RESUMO

Diluted bitumen (dilbit) is an unconventional oil produced by the oil sands industry in Canada. Despite the knowledge available on hydrocarbon toxicity, the effects of diluted bitumen on benthic organisms are still largely unknown. Moreover, in Quebec there are only provisional threshold values of 164 mg/kg C10-C50 for chronic effects and 832 mg/kg for acute effects. The protectiveness of these values for benthic invertebrates has not been tested for heavy unconventional oils such as dilbit. Two benthic organisms, the larvae of Chironomus riparius and Hyalella azteca, were exposed to these two concentrations and to an intermediate concentration (416 mg/kg) of two dilbits (DB1 and DB2) and a heavy conventional oil (CO). The aim of the study was to assess the sublethal and lethal effects of spiked sediment by dilbit. The oil was rapidly degraded in the sediment, especially in the presence of C. riparius. Amphipods were much more sensitive to oil than chironomids. LC50-14d values for H. azteca were 199 mg/kg C10-C50 for DB1, 299 mg/kg for DB2 and 8.42 mg/kg for CO compared to LC50-7d values for C. riparius of 492 mg/kg for DB1, 563 mg/kg for DB2 and 514 mg/kg for CO. The size of the organisms was reduced compared to controls for both species. The defense enzymes (GST, GPx, SOD and CAT) were not good biomarkers in these two organisms for this type of contamination. The current provisional sediment quality criteria seem too permissive for heavy oils and should be lowered.

This study verified the effectiveness and the potential toxic impact of PERACLEAN Ocean ballast water treatment for very cold freshwater (0.1-0.5 degrees C) in real ballast tank (750 m(3)) conditions aboard a ship and in large-volume (4.5 m(3)) polyethylene tanks. Concentrations of peracetic acid (PAA) and hydrogen peroxide (H2O2) gradually dropped by 41-59% over 5 days. The treatment altered the quality of the treated waters by causing a pH drop of 0.9-1.3 units and a fourfold to sevenfold increase in dissolved organic carbon and organophosphates concentrations. More than 90% of the biomass of free-floating micro-organisms and viable phytoplankton were eliminated within 48 h after treatment. The treatment resulted in 100% mortality in caged fish exposed to treated waters but was totally ineffective against adult zebra mussels and some nematods living in tank sediments. Toxic response from ecotoxicological assays indicated that treated waters after 5 days should be diluted by a factor of 1:2 to 1:200 to reduce toxicity below selected endpoints of acute lethality tests. On the basis of PAA degradation rate, fresh waters treated with 100-ppm PERACLEAN Ocean should be kept in ballast tanks for 15-20 days after treatment to reduce toxicity. It is concluded that the treatment can be an effective biocide to rapidly eliminate organisms of the water column inside the ballast tanks over a wide range of environmental conditions, but that the discharge of the toxic treated waters should be properly managed to minimize potential environmental impact.

RESUMO

Solanum phureja clone 1-3 and S. chacoense clone 80-1 have a zero and high leptine content in their foliage, respectively. An F(1) hybrid (CP2) was intermediate for the trait, but self-incompatible. Two reciprocal backcross families, PBCp ( phu 1-3 x CP2) and PBCc (CP2 x phu 1-3), and a family of monoploids derived by anther culture of CP2, were characterized for leptine as the aglycon, acetylleptinidine (ALD), content in leaves by gas chromatography. ALD was present in 43 of 87 genotypes in the PBCp backcross, implying simple genetic control by a dominant gene. However, the ALD levels were low compared to CP2. In the PBCc backcross, only 7 of 42 genotypes expressed ALD at a level generally higher than in PBCp. This ratio was significantly different from the 1:1 segregation observed in the reciprocal backcross and suggests a cytoplasmic influence. ALD levels in the CP2 monoploids ranged from 0 to 8,968 &mgr;g.g(-1) of dry weight (dw) with 18 individuals expressing ALD and five with 0 ALD content. Ten high (mean ALD = 546 &mgr;g.g(-1) of dw) and ten low (mean ALD = 0) individual plants within PBCp and seven high (mean ALD = 3,037 &mgr;g.g(-1) of dw) and eight low (mean ALD = 0) individual plants within PBCc were used for bulk segregant analysis (BSA) using 214 RAPD (randomly amplified polymorphic DNA) primers. Three RAPD primers (OPQ-2, OPT-16 and OPT-20) amplified bands exclusively in bulks containing DNA mixes of high ALD producers in both PBCp and PBCc populations. These results suggest that these markers were associated in coupling to ALD content. ANOVAs for ALD content verified association between the markers and the trait. A CAPS (cleaved amplified polymorphic sequence) marker, GP82A, was also significantly associated with ALD production in both the monoploid and the PBCp populations. None of the RAPD markers was associated to ALD in the monoploids but one was associated in repulsion. The monoploid data indicate the likelihood of a recessive gene(s) that controls leptine production, but the backcross data indicate the action of modifying loci.
