OBJECTIVE: To explore the genetic etiology of a fetus with short limbs identified by prenatal ultrasonography. METHODS: A fetus detected with short limb malformations at Shengjing Hospital Affiliated to China Medical University on October 25, 2021 was selected as the study subject. Prenatal ultrasound and post-abortion imaging were carried out to determine the phenotypic characteristics of the fetus. Amniotic fluid sample of the fetus and peripheral blood samples of its parents were collected. Following extraction of genomic DNA, whole-exome sequencing was carried out. Candidate variants were verified by Sanger sequencing. Online software was used to predict the structural changes of the mutant proteins. RESULTS: Prenatal ultrasound showed that the fetus had a small bell-shaped thorax, markedly shortened limbs, flat midface, a small nose with anteriorly tilted nostrils, and a small mandible. Post-abortion CT showed typical short and wide fetal ribs, cupped metaphyses at both ends, short long bones with wide metaphyses, resulting in a dumbbell-shaped appearance and curved thoracic vertebrae. Whole-exome sequencing revealed that the fetus had harbored compound heterozygous variants of the COL11A1 gene, namely c.2251G>T and c.3790G>T, both of which were predicted to alter the important Gly-X-Y structure of collagen protein. Sanger sequencing confirmed that the variants were respectively inherited from its parents. CONCLUSION: A rare fetus with Fibrochondrogenesis type 1 due to compound heterozygous variants of the COL11A1 gene has been diagnosed. Above finding has enabled genetic counseling and reproductive guidance for this family.

Aqueous Zn-metal battery is considered as a promising energy-storage system. However, uncontrolled zinc dendrite growth is the main cause of short-circuit failure in aqueous Zn-based batteries. One of the most efficient and convenient strategies to alleviate this issue is to introduce appropriate zincophilic nucleation sites to guide zinc metal deposition and regulate crystal growth. Herein, this work proposes Bi2 O3 /Bi nanosheets anchored on the cell wall surface of the 3D porous conductive host as the Zn deposition sites to modulate Zn deposition behavior and hence inhibit the zinc dendrite growth. Density functional theory and experimental results demonstrate that Bi2 O3 has a super zinc binding energy and strong adsorption energy with zinc (002) plane, as a super-zincophilic nucleation site, which results in the deposition of zinc preferentially along the horizontal direction of (002) crystal plane, fundamentally avoids the formation of Zn dendrites. Benefiting from the synergistic effect Bi2 O3 /Bi zincophilic sites and 3D porous structure in the B-BOGC host, the electrochemical performance of the constructed Zn-based battery is significantly improved. As a result, the Zn anode cycles for 1500 cycles at 50 mA cm-2 and 1.0 mAh cm-2 . Meanwhile, the Zn@B-BOGC//MnO2 full cell can operate stably for 2000 cycles at 2.0 A g-1 .

Lightweight porous composite materials (PCMs) with outstanding electromagnetic interference (EMI) shielding performances are ideal for aerospace, artificial intelligence, military, and other fields. Herein, a three-dimensional Ti3C2Tx MXene/sodium alginate (SA)/carbon nanotubes (CNTs) (MSC) PCMs was prepared by a controlled directional freezing process. This method constructs a directionally ordered porous structure, which can make the incident electromagnetic waves reflect and scattered several times in the PCMs. The introduction of CNTs into the MSC PCMs can form three-dimensional conductive networks with MXene, thus improving the conductivity and further improving the electromagnetic shielding performance. Furthermore, the SA with abundant hydrogen bonding can strengthen the interlayer interaction between MXene and CNTs. Profiting from the controlled directional freezing and highly aligned porous structure, the MSC PCMs with 75 wt % CNTs exhibit ultrahigh conductivity of 1630 S m-1, an ultrahigh EMI shielding effectiveness of 48.0 dB in X-band for electromagnetic waves incident perpendicular to the hole growth direction, and compressive strength of 72.3 kPa. The as-prepared MSC PCMs show excellent EMI shielding and mechanical properties and have significant applications in the preparation of an entirely novel type of EMI shielding materials with an absorption-based mechanism.

