RESUMO

Historically, sponge classification is based on the interpretation of morphological characters, whose phylogenetic information content is frequently limited, subject to homoplasies, or prone to environmental plasticity (e.g., Chombard et al. 1998). Therefore, the currently accepted order-level classification of its largest class, Demospongiae, has been largely revised with molecular phylogenetic data (Morrow & Cárdenas 2015). Nevertheless, numerous sponge genera with ambiguous or provisoric phylogenetic placement still await definite classification.

Sponges (Porifera) are a diverse and globally distributed clade of benthic organisms, with an evolutionary history reaching at least the Ediacaran-Cambrian (541 Ma) boundary interval. Throughout their research history, sponges have been subjects of intense studies in many fields, including paleontology, evolutionary biology, and even bioengineering and pharmacology. The skeletons of sponges are mostly characterized by the presence of mineral elements termed spicules, which structurally support the sponge bodies, though they also minimize the metabolic cost of water exchange and deter predators. The description of the spicules' shape and the skeleton organization represents the fundamental basis of sponge taxonomy and systematics. Here, we provide an illustrated catalogue of sponge spicules, which is based on previous works on sponge spicules, for example, and gathers and updates all terms that are currently used in sponge descriptions. Each spicule type is further illustrated through high quality scanning electron microscope micrographs. It is expected to be a valuable source that will facilitate spicule identification and, in certain cases, also enable sponge classification.

In the 10th edition of the Systema Naturae (Linnaeus, 1758), which is the starting point of the Code for Zoological Nomenclature (ICZN Art. 3), Linnaeus named three species of the genus Alcyonium, A. arboreum, A. digitatum, and A. bursa. The genus name Alcyonium was based on the 16th and 17th century pre-Linnaean use for a diversity of marine organisms, including cnidarians, sponges, bryozoans, and algae. In the first valid presentation of the genus name, Linnaeus narrowed this down to comprise two clear cnidarians (A. arboreum, currently Paragorgia arborea, and A. digitatum, still accepted under this name and subsequently assigned as type species), but the pre-Linnaean diversity perhaps explains why the third species, A. bursa, was not recognized as a cnidarian. Linnaeus defined it as 'Alcyonium acaule pulposum subglobosum. Habitat in O. Europaea.' (translated as: Alcyonium without stalk, fleshy, semiglobular. From the European Ocean).' Attempts to fix its identity among contemporary authors at the end of the 18th and beginning of the 19th century followed a checkered course, with opinions varying from algae to tunicates and sponges.

The species of the cosmopolitan sponge genus Mycale occurring in the tropical Indo-West Pacific region and adjacent subtropical waters are reviewed taxonomically. Specimens incorporated in the collections of the Naturalis Biodiversity Center form the basis of this comprehensive study, supplemented by (type) specimens borrowed from or examined in other institutions. Specimens available numbered 351, belonging to 44 species, including 14 species new to science, Mycale (Aegogropila) prognatha sp.nov., Mycale (Carmia) amiri sp.nov., Mycale (Carmia) fungiaphila sp.nov., Mycale (Carmia) monomicrosclera sp.nov., Mycale (Carmia) tenuichela sp.nov., Mycale (Carmia) tubiporicola sp.nov., Mycale (Carmia) tydemani sp.nov., Mycale (Mycale) asigmata sp.nov., Mycale (Mycale) grandoides sp.nov., Mycale (Mycale) sundaminorensis sp.nov., Mycale (Naviculina) mascarenensis sp.nov., Mycale (Paresperella) sceptroides sp.nov., Mycale (Paresperella) seychellensis sp.nov., Mycale (Zygomycale) sibogae sp.nov. Three species, indicated by the designation 'aff.', were not definitely assigned to known or new species due to uncertainty of their identity. The genus Kerasemna, previously considered a junior synonym of Mycale, was revived as an additional subgenus Mycale (Kerasemna). One species, previously assigned to the genus Desmacella as D. lampra De Laubenfels is here reassigned to Mycale, subgenus at present undecided. Additionally, species previously reported from the region but not represented in our collections are briefly characterized and discussed. We propose new names Mycale (Mycale) mauricei nom.nov. for Mycale macrochela Burton (junior primary homonym of Mycale fistulata var. macrochela Hentschel) and Mycale (Mycale) bouryesnaultae nom.nov. for Mycale (Mycale) fibrosa Boury-Esnault Van Beveren (junior primary homonym of Mycale (Aegogropila) adhaerens subsp. fibrosa Koltun). Keys to the species of each subgenus occurring in the region are provided. The opportunity of having studied this comprehensive set of species and specimens from the tropical Indo-West Pacific is taken to review and discuss the morphological and biogeographical data gathered so far on the genus Mycale. The genus currently comprises approximately 255 accepted species, with highest diversity focused in tropical Atlantic and Indo-West Pacific regions as well as in warm-temperate Mediterranean-Atlantic regions.

