OBJECTIVE: To investigate the effect of acupotomy, on mitophagy and the Pink1-Parkin pathway in chondrocytes from rabbits with knee osteoarthritis (KOA). METHODS: A KOA model was established via the modified Videman method. Rabbits were randomly divided into a control group (CON), KOA group and KOA + acupotomy group (Acu). Rabbits in the acupotomy group were subjected to acupotomy for 4 weeks after model establishment. The behavior of the rabbits before and after intervention was recorded. Cartilage degeneration was evaluated by optical microscopy and fluorescence microscopy. The level of mitophagy was evaluated by transmission electron microscopy, immunofluorescence and enzyme-linked immunosorbent assay (ELISA). The expression of phosphatase and tensin homolog (PTEN)-induced kinase 1 (Pink1)-Parkin mitophagy pathway components was evaluated by immunofluorescence, Western blotting and real-time polymerase chain reaction. RESULTS: In rabbits with KOA, joint pain, mobility disorders and cartilage degeneration were observed, the Mankin score was increased, collagen type Ⅱ (Col-Ⅱ) expression was significantly decreased, mitophagy was inhibited, mitochondrial function was impaired, and factors associated with the Pink1-Parkin pathway were inhibited. Acupotomy regulated the expression of Pink1-Parkin pathway-related proteins, the mitophagy-related protein microtubule-associated protein-1 light chain-3, the translocase of the outer membrane, and the inner mitochondrial membrane 23; increased the colocalization of mitochondria and autophagosomes; promoted the removal of damaged mitochondria; restored mitochondrial adenosine-triphosphate (ATP) production; and alleviated cartilage degeneration in rabbits with KOA. CONCLUSIONS: Acupotomy played a role in alleviating KOA in rabbits by activating mitophagy in chondrocytes via the regulation of proteins that are related to the Pink1-Parkin pathway.

Knee osteoarthritis (KOA) is one of the most frequently encountered diseases in the orthopedic department, which seriously reduces the quality of life of people with KOA. Among several pathogenic factors, the biomechanical imbalance of the knee joint is one of the main causes of KOA. Acupotomology believes that restoring the mechanical balance of the knee joint is the key to treating KOA. Clinical studies have shown that acupotomy can effectively reduce pain and improve knee mobility by reducing adhesion, contracture of soft tissues, and stress concentration points in muscles and tendons around the knee joint. In this protocol, we used the modified Videman method to establish a KOA model by immobilizing the left hindlimb in a straight position. We have outlined the method of operation and the precautions related to acupotomy in detail and evaluated the efficacy of acupotomy in conjunction with the theory of "Modulating Muscles and Tendons to Treat Bone Disorders" through the detection of the mechanical properties of quadriceps femoris and tendon, as well as cartilage mechanics and morphology. The results show that acupotomy has a protective effect on cartilage by adjusting the mechanical properties of the soft tissues around the knee joint, improving the cartilage stress environment, and delaying cartilage degeneration.

OBJECTIVE To optimize the processing technology of honey bran-fried Atractylodis Rhizoma, and to compare the anti-gastric ulcer effect before and after processing. METHODS Combing with entropy-weight and technique for order preference by similarity to ideal solution model, L(9 34) orthogonal experiment design was adopted to optimize the processing technology of honey bran-fried Atractylodis Rhizoma using the comprehensive score of the contents of atractylone, β-cineole, atractylenolide Ⅲ and atractylodine as evaluation index, using the ratio of excipients to medicine, frying temperature and frying time as factors. The validation tests were conducted. The gastric ulcer model of mice was induced by intragastrical administration of anhydrous ethanol; using Compound aluminum hydroxide tablet as positive control, anti-gastric ulcer effect of Atractylodis Rhizoma was compared with that of honey bran-fried Atractylodis Rhizoma using the contents of serum inflammatory factors [interleukin-2 (IL-2), IL-6, tumor necrosis factor-α (TNF-α)], ulcer index and inhibitory rate of gastric ulcer as evaluation indexes. RESULTS The optimal processing technology of honey bran-fried Atractylodis Rhizoma was as follows:ratio of adjuvant and medicinal materials of 3∶10 (g/g), frying temperature at 140 ℃ and frying time of 4 min. Results of 3 validation tests showed that the contents of 4 components (including atractylone), in honey bran-fried Atractylodis Rhizoma processed by the optimal technology kept stable (RSDs were 3.47%-5.80%, n=3); the comprehensive scores were 95.53%-95.89% (RSD=0.21%, n=3). Atractylodis Rhizoma and honey bran-fried Atractylodis Rhizoma could increase the serum content of IL-2 in mice, but reduce serum contents of IL-6 and TNF-α to varying degrees; honey bran-fried Atractylodis Rhizoma could significantly decrease its ulcer indexes (P<0.05 or P< 0.01); the improvement effect of honey bran-fried Atractylodis Rhizoma on the above indicators was generally better than that of the same dosage of Atractylodis Rhizoma (P<0.05 or P< 0.01). The inhibitory rates of low-dose, medium-dose and high-dose Atractylodis Rhizoma and honey bran-fried Atractylodis Rhizoma to gastric ulcer in mice were 9.18%, 19.30%, 30.70%, and 50.32%, 61.39%, 53.16%, respectively. CONCLUSIONS The optimal processing technology of honey bran-fried Atractylodis Rhizoma is stable and feasible, and the anti-gastric ulcer effect of Atractylodis Rhizoma has been enhanced after being fried with honey bran.

