RESUMO

Clubfoot is a common birth defect characterized by inward posturing and rigid downward displacement of one or both feet. The etiology of syndromic forms of clubfoot is varied and the causes of isolated clubfoot are not well understood. A microduplication of 2.2 Mb on chromosome 17q23.1q23.2 which includes T-box 4 (TBX4), a hindlimb-specific gene, and 16 other genes was recently identified in 3 of 66 families reported as nonsyndromic clubfoot, but additional non-foot malformations place them in the syndromic clubfoot category. Our study assesses whether variation in or around TBX4 contributes to nonsyndromic clubfoot. To determine whether this microduplication was a common cause of nonsyndromic clubfoot, 605 probands (from 148 multiplex and 457 simplex families) with nonsyndromic clubfoot were evaluated by copy number and oligonucleotide array CGH testing modalities. Only one multiplex family (0.68%) that had 16 with clubfoot and 9 with other foot anomalies, had a 350 kb microduplication, which included the complete duplication of TBX4 and NACA2 and partial duplication of BRIP1. The microduplication was transmitted in an autosomal dominant pattern and all with the microduplication had a range of phenotypes from short wide feet and toes to bilateral clubfoot. Minimal evidence was found for an association between TBX4 and clubfoot and no pathogenic sequence variants were identified in the two known TBX4 hindlimb enhancer elements. Altogether, these results demonstrate that variation in and around the TBX4 gene and the 17q23.1q23.2 microduplication are not a frequent cause of this common orthopedic birth defect and narrows the 17q23.1q23.2 nonsyndromic clubfoot-associated region.

OBJECTIVE: Women who have low cobalamin (vitamin B(12)) levels are at increased risk for having children with neural tube defects (NTDs). The transcobalamin II receptor (TCblR) mediates uptake of cobalamin into cells. Inherited variants in the TCblR gene as NTD risk factors were evaluated. METHODS: Case-control and family-based tests of association were used to screen common variation in TCblR as genetic risk factors for NTDs in a large Irish group. A confirmatory group of NTD triads was used to test positive findings. RESULTS: 2 tightly linked variants associated with NTDs in a recessive model were found: TCblR rs2336573 (G220R; p(corr)=0.0080, corrected for multiple hypothesis testing) and TCblR rs9426 (p(corr)=0.0279). These variants were also associated with NTDs in a family-based test before multiple test correction (log-linear analysis of a recessive model: rs2336573 (G220R; RR=6.59, p=0.0037) and rs9426 (RR=6.71, p=0.0035)). A copy number variant distal to TCblR and two previously unreported exonic insertion-deletion polymorphisms were described. CONCLUSIONS: TCblR rs2336573 (G220R) and TCblR rs9426 represent a significant risk factor in NTD cases in the Irish population. The homozygous risk genotype was not detected in nearly 1000 controls, indicating that this NTD risk factor may be of low frequency and high penetrance. 9 other variants are in perfect linkage disequilibrium with the associated single nucleotide polymorphisms. Additional work is required to identify the disease-causing variant. Our data suggest that variation in TCblR plays a role in NTD risk and that these variants may modulate cobalamin metabolism.

1. Ants are important predators in agricultural systems, and have complex and often strong effects on lower trophic levels. Agricultural intensification reduces habitat complexity, food web diversity and structure, and affects predator communities. Theory predicts that strong top-down cascades are less likely to occur as habitat and food web complexity decrease. 2. To examine relationships between habitat complexity and predator effects, we excluded ants from coffee plants in coffee agroecosystems varying in vegetation complexity. Specifically, we studied the effects of eliminating ants on arthropod assemblages, herbivory, damage by the coffee berry borer and coffee yields in four sites differing in management intensification. We also sampled ant assemblages in each management type to see whether changes in ant assemblages relate to any observed changes in top-down effects. 3. Removing ants did not change total arthropod densities, herbivory, coffee berry borer damage or coffee yields. Ants did affect densities of some arthropod orders, but did not affect densities of different feeding groups. The effects of ants on lower trophic levels did not change with coffee management intensity. 4. Diversity and activity of ants on experimental plants did not change with coffee intensification, but the ant species composition differed. 5. Although variation in habitat complexity may affect trophic cascades, manipulating predatory ants across a range of coffee agroecosystems varying in management intensity did not result in differing effects on arthropod assemblages, herbivory, coffee berry borer attack or coffee yields. Thus, there is no clear pattern that top-down effects of ants in coffee agroecosystems intensify or dampen with decreased habitat complexity.