RESUMO

An inventory of sponges from the shallow subtidal reefs of the west and east coasts of southern India is presented. The specimens offered in this paper were based on in-situ collections unlike the previous records of dry and net-entangled collections. A total of 101 species belonging to 12 orders, 22 families, 5 subfamilies and 44 genera from 4 subclasses of Class Demospongiae and one species from Class Calcarea are recorded. We recorded 18 new records to India, six new species combinations, 37 potential new records, and 40 species for the first time from the southwest coast.

The occurrence of different sponge species bearing the same Linnean binomial name combination, i.e. homonyms, is to be avoided for obvious reasons. In a review of sponge taxon names of the World Porifera Database, we detected 121 homonymic cases (115 species-group names, 6 genus-group names), involving a total of 272 nominal taxa. It is the object of the present study to remove their occurrence by proposing new names for the junior homonyms following the rules of the International Commission of Zoological Nomenclature as laid down in the Code (ICZN, 1999) and the on-line edition http://iczn.org/iczn/index.jsp . Homonym cases are discussed and, where applicable, junior homonyms are either replaced by nomina nova or reassigned to their earliest available synonyms. The order in which the homonyms are treated is alphabetical on original species name, with genus names separately treated at the end. A summary table with all proposed name changes is also presented to allow quick access to the junior homonyms and their proposed new names. A total of 116 nomina nova are proposed, including five new genus names.

Thirteen nitrogen-containing molecules (1a/1b and 2-12) were isolated from the Indonesian sponge Acanthostrongylophora ingens, highlighting the richness of this organism as a source of alkaloids. Their structures were elucidated using one- and two-dimensional NMR spectroscopy and HR-ESI-MS, while the stereochemistry of the diketopiperazines was established using Marfey's method. All compounds were screened in our standard bioactivity assays, including antibacterial, antikinases, and amyloid ß-42 assays. The most interesting bioactivity result was obtained with the known acanthocyclamine A (3), which revealed for the first time a specific Escherichia coli antimicrobial activity and an inhibitory effect on amyloid ß-42 production induced by aftin-5 and no cytotoxicity at the dose of 26 µM. These results highlight the potentiality of a bipiperidine scaffold as a promising skeleton for preventing or reducing the production of amyloid ß-42, a key player in the initiation of Alzheimer's disease.

