RESUMO

Both polycyclic aromatic hydrocarbons (PAHs) and heavy metals persist in the environment and are toxic to organisms. Their co-occurrence makes any of them difficult to remove during bioremediation and poses challenges to environmental management and public health. Microorganisms capable of effectively degrading PAHs and detoxifying heavy metals concurrently are required to improve the bioremediation process. In this study, we isolated a new strain, Sphingobium sp. SJ10-10, from an abandoned coking plant and demonstrated its capability to simultaneously degrade 92.6 % of 75 mg/L phenanthrene and reduce 90 % of 3.5 mg/L hexavalent chromium [Cr(VI)] within 1.5 days. Strain SJ10-10 encodes Rieske non-heme iron ring-hydroxylating oxygenases (RHOs) to initiate PAH degradation. Additionally, a not-yet-reported protein referred to as Sphingobium chromate reductase (SchR), with low sequence identity to known chromate reductases, was identified to reduce Cr(VI). SchR is distributed across different genera and can be classified into two classes: one from Sphingobium members and the other from non-Sphingobium species. The widespread presence of SchR in those RHO-containing Sphingobium members suggests that they are excellent candidates for bioremediation. In summary, our study demonstrates the simultaneous removal of PAHs and Cr(VI) by strain SJ10-10 and provides valuable insights into microbial strategies for managing complex pollutant mixtures.

A taxonomic investigation was conducted on four bacterial strains isolated from soil contaminated with polycyclic aromatic hydrocarbons and heavy metals. Phylogenetic analysis revealed that these strains belonged to the family Chitinophagaceae. Examination of the 16S rRNA genes indicated that their sequence identities were below 97.6 % compared to any known and validly nominated bacterial species. The genomes of the four strains ranged from 4.12 to 8.76 Mb, with overall G + C molar contents varying from 41.28 % to 50.39 %. Predominant cellular fatty acids included iso-C15:0, iso-C15:1 G, and iso-C17:0 3-OH. The average nucleotide identity ranged from 66.90 % to 74.63 %, and digital DNA-DNA hybridization was 12.5-12.8 %. Based on the genomic and phenotypic features of the new strains, four novel species and two new genera were proposed within the family Chitinophagaceae. The ecological distributions were investigated by data-mining of NCBI databases, and results showed that additional strains or species of the newly proposed taxa were widely distributed in various environments, including polluted soil and waters. Functional analysis demonstrated that strains H1-2-19XT, JS81T, and JY13-12T exhibited resistance to arsenite (III) and chromate (VI). The proposed names for the four novel species are Paraflavitalea pollutisoli (type strain H1-2-19XT = JCM 36460T = CGMCC 1.61321T), Terrimonas pollutisoli (type strain H1YJ31T = JCM 36215T = CGMCC 1.61343T), Pollutibacter soli (type strain JS81T = JCM 36462T = CGMCC 1.61338T), and Polluticoccus soli (type strain JY13-12T = JCM 36463T = CGMCC 1.61341T).

There are many unidentified microbes in polluted soil needing to be explored and nominated to benefit the study of microbial ecology. In this study, a taxonomic research was carried out on five bacterial strains which were isolated and cultivated from polycyclic aromatic hydrocarbons, and heavy metals polluted soil of an abandoned coking plant. Phylogenetical analysis showed that they belonged to the phyla Proteobacteria and Actinobacteria, and their 16S rRNA gene sequence identities were lower than 98.5% to any known and validly nominated bacterial species, suggesting that they were potentially representing new species. Using polyphasic taxonomic approaches, the five strains were classified as new species of the families Microbacteriaceae and Sphingomonadaceae. Genome sizes of the five strains ranged from 3.07 to 6.60 Mb, with overall DNA G+C contents of 63.57-71.22 mol%. The five strains had average nucleotide identity of 72.38-87.38% and digital DNA-DNA hybridization of 14.0-34.2% comparing with their closely related type strains, which were all below the thresholds for species delineation, supporting these five strains as novel species. Based on the phylogenetic, phylogenomic, and phenotypic characterizations, the five novel species are proposed as Agromyces chromiiresistens (type strain H3Y2-19aT = CGMCC 1.61332T), Salinibacterium metalliresistens (type strain H3M29-4T = CGMCC 1.61335T), Novosphingobium album (type strain H3SJ31-1T = CGMCC 1.61329T), Sphingomonas pollutisoli (type strain H39-1-10T = CGMCC 1.61325T), and Sphingobium arseniciresistens (type strain H39-3-25T = CGMCC 1.61326T). Comparative genome analysis revealed that the species of the family Sphingomonadaceae represented by H39-1-10T, H39-3-25T, and H3SJ31-1T possessed more functional protein-coding genes for the degradation of aromatic pollutants than the species of the family Microbacteriaceae represented by H3Y2-19aT and H3M29-4T. Furthermore, their capacities of resisting heavy metals and metabolizing aromatic compounds were investigated. The results indicated that strains H3Y2-19aT and H39-3-25T were robustly resistant to chromate (VI) and/or arsenite (III). Strains H39-1-10T and H39-3-25T grew on aromatic compounds, including naphthalene, as carbon sources even in the presence of chromate (VI) and arsenite (III). These features reflected their adaptation to the polluted soil environment.

