RESUMO

Apocynum venetum L. belongs to the Apocynaceae family and is a plant that is highly resistant to stress. It is important in the fields of ecology, feeding, industry and medicine. The molecular mechanism underlying salt tolerance has not been elucidated. In this study, RNA-seq based transcriptome sequencing of A. venetum leaves after 0, 2, 6, 12, 24 and 48 h of treatment with 300 mM NaCl was performed. We conducted a comprehensive analysis of the transcriptome expression profiles of A. venetum under salt stress using the WGCNA method and identified red, black, and brown as the core modules regulating the salt tolerance of A. venetum. A co-expression regulatory network was constructed to identify the core genes in the module according to the correlations between genes. The genes TRINITY_DN102_c0_g1 (serine carboxypeptidase), TRINITY_DN3073_c0_g1 (SOS signaling pathway) and TRINITY_DN6732_c0_g1 (heat shock transcription factor) in the red module were determined to be the core genes. Two core genes in the black module, TRINITY_DN9926_c0_g1 and TRINITY_DN7962_c0_g1, are pioneer candidate salt tolerance-associated genes in A. venetum. The genes in the brown module were mainly enriched in two pathways, namely photosynthesis and osmotic balance. Among them, the TRINITY_DN6321_c0_g2 and TRINITY_DN244_c0_g1 genes encode aquaporin, which is helpful for maintaining the cell water balance and plays a protective role in defending A. venetum under abiotic stress. Our findings contribute to the identification of core genes involved in the response of A. venetum to salt stress.

The grain yield is closely associated with spikelet fertility in rice (Oryza sativa L.) under high temperatures, and nitrogen (N) plays a crucial role in yield formation. To investigate the effect of panicle N application on yield formation under high temperatures at the panicle initiation stage, two rice varieties [Liangyoupeijiu (LYPJ, heat susceptible) and Shanyou63 (SY63, heat tolerant)] were grown and exposed to high daytime temperature (HT) and control temperature (Control) during the panicle initiation stage. Low (LPN) and high (HPN) panicle N applications were conducted. HT markedly decreased the yields by 87% at LPN and 48% at HPN in LYPJ and 31% at LPN and 36% at HPN in SY63. The decrease in grain yield under HT was primarily attributed to the decline in spikelet fertility, HPN increased spikelet fertility. HT resulted in the abnormal development of anthers, which included disordered, enlarged, and broken anther wall layers, degraded and irregularly shaped microspores, delayed tapetum degradation, less vacuolated microspores per locule, abnormal and aborted pollen grains; however, HPN improved the development of anthers under HT, particularly in LYPJ. A high rate of evapotranspiration resulted in an approximately 1°C decrease in panicle temperatures at HPN compared with that at LPN in both varieties under HT. Overall, these results demonstrate that the increased panicle N application favors normal anther development in LYPJ by decreasing the panicle temperature, which results in high pollen viability and spikelet fertility, and consequently less yield loss under HT.

Objective: To investigate the clinical application of high-frequency color Doppler ultrasound (HFCDU) in detecting perforators in the deep adipose layers for harvesting super-thin anterolateral thigh flap (ALTF). Methods: Between August 2019 and January 2023, 45 patients (46 sides) with skin and soft tissue defects in the foot and ankle were treated, including 29 males and 16 females, aged from 22 to 62 years, with an average of 46.7 years. The body mass index ranged from 19.6 to 36.2 kg/m 2, with an average of 23.62 kg/m 2. The causes of injury included traffic accident injury in 15 cases, heavy object crush injury in 20 cases, mechanical injury in 8 cases, heat crush injury in 1 case, and chronic infection in 1 case. There were 20 cases on the left side, 24 cases on the right side, and 1 case on both sides. After thorough debridement, the wound size ranged from 5 cm×4 cm to 17 cm×11 cm. All patients underwent free super-thin ALTF transplantation repair. HFCDU was used to detect the location of the perforators piercing the deep and superficial fascia, as well as the direction and branches of the perforators within the deep adipose layers before operation. According to the preoperative HFCDU findings, the dimensions of the super-thin ALTF ranged from 6 cm×4 cm to 18 cm×12 cm. The donor sites of the flaps were directly sutured. Results: A total of 55 perforators were detected by HFCDU before operation, but 1 was not found during operation. During operation, a total of 56 perforators were found, and 2 perforators were not detected by HFCDU. The positive predictive value of HFCDU for identifying perforator vessels was 98.2%, and the sensitivity was 96.4%. Among the 54 perforators accurately located by HFCDU, the orientation of the perforators in the deep adipose layers was confirmed during operation. There were 21 perforators (38.9%) traveled laterally and inferiorly, 12 (22.2%) traveled medially and inferiorly, 14 (25.9%) traveled laterally and superiorly, 5 (9.3%) traveled medially and superiorly, and 2 (3.7%) ran almost vertically to the body surface. Among the 54 perforators accurately located by HFCDU, 35 were identified as type 1 perforators and 12 as type 2 perforators (HFCDU misidentified 7 type 2 perforators as type 1 perforators). The sensitivity of HFCDU in identifying type 1 perforators was 100%, with a positive predictive value of 83.3%. For type 2 perforators, the sensitivity was 63.2%, and the positive predictive value was 100%. The surgeries were successfully completed. The super-thin ALTF had a thickness ranging from 2 to 6 mm, with an average of 3.56 mm. All super-thin ALTF survived, however, 1 flap experienced a venous crisis at 1 day after operation, but it survived after emergency exploration and re-anastomosis of the veins; 1 flap developed venous crisis at 3 days after operation but survived after bleeding with several small incisions; 3 flaps had necrosis at the distal edge of the epidermis, which healed after undergoing dressing changes. All 45 patients were followed up 6-18 months (mean, 13.6 months). Three flaps required secondary defatting procedures, while the rest had the appropriate thickness, and the overall appearance was satisfactory. Conclusion: Preoperative application of HFCDU to detect the perforator in the deep adipose layers can improve the success and safety of the procedure by facilitating the harvest of super-thin ALTF.