Ammonium vanadate (NVO) often has unsatisfactory electrochemical performance due to the irreversible removal of NH4 + during the reaction. Herein, layered DMF-NVO nanoflake arrays (NFAs) grown on highly conductive carbon cloth (CC) are employed as the binder-free cathode (DMF-NVO NFAs/CC), which produces an enlarged interlayer spacing of 12.6 Å (against 9.5 Å for NH4 V4 O10 ) by effective N, N-dimethylformamide (DMF) intercalation. Furthermore, the strong attraction of highly polar carbonyl and ammonium ions in DMF can stabilize the lattice structure, and low-polar alkyl groups can interact with the weak electrostatic generated by Zn2+ , which allows Zn2+ to be freely intercalated. The DMF-NVO NFAs/CC//Zn battery exhibits an impressive high capacity of 536 mAh g-1 at 0.5 A g-1 , excellent rate capability, and cycling performance. The results of density functional theory simulation demonstrate that the intercalation of DMF can significantly reduce the band gap and the diffusion barrier of Zn2+ , and can also accommodate more Zn2+ . The assembled flexible aqueous rechargeable zinc ion batteries (FARZIBs) exhibit outstanding energy density and power density, up to 436 Wh kg-1 at 400 W kg-1 , and still remains 180 Wh kg-1 at 4000 W kg-1 . This work can provide a reference for the design of cathode materials for high-performance FARZIBs.

This study aims to clarify the effect of Jingfang Mixture on the treatment of chronic urticarial and its mechanism, and investigate the regulatory effect of chronic urticaria on the metabolic disorder of endogenous metabolites in the blood. The mice were randomly divided into normal group, model group, and Jingfang Mixture group, and modeling and administration continued for 21 d. The changes in endogenous small molecules in rat serum were determined by ultra-high performance liquid chromatography-electrospray ionization-Q Exactive-Orbitrap-mass spectrometry(UHPLC-ESI-QE-Orbitrap-MS) metabolomics technology. The change trend of endogenous metabolites in rat serum was analyzed to find potential biomarkers. The results showed that Jingfang Mixture regulate 16 biomarkers, mainly including taurine, glutamate, succinic acid, docosahexaenoic acid, and arachidonic acid. Metabolic pathway analysis was carried out by MetaboAnalyst, and P<0.01 was taken as the potential key metabolic pathway. Ten metabolic pathways were closely related to the treatment of chronic urticarial by Jingfang Mixture, mainly involved in the glutamate metabolism, taurine and hypotaurine metabolism, arginine and proline metabolism, arachidonic acid metabolism, tricarboxylic acid cycle, unsaturated fatty acid biosynthesis, glutathione metabolism, phenylalanine metabolism, alanine, aspartic acid, and glutamate metabolism, and butyric acid metabolism. Glutamate metabolism and butyric acid metabolism involved more metabolic pathways than others. Therefore, it was speculated that Jingfang Mixture had a balanced regulating effect on the related metabolic pathways which caused the serum disorder in the rats with urticaria, and tended to regulate the metabolic differential to the normal level in the rats with urticaria. This paper provides references for studying the mechanism of Jingfang Mixture from the perspective of endogenous metabolites and metabolic pathways in vivo. At the same time, the endogenous substances explored in this paper can be used as important biomarkers for the prevention of urticaria.

