BACKGROUND: Investigating the unexplored territory of lncRNA m6A modification in colorectal cancer (CRC) vasculature, this study focuses on LINC01106 and YTHDF1. METHODS: Clinical assessments reveal upregulated LINC01106 promoting vascular generation via the miR-449b-5p-VEGFA pathway. RESULTS: YTHDF1, elevated in CRC tissues, emerges as an adverse prognostic factor. Functional experiments showcase YTHDF1's inhibitory effects on CRC cell dynamics. Mechanistically, Me-CLIP identifies m6A-modified LINC01106, validated as a YTHDF1 target through Me-RIP. CONCLUSIONS: This study sheds light on the YTHDF1-mediated m6A modification of LINC01106, presenting it as a key player in suppressing CRC vascular generation.

Hepatocellular carcinoma (HCC) poses a significant challenge with dismal survival rates, necessitating a deeper understanding of its molecular mechanisms and the development of improved therapies. Metabolic reprogramming, particularly heightened glycolysis, plays a crucial role in HCC progression. Glycolysis-associated genes (GAGs) emerge as key players in HCC pathogenesis, influencing the tumor microenvironment and immune responses. This study aims to investigate the intricate interplay between GAGs and the immune landscape within HCC, offering valuable insights into potential prognostic markers and therapeutic targets to enhance treatment strategies and patient outcomes. Through the exploration of GAGs, we have identified two distinct molecular glycolytic subtypes in HCC patients, each exhibiting significant differences in both the immune microenvironment and prognosis. A risk model comprising five key GAGs was formulated and subsequently evaluated for their predictive accuracy. Our findings underscore the diverse tumor microenvironment and immune responses associated with the varying glycolytic subtypes observed in HCC. The identified key GAGs hold promise as prognostic indicators for evaluating HCC risk levels, predicting patient outcomes, and guiding clinical treatment decisions, particularly in the context of anticipating responses to immunotherapy drugs.

Site-selective excitation (SSE), which is usually realized by tuning the wavelength of absorbed light, is an ideal way to study bond-selective chemistry, analyze the crystal structure, investigate protein conformation, etc., eventually leading to active manipulation of desired processes. Herein, SSE has been explored in (110)-, (100)-, and (011)-faced rutile TiO2, a prototypical material in both surface science and photocatalysis fields. Using ultraviolet photoelectron spectroscopy and photon energy-, substrate orientation-, and laser polarization-dependent two-photon photoemission spectroscopy (2PPE), intra-atomic 3d → 3d transition from the split Ti3+ 3d orbitals, i.e., band gap states and excited states at ∼1.00 eV below and ∼2.40 eV above the Fermi level, respectively, has been proven for all of the samples, suggesting that it is a common property of this material. The distinct structure of rutile TiO2 results in the anisotropic 3d → 3d transitions with the transition dipole moment along the long axes ([110] and [11̅0]) of TiO6 blocking units. This anisotropy facilitates the selective excitation of Ti3+ ions in the two types of TiO6, which cannot be realized by conventional wavelength tuning, via polarization alignment of the excitation source. Discovery in this work builds the foundation for future investigation of site-selective photophysical and photochemical processes and eventually possible active manipulation in this material at the atomic level.

Postoperative gastrointestinal disorder (POGD) was a common complication after surgery under anesthesia. Strategies in combination with Traditional Chinese Medicine and Western medicine showed some distinct effects but standardized clinical practice guidelines were not available. Thus, a multidisciplinary expert team from various professional bodies including the Perioperative and Anesthesia Professional Committees of the Chinese Association of Integrative Medicine (CAIM), jointly with Gansu Province Clinical Research Center of Integrative Anesthesiology/Anesthesia and Pain Medical Center of Gansu Provincial Hospital of Traditional Chinese Medicine and WHO Collaborating Center for Guideline Implementation and Knowledge Translation/Chinese Grading of Recommendations, Assessment, Development, and Evaluation (GRADE) Center/Gansu Provincial Center for Medical Guideline Industry Technology/Evidence-based Medicine Center of Lanzhou University, was established to develop evidence-based guidelines. Clinical questions (7 background and 12 clinical questions) were identified through literature reviews and expert consensus meetings. Based on systematic reviews/meta-analyses, evidence quality was analyzed and the advantages and disadvantages of interventional measures were weighed with input from patients' preferences. Finally, 20 recommendations were developed through the Delphi-based consensus meetings. These recommendations included disease definitions, etiologies, pathogenesis, syndrome differentiation, diagnosis, and perioperative prevention and treatment.