African sponges, particularly freshwater sponges, are understudied relative to demosponges in most other geographical regions. Freshwater sponges (Spongillida) likely share a common ancestor; however, their evolutionary history, particularly during their radiation into endemic and allegedly cosmopolitan groups, is unclear. Freshwater sponges of at least 58 species of 17 genera and four families are described from Central and Eastern Africa, but the diversity is underestimated due to limited distinguishable morphological features. The discovery of additional cryptic species is very likely with the use of molecular techniques such as DNA barcoding. The Royal Museum of Central Africa (MRAC, Tervuren, Belgium) hosts one of the largest collections of (Central) African freshwater sponge type material. Type specimens in theory constitute ideal targets for molecular taxonomy; however, the success is frequently hampered by DNA degradation and deamination, which are a consequence of suboptimal preservation techniques. Therefore, we genotyped African demosponge holotype material of the MRAC with specific short primers suitable for degenerated tissue and compare the results with the current, morphology-based classification. Our results demonstrate the utility of minimalistic barcodes for identification of sponges, potentially enabling efficient identification of individuals in taxonomic or metabarcoding studies, and highlight inconsistencies in the current freshwater sponge classification.

Past taxonomic studies of Western Indian Ocean and Red Sea Calcarea have been few and sporadic (e.g. Schuffner 1877, Jenkin 1908, Row 1909, Dendy 1913, 1916, Voigt et al. 2017, 2018). Nevertheless, approximately 70 species are known from these studies for the considered region, but the descriptions of the older records often lack sufficient details for reliable identification. We studied the Western Indian Ocean Calcarea collection kept in the Naturalis Biodiversity Center. Available specimens numbered 145, collected in the Red Sea, Seychelles, Maldives, Mayotte and Rodrigues, in addition to incidental samples from Oman, the Lakshadweep Islands, the Mozambique Channel, and Eastern South Africa. Using a combination of techniques (in situ and 'on deck' photography, detailed field notes, light microscopic studies and measurements, SEM microscopy, and selected DNA sequencing) we identified 45 species, divided over the two main classes Calcinea (24 spp.) and Calcaronea (21 spp.). Not all species could be definitely assigned to an already described or a new species, as seven remained qualified as 'spec.' or 'aff.' for reasons of insufficient material or lack of details of in situ habitus. Sixteen species appeared to be new to science: Borojevia voigti sp.nov., Borojevia tubulata sp.nov., Borojevia pirella sp.nov., Clathrina rodriguesensis sp.nov., Clathrina maremeccae sp.nov., Clathrina repens sp.nov., Leucascus schleyeri sp.nov., Leucetta sulcata sp.nov., Ute insulagemmae sp.nov., Leucandra pilula sp.nov., Leucandra mozambiquensis sp.nov., Grantessa woerheidei sp.nov., Sycettusa hirsutissima sp.nov., Vosmaeropsis glebula sp.nov., Paraleucilla erpenbecki sp.nov., and Kebira tetractinifera sp.nov. For a selection of the identified species from the Western Indian Ocean and the Red Sea (30 spp.), as well as from Indonesian material (22 spp.) published previously (see Van Soest De Voogd 2015) we obtained sequences of the partial 28S gene of nuclear rDNA (C2-D2 region, cf. Voigt Wörheide 2016). The sequences of the Western Indian Ocean and Red Sea species were used to assign these to genera and families based on a phylogenetic analysis using MEGA pack vs. 06.6 for Mac of the available dataset. The Indonesian sequences supplemented by partial 28S sequences taken from the Sponge Barcode Project website and the NCBI website were included in the phylogenetic analysis to confirm the assignments. The results were compared and discussed with additional information on regional Calcarea not represented in our material. The latter chapter yielded the discovery of a preoccupied name leading to Sycon oscari nom.nov. for a species described from Mauritius.

