Finding effective antibiotic alternatives is crucial to managing the re-emerging health risk of Clostridium perfringens (CP) type A/G-induced avian necrotic enteritis (NE), a disease that has regained prominence in the wake of governmental restrictions on antibiotic use in poultry. Known for its antimicrobial and immunomodulatory effects, the use of bovine lactoferrin (bLF) in chickens is yet to be fully explored. In this study, we hypothesized that bLF can accumulate in the small intestines of healthy chickens through gavage and intramuscular supplementation and serves as a potential antibiotic alternative. Immunohistochemistry located bLF in various layers of the small intestines and ELISA testing confirmed its accumulation. Surprisingly, sham-treated chickens also showed the presence of bLF, prompting a western blotting analysis that dismissed the notion of cross-reactivity between bLF and the avian protein ovotransferrin. Although the significance of the route of administration remains inconclusive, this study supports the hypothesis that bLF is a promising and safe antibiotic alternative with demonstrated resistance to the degradative environment of the chicken intestines. Further studies are needed to determine its beneficial pharmacological effects in CP-infected chickens.

While the chicken (Gallus gallus) is the most consumed agricultural animal worldwide, the chicken transcriptome remains understudied. We have characterized the transcriptome of 10 cell and tissue types from the chicken using RNA-seq, spanning intestinal tissues (ileum, jejunum, proximal cecum), immune cells (B cells, bursa, macrophages, monocytes, spleen T cells, thymus), and reproductive tissue (ovary). We detected 17,872 genes and 24,812 transcripts across all cell and tissue types, representing 73% and 63% of the current gene annotation, respectively. Further quantification of RNA transcript biotypes revealed protein-coding and lncRNAs specific to an individual cell/tissue type. Each cell/tissue type also has an average of around 1.2 isoforms per gene, however, they all have at least one gene with at least 11 isoforms. Differential expression analysis revealed a large number of differentially expressed genes between tissues of the same category (immune and intestinal). Many of these differentially expressed genes in immune cells were involved in cellular processes relating to differentiation and cell metabolism as well as basic functions of immune cells such as cell adhesion and signal transduction. The differential expressed genes of the different segments of the chicken intestine (jejunum, ileum, proximal cecum) correlated to the metabolic processes in nutrient digestion and absorption. These data should provide a valuable resource in understanding the chicken genome.

The pygmy hippopotamus (Choeropsis liberiensis) is an IUCN Red List Endangered species (CITES Appendix II) that has been housed in zoological collections since 1912. As wild populations continue to decline throughout the species' range, successful ex situ breeding and management, including an understanding of morbidity and mortality, are of utmost importance. This study is the first comprehensive review of mortality data from the captive population since 1982 and significantly expands on previous analyses. We solicited necropsy reports from 129/187 zoological institutions worldwide that currently or previously held pygmy hippos and received data for 404 animals (177 ♂, 220 ♀, 7 undermined sex), representing 43% of pygmy hippos that have died in captivity. Mortality in neonates was primarily due to perinatal causes (51.8%-stillbirth, failure to thrive, weakness, poor suckling reflex, maternal neglect) or parent-inflicted trauma (28%). Common causes of mortality in adult and geriatric animals included cardiovascular disease (16%), degenerative musculoskeletal conditions (10%), obstructive gastrointestinal disease (9%), and renal insufficiency (13%), sometimes associated with advanced polycystic kidney disease (PKD). Although not the direct cause of mortality, a number of adult and geriatric pygmy hippos were also overweight to obese. Infectious causes of mortality in included leptospirosis and encephalomyocarditis virus, the latter usually presenting as acute death due to cardiovascular demise. This comprehensive overview presents a useful guide for recommendations in preventative veterinary care and for improved husbandry and management of pygmy hippos in captivity. The aim of this study was to evaluate the effects of IMUNO-2865(®) on hematological and antioxidative parameters in sea bream. Total of 640 sea bream were fed with diets containing 0 (Group 1), 1 (Group 2), 10 (Group 3) and 25 (Group 4) g of IMUNO-2865(®) kg(-1) feed during 90 days. Samples were taken each month and three months after the supplementation. A significant heterophils increase was observed in group 4 compared to group 1 after two months, and an increase in monocytes number was observed in group 4 compared to the other groups after one month. Glutathione peroxidase (GSH-Px) and paraoxonase-1 (PON1) were significantly increased in groups 3 and 4 compared to the control group three months into the experiment. Superoxide dismutase (SOD) was increased in group 4 compared to the control group from day 60 until the end of the experiment, and in groups 2 and 3 compared to the control after three months. Based on the differences in the cellular immunity and oxidative stress parameters, with an overall absence of mortality, the results of this study suggest that the use of IMUNO-2865(®) in aquaculture is safe and possess a cumulative immunostimulatory effect on sea bream.

