RESUMO

BACKGROUND AND AIMS: Patients with obesity and type 2 diabetes (T2D) have shown alterations in the affinity of IgG anti-leptin antibodies which are possibly related to metabolic alterations. In the present exploratory study, we analyzed serum samples from adults with T2D classified by body mass index (BMI) and evaluated the relationship of IgG anti-leptin antibodies with body composition, metabolic and cardiovascular risk parameters. METHODS: Serum IgG anti-leptin antibodies (total, free and immune complexes fractions) were measured by in-house ELISA. Body composition, metabolic biomarkers (glucose, glycated hemoglobin, lipid profile, insulin, leptin) and cardiometabolic risk indexes (AIP, HOMA-IR, HOMA-ß) were evaluated in one hundred T2D patients. RESULTS: Patients with T2D and obesity presented a decrease in the percentage of IgG anti-leptin immune complexes compared to patients with T2D and overweight (p < 0.0053). Negative correlations of IgG anti-leptin immune complexes with triglycerides (TG) (r=-0.412, p = 0.023) and VLDL-C (r=-0.611, p = 0.017) were found in normal weight T2D patients. Free IgG anti-leptin antibodies correlated positively with TC (r = 0.390, p = 0.032) and LDL-C (r = 0.458, p = 0.011) in overweight individuals with T2D. Finally, total IgG anti-leptin antibodies correlated positively with leptin hormone levels (r = 0.409, p = 0.024) and negatively with HOMA-IR (r =-0.459, p = 0.012) in T2D patients with obesity. CONCLUSIONS: The decrease of IgG anti-leptin immune complexes observed in patients with T2D and obesity suggests a reduction in antibody affinity to the hormone that may impact its transport and signaling, lipid, lipoprotein and insulin metabolism.

The objective was to evaluate the antioxidant and biological potential of eight freeze-dried berry varieties of southern Jalisco using in silico and in vitro approaches. Fourteen tentative phenolic compounds were identified in berries by ESI-QToF, including anthocyanins, phenolic acids, flavanols and flavonols. In silico assays of phytochemicals in the berry inhibiting enzymes related to obesity and diabetes showed predicted binding energy interactions (ranging from -5.4 to -9.3 kcal/mol). Among the cultivars, antioxidant potential for DPPH IC50 ranged from 1.27 to 3.40 mg/mL, ABTS IC50 from 2.26 to 7.32 mg/mL and nitric oxide (NO) inhibition IC50 from 4.26 to 11.07 mg/mL. The potential to inhibit α-amylase IC50 ranged from 4.02 to 7.66 mg/mL, α-glucosidase IC50 from 0.27 to 4.09 mg/mL, lipase IC50 from 1.30 to 4.82 mg/mL and DPP-IV IC50 from 1.36 to 3.31 mg/mL. Blackberry cultivars from the southern Jalisco region showed outstanding biological potential compared to other evaluated berries and could be used in the formulation of functional foods in the prevention of noncommunicable diseases.

RESUMO

Dysfunctional eating patterns include alterations in experiencing and expressing hunger, appetite, and satiety, which may lead to eating disorders or obesity in the long term. Alterations in hormones such as ghrelin have been suggested to influence emotional eating in women with obesity. Ghrelin-reactive autoantibodies (autoAbs) are present both in healthy individuals and those with eating disorders and have been suggested to protect the hormone from degradation and preserve its functional activity. This study aimed to evaluate the relationship between IgG ghrelin-reactive autoAbs with dysfunctional eating patterns, subjective perception of stress, and body composition parameters in young women. This cross-sectional study included 82 women (age 21±2 years) classified according to body fat percentage. Dysfunctional eating patterns were measured with the Spanish version of the Three-factor Eating Questionnaire-R18, and perceived stress was measured with the Spanish version of the Perceived Stress Scale - 10. A validated in-house enzyme-linked immunosorbent assay was performed to measure IgG ghrelin-reactive autoAbs in its free, total, and immune complex fractions. Free IgG ghrelin-reactive autoAbs were positively correlated with weight, BMI, body fat percentage, waist, and hip circumference in women with very high body fat percentage. In this group, a negative correlation was observed between ghrelin immune complexes and uncontrolled eating. This exploratory research shows that IgG ghrelin-reactive autoAbs have a potential role in altered body composition parameters and appetite expression, such as uncontrolled eating in women with very high body fat. Further studies are required to clarify the role of IgG autoAbs in eating behavior.