Objective:To observe the application effect of standard management-flexible teaching-pay attention to comprehensive quality improvement-correct evaluation (SFPC) teaching on the internship teaching of nursing students in department of obstetrics and gynecology.Methods:Forty-five nursing students who interned in the department of obstetrics and gynecology from July to December 2019 were classified as the control group, and the traditional teaching was adopted. Another 47 nursing students who interned from January to June 2020 were taken as research group, and SFPC was applied. The rotation assessment scores, clinical communication ability and comprehensive ability before and after the internship, and satisfaction with teaching of these nursing students were compared between the two groups. SPSS 23.0 was used for t test, chi-square test and rank sum test. Results:The results of theoretical and operational assessment of obstetrics and gynecology in the research group were significantly higher than those in the control group[(94.28±5.77) vs. (83.91±5.19); (91.85±5.27) vs. (81.07±5.24)]. The scores of building harmonious relationship, identifying patients' problems, keen listening, passing effective information, joint participation and verification of feelings after the teaching were significantly higher than those before teaching ( P<0.05). The scores of ideological quality, teaching activities, ability evaluation and other activities in the comprehensive ability assessment were significantly higher than those before teaching ( P<0.05), and those of research group were significantly higher than those of the control group ( P<0.05). The were significant differences in the grade distribution of teaching satisfaction between the two groups ( P<0.05), and the total satisfaction rate of nursing students in the study group was higher than that in the control group (95.74% vs. 80.00%). Conclusion:The application of SFPC teaching in the clinical teaching of nursing students in department of obstetrics and gynecology can significantly improve the performances of nursing students, clinical communication ability, comprehensive ability and nursing students' satisfaction with teaching.

Objective To explore the effect of information-motivation-behavioral (IMB) skills model on self-management behavior and self perceived burden of elderly patients with permanent bladder fistula. Methods A total of 92 elderly patients with permanent urinary bladder fistula were randomly divided into experiment group and control group in our hospital from November 2015 to November 2016, 46 cases in each group. In the control group, taking classes, face-to-face education were done for a total of 6 months once a month, 60 minutes each time. In the experiment group, the IMB model was used for the nursing intervention for a total of 6 months once a month, 60 minutes each time. The two groups of patients were assessed by self management behavior scale and self perceived burden scale. Results After the intervention, the score on self-management ability of the experiment group was (76.32 ± 4.73), higher than (67.22 ± 3.14) of the control group (P<0.05). The score on the self perceived burden of the experiment group was significantly lower than that of the control group (10.32 ± 4.48 vs. 19.98± 6.63, P<0.05). Conclusion The implementation of IMB model can improve the self-management behavior of elderly male patients with bladder fistula, and reduce the burden of self perception.

Aim To study the protection and possible mechanism of 5-hydroxy-1 H-indazole against 1-methyl-4-phenylpyridinium iodide ( MPP+)-induced SH-SY5 Y cell apoptosis. Methods An apoptotic model was es-tablished in human neuroblastoma SH-SY5 Y by MPP+in vitro. MTT analysis was used to evaluate the protec-tive effect of 5-hydroxy-1H-indazole. Immunochemistry and Hoechst33258 nuclear staining were used to ob-serve the neuroprotection and anti-apoptosis of 5-hy-droxy-1H-indazole. Western blot was used to detect the levels of P-tau ( Ser396 ) closely related to neuronal apoptosis and its upstream kinases:P-GSK-3β and CDK5 . Results MPP+ induced activation of GSK-3β, increase of activity of CDK5 , tau hyperphosphory-lation and neuronal cell apoptosis. However,5-hydrox-y-1 H-indazole reduced the activities of GSK-3β and CDK5,then decreased the level of tau hyperphosphory-lation and inhibited MPP+-induced SH-SY5 Y cells ap-optosis. Conclusions 5-hydroxy-1H-indazole could attenuate MPP+-induced SH-SY5 Y neuronal cell apop-tosis. Possible mechanism is that 5-hydroxy-1H-in-dazole inhibits GSK-3βand CDK5 two signal transduc-tion pathways to lower the level of tau phosphorylation, then plays a role of neuroprotection.

Aim To study the protective effect of 6-Hydroxy-1 H-indazole via inhibition of Tau phosphoryla-tion on MPP +-induced SH-SY5Y cells apoptotsis. Methods An apoptotic model induced by 1 -methy-4-phenylpridnium ion (MPP +)was established in cul-tured human neuroblastoma SH-SY5Y in vitro.MTT a-nalysis was used to evaluate the protective effect of pre-treatment of cells with 6-Hydroxy-1 H-indazole for 2 h. Hoechst33258 staining was used to observe apoptotic situation.Immunohistochemistry was used to detect the p-Tau (Ser396)expression.Results The viability of cells exposed to 200 μmol · L -1 MPP + for 48h was (47.80 ±0.84)% (P <0.01 ),MPP + induced phos-phorylation of Tau at Ser396 and neuronal apoptosis, while 6-Hydroxy-1 H-indazole inhibited MPP +-induced cellular apoptosis,increased the number of TH-positive cells via inhibiting Tau phosphorylation.Conclusion These results indicate that Tau may be a new target used to cure neurodegenerative disorders such as PD.

RESUMO

Objective: To observe the clinical effects on advanced esophageal cancer by combined Xihuang Pill with chemotherapy. Methods: Dividing 35 advanced esophageal cancer cases randomly into two groups: 18 cases in the treatment group and 17 cases in the control group. Cases in the control group were treated by nedaplatin 40mg/m2,d1,2; 5-Fu400mg/m2,d1-5; CF200 mg/m2,d1-5. While the treatment with Xihuang Pill combined. The difference of quality of life, hematological toxicity, efficacy and symptoms between the two groups was observed. Results: The quality of life, remission rate of some symptom in the treatment group were much better than the control group, and the hematological toxicity and efficacy were the same. Conclusion: Xihuang Pill could improve the quality of life of advanced esophageal cancer patient, and alleviate some symptoms.