BACKGROUND: The platelet-derived growth factor (PDGF) family of ligands exerts their cellular effects by binding to α- and ß-tyrosine kinase receptors (PDGFRα and PDGFRß, respectively). SUMOylation is an important posttranslational modification (PTM) which regulates protein stability, localization, activation and protein interactions. A mass spectrometry screen has demonstrated SUMOylation of PDGFRα. However, the functional role of SUMOylation of PDGFRα has remained unknown. RESULTS: In the present study, we validated that PDGFRα is SUMOylated on lysine residue 917 as was previously reported using a mass spectrometry approach. Mutation of lysine residue 917 to arginine (K917R) in PDGFRα substantially decreased SUMOylation, indicating that this amino acid residue is a major SUMOylation site. Whereas no difference in the stability of wild-type and mutant receptor was observed, the K917R mutant PDGFRα was less ubiquitinated than wild-type PDGFRα. The internalization and trafficking of the receptor to early and late endosomes were not affected by the mutation, neither was the localization of the PDGFRα to Golgi. However, the K917R mutant PDGFRα showed delayed activation of PLC-γ and enhanced activation of STAT3. Functional assays showed that the mutation of K917 of PDGFRα decreased cell proliferation in response to PDGF-BB stimulation. CONCLUSIONS: SUMOylation of PDGFRα decreases ubiquitination of the receptor and affects ligand-induced signaling and cell proliferation.

Microplastics in remote areas has received increasing concern in recent years. However, studies on microplastics in alpine rivers and their affecting factors are still limited. In this study, we investigate the abundance and characteristics of microplastic in the surface water of five alpine rivers in Qilian Mountain, China. Utilizing sieve collection, digestion and density separation, along with microscopy and Raman spectroscopy analyses, microplastics were observed in all the water samples and the average abundance of microplastics was 0.48 ± 0.28 items/L, which was lower than in other freshwaters. Transparent (37.3%) and fibrous (72.1%) microplastics were predominant. Polypropylene (53.8%) was the most frequently identified polymer type. Analysis of similarities (ANOSIM) and linear discriminant analysis (LDA) based on microplastic shape, color, and polymer type showed that there was no significant difference in the microplastic characteristics among rivers of Qilian Mountain. The distance decay models revealed that the similarity in microplastics characteristics was not affected by changes in watershed characteristics, such as geographical distance, elevation, water quality, and land use. This finding suggests that the primary source of microplastics in Qilian Mountain rivers could be from dispersed origins. The results of this study indicated that despite remote alpine rivers suffering limited anthropogenic impacts, they were not immune to microplastics. However, in watersheds with lower intensity of human activity, the abundance and characteristics of microplastics in water bodies may be more uniformly distributed and controlled by diffusion conditions such as atmospheric transport or riverine transport. Our investigation unveils novel understanding of microplastic dispersion in secluded alpine territories, emphasizing the crucial need for managing atmospheric transport of microplastics within conservation areas.

Land use influences the variation of river water quality. This effect varies depending on the region of the river and the spatial scale at which land use is calculated. This study investigated the influence of land use on river water quality in Qilian Mountain, an important alpine river region in northwestern China, on different spatial scales in the headwaters and mainstem areas. Redundancy analysis and multiple linear regression were used to identify the optimal scales of land use for influencing and predicting water quality. Nitrogen and organic carbon parameters were more influenced by land use than phosphorus. The impact of land use on river water quality varied according to regional and seasonal differences. Water quality in headwater streams was better influenced and predicted by land use types on the natural surface at the smaller buffer zone scale, while water quality in mainstream rivers was better influenced and predicted by land use types associated with human activities at the larger catchment or sub-catchment scale. The impact of natural land use types on water quality differed with regional and seasonal variations, while the impact of land types associated with human activities on water quality parameters mainly resulted in elevated concentrations. The results of this study suggested that different land types and spatial scales needed to be considered to assess water quality influences in different areas of alpine rivers in the context of future global change.