RESUMO

Studies have demonstrated the presence of low-affinity immunoglobulins (Igs) directed to leptin, a key hormone of the neuroendocrine axis that regulates appetite and metabolism, in adult healthy subjects, patients with obesity, and type 2 diabetes mellitus. In the present exploratory study, IgG leptin-reactive antibodies were analyzed for the first time in children and adolescents according to body mass index (BMI) and were correlated with biochemical profile (lipid profile, insulin, glucose, and leptin) and metabolic risk indexes [homeostasis model assessment for insulin resistance (HOMA-IR), homeostasis model assessment for ß-cell function (HOMA-ß), atherogenic index of plasma (AIP)]. One hundred and thirty-six participants were included (children n = 63, adolescents n = 73). An in-house enzyme-linked immunosorbent assay (ELISA) test was performed to measure IgG anti-leptin antibodies (free, total, and immune complexes). In adolescents, free and total IgG anti-leptin antibodies levels were higher in groups with overweight or obesity than in normal-weight group (P < 0.01), while in children, the total fractions were lower in groups with overweight and obesity than in normal weight (P < 0.02). Immune complexes percentage showed opposite correlations with BMI in children (r = 0.4004, P = 0.0473) and adolescents (r = -0.3983, P = 0.0133). IgG anti-leptin antibodies were also correlated with HOMA-IR in children (r = -0.4569, P = 0.0217) and adolescents (r = -0.3589, P = 0.0316), and with AIP (r = -0.3608, P = 0.0261) in adolescents. Our data suggest that the production and affinity of IgG anti-leptin antibodies can be affected by age, body composition, and metabolic conditions; additionally, in normal conditions, IgG anti-leptin antibodies may have a protective role in insulin resistance and cardiovascular events.

RESUMO

The aim of this research was to compare food intake dysfunctional patterns score with the subjective perception of anxiety and sociodemographic characteristics of the participants in isolation by COVID-19 from Chile, Colombia, and Mexico. A cross-sectional research was carried out, with a virtual questionnaire of subjective perception of anxiety and the questionnaire of three 18-item feeding factors. 958 people of both sexes participated (F = 83%, M = 17%), mainly in the 18 to 35 age range. Dysfunctional eating patterns presented high scores in people who perceived anxiety, as well as in participants from Chile. Additionally, it was found that women present greater cognitive restriction and emotional intake, and college students showed greater disinhibition. In conclusion, the scores of the three dysfunctional eating patterns were higher in people with subjective perception of anxiety during social isolation due to COVID-19, and there were also differences according to country, sex, and educational level.

ABSTRACT Physical activity is considered an anxiety-regulating behavior and is effective in reducing this emotional condition. Due to the coronavirus pandemic (COVID-19), governments of Latin America have applied isolation strategies to reduce the probability of contagion and thus control health effects; however, an unfavorable effect of this situation has been the increase in anxiety levels that can affect food intake. The objective of the current study was to analyze the association between physical activity and differences in dysfunctional patterns of intake in Latin American people who were in isolation. A questionnaire was applied to 1.035 people from different countries. We observed that those performing physical activity presented a 50% lower probability of perceived anxiety. Additionally, the subjects who performed physical activity presented greater cognitive restriction, and those who did not perform physical activity registered a higher level of disinhibition and emotional intake. We conclude that physical activity is an anxiety-regulating behavior in this time of isolation or quarantine and has a positive influence on individuals, since it reduces disinhibition and emotional intake, and could prevent weight gain and possible development of food pathologies. This finding is important for the promotion of physical activity in this time of confinement and similar situations, and it is also useful for interventions on subjects with emotional feeding.