It is a mainstay of community ecology that local exclusion of species will result if competitive pressures become too large. The pattern of exclusion may be complicated, but the qualitative orthodoxy has changed little since the pioneering work of Lotka, Volterra, and Gause--no two species can occupy the same niche. Stated in a more precise form, the higher the intensity of interspecific competition in an assemblage of species, the fewer the number of species that can coexist in perpetuity. We suggest that this orthodoxy results from "linear" thinking, and that if the classical equations are formulated more realistically with attendant nonlinearities, the orthodoxy breaks down and higher levels of competition may actually increase the likelihood that species will avoid competitive exclusion. Furthermore, this increased probability of coexistence at higher levels of competition is accompanied by characteristic dynamic patterns: (i) at lower levels of competition, after all extinction events have occurred, remaining species follow irregular chaotic patterns; (ii) at higher levels of competition, when most species coexist, all species are entrained in a single large limit cycle; (iii) the transient behavior appears to correspond to a special case of chaos, uniform phase chaotic amplitude.

In both strictly theoretical and more applied contexts it has been historically assumed that metapopulations exist within a featureless, uninhabitable matrix and that dynamics within the matrix are unimportant. In this article, we explore the range of theoretical consequences that result from relaxing this assumption. We show, with a variety of modeling techniques, that matrix quality can be extremely important in determining metapopulation dynamics. A higher-quality matrix generally buffers against extinction. However, in some situations, an increase in matrix quality can generate chaotic subpopulation dynamics, where stability had been the rule in a lower-quality matrix. Furthermore, subpopulations acting as source populations in a low-quality matrix may develop metapopulation dynamics as the quality of the matrix increases. By forcing metapopulation dynamics on a formerly heterogeneous (but stable within subpopulations) population, the probability of simultaneous extinction of all subpopulations actually increases. Thus, it cannot be automatically assumed that increasing matrix quality will lower the probability of global extinction of a population.

RESUMO

The debate over the maintenance of high diversity of tree species in tropical forests centers on the role of tree-fall gaps as a primary source of disturbance. Using a 10-year data series accumulated since Hurricane Joan struck the Caribbean coast of Nicaragua in 1988, we examined the pattern of species accumulation over time and with increased sampling of individuals. Our analysis shows that the pattern after a hurricane differs from the pattern after a simple tree-fall disturbance, and we conclude that pioneers are limited in large disturbances and thus do not suppress other species the way they do in smaller disturbances.

The Pain Coping Style Inventory (PCSI) is an instrument designed to measure the pain coping style of individuals. It is based on the pain coping model and typology described by Copp. A study with a convenience sample of 145 university graduates, undergraduates, and faculty revealed a Cronbach's alpha reliability coefficient of 0.90, adequate stability, and a nine factor analysis solution representing 57% of the variance. Four of Copp's five coping styles are clearly defined by the factor analysis: interactive, reactive, combatant, and victim. Five more coping styles are named: contractor, distractor, spiritual coping, substance users, and mind over matter. The development and piloting of the PCSI is a step towards empowering the nurse with the ability to provide patients with individualized pain coping strategies.

Recent changes in the coffee agroecosystem of Costa Rica were used to study the mechanism of biodiversity loss in transforming agroecosystems, focusing on the ground-foraging ant community. Coffee farms are being transformed from vegetationally diverse shaded agroforestry systems to unshaded coffee monocultures. We tested the hypothesis that the high-light environment and lack of leaf litter cover in the unshaded system are the determinants of the differences in ground-foraging ant diversity. Four treatments were established within the light gaps of a shaded plantation: shade, leaf litter, shade plus leaf litter, and a control (no shade or leaf litter added). Ants were sampled using tuna fish baits and light and temperature were measured. Shade and leaf litter had a significant effect on the ant fauna but probably for indirect reasons having to do with species interactions. In both shade treatments, Solenopsis geminata, the tropical fire ant, decreased significantly while the other species increased. The possibility that the physical factor changes the nature of competitive interactions between the most abundant species is discussed.

Health care for rape victims traditionally has been crisis oriented, focused solely upon the victim, and provided either by a gynecologist or psychiatrist. A family practice health team, working in a county hospital, offered rape victims and their families crisis counseling in the emergency room, and follow-up care in a family practice clinic. Follow-up of victims markedly increased from 8% before initiation of the program to 86% afterward. Family members of more than half of the victims seen on follow-up received treatment. A wide range of health needs of both victims and their families, not identified in the emergency room, was identified on follow-up visits. In this way emergency care of the rape victim often became an entry point into the health care system. Rape must be viewed as a family's as well as victim's health problem, often demanding a broader service than can be offered by a gynecologist or psychiatrist alone.
