RESUMO

Tuberculosis (TB) is the leading cause of human death worldwide due to Mycobacterium tuberculosis (Mtb) infection. Multiple lines of evidences have illuminated the emerging role of NLRP3 inflammasome-mediated pyroptosis in the clearance of pathogenic infection. In the current study, we sought to investigate the functional role and feasible potential mechanism of BRD4 in Mtb-infected macrophages. We observed that BRD4 was distinctly ascended in THP-1 macrophages upon Mtb infection. Functionally, intervention of BRD4 or pretreated with JQ1 obviously restricted Mtb-triggered cell pyroptosis, as evidenced by declination of protein level of the specific pyroptosis markers including Cleaved Caspase 1, gasdermin D (GSDMD-N) and Cleaved-IL-1ß. In the meanwhile, disruption of BRD4 or JQ1 application remarkably prohibited excessive inflammatory responses as characterized by reduce the production of the inflammatory factors such as IL-1ß and IL-18. Concomitantly, disruption of BRD4 or administrated with JQ1 manifestly repressed Mtb-aroused Nod-like receptor family pyrindomain-containing 3 (NLRP3) inflammasome activation, as witnessed by attenuation of protein levels of NLRP3, Pro-Caspase1 and apoptosis-associated speck-like protein (ASC). The above findings clearly demonstrated that suppression of BRD4 exerted great influence on regulating Mtb-elicited inflammatory response by coordinating NLRP3 inflammasome-mediated pyroptosis. More importantly, perturbation of BRD4 or JQ1 employment notably restrained endoplasmic reticulum (ER) stress triggered by Mtb-infection, as reflected by noticeably lessened the levels of GRP78, CHOP and ATF6. In terms of mechanism, ER stress agonist tunicamycin profoundly abrogated the favorable effects of BRD4 inhibition on Mtb-triggered pyroptosis, inflammation reaction and inflammasome activation. Collectively, these preceding outcomes strongly illuminated that inhibition of BRD4 targeted ER stress to retard NLRP3 inflammasome activation and subsequent cell pyroptosis and prevention of inflammatory response in Mtb-infected macrophages, highlighting that blocking BRD4 might serve as a promising candidate for protection against Mtb-triggered inflammatory injury.

Influenced by heating, the concentration of atmospheric fine particulate matter (PM2.5) rises in autumn and winter in northern cities. In this study, Q-ACSM, AE33, and Xact 625 were used to carry out online monitoring of PM2.5 chemical components with high time resolution in Xi'an from October 25 to November 17, 2019, to analyze the characteristics of PM2.5 pollution during the transition period of the heating season. Additionally, we analyzed the sources of PM2.5 in combination with the positive matrix factorization model. The results showed that the average PM2.5 concentration during the observation period was (78.3 ± 38.5) µg·m-3, and the main chemical components were organic matter (OA), secondary inorganic ions (SIA), and dust, which accounted for 38.7%, 31.6%, and 21.2%, respectively. The average concentrations of sulfate, nitrate, and ammonium were (4.0 ± 3.1), (14.9 ± 13.7), and (5.8 ± 4.8) µg·m-3, and the average concentrations of the major metals potassium, calcium, and iron were (1.0 ± 0.4), (1.5 ± 1.1), and (1.4 ± 0.9) µg·m-3. Black carbon, chloride ions, and trace elements contributed relatively little to PM2.5 (5.7%, 1.3%, and 1.5%, respectively). In the pollution development and maintenance stage, the concentration of OA and SIA increased by 137.7% to 537.0%, whereas in the pollution dissipation stage, only the concentration of dust gradually increased. The source apportionment results showed that secondary sources, biomass burning, dust, vehicle emission, industrial emission, and coal combustion were the main sources of PM2.5 during the observation period, contributing 29.1%, 21.1%, 15.3%, 12.9%, 11.4%, and 10.2%, respectively. The contribution rate of secondary sources and biomass burning was higher in the pollution development and maintenance stage, and dust was higher in the pollution dissipation stage.