RATIONALE: In recent years, recreational use of inhaled nitrous oxide (N2O) is on the increase among young people, accompanied by a corresponding rise in reports about its toxicity. Subacute combined degeneration of the spine cord (SCD) is the typical clinical picture of the nervous system disorder caused by N2O intoxication, as a result of metabolic disturbance of vitamin B12. PATIENT CONCERNS, DIAGNOSES, INTERVENTIONS AND OUTCOMES: We report a 28-year-old female of SCD due to prolonged use of N2O, presented with paresthesia and unsteady in walking progressing within 1 month. Symptoms gradually improved with the treatment of intramuscular injections of hydroxocobalamin combined with N2O abstinence, and the patient recovered completely with normal neurological examination after 4 months of follow-up. LESSONS: Clinicians should be aware of the clinical features and pathogenesis of SCD caused by N2O intoxication in order to lead effective treatment as soon as possible. Recreational N2O use should always be considered as an etiology when dealing with patients presented with myelopathy and/or neuropathy suspected of vitamin B12 deficiency.

BACKGROUND: Angiogenesis is described as a complex process in which new microvessels sprout from endothelial cells of existing vasculature. This study aimed to determine whether long non-coding RNA (lncRNA) H19 induced the angiogenesis of gastric cancer (GC) and its possible mechanism. METHODS: Gene expression level was determined by quantitative real-time polymerase chain reaction and western blotting. Cell counting kit-8, transwell, 5-Ethynyl-2'-deoxyuridine (EdU), colony formation assay, and human umbilical vein endothelial cells (HUVECs) angiogenesis assay as well as Matrigel plug assay were conducted to study the proliferation, migration, and angiogenesis of GC in vitro and in vivo . The binding protein of H19 was found by RNA pull-down and RNA Immunoprecipitation (RIP). High-throughput sequencing was performed and next Gene Ontology (GO) as well as Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis was conducted to analyze the genes that are under H19 regulation. Methylated RIP (me-RIP) assay was used to investigate the sites and abundance among target mRNA. The transcription factor acted as upstream of H19 was determined through chromatin immunoprecipitation (ChIP) and luciferase assay. RESULTS: In this study, we found that hypoxia-induced factor (HIF)-1α could bind to the promoter region of H19, leading to H19 overexpression. High expression of H19 was correlated with angiogenesis in GC, and H19 knocking down could inhibit cell proliferation, migration and angiogenesis. Mechanistically, the oncogenic role of H19 was achieved by binding with the N 6 -methyladenosine (m 6 A) reader YTH domain-containing family protein 1 (YTHDF1), which could recognize the m 6 A site on the 3'-untransated regions (3'-UTR) of scavenger receptor class B member 1 (SCARB1) mRNA, resulting in over-translation of SCARB1 and thus promoting the proliferation, migration, and angiogenesis of GC cells. CONCLUSION: HIF-1α induced overexpression of H19 via binding with the promoter of H19, and H19 promoted GC cells proliferation, migration and angiogenesis through YTHDF1/SCARB1, which might be a beneficial target for antiangiogenic therapy for GC.

RESUMO

The cases of feeling comfort during acupuncture and moxibustion treatment in literature were summarized and its biological basis was explored. A simple classification of comfort was made, and the importance of obtaining comfort in acupuncture treatment was pointed out. Considering the pursuit of less pain and harmlessness in modern clinical treatment, sugar needle should be advocated and popularized in current clinical practice of acupuncture and moxibustion.

