RESUMO

Glaciers are dwindling archives, releasing animal mummies preserved in the ice for centuries due to climate changes. As preservation varies, residual soft tissues may differently expand the biological information content of such mummies. DNA studies have proven the possibility of extracting and analyzing DNA preserved in skeletal residuals and sediments for hundreds or thousands of years. Paleoradiology is the method of choice as a non-destructive tool for analyzing mummies, including micro-computed tomography (micro-CT) and magnetic resonance imaging (MRI). Together with radiocarbon dating, histo-anatomical analyses, and DNA sequencing, these techniques were employed to identify a 350-year-old Austrian Ardea purpurea glacier mummy from the Ötztal Alps. Combining these techniques proved to be a robust methodological concept for collecting inaccessible information regarding the structural organization of the mummy. The variety of methodological approaches resulted in a distinct picture of the morphological patterns of the glacier animal mummy. The BLAST search in GenBank resulted in a 100% and 98.7% match in the cytb gene sequence with two entries of the species Purple heron (Ardea purpurea; Accession number KJ941160.1 and KJ190948.1) and a 98% match with the same species for the 16 s sequence (KJ190948.1), which was confirmed by the anatomic characteristics deduced from micro-CT and MRI.

Mid-infrared (MIR) microscopic imaging of indolent and aggressive lymphomas was performed including formalin-fixed and paraffin-embedded samples of six follicular lymphomas and 12 diffuse large B-cell-lymphomas as well as reactive lymph nodes to investigate benefits and challenges for lymphoma diagnosis. MIR images were compared to defined pathological characteristics such as indolent versus aggressive versus reactive, germinal centre versus activated cell-of-origin (COO) subtypes, or a low versus a high proliferative index and level of PD-L1 expression. We demonstrated that MIR microscopic imaging can differentiate between reactive lymph nodes, indolent and aggressive lymphoma samples. Also, it has potential to be used in the subtyping of lymphomas, as shown with the differentiation between COO subtypes, the level of proliferation and PD-L1 expression. MIR microscopic imaging is a promising tool for diagnosis and subtyping of lymphoma and further evaluation is needed to fully explore the advantages and disadvantages of this method for pathological diagnosis.

The purpose of this study was to analyze the effect of surfaces with different bearing ratios, but similar roughness heights, on the friction between ultrahigh molecular weight polyethylene (UHMWPE) and snow. On a linear tribometer positioned inside a cold chamber, the different samples were tested over a wide range of velocities and snow temperatures. The surface roughness was measured with a focus variation microscope and analyzed using the bearing ratio curve and its parameters. The surface energy was investigated by measuring the contact angles of a polar (water) and nonpolar (diiodmethane) liquid. The friction tests showed that the bearing ratio had a major effect on the friction between UHMWPE and snow. For temperatures close to the melting point a surface with wide grooves and narrow plateaus (nonbearing surface) performed well. For cold conditions, the friction was less for a surface with narrow grooves and wide plateaus (bearing surface). Interpretations of the results are given on the basis of mixed friction, with lubricated friction being dominant at higher snow temperatures and solid-solid interaction at lower ones.
