RESUMO

This comprehensive review is focused on a serious protozoan disease, amebiasis. This disease is caused by the human parasite Entamoeba histolytica (E. histolytica), the second leading cause of mortality due to protozoan disease worldwide (the leading cause is malaria). The incidence of amebiasis in the Czech Republic is very low, but it may be underreported as the disease often escapes diagnosis. Intestinal colonisation by E. histolytica may be asymptomatic. The clinical picture ranges from diarrhea to colitis or fulminant colitis when the parasite progresses to the trophozoite stage. Secondary dissemination in the blood or lymph system may induce systemic signs of the disease. Liver abscess is the most common extraintestinal form of amebiasis. The diagnosis of intestinal amebiasis is based on the clinical picture and parasitological examination of the stool. To diagnose extraintestinal amebiasis, serology tests are used to detect antibodies in the blood. Recently, molecular methods have been increasingly used for the detection of the nucleic acids of the pathogen in biological specimens. The first line therapy for amebiasis are 5-nitroimidazole drugs, currently available in the Czech Republic. However, surgical intervention should also be considered in patients with a severe course of the disease. Included in the review are the case reports of patients with severe concomitant intestinal and extraintestinal amebiasis.

Residues of printing ink components were determined in 94 samples of packaging materials commercially used in the Czech Republic for food packaging. The samples tested included printed polyethylene and polypropylene films, co-extruded and laminated films, paperboard beverages boxes, foils for thermo sealing of polystyrene cups, and polypropylene cups. Printing ink components were extracted with diethylether, then separated and determined using gas chromatography-mass spectrometry (GC/MS). Fifty compounds potentially originating from printing were isolated, identified and quantified. No acute health risk for consumers were identified, even though several findings of high levels of photo-initiators (e.g. in polyethylene terephthalate (PET) films for thermo sealing of polystyrene cups) as well as plasticizers (acetyl tributyl citrate, tributyl aconitate, 2-butoxyethyl oleate and 2-ethylhexyl diphenyl phosphate in co-extruded films) indicate that their real migration into food and/or food simulants needs to be undertaken for a proper safety evaluation.

RESUMO

The authors present a case-review of a 20-year-old male who suffered from a car accident polytruma (the ISS 22, brain contusion, lung contusion, serial ribs fructures, fracture of the Th8, haematoma of the left kidney with haematuria). Surgery was not indicated, his thorax and abdomen were repeatedly examined with x-rays, ultrasound and CT scans, during the patient's hospitalization and during the follow-up period. After some time, the stomach expansion in the pleural cavity imitated the clinical and roentgenological findings typical for the tension pneumothorax. Urgent drainage solved the cardiopulmonary failure and cleared up the diagnosis. This atypical case is followed by a short literature overview.

RESUMO

Silymarin is an antihepatotoxic substance isolated from fruits of Silybum marianum. Possibly due to their antioxidant and membrane stabilizing properties, the compounds have been shown to protect different organs and cells against a number of insults. The content and composition of main silymarin components (silybin, isosilybin, silydianin and silychristin) in various pharmaceuticals were analysed using HPLC and newly developed capillary zone electrophoresis method. Antioxidant properties expressed as total antioxidant status (TAS) of silymarin components were studied. Results of TAS were correlated with electropherograms and chromatograms.

RESUMO

A capillary isotachophoretic method (CITP) to determine the creatinine concentration in meat and meat products is described. A clear separation of the creatinine from other components of an acidic extract of sample was achieved within 20 min. Method characteristics (linearity, accuracy, precision and detection limit) were determined. The developed method was successfully applied to analyze real samples and to determine creatinine and creatine content (indirect determination after acidic hot extraction) in meat and meat products.

An on-line coupled capillary isotachophoresis-capillary zone electrophoresis (cITP-CZE) method for the determination of the fumaric acid content in apple juice is presented. A clear separation of fumaric acid in real samples is achieved within 20 min. The leading, terminating and background electrolyte of the employed system consist of 10 mM HCl+beta-alanine+5 mM beta-cyclodextrin+0.05% hydroxypropylmethylcelullose (HPMC), pH 3, 10 mM citric acid and 20 mM citric acid+beta-alanine+5 mM beta-cyclodextrin+0.1% HPMC, pH 3.3, respectively. The linearity, recovery, repeatability and detection limit of the developed method are 25-1000 ng/ml, 1.07%, 95.4+/-3.5 (+/-s)% and 10 ng/ml, respectively. Low laboriousness (no sample pretreatment), sufficient sensitivity and low running cost are the important attributes of the cITP-CZE method which was successfully applied to analyses of real samples of apple juices.

Benzoic anhydride and ethyl and propyl esters of 4-hydroxybenzoic acid (ETP and PRP, respectively, also termed parabens) incorporated into low density polyethylene (LDPE) film were studied with regard to migration into food and food simulants at 6 degrees C and 25 degrees C, and changes in selected properties of the film were investigated. Antimicrobials were incorporated into polymer film in concentrations of 5 g/kg and 10 g/kg. The addition of parabens into the polymer was more difficult than benzoic anhydride due to their volatility. For benzoic anhydride, 30-40% and 10-20% of the added amount was found to leach from the film into aqueous and olive oil food simulants, respectively. The migration into both water and olive oil followed a very similar course in the case of parabens. Migration levels over 90% and in the range of 70% to 80%, relative to the amount of agent in the film, were determined for ETP and PRP respectively. The incorporation of antimicrobials into the film significantly changed the functional characteristics of the packaging material, i.e. permeability of oxygen, carbon dioxide and water vapour, tensile strength, coefficient of friction, sealing strength and transparency. Shelf life tests with packaged cheese and toasted bread demonstrated the efficiency of the film containing 10 g/kg of BA against mould growth on the food surface during storage at 6 degrees C.

The main factors affecting the cellular fatty acid composition such as cultivation temperature, pH and NaCl content of cultivation medium, growth stage and the method of fat isolation are summarised. The effects of these factors on the fatty acids patterns are compared with those found after the cultivation of several strains of Lactobacillus sake and L. pentosus.
