RESUMO

BACKGROUND: We have used the advantages of the zebrafish model system to demonstrate which of the vertebrate myosin light chain kinase (MLCK) genes is expressed in thrombocytes and important for thrombus formation. METHODS AND RESULTS: Here we report that Mlck1a is an essential component of thrombus formation. Phylogenetic data revealed four zebrafish orthologous for three human MLCK genes. To investigate expression of the zebrafish mlck genes in thrombocytes we compared GFP-tagged platelets with other cells by microarray analysis, and showed that mlck1a expression was 4.5-fold enriched in platelets. Furthermore, mlck1a mRNA and mRNA for the platelet-specific cd41 co-localized in thrombi. Expression of other mlck subtypes was lower in GFP-tagged platelets (mlck1b; 0.77-fold enriched) and absent in thrombi (mlck1b, -2, -3). To investigate the role of Mlck1a in thrombus formation, we knocked down mlck1a using two morpholinos. This resulted in impaired morphology changes of platelets adhering on fibrinogen. In a thrombosis model, in which thrombocytes adhere to the vessel wall damaged by laser irradiation, thrombus formation was slowed down in mlck1a-deficient embryos. CONCLUSION: We conclude that Mlck1a is the subtype of MLCK that contributes to platelet shape change and thrombus formation.

RESUMO

Classical ballet is a demanding professional occupation, with participants who are often underserved in terms of accurate diagnosis and appropriate comprehensive medical care. The view that follows is designed to be as global and insightful as published to date. Specific rehabilitation considerations, dance mechanics, idiosyncratic differential diagnosis, and personality and equipment issues are discussed, and a rational view of dogma is presented.

RESUMO

Healthcare economics has caused dramatic changes in resource assessment, utilization, and reorganization in hospitals. To address the mismatch between demand and resources and to ensure quality at the bedside, staff members at a 354-bed acute care facility designed and implemented the Collaborative Care Model (CCM). The authors describe this theory-based, collaborative, multidisciplinary professional practice model.

RESUMO

Employed mothers must constantly juggle and balance the multiple roles they face on a daily basis, which can be overwhelming and may adversely affect well-being and health. Findings are presented from a naturalistic-inquiry study that explored how employed mothers with preschool-age children experience positive well-being in their lives. Interviews were conducted with 16 mothers who worked full time and had at least one preschool-age child living at home. Findings included seven categories reflective of experiential well-being: mutuality, spirituality, child-centeredness, acceptance, happiness, security, and enrichment of the world. Results of this study are anticipated to contribute to a foundation for theory development and subsequent theory testing for promoting well-being.

RESUMO

In today's rapidly changing practice, nursing diagnosis is a central and imperative process within nursing practice. However, a review of literature reveals many ambiguities in the description and application of nursing diagnosis. This lack of understanding of how nursing diagnoses function is the reason that many clinical nurses and educators do not use nursing diagnoses as the North American Nursing Diagnosis Association (NANDA) originally intended. Consequently, breakdowns in communication and continuity of care occur at all levels within the health care system. The author discusses descriptions and integration of nursing diagnoses in eight major nursing texts which can be a reference in selecting an appropriate resource for learning.