Motor imagery-based brain-computer interfaces (BCI) have been widely recognized as beneficial tools for rehabilitation applications. Moreover, visually guided motor imagery was introduced to improve the rehabilitation impact. However, the reported results to support these techniques remain unsatisfactory. Electroencephalography (EEG) signals can be represented by a sequence of a limited number of topographies (microstates). To explore the dynamic brain activation patterns, we conducted EEG microstate and microstate-specific functional connectivity analyses on EEG data under motor imagery (MI), motor execution (ME), and guided MI (GMI) conditions. By comparing sixteen microstate parameters, the brain activation patterns induced by GMI show more similarities to ME than MI from a microstate perspective. The mean duration and duration of microstate four are proposed as biomarkers to evaluate motor condition. A support vector machine (SVM) classifier trained with microstate parameters achieved average accuracies of 80.27% and 66.30% for ME versus MI and GMI classification, respectively. Further, functional connectivity patterns showed a strong relationship with microstates. Key node analysis shows clear switching of key node distribution between brain areas among different microstates. The neural mechanism of the switching pattern is discussed. While microstate analysis indicates similar brain dynamics between GMI and ME, graph theory-based microstate-specific functional connectivity analysis implies that visual guidance may reduce the functional integration of the brain network during MI. Thus, we proposed that combined MI and GMI for BCI can improve neurorehabilitation effects. The present findings provide insights for understanding the neural mechanism of microstates, the role of visual guidance in MI tasks, and the experimental basis for developing new BCI-aided rehabilitation systems.

Lead-free perovskite is one of the ideal solutions for the toxicity and instability of lead halide perovskite quantum dots. As the most ideal lead-free perovskite at present, bismuth-based perovskite quantum dots still have the problem of a low photoluminescence quantum yield, and its biocompatibility also needs to be explored. In this paper, Ce3+ ions were successfully introduced into the Cs3Bi2Cl9 lattice using a modified antisolvent method. The photoluminescence quantum yield of Cs3Bi2Cl9:Ce is up to 22.12%, which is 71% higher than that of undoped Cs3Bi2Cl9. The two quantum dots show high water-soluble stability and good biocompatibility. Under the excitation of a 750 nm femtosecond laser, high-intensity up-conversion fluorescence images of human liver hepatocellular carcinoma cells cultured with the quantum dots were obtained, and the fluorescence of the two quantum dots was observed in the image of the nucleus. The fluorescence intensity of cells cultured with Cs3Bi2Cl9:Ce was 3.20 times of that of the control group and 4.54 times of the control group for the fluorescence intensity of the nucleus, respectively. This paper provides a new strategy to develop the biocompatibility and water stability of perovskite and expands the application of perovskite in the field.

In this study, the photothermal effect and up-conversion florescence imaging effect of gold nanobipyramids in liver cancer cells are investigated theoretically and experimentally to explore the photothermal ablation tumor therapy with higher photothermal conversion efficiency, shorter laser action time, smaller action range and lower laser power. The small-size gold nanobipyramids with good biocompatibility and infrared absorption peak located in the first biological window are synthesized. Femtosecond laser is focused on the nanobipyramids clusters in cells and the cells die after being irradiated for 20 s at a power as low as 3 mW. In contrast, the control cells die after irradiation with 30 mW laser for 3 min. The theoretical simulation results show that: under femtosecond laser irradiation, the local thermal effect of gold nanoclusters is produced in the range of hundreds of square nanometers and the temperature rises by 516 °C in 106 picoseconds. This therapy reduces the treatment time to seconds level, and the treatment range to square micrometer level, the power to milliwatt level. In this treatment, cells die by apoptosis rather than necrosis, which reduces inflammation. This result opens up a new way to develop photothermal ablation therapy with less side effects and more minimally invasive.