Background: To improve the dissolution and bioavailability of the component-based Chinese medicine of Ginkgo biloba leaves (GBCCM), a novel nanocrystalline solid dispersion of GBCCM (GBCCM NC-SD) was first prepared. Methods: GBCCM mainly containing high pure flavonoid aglycones (FAs) and terpenoid lactones (TLs) was used as the model drug. PVP K30 and SDS were used as solubilizers, combined stabilizers and carriers, and GBCCM NC-SD was prepared by high-pressure homogenization combined with freeze-dryer. Morphology and crystal characteristic of GBCCM NC-SD were analyzed. The dissolution and bioavailability evaluation were performed to investigate the feasibility of GBCCM NC-SD by in vitro dissolution and in vivo integrated pharmacokinetic models. Results: After homogenizing for 30 cycles under the pressure of 650 bar and freeze-drying, GBCCM NC-SD with uniform quality would be obtained. The particle size, PDI and zeta potential were found to be 335.9 ± 32.8 nm, 0.29 ± 0.02 and -28.4 ± 0.7 mV respectively. Based on charged aerosol detector (CAD) technology, a new chromatographic method for simultaneous detection of eight components in GBCCM was developed. In vitro drug release study showed that the cumulative dissolution of FAs and TLs in GBCCM NC-SD increased from 12.77% to 52.92% (P < 0.01) and 90.91% to 99.21% (P < 0.05) respectively. In comparison with physical mixture of GBCCM and stabilizer (PM), the integrated pharmacokinetics AUC0-t of FAs and TLs in GBCCM NC-SD were significantly increased (P < 0.05), and the T1/2 of TLs was also significantly prolonged (P < 0.05). Conclusion: This study demonstrated that novel GBCCM NC-SD was prepared using Polyvinylpyrrolidone K30 (PVP K30) and Sodium dodecyl sulfate (SDS) as a synergetic stabilizer and also provided a feasible way to improve the dissolution and oral bioavailability of poorly soluble candidate antihypertensive drugs.

Configurational entropy (Sconf) is known to be a key thermodynamic factor governing a glass transition process. However, this significance remains speculative because Sconf is not directly measurable. In this work, we demonstrate the role of Sconf theoretically and experimentally by a comparative study of a Zr-Ti-Cu-Ni-Be high-entropy metallic glass (HE-MG) with one of its conventional MG counterparts. It is revealed that the higher Sconf leads to a glass that is energetically more stable and structurally more ordered. This is manifested by ab initio molecular dynamics simulations, showing that ∼60% fewer atoms are agitated above Tg, and experimental results of smaller heat capacity jump, inconspicuous stiffness loss, insignificant structural change during glass transition, and a more depressed boson peak in the HE-MG than its counterpart. We accordingly propose a model to explain that a higher Sconf promotes a faster degeneracy-dependent kinetics for exploration of the potential energy landscape upon glass transition.

Karst groundwater is an important water resource but it is vulnerable to contaminants, due to the distinctive geological features of abundant transmissive fractures and conduits in the karst area which connect the surface to the underground systems. Anthropogenic activity-derived polycyclic aromatic hydrocarbons (PAHs) on the surface environment could enter groundwater easily and rapidly and threaten water security in karst areas. Samples in the multimedia environment from 10 specific karst spring systems from Western Hubei of Central China were collected to analyze 16 priority PAHs and to investigate their transport in these karst spring systems. The total concentrations of PAHs in the soil, river water, river sediments, spring water, and spring sediments ranged between 6.04 and 67.7 ng g-1, 4.56 and 11.4 ng L-1, 29.9 and 1041 ng g-1, 4.09 and 222 ng L-1, and 5.88 and 83.0 ng g-1, respectively. Levels of PAHs in this area were relatively low when compared to other karst areas. Proportions of low-molecular-weight (LMW)-PAHs in the water, sediments and soil (average 58.2-78.8%) were much higher than those of high-molecular-weight (HMW)-PAHs. The proportion of LMW-PAHs in the sediments (especially in river sediments) was higher than that in the soil. Characteristic ratio analysis and principal component analysis showed that PAHs were from high-temperature combustion of the mixture of coal and biomass, and vehicle emission, where coal and biomass combustion were the dominant sources. Significant correlations of PAH compositions in different media of karst spring systems were observed, especially in the Yuquangdong (YQD)-Migongquan (MGQ), Jiuzhenziquan (JZZQ), Xianyudong (XYD) and Fengdong (FD) karst spring systems, indicating the rapid PAH transport from the recharge area soil to the discharge area of spring water and sediments.