Contaminated sites from electronic waste (e-waste) dismantling and coking plants feature high concentrations of heavy metals (HMs) and/or polycyclic aromatic hydrocarbons (PAHs) in soil. Mixed contamination (HMs + PAHs) hinders land reclamation and affects the microbial diversity and function of soil microbiomes. In this study, we analyzed HM and PAH contamination from an e-waste dismantling plant and a coking plant and evaluated the influences of HM and PAH contamination on soil microbiomes. It was noticed that HMs and PAHs were found in all sites, although the major contaminants of the e-waste dismantling plant site were HMs (such as Cu at 5,947.58 ± 433.44 mg kg-1, Zn at 4,961.38 ± 436.51 mg kg-1, and Mn at 2,379.07 ± 227.46 mg kg-1), and the major contaminants of the coking plant site were PAHs (such as fluorene at 11,740.06 ± 620.1 mg kg-1, acenaphthylene at 211.69 ± 7.04 mg kg-1, and pyrene at 183.14 ± 18.89 mg kg-1). The microbiomes (diversity and abundance) of all sites were determined via high-throughput sequencing of 16S rRNA genes, and redundancy analysis was conducted to investigate the relations between soil microbiomes and contaminants. The results showed that the microbiomes of the contaminated sites divergently responded to HMs and PAHs. The abundances of the bacterial genera Sulfuritalea, Pseudomonas, and Sphingobium were positively related to PAHs, while the abundances of the bacterial genera Bryobacter, Nitrospira, and Steroidobacter were positively related to HMs. This study promotes an understanding of how soil microbiomes respond to single and mixed contamination with HMs and PAHs.

Quantifying microplastic accumulation in aquatic organisms is important for risk assessments in natural habitats. However, studies on the factors influencing microplastic accumulation by fish, particularly in natural water bodies, remain limited. In this study, we monitored the microplastic pollution characteristics in the fish digestive tract and surface water of the Nandu River, the largest river in Hainan, China. Among the 179 fish examined, microplastics were detected in 90.5 %, covering 27 fish species. Moreover, low abundance of microplastics occurred among the different fish, with an average of 3.20 ± 2.60 particles per fish. Fibrous microplastics with a large size (>500 µm) were dominant in both the water column and fish. In this study, fish size, feeding habits, and living habitats did not affect microplastic abundance in fish. Based on the analysis of differences in the abundance and morphological characteristics of microplastics in water column and fish, we suggested that the random intake and differential excretion of microplastics with different characteristics by fish might lead to the homogeneity of microplastic accumulation in different fish. Our study suggest that more attention should be given to the negative effects of microplastics on small fish owing to the higher microplastic exposure per gram weight, and further improvement in microplastic risk assessment methods is required.

Microplastics have been reported in a wide range of aquatic habitats. The wetlands are considered to be important roles in microplastic migration in water bodies. Nevertheless, knowledge about the occurrence and fate of microplastics in urban natural wetland is still limited for us to better understand how they become a sink of microplastics. In this study, the distribution and characteristics of microplastics in surface water, surface sediments, and sediment cores of the Lalu Wetland watershed, China's highest urban wetland, were investigated in August 2020 and January 2021. The abundances of microplastics in the surface water were 0.06-3.05 MPs/L. Microplastic abundance in the surface sediment and sediment core was 0.01-1.10 MPs/g and 0-16.23 MPs/g, respectively. The abundance of microplastics in the water was significantly lower in the wetland than that in the channel in the watershed. Comparing the wetland inlet and outlet water, the microplastic interception rates were 53% in January and 95% in August. The characteristics and seasonal variation of microplastics in the Lalu Wetland implied that urban natural wetlands were good at intercepting microplastics, and vegetation growth might play an important role on the interception of microplastics by the wetland. The increasing of microplastics from bottom to top in the sediment cores of Lalu Wetland also indicated that the ecological risks of microplastics accumulation in sediments of urban natural wetland required further attention.

