways and allograft rejection. Univariate Cox analysis found that 53 immune-related genes were associated with prognosis, and eight prognostic immune-related genes cytochrome P450 1A2(CYP1A2), ficolin 3(FCN3), hepatoma derived growth factor-like 1(HDGFL1), lipocalin 2(LCN2), mitochondrially encoded cytochrome C oxidase II pseudogene 12(MTCO2P12), peptidyl arginine deiminase 3(PADI3) and regulator of G protein signaling 16(RGS16) were further screened by LASSO and multivariate Cox regression analysis. Subsequently, a prognostic nomogram based on risk score was established. The ROC curve, calibration curve and decision curve confirmed that the model has good discrimination, accuracy and clinical value. Furthermore, stratified analysis showed that patients with higher risk scores had poorer prognosis. Conclusion We establish a prognostic nomogram model using eight immune-related genes, which can reliably predict the prognosis of HCC patients.

Background: Ovarian cancer (OC) is the most lethal gynecological malignancy worldwide. Increasing evidence indicates that TBC domain family is implicated in various cellular events contributing to initiation and development of different cancers, including OC. However, the role of TBC1D2, a crucial member of TBC domain family, remains unclear in OC. Methods: IHC and qRT-PCR were employed to determine TBC1D2 expression in OC tissues and cells. In vitro and in vivo assays involving proliferation, migration, invasion were performed to explore the role of TBC1D2 in OC development. The underlying mechanism by which TBC1D2 promotes OC metastasis were elucidated using bioinformatics analysis, western blotting and co-immunoprecipitation. Results: Upregulation of TBC1D2 was found in OC and was associated with a poor prognosis. Meanwhile, TBC1D2 promoted OC cell proliferation, migration, and invasion in vitro and facilitated tumor growth and metastasis in vivo. Moreover, TBC1D2 contributed to OC cell invasion by E-cadherin degradation via disassembling Rac1-IQGAP1 complex. In addition, miR-373-3p was screened out and identified to inhibit OVCAR3 invasion via negative regulation of TBC1D2. Conclusion: Our findings indicated that TBC1D2 is overexpressed in OC and contributes to tumor metastasis via E-cadherin degradation. This study suggests that TBC1D2 may be an underlying therapeutic target for OC.

Hepatoblastoma is the most common liver cancer in children, and the aggressive subtype often has a poor prognosis and lacks effective targeted therapy. Although aggressive hepatoblastoma (HB) is often accompanied by abnormally high expression of the transcription factor c-Myc, the underlying mechanism remains unclear. In this study, we found that mitochondrial fragmentation was enhanced by c-Myc overexpression in human aggressive HB tissues and was associated with poor prognosis. Then, a mouse model resembling human HB was established via hydrodynamic injection of c-Myc plasmids. We observed that liver-specific knockout of the mitochondrial fusion molecule MFN1 or overexpression of mitochondrial fission molecule DRP1 promoted the occurrence of c-Myc-driven liver cancer. In contrast, when MFN1 was overexpressed in the liver, tumor formation was delayed. In vitro experiments showed that c-Myc transcriptionally upregulated the expression of DRP1 and decreased MFN1 expression through upregulation of miR-373-3p. Moreover, enhanced mitochondrial fragmentation significantly promoted aerobic glycolysis and the proliferation of HB cells by significantly increasing reactive oxygen species (ROS) production and activating the RAC-alpha serine/threonine-protein kinase (AKT)/mammalian target of rapamycin (mTOR) and nuclear factor κB (NF-κB) pathways. Taken together, our results indicate that c-Myc-mediated mitochondrial fragmentation promotes the malignant transformation and progression of HB by activating ROS-mediated multi-oncogenic signaling.

ABSTRACT: Gastroesophageal junction adenocarcinoma (GEJAC) is a malignant tumor with high mortality. Its incidence has increased sharply all over the world in recent years. The study aims to search for potential biomarkers for the diagnosis and prognosis of GEJAC based on the Gene Expression Omnibus database (GEO) database and The Cancer Genome Atlas (TCGA) database.Microarray dataset (GSE96668 and GSE74553) of GEJAC was downloaded from the GEO. After screening overlapping differentially expressed genes (DEGs) by GEO2R and Wayne map, functional enrichment analysis of the DEGs was performed by the DAVID database. Then, a protein-protein interaction (PPI) network was constructed, and the hub gene was identified by using STRING and Cytoscape, as well as the diagnostic value of hub genes was evaluated by the receiver operating characteristic (ROC) curves. Finally, the gene transcriptome profiles of gastric cancer named TCGA-STAD were downloaded from TCGA database to screen the potential prognostic genes and construct the prognostic risk model using Cox proportional hazards regression. Meanwhile, the Kaplan-Meier curve and time-dependent ROC curve were adopted to test the prognostic value of the prognostic gene signature.In this study, we identified 10 hub genes that might have high diagnostic value for GEJAC, and inferred that they might be involved in the occurrence and development of GEJAC. Moreover, we conducted a survival prediction model consisting of 6 genes and proved that they have value to some extent in predicting prognosis for GEJAC patients.

BACKGROUND: It has been reported that the prevalence of chronic diseases is high among old people and they have poor chronic diseases knowledge. This study was therefore designed to evaluate the awareness rate of chronic diseases knowledge among people aged over 60 years, to explore its related factors and to provide evidence for future health education. METHODS: A cross-sectional study was conducted from April to August in 2011. People aged 60 years and above from 3 communities in Jinan were selected by cluster sampling. Nine hundred and twenty five participants were interviewed face-to-face using a structured questionnaire. RESULTS: The awareness rates of chronic diseases knowledge varied from 29.5% to 90.2%. Four healthy lifestyles including quitting smoking and less drinking, keeping broad-minded, maintaining balanced diet and moderate physical activity were best known (from 86.3% to 90.2%). The least known knowledge were 2 complications of hypertension: nephropathy (29.5%) and retinopathy (37.2%). Participants with the following characteristics or behaviors were more likely to have higher chronic diseases knowledge: younger age, female, Han Chinese, higher level of education, having health insurance, participating in societies, having family history of chronic diseases, frequently gathering with friends/relatives, usually going to provincial hospitals/hospitals affiliated with medical universities, usually going to municipal hospitals and usually going to community health center/station. CONCLUSIONS: Old people in Jinan had incomplete chronic diseases knowledge and the overall awareness rate was not high. The older people's chronic diseases knowledge should be improved and health education programs should target males, older people with lower educational level, having no health insurance, having no family history of chronic diseases, participating in no societies, and less frequently gathering with friends/relatives. Also, lower level medical facilities should improve their skills of health education.