RATIONALE: Hypertrophic subolivine degeneration (HOD) was destroyed by Guillain-Mollaret triangle (GMT) due to various injuries, resulting in anterograde cavity-like degeneration of the lower olive nucleus. In addition, the brain stem is related to the muscle coordination of the upper respiratory tract. Obstructive sleep apnea hypopnea syndrome may affect the respiratory center due to the injury of the brain stem. Brain stem hemorrhage damage GMT, resulting in transsynaptic neuronal degeneration. Clinical manifestations can be complex, and enhanced magnetic resonance imaging can be helpful in distinguishing them. PATIENT CONCERNS: HOD is a self-limiting disease with no effective treatment and a long course of disease. Most patients can improve their symptoms after symptomatic treatment, and some patients can relieve their symptoms by themselves after 3 to 4 years. DIAGNOSIS INTERVENTIONS: The limbs wobble involuntarily. His clinical symptoms and signs are consistent with HOD. Imaging with a clear primary lesion confirmed HOD. After treatment with antiepileptic drugs, the patient's symptoms were relieved. Moreover, the patient had snoring and apnea, and respiratory sleep monitoring showed moderate obstructive sleep apnea hypopnea syndrome, which was treated with noninvasive ventilator. OUTCOMES: After treatment with antiepileptic drugs and noninvasive ventilator, the patient's symptoms were significantly relieved. LESSONS: HOD is a rare clinical disorder. Therefore, for similar patients, more attention should be paid to early diagnosis and treatment to avoid missed diagnosis, misdiagnosis and unnecessary intervention measures. The diagnosis can be confirmed by primary disease, clinical symptoms, and imaging based on GMT.

Pre-eclampsia (PE) is a serious complication in pregnant women characterized by failure of placental remodeling and is one of the primary causes of changes in the placental structure and function. The aberrant expression of long noncoding RNA is associated with the occurrence and progression of PE. This study found that linc01116 expression was significantly downregulated in PE patients and was related to poor uterine spiral artery remodeling. Knockdown of linc01116 remarkably decreased the angiogenesis of trophoblast cells in vitro and in vivo. Mechanistically, IGF2BP2 regulated linc01116 RNA stability via m6 A methylation. Bioinformatics and other experiments further revealed that linc01116 upregulates AAMP expression by adsorbing miR-210-3p in trophoblast cells. In conclusion, this study revealed the critical role of linc01116 in regulating trophoblast angiogenesis. Furthermore, the study provides new clues for detecting placental pathology in PE.

RATIONALE: Due to neuronal destruction caused by Epstein-Barr virus (EBV) infection, exposure to neuronal surface antigens may lead to an imbalance in immune tolerance, which in turn triggers an autoimmune response. In addition, due to the involvement of nonspecific B-cell activation or molecular mimicry, EBV and Glial Fibrillary Acidic Protein (GFAP) receptors may have common epitopes. Viral infection triggers activation of B-cell and cross-reaction with viral antibodies, resulting in autoimmune encephalomyelitis. The clinical presentation may be complex or even atypical. A small number of patients may develop autoimmune reactions. PATIENT CONCERNS: Most patients with EBV encephalomyelitis have a good prognosis, with the disease generally having a short course, few complications, and a good prognosis. In most patients, after treatment, their neurological function basically recovers within a few weeks or months. DIAGNOSIS INTERVENTIONS: The patient had fever and headache. His 3 tests for cerebral spinal fluid (CSF) are consistent with the features of viral encephalomyelitis. Pathogenic examination of CSF confirmed EBV, and imaging suggested brain and spinal cord involvement. After antiviral treatment, the patient's symptoms relieved. The diagnosis of EBV encephalomyelitis was considered. However, the patient's temperature continued to increase. He was transferred to a superior hospital and was given GFAP-Ab in CSF, which was strongly positive. The patient was given immunoglobulin and antiviral therapy. This supports the diagnosis of GFAP-IgG antibody positive with EBV encephalomyelitis. OUTCOMES: After treatment with antiviral drugs and immunoglobulins, the patient's symptoms improved and he was able to function. LESSONS: EBV encephalomyelitis is a rare clinical disease. Therefore, more attention should be paid to the early diagnosis and treatment of similar patients to avoid misdiagnosis. CSF tests, genetic tests, and imaging tests can confirm the diagnosis.