Interaction of platelet-derived growth factor (PDGF) isoforms with their receptors results in activation and internalization of receptors, with a concomitant activation of downstream signalling pathways. Ubiquitination of PDGFRs serves as a mark to direct the internalization and sorting of the receptors. By overexpressing a panel of deubiquitinating enzymes (DUBs), we found that USP17 and USP4 efficiently deubiquitinate PDGF receptor ß (PDGFRß) and are able to remove both Lys63 and Lys48-linked polyubiquitin chains from the receptor. Deubiquitination of PDGFRß did not affect its stability, but regulated the timing of its trafficking, whereby USP17 prolonged the presence of the receptor at the cell surface, while USP4 affected the speed of trafficking towards early endosomes. Induction of each of the DUBs in BJhTERT fibroblasts and U2OS osteosarcoma cells led to prolonged and/or shifted activation of STAT3 in response to PDGF-BB stimulation, which in turn led to increased transcriptional activity of STAT3. Induction of USP17 promoted acute upregulation of the mRNA expression of STAT3-inducible genes STAT3, CSF1, junB and c-myc, while causing long-term changes in the expression of myc and CDKN1A. Deletion of USP17 was lethal to fibroblasts, while deletion of USP4 led to a decreased proliferative response to stimulation by PDGF-BB. Thus, USP17- and USP4-mediated changes in ubiquitination of PDFGRß lead to dysregulated signalling and transcription downstream of STAT3, resulting in defects in the control of cell proliferation.

TC31 is a new type of α+ß dual phase high temperature titanium alloy, which has a high specific strength and creep resistance at temperatures from 650 °C to 700 °C. It has become one of the competitive candidates for the skin and air inlet components of hypersonic aircraft. However, it is very difficult to obtain the best forming windows for TC31 and to form the corresponding complex thin-walled components. In this paper, high temperature tensile tests were carried out at temperatures ranging from 850 °C to 1000 °C and strain rates ranging from 0.001 s-1 to 0.1 s-1, and the microstructures before and after deformation were characterized by an optical microscope, scanning electron microscope, and electron back-scatter diffraction. The dynamic softening and hardening behaviors and the corresponding micro-mechanisms of a TC31 titanium alloy sheet within hot deformation were systematically studied. The effects of deformation temperature, strain rate, and strain on microstructure evolution were revealed. The results show that the dynamic softening and hardening of the material depended on the deformation temperature and strain rate, and changed dynamically with the strain. Obvious softening occurred during hot tensile deformation at a temperature of 850 °C and a strain rate of 0.001 s-1~0.1 s-1, which was mainly caused by void damage, deformation heat, and dynamic recrystallization. Quasi-steady flowing was observed when it was deformed at a temperature of 950 °C~1000 °C and a strain rate of 0.001 s-1~0.01 s-1 due to the relative balance between the dynamic softening and hardening. Dynamic hardening occurred slightly with a strain rate of 0.001 s-1. Mechanisms of dynamic recrystallization transformed from continuous dynamic recrystallization to discontinuous dynamic recrystallization with the increase in strain when it was deformed at a temperature of 950 °C and a strain rate of 0.01 s-1. The grain size also decreased gradually due to the dynamic recrystallization, which provided an optimal forming condition for manufacturing thin-walled components with the desired microstructure and an excellent performance.

In this paper, the fatigue performance of the aluminide layer-coated and as-received MAR 247 nickel superalloy with three different initial microstructures (fine grain, coarse grain and column-structured grain) was monitored using nondestructive, eddy current methods. The aluminide layers of 20 and 40 µm were obtained through the chemical vapor deposition (CVD) process in the hydrogen protective atmosphere for 8 and 12 h at the temperature of 1040 °C and internal pressure of 150 mbar. A microstructure of MAR 247 nickel superalloy and the coating were characterized using light optical microscopy (LOM), scanning electron microscopy (SEM) and X-ray energy dispersive spectroscopy (EDS). It was found that fatigue performance was mainly driven by the initial microstructure of MAR 247 nickel superalloy and the thickness of the aluminide layer. Furthermore, the elaborated methodology allowed in situ eddy current measurements that enabled us to localize the area with potential crack initiation and its propagation during 60,000 loading cycles.