Sponges collected on the Guyana Shelf, predominantly in Suriname offshore waters, by Dutch HMS 'Snellius' O.C.P.S. 1966, HMS 'Luymes' O.C.P.S. II 1969, and HMS 'Luymes' Guyana Shelf 1970 expeditions are described in this study. Sponges were obtained by trawling, dredging or grabbing on sandy, muddy, shelly, and fossil reef bottoms at 88 stations between 19 and 681 m depth. A total of 351 samples were identified to species level, each consisting of one or more specimens of a given species from each individual station (together comprising 547 individuals and fragments). The collection yielded 119 species together belonging to all sponge classes, but in large majority are Demospongiae. All species are identified to species level, occasionally tentatively, and all are described and illustrated. A new subgenus is proposed, Tedania (Stylotedania) subgen. nov. and a previously synonymized genus, Tylosigma Topsent, 1894 is revived. Thirtysix species were found to be new to science, excluding the first Central West Atlantic record of the genus Halicnemia, not named at the species level because of lack of sufficient material. The new species erected are, in alphabetical order: Amphoriscus ancora sp. nov., Biemna rhabdotylostylota sp. nov., Callyspongia (Callyspongia) scutica sp. nov., Chelonaplysilla americana sp. nov., Cladocroce guyanensis sp. nov., Clathria (Axosuberites) riosae sp. nov., Clathria (Clathria) gomezae sp. nov., Clathria (Microciona) snelliusae sp. nov., Clathria (Thalysias) complanata sp. nov., Clathria (Thalysias) zeai sp. nov., Coelosphaera (Coelosphaera) lissodendoryxoides sp. nov., Craniella crustocorticata sp. nov., Diplastrella spirastrelloides sp. nov., Epipolasis tubulata sp. nov., Erylus rhabdocoronatus sp. nov., Erylus surinamensis sp. nov., Geodia pocillum sp. nov., Geodia sulcata sp. nov., Hemiasterella camelus sp. nov., Hymedesmia (Stylopus) alcoladoi sp. nov., Hymenancora cristoboi sp. nov., Penares sineastra sp. nov., Hymerhabdia kobluki sp. nov., Leucosolenia salpinx sp. nov., Petrosia (Strongylophora) devoogdae sp. nov., Placospongia ruetzleri sp. nov., Pyloderma tropicale sp. nov., Raspailia (Parasyringella) thamnopilosa sp. nov., Raspailia (Raspailia) johnhooperi sp. nov., Sphaerotylus bouryesnaultae sp. nov., Spirastrella erylicola sp. nov., Stelletta vervoorti sp. nov., Suberites crispolobatus sp. nov., Tedania (Stylotedania) folium subgen. nov. sp. nov., Timea tylasterina sp. nov., and Tylosigma ostreicola sp. nov. Two new combinations are proposed: Amphimedon nanaspiculata (Hartman, 1955) comb. nov. and Oceanapia ascidia (Schmidt, 1870) comb. nov. In addition, for two preoccupied combinations, new names, Coelosphaera (Coelosphaera) barbadensis nom. nov. and Hymedesmia (Hymedesmia) rowi nom. nov., are erected. The spatial distribution of the collected specimens over the Guyana Shelf was traced and the results were compared with existing information on bottom conditions. There was a convincing correlation between the location of fossil reefs and other hard substrates such as shell ridges with peaks in the occurrence of species diversity and specimen numbers. Stations made on sand and mud bottoms away from these zones of hard substrates were generally poor in species, but some were found to be rich in individuals of specialized soft bottom dwellers such as Tetilla pentatriaena, Tribrachium schmidtii, Fangophilina submersa, and Oceanapia species. The species composition of the Guyana Shelf was compared with that of neighbouring regions of the Caribbean and from North and East Brazil (Díaz in Miloslavich et al. 2010; Muricy et al. 2011). About 35% of the species encountered are widespread in the Central West Atlantic, occurring both to the north and to the south. Indications that the Guyana Shelf sponge fauna is clearly transitional are southernmost occurrences for Caribbean species (about 30%), and northernmost occurrences of Brazilian species (13%), with a high proportion (25%) of new species, which may be expected to have distributions extending to the Caribbean, to Brazil or both regions.