ABSTRACT Deaths from heart disease and other cardiovascular diseases have increased in recent decades. Among the main factors that lead to the development of heart disease is dyslipidemia, which are disorders that directly affect the blood supply through the development of atheroma in the cardiovascular system. It has been proposed that phenolic compound intake has an important antilipidemic effect, a fact that suggests a preventive and protective characteristic against these diseases. The objective of this work is to present an analysis of the information related to treatment that includes the use of phenolic compounds on dyslipidemia parameters in humans; comparing samples, the components and the results of these types of disorders. Although the number of articles exploring this relationship is diverse, this systematic review explores the data related to a possible beneficial reaction of human metabolism. The search was carried out in two databases: PubMed and Web of Science, where a total of 2229 potential articles were obtained, of which 2194 were excluded, leaving 35 articles for review. Altogether, the results showed that, although there are certain beneficial properties of phenolic compounds such as isoflavones, tannins, theaflavins and anthocyanins that have been proven beneficial for cardiovascular diseases, for dyslipidemia specifically, it is still difficult to combine conclusive results. In these circumstances, the available information indicates the necessity to continue investigating the action of phenolic compounds on these types of pathologies.

RESUMO

CD36 glycoprotein is a candidate receptor involved in the gustatory detection of lipids and emerging evidence has suggested that genetic variations in CD36 may modulate the oral perception threshold to fatty acids. Here, we analyzed the association of -31118 G > A polymorphism in CD36 gene with nutritional status and preferences for fatty foods in Mexican children. Genotyping of SNP rs1761667 was performed in school-age children (n = 63) in addition to sensory tests evaluating the preference and satisfaction score assigned to oil-based sauces of different fatty acid composition. The G allele was associated with high BMI z-score in children (OR = 2.43, 95% (CI 1.02-5.99); p = 0.02) but CD36 genotypes (AA, GA, and GG) did not show significant association with the preference and satisfaction scores assigned to oil-based sauces. The BMI z-score showed no association with the preference to oil-based sauces; however, children with normal weight gave higher satisfaction scores to sauces with a high content of unsaturated fatty acids than to sauces rich in saturated fatty acids (0.56 ± 1.26 vs. 0.06 ± 1.22; p = 0.02). Therefore, the G allele of -31118 G > A SNP in CD36 gene is associated with overweight and obesity in Mexican children but do not appear to modulate the preferences and satisfaction scores to fat.

RESUMO

The role of gut microbiota on immune regulation and the development of autoimmune diseases such as rheumatoid arthritis (RA) is an emerging research topic. Multiple studies have demonstrated alterations on gut microbiota composition and/or function (referred to as dysbiosis) both in early and established RA patients. Still, research delineating the molecular mechanisms by which gut microorganisms induce the loss of immune tolerance or contribute to disease progression is scarce. Available data indicate that gut microbiota alterations are involved in RA autoimmune response by several mechanisms including the post-translational modification of host proteins, molecular mimicry between bacterial and host epitopes, activation of immune system and polarization toward inflammatory phenotypes, as well as induction of intestinal permeability. Therefore, in this review we analyze recent clinical and molecular evidence linking gut microbiota with the etiopathogenesis of RA. The potential of the gut microbiota as a diagnostic or severity biomarker is discussed, as well as the opportunity areas for the development of complementary therapeutic strategies based on the modulation of gut microbiota in the rheumatic patient.