This study primarily focused on the efficient transformation of low-priced blue coke powder into a high-capacity adsorbent and aimed to address the pollution issue of hexavalent chromium (Cr (VI))-laden wastewater and to facilitate the effective utilization of blue coke powder. A two-step method was utilized to fabricate a blue coke-based nitric acid-modified material (LCN), and the impact of nitric acid modification on the material's structure and its efficacy in treating Cr (VI)-contaminated wastewater was evaluated. Our experimental results illustrated that, under identical conditions, LCN exhibited superior performance for Cr (VI) treatment compared to the method employing only potassium hydroxide (LCK). The specific surface area and pore volume of LCN were 1.39 and 1.36 times greater than those of LCK, respectively. Further chemical composition analysis revealed that the functional group structure on the LCN surface was more conducive to Cr (VI) adsorption. The highest amount of Cr (VI) that LCN could bind was measured at 181.962 mg/g at 318 K. This was mostly due to chemisorption, which is dominated by redox reactions. The Cr (VI) removal process by LCN was identified to be a spontaneous, exothermic, and entropy-increasing process. Several tests on recycling and reuse showed that LCN is a stable and effective chromium-containing wastewater adsorbent, showing that it could be used in many situations.

Extracellular vesicle (EV) secretion is a dynamic process crucial to cellular communication. Temporally sorting EVs, i.e., separating the newly-produced ones from the pre-existing, can allow not only deep understanding of EV dynamics, but also the discovery of potential EV biomarkers that are related to disease progression or responsible to drug intervention. However, the high similarity between the nascent and pre-existing EVs makes temporal separation extremely challenging. Here, by co-translational introduction of azido groups to act as a timestamp for click chemistry labelling, we develop a microfluidic-based strategy to enable selective isolation of nascent EVs stimulated by an external cue. In two mouse models of anti-PD-L1 immunotherapy, we demonstrate the strategy's feasibility and reveal the high positive correlation of nascent PD-L1+ EV level to tumor volume, suggesting an important role of nascent EVs in response to immunotherapy in cancer treatment.

Hepatitis E infection poses a serious health problem in developing countries. Hepatitis E vaccination is important for prevention, but it is influenced by residents' knowledge. Qingdao residents' knowledge of hepatitis E remains unknown. This study used an online survey on the WeChat platform. The chi-square test was used to compare the hepatitis E influencing factors between the subgroups. Binary logistic regression was used for multiple factor analysis to explore the hepatitis E influencing factors. The total awareness rate of hepatitis E was 60.51%. Females aged between 51 and 60, aged 61 and above, and working in government-affiliated departments were found to have higher awareness rates than other subgroups. Participants with family members infected with hepatitis E had a lower awareness rate. The government and relevant departments should focus on education regarding the hepatitis E vaccination and disease process.

RESUMO

SARS-CoV-2 mutates rapidly as evidenced by the emergence of Omicron which causes changes in the recognition epitopes of most current neutralizing antibodies and immune evasion. Although aptamers are potential neutralizing agents for SARS-CoV-2 due to their unique molecular properties, it is difficult to compare their performances as assay conditions vary greatly, and their activity levels against variants remain unknown. Here, we evaluated the performances of 14 SARS-CoV-2 aptamers and provided a comprehensive analysis them, which we expect will improve the development of aptamer tools for SARS-CoV-2 diagnostics and therapeutics.