High-efficiency and low-cost hot forming technologies for titanium alloys have been developed for producing complex-shaped, thin-walled tubular components under non-superplastic forming conditions. Under these forming conditions, there exist complex and highly integrated material evolution processes including microscopic heterogeneous deformation, microstructure evolution and damage behaviour. This paper presents an integrated crystal plasticity finite element model of near-α titanium alloys during non-superplastic hot deformation conditions considering grain boundary sliding (GBS), dynamic recrystallisation (DRX), as well as void evolution. The polycrystalline model of a near-α TA15 titanium alloy was established, containing α phase, ß phase and grain boundary (GB) regions, in which the GB region was a visualised representation of GBS. The quantitative strength ratio between the GB regions and α phase was calculated according to the Zener-Holloman parameter Z and grain size, which determined the microscopic deformation behaviour. There were found to be two high microscopic strain regions in the α phase: intragranular deformation bands through the most favourable slipping and near the GBs through multiple slipping, which promoted continuous and discontinuous DRX, respectively. With the decrease in parameter Z or grain size, the activated dislocations accommodating GBS were found to no longer pile up inside the grain, but instead travel across the grain interior. Finally, methods to improve the macroscopic plastic formability were proposed for the difficult-to-form titanium alloys experiencing non-superplastic hot deformation.

In this paper, hot gas pressure forming (HGPF) of Ti-55 high temperature titanium alloy was studied. The hot deformation behavior was studied by uniaxial tensile tests at temperatures ranging from 750 to 900 °C with strain rates ranging from 0.001 to 0.05 s-1, and the microstructure evolution during tensile tests was characterized by electron backscatter diffraction. Finite element (FE) simulation of HGPF was carried out to study the effect of axial feeding on thickness distribution. Forming tests were performed to validate this process for Ti-55 alloy. Results show that when the temperature was higher than 750 °C, the elongation was large enough for HGPF of Ti-55 alloy. Dynamic recrystallization (DRX) occurred during the tensile deformation, which could refine the microstructure. The thickness uniformity of the formed part could be improved by increasing feeding length. The maximum thinning ratio decreased from 27.7% to 11.5% with the feeding length increasing from 0 to 20 mm. A qualified Ti-55 alloy component was successfully formed at 850 °C, the microstructure was slightly refined after forming, and the average post-form yield strength and peak strength were increased by 8.7% and 6.9%, respectively. Near- α titanium alloys have extensive applications in high temperature structural components of aircrafts. To manufacture complex-shaped titanium alloy panel parts with desired microstructure and good properties, an innovative low-cost hot stamping process for titanium alloy was studied in this paper. Firstly, a series of hot tensile tests and Scanning Electron Microscope (SEM) observations were performed to investigate hot deformation characteristics and identify typical microstructural evolutions. The optimal forming temperature range is determined to be from 750 °C to 900 °C for hot stamping of TA15. In addition, a unified mechanisms-based material model for TA15 titanium alloy based on the softening mechanisms of recrystallization and damage was established, which enables to precisely predict stress-strain behaviors and potentially to be implemented into Finite Element (FE) simulations for designing the reasonable processing window of structural parts for the aerospace industry.

Tobacco smoke is an important risk factor of gastric cancer. Epithelial-mesenchymal transition is a crucial pathophysiological process in cancer development. ERK5 regulation of epithelial-mesenchymal transition may be sensitive to cell types and/or the cellular microenvironment and its role in the epithelial-mesenchymal transition process remain elusive. Epigallocatechin-3-gallate (EGCG) is a promising chemopreventive agent for several types of cancers. In the present study we investigated the regulatory role of ERK5 in tobacco smoke-induced epithelial-mesenchymal transition in the stomach of mice and the preventive effect of EGCG. Exposure of mice to tobacco smoke for 12 weeks reduced expression of epithelial markers E-cadherin, ZO-1, and CK5, while the expression of mesenchymal markers Snail-1, Vimentin, and N-cadherin were increased. Importantly, we demonstrated that ERK5 modulated tobacco smoke-mediated epithelial-mesenchymal transition in mice stomach, as evidenced by the findings that tobacco smoke elevated ERK5 activation, and that tobacco smoke-triggered epithelial-mesenchymal transition was reversed by ERK5 inhibition. Treatment of EGCG (100 mg/kg BW) effectively attenuated tobacco smoke-triggered activation of ERK5 and epithelial-mesenchymal transition alterations in mice stomach. Collectively, these data suggested that ERK5 was required for tobacco smoke-triggered gastric epithelial-mesenchymal transition and that EGCG suppressed ERK5 activation to reverse tobacco smoke-triggered gastric epithelial-mesenchymal transition in BALB/c mice. These findings provide new insights into the mechanism of tobacco smoke-associated gastric tumorigenesis and the chemoprevention of tobacco smoke-associated gastric cancer.