The development of cost-effective, high-performance and flexible electrocatalysts for hydrogen production is of scientific and technological importance. Catalysts with a core-shell structure for water dissociation have been extensively investigated. However, most of them are nanoparticles and thus their catalytic properties are inevitably limited by the use of binders in practice. Herein, this work reports a physical-metallurgy-based structural design strategy to develop a self-supported and unique nanoporous structure with core-shell-like ligaments, i.e., a Cu core surrounded by a NiO shell, formed on a metallic glass (MG) substrate. These newly developed noble metal-free catalysts exhibit outstanding HER performance; the overpotential reaches 67 mV at a current density of 10 mA cm-2, accompanied by a low Tafel slope of 40 mV dec-1 and good durability. More importantly, the current strategy could be readily applied to fabricate other nanoporous metals, which opens a new space for designing advanced catalysts as cost-effective electrode materials.

Introducing regions of looser atomic packing in bulk metallic glasses (BMGs) was reported to facilitate plastic deformation, rendering BMGs more ductile at room temperature. Here, we present a different alloy design approach, namely, doping the nonmetallic elements to form densely packed motifs. The enhanced structural fluctuations in Ti-, Zr- and Cu-based BMG systems leads to improved strength and renders these solutes' atomic neighborhoods more prone to plastic deformation at an increased critical stress. As a result, we simultaneously increased the compressive plasticity (from ∼8% to unfractured), strength (from ∼1725 to 1925 MPa) and toughness (from 87 ± 10 to 165 ± 15 MPa√m), as exemplarily demonstrated for the Zr20Cu20Hf20Ti20Ni20 BMG. Our study advances the understanding of the atomic-scale origin of structure-property relationships in amorphous solids and provides a new strategy for ductilizing BMG without sacrificing strength.

This study aims to evaluate HR-HPV viral load in the cervical lesion assessment and its diagnostic value on the triage of ASCUS. The three-step protocol for cervical cancer screening was carried out in 5171 patients from June 2017 to August 2019, and 1620 histopathological results were obtained. The positive rate of HR-HPV and TCT increased with the aggravation of pathological grades of cervical lesions. The sensitivity and specificity of HR-HPV (DH3) to detect CIN II+ were 91.91% and 84.46%, respectively. In comparison, the corresponding results of the cytology test were 80.51% and 83.12%. HPV16/18 viral load was positively correlated with the grade of cervical lesions (p < 0.001, r = 0.321). The diagnostic efficiency of AUC by applying HPV16/18 viral load was 0.682 for the diagnosis of CIN II+. The optimal HPV16/18 viral load for predicting CIN II+ was 6.80 RLU/CO (relative light units/cut-off), with corresponding sensitivity of 48.6%, specificity of 79.7%, and Youden index of 0.283. In the ASCUS population, viral loads were statistically different in HPV16/18 and the other 12 HR-HPV when compared cervicitis group with CIN I group and CIN II+ group (all p < 0.05). Statistical differences were detected concerning HPV16/18 viral load, contact bleeding status, and smoking status when compared cervicitis group with CIN I group and CIN II+ group (p < 0.05), with a corresponding odds ratio of 1.004, 1.533, and 5.513, respectively. Our findings suggest that HR-HPV viral load can be regarded as a useful tool to predict the grade of cervical lesions for ASCUS triage. ClinicalTrials.gov ID: NCT03178136.

BACKGROUND: This study aims to investigate the effect of hydrogen sulfide on the mitogen-activated protein kinases signaling pathway in in vitro cultured skin macrophages of burned rats. MATERIALS AND METHODS: Thirteen healthy Sprague-Dawley rats were divided into five groups: normal control group, burned control group, sodium hydrogen sulfide group, glibenclamide group, and sodium hydrogen sulfide + glibenclamide group. The burned rats were made into a deep II° 5% total body surface area flame burn injury model. The skin basement macrophages were separated from the skin of normal rats and the wound skin of burned rats and cultured. At 1, 6, and 12 h after intervention, extracellular signal-regulated kinase (ERK), c-Jun N-terminal kinase (JNK), and p38 protein levels were detected by Western blot, and ERK, p38, and JNK messenger RNA (mRNA) levels were detected by reverse transcription polymerase chain reaction. RESULTS: Differences in ERK, p38, and JNK mRNA and protein levels between the normal control group and burned control group were statistically significant (P < 0.05). At the same time point, the ERK, p38, and JNK mRNA and protein levels in the NaSH group were different from those in other groups, and the differences were statistically significant (P < 0.05). CONCLUSIONS: Hydrogen sulfide has a regulatory effect on ERK, JNK, and p38 in the mitogen-activated protein kinases signaling pathway in macrophages of burned rats.