Extracellular vesicle PD-L1 (programmed death-1 ligand 1) is of greater value in tumor diagnosis, prognosis, and efficacy monitoring of anti-PD-1/PD-L1 immunotherapy. However, soluble PD-L1 interferes with the accurate detection of extracellular vesicle (EV) PD-L1. Here, we developed a microfluidic differentiation method for the detection of extracellular PD-L1, without the interference of soluble, by DNA computation with lipid probes and PD-L1 aptamer as inputs (DECLA). For the developed DECLA method, a cholesterol-DNA probe was designed that efficiently embeds into the EV membrane, and an aptamer-based PD-L1 probe was used for PD-L1 recognition. Due to the stable secondary structure of the designed connector, only cobinding of cholesterol-DNA and PD-L1 affinity probe induced biotin-labeled connector activation, while soluble PD-L1 cannot hybridize. As a result, PD-L1 EVs can be efficiently captured by streptavidin-functioned herringbone chip and quantified by anti-CD63-induced fluorescence signal. The high specificity of dual-input DNA computation allied to the high sensitivity of microfluidic-based detection was suitable for distinguishing lung cancer patients from healthy donors, highlighting its potential translation to clinical diagnosis and therapy monitoring.

In the production process of puree cans such as chestnuts cans, it is easy to browning due to excessive heating, which causes a lot of waste every year. The heat and mass transfer model of Chinese Chestnut Puree was established through the finite element method. The model simulated the change process of the temperature field, heat flow velocity field and F value during the production of Chinese Chestnut Puree. After comparing and confirming the effectiveness of the model through the thermal penetration test, the model was used to adjust and optimize the production process. For #9121 cans, the two-stage sterilization method was adopted. Through the sterilization method at 10-65-48-14/118-110°C, a sterilization effect equivalent to that of the original process at 10-86-24/121°C was achieved, the browning problem of the product was alleviated, and the product quality was improved. This practice can also provide a reference for canning enterprises to adjust their production processes in the future.

To solve the issue of hexavalent chromium (Cr(VI)) contamination in water bodies, blue coke powder (LC) was chemically changed using potassium hydroxide to create the modified material (GLC), which was then used to treat a Cr(VI)-containing wastewater solution. The differences between the modified and unmodified blue coke's adsorption characteristics for Cr(VI) were studied, and the impact of pH, starting solution concentration, and adsorption period on the GLC's adsorption performance was investigated. The adsorption behavior of the GLC was analyzed using isothermal adsorption models, kinetic models, and adsorption thermodynamic analysis. The mechanism of Cr(VI) adsorption by the GLC was investigated using characterization techniques such as Fourier Transform Infrared Spectroscopy (FTIR), Field Emission Scanning Electron Microscope (FE-SEM), X-Ray Diffraction (XRD), and X-Ray Photoelectron Spectroscopy (XPS). With the biggest difference in removal rate at pH = 2, which was 2.42 times that of LC, batch adsorption experiments revealed that, under the same adsorption conditions, the GLC always performed better than LC. With a specific surface area that was three times that of LC and an average pore diameter that was 0.67 times that of LC, GLC had a more porous structure than LC. The alteration significantly increased the number of hydroxyls on the surface of GLC by altering the structural makeup of LC. The ideal pH for removing Cr(VI) was 2, and the ideal GLC adsorbent dosage was 2.0 g/L. Pseudo-second-order kinetic (PSO) model and Redlich-Peterson (RP) model can effectively describe the adsorption behavior of GLC for Cr(VI). Physical and chemical adsorption work together to remove Cr(VI) by GLC in a spontaneous, exothermic, and entropy-increasing process, with oxidation-reduction processes playing a key role. GLC is a potent adsorbent that can be used to remove Cr(VI) from aqueous solutions.

Conjugate addition is among the most important synthetic protocols for constructing carbon skeletons and is widely used to synthesize natural products and drugs. However, asymmetric catalysis studies have mainly focused on constructing stereogenic centers arising from conjugate alkenes. Here, we report the first photoinduced cobalt-catalyzed dynamic kinetic reductive conjugate addition reaction that enables the formation of heterobiaryls with axial chirality (45 examples, up to 91% yield and 97% ee). This method features mild reaction conditions, good functional-group tolerance, and excellent enantiomeric control. Significantly, large amounts of metal waste and precious metal catalysts can be avoided under these conditions. Migration of the chiral arylcobalt species into the alkene might be the rate-determining step based on kinetic studies.