Studies have revealed the neuropathological mechanism of the cognitive impairment associated with neurodegenerative diseases.However,the therapies for these cognitive disorders are limited,and the prevalence of cognitive impairment is expected to increase significantly in the future,which proves the necessity of new therapeutic agents.In recent years,the pharmacological activity of ß2-adrenergic receptor(ß2-AR)has been extensively studied,which has demonstrated that ß2-AR agonist has therapeutic effects on the cognitive impairment associated with several common neurodegenerative diseases,including Alzheimer's disease,vascular dementia,Parkinson's disease with dementia,and Lewy body dementia.We reviewed the neuropathological features of cognitive impairment in several common neurodegenerative diseases and expounded the pharmacological effects of ß2-AR on related diseases.

RATIONALE: Anti-LGI1 antibody encephalitis and anti-mGluR5 are both uncommon encephalitis, and we report the first case of autoimmune encephalitis (AE) with dual seropositive antibodies of leucine-rich glioma-inactivated 1 (LGI1) and mGluR5. PATIENT CONCERNS: We present a case of AE with dual seropositive antibodies of LGI1 and mGluR5 in a 65-year-old woman who presented with sudden onset left faciobrachial dystonic seizures and unresponsive for 5 hours. DIAGNOSIS: The patient was diagnosed with anti-LGI1 AE and anti-mGluR5 AE mainly based on the clinical symptoms and further test of the antibody in serum and cerebral spinal fluid (CSF). INTERVENTIONS AND OUTCOMES: The patient was treated with glucocorticoid intravenous drip. We also gave her the therapy of immunoglobulin (25 g q.d) for 5 days and anti-epileptic therapy. She had no more convulsions on the left side of the face and limbs. She did not complain of any uncomfort until July 18. LESSONS: Early recognition of AE is crucial. Specific autoantibodies are associated with corresponding syndromes. Our patient was initially diagnosed with acute ischemic stroke. Therefore, we should conduct further study on the related symptoms of AE.

PURPOSE: The angiogenesis is among the primary factors that affect tumor recurrence and distant organ metastasis in colorectal cancer (CRC). N6-methyladenosine (m6A) modification is one of the most common chemical modifications in eukaryotic mRNA, especially at the post-transcriptional level. Methyltransferase-like 3 (METTL3) promoting angiogenesis in a variety of tumors has been reported. However, the mechanism of how METTL3 dual-regulates the stability of long non-coding RNAs (lncRNAs) and vascular-related factor RNAs to affect angiogenesis in CRC is unclear. METHODS: 64 paired CRC and adjacent normal tissues were collected. In vitro, quantitative real-time polymerase chain reaction (qRT-PCR), immunohistochemistry (IHC), actinomycin assay, methylated RNA immunoprecipitation (MeRIP) experiment,3-(4,5)-dimethylthiahiazo(-z-y1)-3,5-di-phenytetrazoliumromide (MTT) and colony formation assay were performed. The functions were also studied in zebrafish model animals in vivo. RESULTS: We found that the vascular endothelial growth factor A(VEGFA), METTL3 and LINC00662 RNAs were highly expressed in CRC, and that METTL3 was significantly positively correlated with LINC00662 and VEGFA. The protein expression levels of CD31, CD34, VEGFA, m6A and METTL3 were all significantly increased in the CRC tissues. The angiogenesis experiments both in vivo and in vitro found that METTL3 and LINC00662 promoted angiogenesis in CRC. The actinomycin assay indicated that METTL3 maintained the stability of LINC00662 and VEGFA RNAs. In addition, the MeRIP experiment confirmed that the LINC00662 and VEGFA RNAs had METTL3-enriched sites. CONCLUSION: These findings suggest that METTL3 and LINC00662 may both serve as diagnostic and prognostic predictive biomarkers for CRC and potential targets for anti-vascular therapy.