The halogenated alkaloid chloromethylhalicyclamine B (1), together with the known natural compound halicyclamine B (2), was isolated from the extract of the sponge Acanthostrongylophora ingens. The structure of compound 1 was determined by spectroscopic means, and it was shown that 1 is produced by reaction of 2 with CH2Cl2 used for extraction. Compound 1 was a selective CK1δ/ε inhibitor with an IC50 of 6 µM, while the natural compound 2 was inactive. The absolute configuration of 1 was determined by quantum mechanical calculation of its ECD spectrum, and this also determined the previously unknown absolute configuration of the parent halicyclamine B (2). Computational studies, validated by NOESY data, showed that compound 1 can efficiently interact with the ATP-binding site of CK1δ in spite of its globular structure, very different from the planar structure of known inhibitors of CK1δ. This opens the way to the design of a new structural type of CK1δ/ε inhibitors.

Marine sponges are found to be a rich source of bioactive compounds which show a wide range of biological activities including antiviral, antibacterial, and anti-inflammatory activities. This study aimed to investigate the possible anti-inflammatory, antioxidant and immunomodulator effects of the methanolic extract of the Red Sea marine sponge Xestospongia testudinaria. The chemical composition of the Xestospongia testudinaria methanolic extract was determined using Gas chromatography-mass spectroscopy (GC-MS) analysis. DPPH (2, 2-diphenyl-1-picryl-hydrazyl) was measured to assess the antioxidant activity of the sponge extract. Carrageenan-induced rat hind paw edema was adopted in this study. Six groups of rats were used: group1: Control, group 2: Carrageenan, group 3: indomethacin (10 mg/kg), group 4-6: Xestospongia testudinaria methanolic extract (25, 50, and 100 mg/kg). Evaluation of the anti-inflammatory activity was performed by both calculating the percentage increase in paw weight and hisopathologically. Assessment of the antioxidant and immunomodulatory activity was performed. GC-MS analysis revealed that there were 41 different compounds present in the methanolic extract. Sponge extract exhibited antioxidant activity against DPPH free radicals. Xestospongia testudinaria methanolic extract (100 mg/kg) significantly decreased % increase in paw weight measured at 1, 2, 3 and 4 h after carrageenan injection. Histopathologically, the extract caused a marked decrease in the capillary congestion and inflammatory cells infiltrate. The extract decreased paw malondialdehyde (MDA) and nitric oxide (NO) and increased the reduced glutathione (GSH), glutathione peroxidase (GPx), and catalase (CAT) activity. It also decreased the inflammatory cytokines, tumor necrosis factor-α (TNF-α), interleukin-1 ß(IL-1ß) and IL-6. The results of this study demonstrated the anti-inflammatory, antioxidant, and immunomodulatory effects of the methanolic extract of the Red Sea sponge Xestospongia testudinaria (100 mg/kg).

The calcareous sponges collected during Indonesian-Dutch research projects, incorporated in the collections of the Naturalis Biodiversity Center (formerly the Rijksmuseum van Natuurlijke Historie and the Zoölogisch Museum of the University of Amsterdam), are described and discussed. A total of 37 species were distinguished, of which 16 are new to science, while several others are very poorly known. The new species are Clathrina purpurea sp.nov., Clathrina beckingae sp.nov., Clathrina sororcula sp.nov., Arthuria tubuloreticulosa sp.nov., Ernstia indonesiae sp.nov., Ernstia chrysops sp.nov., Ernstia klautauae sp.nov., Ernstia naturalis sp.nov., Ascandra kakaban sp.nov., Ascandra crewsi sp.nov., Ascaltis angusta sp.nov., Pericharax orientalis sp.nov., Sycetta vinitincta sp.nov., Anamixilla singaporensis sp.nov., Grantessa borojevici sp.nov. and Grantessa tenhoveni sp.nov. An additional six species reported from Indonesia, but not represented in our material, are briefly characterized.

RESUMO

Paragrantia waguensis Hôzawa is reported from coastal reefs of the island of Okinawa. This rare species was previously known only from Central Japan, Mie Prefecture. It has peculiar apopylar tetractine spicules, so far unique among Calcarea. We present in situ images of the species and a full description including SEM images of skeletal structure and spicule complement. The status of Paragrantia as a separate genus of the family Grantiidae distinct from Grantia Fleming is confirmed on the basis of a morphological and molecular comparison with the European type species of Grantia, Grantia compressa (Fabricius).