PURPOSE: Pleomorphic xanthoastrocytoma (PXA) is an uncommon astrocytoma that tends to occur in children and young adults and has a relatively favorable prognosis. The 2021 WHO classification of tumors of the central nervous system (CNS WHO), 5th edition, rates PXAs as grade 2 and grade 3. The histological grading was based on mitotic activity (≥2.5 mitoses/mm2). This study specifically evaluates the clinical, morphological, and, especially, the molecular characteristics of grade 2 and 3 PXAs. METHODS: Between 2003 and 2021, we characterized 53 tumors with histologically defined grade 2 PXA (n = 36, 68%) and grade 3 PXA (n = 17, 32%). RESULTS: Compared with grade 2 PXA, grade 3 PXA has a deeper location and no superiority in the temporal lobe and is more likely to be accompanied by peritumoral edema. In histomorphology, epithelioid cells and necrosis were more likely to occur in grade 3 PXA. Molecular analysis found that the TERT promoter mutation was more prevalent in grade 3 PXA than in grade 2 PXA (35% vs. 3%; p = 0.0005) and all mutation sites were C228T. The cases without BRAF V600E mutation or with necrosis in grade 3 PXA had a poor prognosis (p = 0.01). CONCLUSION: These data define PXA as a heterogeneous astrocytoma. Grade 2 and grade 3 PXAs have different clinical and histological characteristics as well as distinct molecular profiles. TERT promoter mutations may be a significant genetic event associated with anaplastic progression. Necrosis and BRAF V600E mutation play an important role in the prognosis of grade 3 PXA.

OBJECTIVE: To investigate the accuracy and safety of pedicle screw placement assisted by orthopedic robot and C-arm fluoroscopy. METHODS: The clinical data of 36 patients with spinal diseases underwent surgical treatment from January 2019 to August 2020 was retrospectively analyzed. Among them, 18 cases were implanted pedicle screws assisted by orthopaedic robot(observation group), including 12 males and 6 females, aged from 16 to 61 years with an average of (38.44±3.60) years;there were 1 case of adolescent scoliosis, 1 case of spinal tuberculosis, 7 cases of lumbar spondylolisthesis, 4 cases of thoracic fracture and 5 cases of lumbar fracture. Another 18 cases were implanted pedicle screws assisted by C-arm fluoroscopy(control group), including 10 males and 8 females, aged from 18 to 58 years with an average of (43.22±2.53) years;there were 1 case of adolescent scoliosis, 6 cases of lumbar spondylolisthesis, 6 cases of thoracic fracture and 5 cases of lumbar fracture. The intraoperative fluoroscopy times, nail placement time and postoperative complications were recorded in two groups. CT scan was performed after operation. The Gertzbein-Robbins standard was used to evaluate the accuracy of pedicle screw placement which was calculated. RESULTS: The number of intraoperative fluoroscopy in observation group was(6.89±0.20) times, which was significantly higher than that in control group(14.00±0.18)times(P<0.05). The placement time of each screw in observation group was(2.56±0.12) min, which was significantly different from that in control group(4.22±0.17) min (P<0.05). One case of incision infection occurred in control group after operation, and recovered after active dressing change. During the follow-up period, no serious complications such as screw loosening and fracture occurred in two groups, and there was no significant difference in complications between two groups(P>0.05). A total of 107 screws were placed in observation group, including 101 screws in class A, 4 in class B, 2 in class C, 0 in class D and 0 in class E, the accuracy rate of pedicle screw placement=[(number of screws in class A+B) / the number of all screws placed in the group] ×100%=98.1%(105/107); and a total of 104 screws were placed in control group, including 90 screws in class A, 4 in class B, 5 in class C, 5 in class D and 0 in class E, the accuracy rate of pedicle screw implantation=[(number of screws in class A+B/the number of all screws placed in the group]×100%=90.3% (94/104); there was significant difference between two groups (P<0.05). CONCLUSION: Orthopaedic robot assisted pedicle screw placement has the advantages of less fluoroscopy times, shorter screw placement time and higher accuracy, which can further improve the surgical safety and has a broad application prospect in the orthopaedic.

We present a study of the light-absorbing properties of water-soluble brown carbon (WS-BrC) and methanol-soluble brown carbon (MeS-BrC) at a remote site (Lulang, 3326m above sea level) in the southeastern Tibetan Plateau during the period 2015-2016. The light absorption coefficients at 365nm (babs365) of WS-BrC and MeS-BrC were the highest during winter and the lowest during monsoon season. MeS-BrC absorbs about 1.5 times higher at 365nm compared to WS-BrC. The absorption at 550nm appears lower compared to that of 365nm for WS-BrC and MeS-BrC, respectively. Higher average value of the absorption Ångström exponent (AAE, 365-550nm) was obtained for MeS-BrC (8.2) than that for WS-BrC (6.9). The values of the mass absorption cross section at 365nm (MAC365) indicated that BrC in winter absorbs UV-visible light more efficiently than in monsoon. The results confirm the importance of BrC in contributing to light-absorbing aerosols in this region. The understanding of the light absorption properties of BrC is of great importance, especially in modeling studies for the climate effects and transport of BrC in the Tibetan Plateau.