RESUMO

A catalyst-free method for the highly regioselective chloroboration of allenylsilanes is described. In the presence of BCl3 and 2,6-lutidine, chloroboration of allenylsilanes proceeds smoothly without any catalyst, and the product could be treated with pinacol to afford the corresponding pinacol borates in one-pot reaction. This reaction provides a direct approach to construct valuable 2-silylallylboronate frameworks with operational simplicity and high atom-economy.

In this paper, effects of cryogenic thermal cycling on deformation behavior and thermal stability of the Zr46Cu46Al8 bulk metallic glass (BMG) were studied. The results show that with the increase of the number of cryogenic thermal cycles (CTC), thermal stability remains almost unchanged, while the plasticity is increased, indicating that the cryogenic thermal cyclic treatment is an effective way to improve plasticity of metallic glasses without distinctly deteriorating thermal stability. Our analysis suggests that the increase in the defect density resulted from the cryogenic thermal treatments are responsible for the plasticity increment. Variation of yield strength can be well interpreted from microstructural percolation which affected by both density and characteristic volume of the defect sites.

Although arctigenin (AG) has diverse bioactivities, such as anti-oxidant, anti-inflammatory, anti-cancer, immunoregulatory and neuroprotective activities, its pharmacokinetics have not been systematically evaluated. The purpose of this work was to identify the pharmacokinetic properties of AG via various experiments in vivo and in vitro. In this research, rats and beagle dogs were used to investigate the PK (pharmacokinetics, PK) profiles of AG with different drug-delivery manners, including intravenous (i.v), hypodermic injection (i.h), and sublingual (s.l) administration. The data shows that AG exhibited a strong absorption capacity in both rats and beagle dogs (absorption rate < 1 h), a high absorption degree (absolute bioavailability > 100%), and a strong elimination ability (t1/2 < 2 h). The tissue distributions of AG at different time points after i.h showed that the distribution of AG in rat tissues is rapid (2.5 h to reach the peak) and wide (detectable in almost all tissues and organs). The AG concentration in the intestine was the highest, followed by that in the heart, liver, pancreas, and kidney. In vitro, AG were incubated with human, monkey, beagle dog and rat liver microsomes. The concentrations of AG were detected by UPLC-MS/MS at different time points (from 0 min to 90 min). The percentages of AG remaining in four species' liver microsomes were human (62 ± 6.36%) > beagle dog (25.9 ± 3.24%) > rat (15.7 ± 9%) > monkey (3.69 ± 0.12%). This systematic investigation of pharmacokinetic profiles of arctigenin (AG) in vivo and in vitro is worthy of further exploration.