RESUMO

This report describes the use of α-glucosidase to evaluate the anti-diabetic potential of extracts from marine sponges collected in the Mauritius waters. Initial screening at 1.0 mg/mL of 141 extracts obtained from 47 sponge species revealed 10 extracts with inhibitory activity greater than 85%. Seven of the 10 extracts were further tested at 0.1 and 0.01 mg/mL and only the methanol extract of two sponges namely Acanthostylotella sp. (ASSM) and Echinodictyum pykei (EPM) showed inhibition activity greater than 60% at 0.1 mg/mL with an IC50 value of 0.16 ± 0.02 and 0.04 ± 0.01 mg/mL, respectively, while being inactive at 0.01 mg/mL.

Four submersible dives off the coast of Bonaire (Caribbean Netherlands) and Klein Curaçao (Curaçao) to depths of 99.5-242 m, covering lower mesophotic and upper dysphotic zones, yielded 52 sponge specimens belonging to 31 species. Among these we identified 13 species as new to science. These are Plakinastrella stinapa n. sp., Pachastrella pacoi n. sp., Characella pachastrelloides n. sp., Geodia curacaoensis n. sp., Caminus carmabi n. sp., Discodermia adhaerens n. sp., Clathria (Microciona) acarnoides n. sp., Antho (Acarnia) pellita n. sp., Parahigginsia strongylifera n. sp., Calyx magnoculata n. sp., Neopetrosia dutchi n. sp., Neopetrosia ovata n. sp. and Neopetrosia eurystomata n. sp. We also report an euretid hexactinellid, which belongs to the rare genus Verrucocoeloidea, recently described (2014) as V. liberatorii Reiswig & Dohrmann. The remaining 18 already known species are all illustrated by photos of the habit, either in situ or 'on deck', but only briefly characterized in an annotated table to confirm their occurrence in the Southern Caribbean. The habitat investigated-steep limestone rocks, likely representing Pleistocene fossil reefs--is similar to deep-water fossil reefs at Barbados of which the sponges were sampled and studied by Van Soest and Stentoft (1988). A comparison is made between the two localities, showing a high degree of similarity in sponge composition: 53% of the present Bonaire-Klein Curaçao species were also retrieved at Barbados. At the level of higher taxa (genera, families) Bonaire-Klein Curaçao shared approximately 80% of its lower mesophotic and upper dysphotic sponge fauna with Barbados, despite a distance between them of 1000 km, indicating high faunal homogeneity. We also preliminarily compared the shallow-water (euphotic) sponge fauna of Curaçao with the combined data available for the Barbados, Bonaire and Klein Curaçao mesophotic and upper dysphotic sponges, which resulted in the conclusion that the two faunas show only little overlap.