Gastrointestinal nematodes (GIN) are a serious health problem and represent the most significant constraint in sheep grazing operations. Problems tend to be worse in organic sheep farming systems, as a consequence of a less restricted access of animals to outdoor environment with a higher exposure to infective larvae. In domestic animals, GIN are effectively controlled by an aggressive prophylactic administration of commercially available anthelmintics. As a consequence to a common overdose and misuse of readily available antiparasitic treatments, there is an inevitable development of populations of GIN resistant to all major classes of anthelmintics. Also, the control of GIN that is based entirely on the anthelmintic use, threatens sustainability of the sheep farming worldwide. The combination of the optimized use of anthelmintic drugs and alternative approaches seem to be a reasonable choice in sustainable parasitic control programs that offer a substantial reduction of anthelmintic treatments and conservation of anthelmintic efficacy. In that aspect, a "targeted selective treatment (TST)" directed towards animals clinically diagnosed with GIN, seems to be an effective approach to leave some parasite populations unexposed to anthelmintics (refugia) and to reduce development of anthelmintic resistance. Also, many current research efforts aim to find and validate sustainable non-chemotherapeutic approaches to GIN control, including changes in grazing management, optimized nutrition, dietary supplementation, consumption of plants with anthelmintic properties, biological control by nematophagous fungi, copper oxide wire particles (COWP), and homeopathic treatments. This manuscript outlines (outlines) and discusses relevant alternative approaches for GIN control in modern sheep farming systems.

Recurring infection of reticuloendotheliosis virus (REV), an avian oncogenic gammaretrovirus, has been a major obstacle in attempts to breed and release the endangered Attwater's prairie chicken (Tympanicus cupido attwateri). The aim of this study was to develop a DNA vaccine that protects the birds against REV infection. A plasmid was constructed expressing fusion proteins of REV envelope (env) and VP22 of Gallid herpesvirus 2 or REV gag and VP22. Birds vaccinated with these recombinant plasmids developed neutralizing antibodies; showed delayed replication of virus; and had significantly less infection of lymphocytes, specifically CD4+ lymphocytes. Although the vaccine did not prevent infection, it offered partial protection. Birds in field conditions and breeding facilities could potentially benefit from increased immunity when vaccinated.

Clinical respiratory illness was compared in five homozygous chicken lines, originating from homozygous B2, B8, B12 and B19, and heterozygous B2/B12 birds after infection with either of two strains of the infectious bronchitis virus (IBV). All chickens used in these studies originated from White Leghorn and Ancona linages. IBV Gray strain infection of MHC homozygous B12 and B19 haplotype chicks resulted in severe respiratory disease compared to chicks with B2/B2 and B5/B5 haplotypes. Demonstrating a dominant B2 phenotype, B2/B12 birds were also more resistant to IBV. Respiratory clinical illness in B8/B8 chicks was severe early after infection, while illness resolved similar to the B5 and B2 homozygous birds. Following M41 strain infection, birds with B2/B2 and B8/B8 haplotypes were again more resistant to clinical illness than B19/B19 birds. Real time RT-PCR indicated that infection was cleared more efficiently in trachea, lungs and kidneys of B2/B2 and B8/B8 birds compared with B19/B19 birds. Furthermore, M41 infected B2/B2 and B8/B8 chicks performed better in terms of body weight gain than B19/B19 chicks. These studies suggest that genetics of B defined haplotypes might be exploited to produce chicks resistant to respiratory pathogens or with more effective immune responses.

As detailed in the Association of Schools of Public Health / Association of American Veterinary Medical Colleges 2007 Joint Symposium on Veterinary Public Health, veterinary public health (VPH) can no longer be viewed as a unique sub-specialty of veterinary medicine. Rather, its practice pervades nearly every aspect of the veterinary profession, regardless of whether the practitioner is engaged in small-animal, large-animal, research, corporate, or military practice. In congruence with the practice of VPH, the teaching of VPH should also pervade nearly every aspect of veterinary education. Accordingly, at Western University of Health Sciences, College of Veterinary Medicine (WU-CVM), public health is not simply taught as an individual course but, rather, is interwoven into almost every aspect of the curriculum, continually emphasizing the relevance of this discipline to the practice of veterinary medicine. This article outlines the teaching philosophy of WU-CVM, provides an overview of the curriculum, and describes the integral nature of public health throughout all four years of the educational program.