We present a study of aerosol light absorption by using a 7-wavelength Aethalometer model AE33 at an urban site (Lhasa) and a remote site (Lulang) in the Tibetan Plateau. Approximately 5 times greater aerosol absorption values were observed at Lhasa (53±46Mm-1 at 370nm and 20±18Mm-1 at 950nm, respectively) in comparison to Lulang (15±19Mm-1 at 370nm and 4±5Mm-1 at 950nm, respectively). Black carbon (BC) was the dominant light absorbing aerosol component at all wavelengths. The brown carbon (BrC) absorption at 370nm is 32±15% of the total aerosol absorption at Lulang, whereas it is 8±6% at Lhasa. Higher value of absorption Ångström exponent (AAE, 370-950nm) was obtained for Lulang (1.18) than that for Lhasa (1.04) due to the presence of BrC. The AAEs (370-950nm) of BrC were directly extracted at Lulang (3.8) and Lhasa (3.3). The loading compensation parameters (k) increased with wavelengths for both sites, and lower values were obtained at Lulang than those observed at Lhasa for all wavelengths. This study underlines the relatively high percentage of BrC absorption contribution in remote area compared to urban site over the Tibetan Plateau.

PM2.5 mass and selected chemical species are measured in 24-h integrated PM2.5 samples collected simultaneously at the urban and rural regions of Xi'an (six sites in total), China in the four seasons of 2010. The analytes include organic carbon and elemental carbon (OC+EC = total carbon, TC), seven water-soluble inorganic ions (NH4(+), K(+), Mg(2+), Ca(2+), Cl(-), SO4(2-), NO3(-)) and six trace elements (Ti, Mn, Fe, Zn, As, Pb). The average PM2.5 mass for the entire measurement period is 142.6 ± 102.7 µg m(-3), which is more than four times that of the Chinese national ambient air quality standard. Spatial variations in PM2.5 mass are not pronounced. The PM2.5 mass and those species measured show a similar seasonal pattern in all six measurement sites, i.e., in the order of winter > autumn > spring > summer. The dominant PM2.5 composition is OC in winter, soil dust in spring, and sulfate, nitrate, and ammonium in summer and autumn. Seasonal variations of TC/PM2.5 and OC/EC ratios follow the PM2.5 changes. Seasonal distributions of (SO4(2-)+NO3(-)+NH4(+))/PM2.5 showed increase in autumn and decrease in winter, while NO3(-)/SO4(2-) ratios increased in autumn and decreased in summer. Eight main PM2.5 sources are identified based on the positive matrix factorization (PMF) analysis and emissions from fossil fuel combustion (traffic and coal burning) are founded to be the main source responsible for the fine particle pollution in Xi'an. In addition, a decreasing trend in OC/PM2.5 is observed in comparison with previous studies in Xi'an.

A total of 131 samples of PM2.5 were collected at urban area of Chengdu from April, 2009 to January, 2010. The secondary water-soluble inorganic ions (NH4(+), NO3(-) and SO4(2-)) in PM2.5 were determined to study the pollution characteristics by Ion Chromatography method. The results showed the NH4(+), NO3(-) and SO4(2-) concentrations were (10.4 ± 8.6), (19.7 ± 14.6) and (32.8 ± 21.8) µg x m(-3), accounting for (5.5 ± 2.8)%, (11.1 ± 3.5)% and (19.3 ± 6.4)% of PM2.5, respectively, and the sum of these three ions accounted for (35.9 ± 12.7)%. Meanwhile, the seasonal characteristics of secondary inorganic ions were obvious. The seasonal concentrations order of secondary inorganic components was SO4(2-) > NO3(-) > NH4(+) in summer and winter, and the proportion of secondary inorganic ions of PM2.5 was 39.4% in summer and 44.3% in winter. The results of Correlation Analysis showed that most of NH4(+), NO3(-) and SO4(2-) existed as NH4HSO4, (NH4 )2SO4 and NH4NO5 in Chengdu. And the average ratio of NO3(-)/SO4(2-) indicated that the main source of sulfur and nitrogen in the atmosphere was given priority to a fixed source. In addition, the sulfate oxidizing rate (SOR) and nitrate oxidizing rate (NOR) were 0.33 ± 0.12 and 0.19 ± 0.09, respectively. It showed that SO4(2-) and NO3(-) in PM2.5 were due to the secondary transformation.

The racemic title compound, C(14)H(12)ClNO, contains two mol-ecules in the asymmetric unit. The dihedral angles between the phenyl and benzene rings are 84.03 (15) and 83.92 (13)°. The crystal structure involves inter-molecular N-H⋯O, C-H⋯Cl and C-H⋯O hydrogen bonds, linking mol-ecules into layers parallel to the (100) plane.