OBJECTIVE: To study the vascular endothelial function recovery and its mechanism of Banxia Baizhu Tianma Decoction (BBTD). METHODS: 54 SH rats were randomly divided into three groups: the blank control group, BBTD group and Captopril treatment group. BBTD (at the daily dose of 4. 320 g crude drug/kg) and Captopril (at the daily dose of 3.375 g/kg) was administered from the 7th week to the 24th week. Another eighteen Wistar-Kyoto rats of the same ages were taken as the control. Medication was discontinued and effects were observed until the 32nd week. The blood pressure was determined by arterial carotis cannula. The concentration of serum NO2(-) and total anti-oxidation were determined by Griess and fluorescence recovery after photobleaching (FRAP). The acetylcholine (Ach)-dependent relaxation of superior mesenteric artery was detected using in vitro vascular ring. The mRNA expressions of IL-1, IL-6 and iNOS were detected by Real-time PCR at the 18th, 24th, and 32nd week. RESULTS: BBTD could significantly lower blood pressure of SHR and the concentration of serum NO2(-) at the 18th and 24th week (P<0.05). The total anti-oxidation of SH rats increased at the 18th week (P<0.01), and ACh-dependent relaxation of superior mesenteric artery increased at the 24th week. The mRNA expressions of IL-1 was markedly suppressed by BBTD at the 18th, 24th, and 32nd week (P<0.05), while IL-6 and iNOS mRNA expression were significantly lowered only at the 32nd week (P< 0.01). Captopril could significantly lower blood pressure of SHR at the 18th and 24th week (P<0.05). It significantly increased the total anti-oxidation of SH rats at the 18th week (P<0.01). However, it could not increase ACh-dependent relaxation of superior mesenteric artery and regulate the concentration of NO2(-) at the 18th, 24th, and 32nd week. The mRNA expression of iNOS was markedly suppressed by Captopril at the 24th and 32nd week, while mRNA expressions of IL-1 and IL-6 were significantly lower only at the 32nd week (P<0.01). CONCLUSIONS: BBTD showed similar effect in decreasing the blood pressure to captopril, but it showed better effect in improving the mesenteric endothelial dysfunction of SHR, which may be associated with its inhibition on NO and IL-1 expression, and improvement of the oxidative stress state.

RESUMO

OBJECTIVE: To investigate the changes in renin angiotensin system (RAS) in hypertrophied myocardium of spontaneous hypertensive rat (SHR), and the effect of Banxia Baizhu Tianma Decoction (BBTD) on the changes in haemodynamic parameters and mRNA of signaling molecules of RAS at different periods. METHODS: Fifty-four male SHRs of 6 weeks old were randomly and equally divided into three groups: the untreated control group, the test group, and the positive control group, and they were treated respectively with distilled water, BBTD and captopril by dissolving in equal volume of water administrated via gavage for 18 weeks. Besides, 18 age matched Wistar-Kyoto (WKY) rats treated with distilled water were allocated in a normal control group. Rats were managed in batches at their age of 18, 24, and 32 weeks old. Rat's hemodynamic parameters were measured through carotid artery catheterization, myocardial pathology was observed, and their mRNA expressions of angiotensin (AGT), angiotensin-converting (ACE) and angiotension-converting 2 (ACE2) were determined by Real-time PCR. RESULTS: Compared with WKY rats, the arterial pressure and left ventricular mass index (LVMI)in SHR were significantly higher at 18, 24 and 32 weeks respectively (P < 0.01); average cycle rate showed in electrocardiogram was higher (P < 0.05), though the blood stream was similar; mRNA expressions of AGT and ACE in heart tissue were markedly higher (P < 0.01), but that of ACE2 at 18 and 24 weeks were lower (P < 0.01). Compared with untreated SHR, arterial pressure at 18 and 24 weeks was lower (P < 0.05); cardiac muscle structure was improved; LVMI at 24 weeks was improved (P < 0.05); the mRNA expressions of AGT and ACE were suppressed but that of ACE2 increased at 18, 24,and 32 weeks significantly in the test group after BBTD treatment (P < 0.05). CONCLUSIONS: Changes in RAS in the hypertrophied myocardium of SHR may be one of the molecular mechanisms for hypertension leading to left ventricular hypertrophy. BBTD can improve the hemodynamic parameters, regulate RAS, so as to lower the arterial pressure.

RESUMO

OBJECTIVE: To investigate the effects of total saponins from rhizomes and flowers of Panax notoginseng on tumor cell induced platelet aggregation (TCIPA). METHODS: MDA-MB-231 breast carcinoma was applied as inductor and the platelet aggregation were investigated by Born's method in vitro and in vivo. RESULTS: Saponins from the flowers of Panax notoginseng inhibited TCIPA at the dose of 120, 240, 480 mg/L respectively in vitro and in vivo. Saponins from rhizomes of Panax notoginseng inhibited TCIPA only at its high dose in vivo. CONCLUSION: The saponins from Panax notoginseng has a dose-dependent inhibition of TCIPA. It may be a new class of antimetastatic agent.