Aging is an important risk factor for neurodegenerative diseases. The activation of α7 nicotinic acetylcholine receptor (α7nAChR) is involved in inflammation and cognition, but the specific role it plays in aging remains unknown. This study aimed to investigate the anti-aging effect of the activation of α7nAChR on aging rats and BV2 cells induced by D-galactose, as well as its potential mechanism. D-galactose induced an increase in the SA-ß-Gal positive cells, expression of p16 and p21 in vivo and in vitro. α7nAChR selective agonist PNU282987 decreased levels of pro-inflammatory factors, MDA, and Aß, enhanced SOD activity and levels of anti-inflammatory factor (IL10) in vivo. PNU282987 enhanced the expression of Arg1, decreased the expression of iNOS, IL1ß and TNFα in vitro. PNU282987 upregulated the levels of α7nAChR, Nrf2 and HO-1 in vivo and in vitro. The results of Morris water maze and novel object recognition tests showed that PNU282987 improved cognitive impairment in aging rats. Furthermore, α7nAChR selective inhibitor methyllycaconitine (MLA) results were opposite with PNU282987. PNU282987 improves cognitive impairment through inhibiting oxidative stress and neuroinflammation in D-galactose induced aging via regulating the α7nAChR/Nrf2/HO-1 signaling pathway. Therefore, targeting the α7nAChR may be a viable therapeutic approach for anti-inflammaging and neurodegenerative diseases.

To provide guidance for the selection of woody-activated carbon in the treatment of wastewater containing hexavalent chromium (Cr(VI)), the adsorption tests on two varieties of commercial woody-activated carbon powder from different manufacturers were carried out. The physicochemical properties and structural characteristics of activated carbon were studied by using elemental, chemical, and instrumental analyses. The adsorption mechanism of Cr(VI) was discussed by investigating the factors affecting the removal of hexavalent chromium. The two kinds of woody-activated carbon have microporous and mesoporous structures. Commercial woody-activated carbon No.1 (ACI) has a more extensive specific surface area and a better-developed pore structure. While ACI exhibits a higher adsorption capability when the content of Cr(VI) is high, commercial woody-activated carbon No.2 (AC) can remove hexavalent chromium fast when the concentration is low. A rise in pH value is not helpful for the materials to remove Cr(VI) from solutions. For Cr(VI) removal, the optimum pH value is 2. The adsorption of Cr(VI) by AC and ACI followed the pseudo-second-order kinetic model and Langmuir isothermal adsorption equation. The maximum adsorption value of Cr(VI) is 154.56 mg/g for AC and 241.55 mg/g for ACI. There is chemical adsorption during the Cr(VI) removal. A lot of Cr (Ⅲ) was formed by Cr(VI). The abundance of pores and the reducing ability of the materials are essential for the removal of Cr(VI).

Oxidative stress and neuroinflammation play crucial roles in aging. S-adenosylmethionine (SAM), a popular supplement, is a potential antioxidant and candidate therapy for depression. This study aimed to evaluate the neuroprotective effects of SAM on D-galactose-induced brain aging and explore its underlying mechanisms. Brain aging model was established with D-galactose (180 mg/kg/day) for 8 weeks. During the last 4 weeks, SAM (16 mg/kg) was co-administrated with D-galactose. Behavior tests were used to assess cognitive function and depression-like behaviors of rats. Results showed that cognitive impairment and depression-like behaviors were reversed by SAM. SAM reduced neuronal cell loss, increased brain-derived neurotrophic factor level in the hippocampus, inhibited amyloid-ß level and microglia activation, as well as pro-inflammatory factors levels in the hippocampus and serum. Further, SAM enhanced antioxidant capacity and attenuated cholinergic damage by reducing malondialdehyde levels, increasing acetylcholine levels, expression levels of α7 nicotinic acetylcholine receptor (α7nAChR), nuclear factor erythrocyte 2-related factor 2 (Nrf2) and heme oxygenase 1 (HO-1) in the hippocampus. Above all, SAM has a potential neuroprotective effect on ameliorating cognitive impairment in brain aging, which is related to inhibition of oxidative stress and neuroinflammation, as well as α7nAChR signals. For Reeb graph extraction on surfaces, existing methods always use the isolines of a function defined on the surface to detect the surface components and the neighboring relationships between them. Since such detection is unstable, it is still a challenge for the extracted Reeb graphs to stably and concisely encode the topological information of the surface. In this article, we address this challenge by using foliation leaves to extract Reeb graphs. In particular, we employ a method for generating measured harmonic foliations by defining loops for foliation initialization and diffusing leaves from loops over the surface. We demonstrate that when the loops are determined, the neighboring relationships between the leaves from different loops are fixed. Thus, we can use loops to represent surface components for robustly detecting the interrelationships between surface components. As a result, we are able to extract stable and concise Reeb graphs. We developed novel measures for loop determination and improved foliation generation, and our method allows the user to manually prescribe loops for generating Reeb graphs with desired structures. Therefore, the potential of Reeb graphs for representing surfaces is enhanced, including conveniently representing the symmetries of the surface and ignoring topological noise. This is verified by our experimental results which indicate that our Reeb graphs are compact and expressive, promoting shape analysis.