Abstract Obesity is a global health problem and one of the main causes of chronic diseases. There are several factors that influence eating behavior and obesity, which can be genetic, physiological, nutritional, psychological or sociocultural. In recent decades, the change in lifestyle has caused inadequate eating patterns, coupled with a decrease in energy expenditure. This review describes the main physiological, nutritional and psychological factors that intervene in energy intake and eating behavior and how they can -by regulating appetite and body weight- influence the development of obesity. The neuropsychology of food reward, preferences, choice and eating behavior are aspects not yet understood. Future research needs to focus on elucidating the pathways involved in the rewarding aspects of food and, as an alternative to control obesity, replicate this experience with healthier foods. However, understanding the link between food, energy intake and body weight regulation will only be achieved through multidisciplinary work.

RESUMO

Pomegranate juice (PJ) has gained popularity attributed to its phenolic compounds and their medicinal properties. Its potential hypoglycemic effect has been related to enzymatic inhibition, insulin release, and the protection of pancreatic tissue. These effects depend on several aspects, such as the content and composition of phenols in pomegranate and characteristics of the organism that consumes the juice. The objective of this study was to systematically review scientific evidence investigating the hypoglycemic effect of PJ; the factors that affect bioactive compounds; and the mechanisms of action attributed to this effect. Human and rodent in vivo and in vitro studies were retrieved from PubMed, Scopus, and ScienceDirect databases. After reviewing the articles, it was identified that the methodologies and results among the scientific evidence were quite heterogeneous. Despite these limitations, many of the in vivo and in vitro studies found important hypoglycemic effects from PJ, as well as an increase in the function of ß-cell, insulin secretion, a significantly lower activity of α-amylase enzyme, an inhibition of the enzyme α-glucosidase and dipeptidyl peptidase-4 (DPP-4), and the protection against DNA damage. Determining the potential health benefits of polyphenols contained in the pomegranate is limited for multiple factors that could affect the efficacy of PJ. Overall, the results of this review suggest the need for further experimentation, using controlled variable factors and testing the effect of PJ under similar experimental conditions.

RESUMO

As a source of bioactive compounds, species of the genus Lupinus are interesting legumes from a nutritional point of view. Although wild species are abundant and represent a potential source of nutrients and biologically active compounds, most research has focused on domesticated and semi-domesticated species, such as Lupinus angustifolius, Lupinus albus, Lupinus luteus, and Lupinus mutabilis. Therefore, in this review, we focus on recent research conducted on the wild Lupinus species of Mexico. The nutritional content of these species is characterized (similar to those of the domesticated species), including proteins (isolates), lipids, minerals, dietary fiber, and bioactive compounds, such as oligosaccharides, flavonoids, and alkaloids.

The purpose of this work was to determine the preference and degree of satisfaction of four bakery products (cookies and grissinis) made from a mixture of wheat and common beans (Phaseolus vulgaris) or wheat and lupine (Lupinus albus) in adults from Chile, and compare the results with the results recently obtained for these products in the Mexican population. The sensory analysis was performed using two hedonic sensory tests, one of preference and the other of degree of satisfaction. The participants were 60 adults from Concepción, Chile. The results showed that the four products were liked by the participants, the versions made from wheat and lupine flour were most preferred and highest rated (2.05 ± 1.05 cookie and 1.28 ± 1.22 grissini ) regarding those made of wheat flour and beans (1.90 ± 1.22 cookie and 1.28 ± 1.22 grissini). When comparing the values obtained from the four products, significant statistical difference was found in the scores given to the cookies. The grissinis scores were identical. The results of the Mexican participants showed a greater preference and degree of satisfaction for products made from wheat flour and beans. According to the results, it can be concluded that bakery products made with mixtures of cereals and legumes are an option by which the consumption of legumes can be favored, which has declined in recent years(AU)