Preeclampsia (PE) is the predominant medical condition leading to maternal and fetal mortality, and the lack of effective treatment increases its risk to the public health. Among the numerous predisposing factors, the ineffectual remodeling of the uterine spiral arteries, which can induce abnormal placental angiogenesis, has been focused to solve the pathogenesis of PE. According to the preceding research results, abnormal expression of long non-coding RNAs (lncRNA)s could be associated with the pathological changes inducing PE. To be more specific, lncRNA HIF1A-AS2 was proposed for its potential to participate in the molecular mechanisms underlying PE. In vitro, in trophoblast cell lines HTR-8/SVneo and human umbilical vein endothelial cells HUVECs, HIF1A-AS2 knockdown inhibited cell proliferation, migration and tube formation. Mechanistically, transcription factor FOXP1 could regulate the expression of HIF1A-AS2. Moreover, a series of assays, including RNA pull down and mass spectrometry, RNA immunoprecipitation and chromatin immunoprecipitation assay, revealed that HIF1A-AS2 interacted with Lamin A/C (LMNA) to inhibit ANGPTL4 expression in trophoblast cells, thus further participating in the progression of PE. Taken together, these findings suggested that further analysis on HIF1A-AS2 could contribute to the development of prospective therapeutic strategy for PE.

Band structure dictates optical and electronic properties of solids and eventually the efficiency of the semiconductor based solar conversion. Compared to numerous theoretical calculations, the experimentally measured band structure of rutile TiO2, a prototypical photocatalytic material, is rare. In this work, the valence band structure of rutile TiO2(110) is measured by angle-resolved photoelectron spectroscopy using polarized extreme ultraviolet light. The effective mass of the hole, which has never been measured before, is determined to be 4.66-6.87 m0 (free electron mass) and anisotropic. The dependence of photoemission intensities on excitation light polarization is analyzed by taking into account of the parity symmetry of molecular orbitals in the blocking unit of rutile TiO2. This work reports a direct measurement of valence band structure and hole effective mass of rutile TiO2(110), which will deepen our understanding of the electronic structure and charge carrier properties of the model material and provide reference data for future theoretical calculations.

Brain vascular inflammation plays a crucial role in the pathogenesis of Alzheimer's disease (AD). As a central pathogenic factor in AD, the extracellular buildup of amyloid-ß (Aß) induces brain microvascular endothelial cells activation, impairs endothelial structure and function. Formononetin (FMN) has been reported to protect against Alzheimer's disease (AD) and attenuates vascular inflammation in atherosclerosis. However, its involvement in regulating vascular inflammation of AD has not been investigated. In the study, we found that FMN significantly attenuates Aß25-35-induced expression of adhesion molecules, including intracellular adhesion molecule-1 (ICAM-1) and vascular cell adhesion molecule-1 (VCAM-1) in the human brain microvascular endothelial cells (HBMECs), suggesting that FMN inhibits Aß25-35-induced brain endothelial cells inflammatory response. Moreover, we observed that FMN attenuates Aß25-35-induced translocation of NFκB (p65) into the nucleus of HBMECs, and found that FMN treatment induces Nrf2 expression and attenuates Nrf2-Keap1 association in a dose-dependent manner in HBMECs. Furthermore, we demonstrated that Nrf2 silencing significantly attenuates FMN-reduced NFκB (p65) activation and nuclear translocation. Lastly, our results showed that FMN treatment attenuates Aß25-35-induced adhesion of THP-1 cell to endothelial cell monolayer. Collectively, these findings suggest that FMN attenuates Aß25-35-induced activation in human brain microvascular endothelial cells, which at least in part was mediated through Nrf2 pathways.