The fecal shedding and pathogenicity of enterohemorrhagic E. coli (EHEC) O26:H11, EHEC O111:NM, and EHEC O157:H7 in weaned calves (8 to 10 weeks of age) were compared with and without treatment with a three-strain mixture of probiotic bacteria (competitive-exclusion E. coli). Three groups of 12 calves were each perorally given a five-strain mixture of one of the EHEC serotypes (10(10) CFU of total bacteria per calf). Seventy-two hours later, six calves from each group were each administered 10(10) CFU of probiotic bacteria. None of the EHEC serotypes caused significant clinical disease, although a few calves developed mild transient diarrhea or pyrexia. Gross or microscopic lesions attributable to EHEC were not detected in control or probiotic-treated calves at necropsy. For probiotic-treated calves given E. coli O157:H7 and for probiotic-treated calves given E. coli O111:NM, fecal shedding was reduced compared with that for untreated calves. For the probiotic-treated calves given E. coli O157:H7, the reductions in fecal shedding on days 8, 12, 14, 16, 20, 22, 28, and 30 after peroral administration were statistically significant (P<0.05). For probiotic-treated calves given E. coli O111:NM, there were statistically significant reductions (P<0.05) in fecal shedding on days 6, 8, 10, and 12. In contrast, there was no reduction in fecal shedding for calves administered E. coli O26:H11 and treated with the probiotic bacteria. In fact, calves in both the treated and the nontreated groups continued to shed large populations of E. coli O26:H11 throughout the 32-day trial. At necropsy, E. coli O157:H7 was isolated from five of six untreated calves and from only two of six probiotic-treated calves. E. coli O111:NM was isolated from four of six untreated calves at necropsy and from two of six probiotic-treated calves. However, E. coli O26:H11 was isolated from five of six untreated calves and from all six probiotic-treated calves. The results obtained in this study indicate that probiotic E. coli substantially reduced or eliminated fecal shedding of E. coli O157:H7 and E. coli O111:NM 8 to 30 days and 6 to 12 days after the administration of the probiotic culture, respectively, and reduced the persistence of E. coli O157:H7 in the gastrointestinal tract at necropsy (31 to 33 days after the administration of the probiotic culture). The probiotic E. coli did not reduce fecal shedding or gastrointestinal persistence of E. coli O26:H11.

The pathogenicity and fecal shedding of enterohemorrhagic Escherichia coli (EHEC) O26:H11, O111:NM, and O157:H7 were compared in calves (< 1 week of age) with or without prior treatment with probiotic bacteria (competitive exclusion E. coli). Three groups of 12 to 14 calves were used for these treatments. Half of the calves in each group were perorally administered 10(10) CFU of probiotic bacteria per calf, and, 2 days thereafter, 10(8) CFU of a five-strain mixture with one of the three EHEC serotypes per calf were administered to each calf. None of the EHEC serotypes caused clinical disease,and neither gross nor microscopic lesions attributable to EHEC were detected in control or probiotic-treated calves at necropsy. In calves administered E. coli O157:H7, fecal shedding was greatly reduced (> 6 log10 CFU/g) by 8 days after administration, and there was no significant difference (P > 0.05) in fecal shedding of E. coli O157:H7 between probiotic-treated and untreated control groups at that time. In contrast, control calves perorally administered E. coil of serotypes O111:NM or O26:H11 continued to shed substantial populations (10(2.1) to 10(6) CFU/g of feces and 10(2.5) to 10(4.9) CFU/g of feces, respectively) throughout 7 days postadministration of EHEC. In both groups administered either E. coli O111:NM or O26:H11, significantly less (P < 0.05) EHEC was isolated from feces at 7 days postadministration of EHEC and at necropsy from theprobiotic-treated group than from the untreated control group. Overall, neonatal calves shed in the feces from 1 to 7 days following peroral administration of EHEC greater populations of E. coli O111:NM and O26:H111 than E. coli O157:H7. In addition, treatment of calves with probiotic E. coli reduced fecal shedding of E. coli O111:NM and O26:H11 in most calves.

The emergence of antibiotic resistance among important foodborne pathogens like Escherichia coli O157:H7 has become an important issue with regard to food safety. In contrast to the case for Salmonella, antibiotic resistance has been slow in its development in E. coli O157:H7 despite the presence of mobile antibiotic resistance genes in other E. coli organisms that inhabit the same animal host. We set out to determine if rumen fluid influences the transfer of plasmid-mediated, antibiotic resistance to E. coli O157:H7. A commensal E. coli strain from a dairy cow was transformed with conjugative R plasmids and served as the donor in matings with naladixic acid-resistant E. coli O157:H7. R plasmids were transferred from the donor E. coli strain to E. coli O157:H7 in both Luria-Bertani (LB) broth and rumen fluid. R plasmids were transferred at a higher frequency to E. coli O157:H7 during 6 h of incubation in rumen fluid at rates comparable to those in LB broth, indicating that conditions in rumen fluid favor the transfer of the plasmids to E. coli O157. This finding suggests that the cow's rumen is a favorable environment for the genetic exchange of plasmids between microflora and resident E. coli O157:H7 in the bovine host.