Diverse bioactive compounds of foods have been used in the alternative treatment of diabetes mellitus. Pomegranate juice has a high content of phenolic compounds to which biological properties are attributed as lipid-lowering, hypoglycemic and protective of pancreatic tissue. The objective of this study was to evaluate the effect of free access of pomegranate juice on the levels of glucose, cholesterol, triglycerides and the integrity of pancreatic tissue in vivo. Eighteen male Wistar rats were induced to hyperglycemia with 60 mg streptozotocin/kg of body weight intraperitoneal (IP). Three experimental groups were formed. The HS group exposed to sucrose solution, the HJG group exposed to pomegranate juice and the HSI group exposed to sucrose solution and insulin treatment. The results showed that the rats of the HJG group consumed pomegranate juice for 21 days, which resulted in the reduction of glucose levels from its initial value of 417 to 356 mg/dL, not significantly (p>0.05). Lipid levels showed a non-significant reduction at the end of the intervention (p>0.05). The histological study of the pancreas in the HJG group showed conservation of the pancreatic architecture and the presence of islets of Langerhans; while the HS group showed extensive pancreatic necrosis and the HSI group showed intermediate damage with few islets of Langerhans. It is suggested that pomegranate juice has hypoglycaemic effects and protects pancreatic tissue in induced hyperglycemic rats(AU)

RESUMO

ABSTRACT Objective To evaluate iron bioavailability of roots and cooked seeds of Lupinus rotundiflorus for human consumption using a low-iron-diet rat model. Methods A hemoglobin depletion-repletion test was performed using rats. A standard diet containing 8mg kg-1 of iron was used. Three experimental diets were prepared based on the standard diet: 2.3g of root flour added to D1, 21.5g cooked seed flour added to D2, and 0.03g of ferrous sulfate added to D3 (control diet), adjusting iron concentration of the diets to 24mg kg-1. Hemoglobin regeneration efficiency was used to measure iron bioavailability. Results Hemoglobin regeneration efficiency showed values of 13.80+2.49%, 13.70+1.60% and 18.38+1.56 in D1, D2 and D3 respectively, and biological relative values of 72.8, 75.08, and 100.00% (.<0.05). Conclusion Roots and cooked seeds of Lupinus rotundiflorus showed potential iron bioavailability, despite being a vegetal source, which could also encourage the study of other species of lupin as a source of iron.

Abstract: Sedentary behavior's role on health damage has been documented as a promoting factor of pathologies. Scientific evidence shows an increasing percentage of people with sedentary activity especially in large cities causing overweight, obesity, diabetes, hypertension etc. Effects of interrupting sedentary behavior periods with physical activity have been recently evaluated, demonstrating increases on energy expenditure. The objective of this experiment was to compare the effect of two interruption programs of sedentary behavior pattern on caloric expenditure. Participants were exposed to two interruption programs of sedentary behavior. For energy expenditure, heart monitors brand Beurer PM18 model used. First program consisted on 15-minutes periods of sedentary behavior followed by a 2.5 minutes walking break as physical activity. Second program consisted on periods of 30 minutes of sedentary behavior followed by a 5 minutes break of physical activity. Analysis used Student t test for paired samples showed a significant difference in caloric expenditure during sedentary behavior between program 1 and 2. Concluding that increasing the number of interruptions of periods of sedentary behavior has a direct effect on caloric expenditure.

RESUMO

Wild lupine consumption has restrictions due to the presence of alkaloids; however, these components can be reduced with a suitable thermal treatment. The aim of this research was to determine the thermal effect on chemical composition and minerals of wild lupine. Lupinus mexicanus had a reduction in protein and fat contents of 34.76 to 33.11 and 6.10 to 5.41 g/100 g of sample respectively, and an increase in ash and dietary fiber of 3.84 to 4.53 g/100 g and 20.9 to 28.48 g/100 g respectively. L. mexicanus raw seeds revealed the highest Ca content (3,252 mg/kg), L. elegans was the highest in Mg with 2,656 mg/kg. Highest Fe content was found in Lupinus rotundiflorus (82.8 mg/kg), and Lupinus exaltatus in Cu (184.4 mg/kg). All species showed similar Zn content of 73.3 mg/kg (Lupinus montanus) to 89.6 mg/kg (L. exaltatus). In all species the Cu content decreased, mainly in Lupinus elegans with a loss of 76,71 %.