The genus Agelas comprises a group of tropical and subtropical reef sponges that contains large, long-lived, often brightly colored and conspicuous species, distributed throughout the tropica l western Atlantic, temperate northern Atlantic (Mediterranean Sea), and western and central Indo-Pacific Realms. Among tropical sponge genera, Agelas is one with similar species richness in the Greater Caribbean in comparison to the Indo Pacific. The presence of verticillated acanthostyle spicules and a fibroreticulate skeleton of spongin fibres cored and/or echinated by spicules characterize this group. Taxonomic identification relies on a combination of characters, where external morphology and color play a key role, owing to the paucity of microscopical characters. Thus, there is still a great deal of taxonomic confusion, even for the more common species. We carried out a detailed revision of Agelas species throughout the Greater Caribbean area using classic taxonomic tools. Samples and observations covered Colombia, Belize, Jamaica, the Bahamas, Barbados, Curaçao and Venezuela, and included type material from major museum collections. According to our results, the genus Agelas in the Caribbean has at least thirteen valid species, viz. Agelas sceptrum (Lamarck, 1815); A. dispar Duchassaing & Michelotti, 1864;  A. dilatata Duchassaing & Michelotti, 1864; A. clathrodes (Schmidt, 1870);  A. cervicornis (Schmidt, 1870); A. conifera (Schmidt, 1870); A. schmidti Wilson, 1902;   A. tubulata Lehnert & van Soest, 1996; A. wiedenmayeri Alcolado, 1984;  A. citrina Gotera & Alcolado, 1987; A. sventres Lehnert & van Soest, 1996; A. repens Lehnert & van Soest, 1998; and A. cerebrum Assmann et al., 2001. We found that variation of microscopic characteristics like skeleton arrangement, number of verticills and their spines, and spicule length and width, can be used as taxonomic tools, but only in a thorough comparison with other species in the same sub-regional context. Thus, a certain degree of familiarity with the genus' regional variation is often required. The richness and distribution of these species in the Caribbean area show north/south differences and other ecological patterns are evident.

The Caribbean barrier reef near Carrie Bow Cay, Belize, has been a focus of Smithsonian Institution (Washington) reef and mangrove investigations since the early 1970s. Systematics and biology of sponges (Porifera) were addressed by several researchers but none of the studies dealt with cryptic habitats, such as the shaded undersides of coral rubble, reef crevices, and caves, although a high species diversity was recognized and samples were taken for future reference and study. This paper is the result of processing samples taken between 1972 and 2012. In all, 122 species were identified, 14 of them new (including one new genus). The new species are Tetralophophora (new genus) mesoamericana, Geodia cribrata, Placospongia caribica, Prosuberites carriebowensis, Timea diplasterina, Timea oxyasterina, Rhaphidhistia belizensis, Wigginsia curlewensis, Phorbas aurantiacus, Myrmekioderma laminatum, Niphates arenata, Siphonodictyon occultum, Xestospongia purpurea, and Aplysina sciophila. We determined that about 75 of the 122 cryptic sponge species studied (61%) are exclusive members of the sciophilic community, 47 (39 %) occur in both, light-exposed and shaded or dark habitats. Since we estimate the previously known sponge population of Carrie Bow reefs and mangroves at about 200 species, the cryptic fauna makes up 38 % of total diversity.

Eight cyclic peroxide norterpenoids, compounds 1-8, have been isolated and characterized from the Red Sea sponge Diacarnus erythraeanus, including two new norsesterterpene derivatives (3, 4). Among these metabolites, (-)-muqubilin A (5) (nine cell lines analyzed) and the new compounds 3 and 4 (seven cell lines analyzed) displayed mean IC50 growth inhibitory concentrations in vitro of <10 µM, while the remaining compounds (1, 6-8) were inactive in these cancer cell lines. Compound 5 displayed no selectivity between normal and cancer cells in terms of in vitro growth inhibition. Quantitative video microscopy analysis carried out on (-)-muqubilin A-treated cells validated the data obtained by means of the MTT colorimetric assay, while flow cytometry analysis revealed ROS production but no induction of apoptosis in cancer cells.

An optically inactive C-20 bisacetylenic alcohol, (4E,16E)-icosa-4,16-diene-1,19-diyne-3,18-diol, was isolated from a marine sponge Callyspongia sp. as a result of screening of antilymphangiogenic agents from marine invertebrates. An optical resolution using chiral-phase HPLC gave each enantiomer, (-)-1 and (+)-2. Because the natural and synthetic enantiomers 1 and 2 showed different biological properties, we investigated the structure-activity relationships of bisacetylenic alcohols using 11 synthetic derivatives, and it is clarified that the essential structural unit for antiproliferative activity is the "1-yn-3-ol" on both termini and that there is a minimum chain length that connects the "1-yn-3-ol" moieties.