Little concern has been paid to the relationship between temperature and varicella among adults. Daily meteorological data and varicella cases in Qingdao among adults from 1 January 2008 to 31 December 2019 were collected. A combination of quasi-Poisson generalized additive model (GAM) and distributed lag non-linear model (DLNM) was conducted to assess the temperature-lag-varicella relationship. We also estimated the lag-response curves for different temperatures and the exposure-response relationships for different lag days. The number of varicella cases was 10,296. Compared with the minimum-varicella temperature (25°C), we found the largest effect of temperature on varicella within 21 lag days was at 1°C (RR, 6.72; 95% CI, 2.90-15.57), and then the effect declined as the temperature increased. A similar trend of rising first and then falling was found in temperature-response curves for different lag days. A reverse U-shape lag pattern was found for different levels of temperatures. Temperature may affect varicella.

Extracting concise 3D curve skeletons with existing methods is still a serious challenge as these methods require tedious parameter adjustment to suppress the influence of shape boundary perturbations to avoid spurious branches. In this paper, we address this challenge by enhancing the capture of prominent features and using them for skeleton extraction, motivated by the observation that the shape is mainly represented by prominent features. Our method takes the medial mesh of the shape as input, which can maintain the shape topology well. We develop a series of novel measures for simplifying and contracting the medial mesh to capture prominent features and represent them concisely, by which means the influences of shape boundary perturbations on skeleton extraction are suppressed and the quantity of data needed for skeleton extraction is significantly reduced. As a result, we can robustly and concisely extract the curve skeleton based on prominent features, avoiding the trouble of tuning parameters and saving computations, as shown by experimental results.

RESUMO

A high efficiency protocol was developed for the synthesis of 2,5-disubstituted oxazoles via iodine-promoted oxidative domino cyclization. These reactions were performed with readily available methyl azaarenes and α-amino ketones under metal-free conditions. This protocol is a simple method with high functional group compatibility, a wide range of substrates, and excellent yield, providing a new way to synthesize azaarene-attached oxazoles.

Phloem unloading plays an important role in photoassimilate partitioning and grain yield improvements in cereal crops. The phloem unloading strategy and its effects on photoassimilate translocation and yield formation remain unclear in rice. In this study, plasmodesmata were observed at the interface between the sieve elements (SEs) and companion cells (CCs), and between the SE-CC complex and surrounding parenchyma cells (PCs) in phloem of the dorsal vascular bundle in developing caryopses. Carboxyfluorescein (CF) signal was detected in the phloem of caryopses, which showed that CF was unloaded into caryopses. These results indicated that the SE-CC complex was symplasmically connected with adjacent PCs by plasmodesmata. Gene expression for sucrose transporter (SUT) and cell wall invertase (CWI), and OsSUT1 and OsCIN1 proteins were detected in developing caryopses, indicating that rice plants might actively unload sucrose into caryopses by the apoplasmic pathway. Among three rice recombinant inbred lines, R201 exhibited lower plasmodesmal densities at the boundaries between cell types (SE-CC, SE-PC and CC-PC) in developing caryopses than R91 and R156. R201 also had lower expression of SUT and CWI genes and lower protein levels of OsSUT1 and OsCIN1, as well as CWI activity, than R91 and R156. These data agreed with stem non-structural carbohydrate (NSC) translocation and grain yields for the three lines. The nitrogen application rate had no significant effect on plasmodesmal densities at the interfaces between different cells types, and did not affect CF unloading in the phloem of developing caryopses. Low nitrogen treatment enhanced expression levels of OsSUT and OsCIN genes in the three lines. These results suggested that nitrogen application had no substantial effect on symplasmic unloading but affected apoplasmic unloading. Therefore, we concluded that poor symplasmic and apoplasmic unloading in developing caryopses might result in low stem NSC translocation and poor grain yield formation of R201.
