RESUMO

Effects of nutrition on insulin-like growth factor-I (IGF-I), IGF binding proteins (IGFBP), and insulin in plasma and dominant follicles were evaluated at day 72 and 56 (Exp. 1, n = 12 and Exp. 2, n = 28, respectively) postpartum in anovulatory primiparous beef cows. Cows were stratified based on body condition score at calving and randomly assigned to nutritional treatments: maintain (M), 2.27 kg of a 40 % CP supplement per day and ad libitum hay; or gain (G), ad libitum access to a 50 % concentrate diet and ad libitum hay. Blood samples were collected twice weekly starting 30 days postpartum. Ovarian follicles were evaluated using ultrasonography commencing 42 (Exp. 1) or 30 (Exp. 2) days postpartum. Body weight and condition score were greater (P < 0.05) for cows of G than M groups and postpartum interval to luteal function was longer for cows of the M than G group. Insulin and IGF-I concentrations in follicular fluid (FF) and plasma were greater (P < 0.05) for cows of the G than M group at follicular aspiration. Plasma and FF IGFBP4 and IGFBP5 concentrations were greater (P <  0.05) in Exp. 2, and IGFBP5 was greater in Exp. 1 for cows of the G than M group. Treatment did not affect FF steroid concentrations or granulosal cell CYP19A1, PAPPA, IGFBP4, and IGFBP5 mRNA abundance. These results indicate concentrations of IGF-I, insulin, IGFBP4, and IGFBP5 in FF and plasma are affected by nutritional intake and may be related to follicular function.

Fall-calving Angus cows were used to evaluate the effect of ambient temperature on duration of gestation. In Exp. 1, cows were AI and calved in August (n = 14) or October (n = 10). Cows grazed native prairie pasture in Oklahoma and had a BCS of 6.0 ± 0.5 (1 = emaciated, and 9 = obese) at parturition. Commencing 2 wk before the expected calving date, blood samples were taken from the coccygeal vein every 2 to 3 d until calving. Cows that calved in August tended to have shorter gestations (P = 0.07) compared with cows that calved in October. Maximum daily ambient temperature during the last 14 d of gestation was greater for August-calving cows (P < 0.001) compared with October cows. Concentrations of cortisol in plasma during the last 4 d of gestation were greater in cows that calved in August (P < 0.04) compared with cows that calved in October. In Exp. 2, cows were AI and calved in either mid-August (n = 7), late-August (n = 6), September (n = 6), or October (n = 8) to evaluate the effects of elevated ambient temperature on duration of gestation, ruminal temperature at parturition, and plasma cortisol, progesterone, and estradiol. Temperature boluses (SmartStock, LLC, Pawnee, OK) programmed to transmit temperature every hour were place in the rumen at 255 d of gestation. Cows grazed native prairie pasture in Oklahoma and had a BCS of 6.5 ± 0.4 at calving. Maximum ambient temperatures during d 263 to 273 of gestation were influenced by month of calving × day (P < 0.001). Duration of gestation was shorter for mid-August cows (P < 0.05) compared with October cows, but did not differ compared with late-August (P = 0.29) and September (P = 0.50) cows. Ruminal temperature during the 4 d before calving was not influenced by month of calving (P = 0.76). Ruminal temperature was decreased during the 24 h before parturition for cows in all months (P < 0.01) compared with 2 to 4 d before parturition. Concentrations of cortisol in plasma during d 271 to 276 of gestation were less (P < 0.05) for late-August compared with cows that calved during the other months. Concentrations of progesterone were greater during 7 d before parturition in October compared with cows that calved in September. Estradiol in plasma of cows during late gestation was not affected by month of calving (P = 0.76). Exposure of beef cows to elevated ambient temperature resulted in shorter gestations. Ruminal temperature in cows decreased ≥ 0.3°C the day before parturition.

Gestating Angus, nonlactating, spring-calving cows were used to determine variation in maintenance energy requirements (MR); to evaluate the relationship among MR and cow and calf performance, plasma concentrations of IGF-I, T4, glucose, insulin, and ruminal temperature; and to describe the LM proteome and evaluate protein abundance in cows with different MR. Cows (4 to 7 yr of age) with a BCS of 5.0 ± 0.2 and BW of 582 ± 37 kg in the second to third trimester of gestation were studied in 3 trials (trial 1, n = 23; trial 2, n = 32; trial 3, n = 38). Cows were individually fed a complete diet in amounts to meet predicted MR (Level 1 Model of NRC), and feed intake was adjusted weekly until constant BW was achieved for at least 21 d (maintenance). Cows were classified on the basis of MR as low (>0.5 SD less than mean, LMR), moderate (±0.5 SD of mean, MMR), or high (>0.5 SD more than mean, HMR) MR. Blood samples were taken at maintenance and at 2 mo postpartum in trial 2. Muscle biopsies were taken from LMR and HMR after cows consumed actual MR for 28 d (trial 2) or 21 d (trial 3). Proteins from LM were separated by 2-dimensional difference gel electrophoresis and were identified, and abundance was quantified and compared. The greatest differences in MR between cows were 29%, 24%, and 25% in trials 1, 2, and 3, respectively. Daily MR (NEm, kcal·BW(-0.75)·d(-1)) averaged 89.2 ± 6.3, 93.0 ± 4.9, and 90.4 ± 4.6 in trials 1, 2, and 3, respectively. Postpartum BW and BCS, calf birth and weaning weights, postpartum luteal activity, and ruminal temperature were not influenced by MR of the cows. Concentrations of IGF-I were greater (P = 0.001) in plasma of MMR compared with LMR cows consuming predicted MR diets, and MR was negatively correlated with concentrations of IGF-I in plasma (r = -0.38; P = 0.05) at 2 mo postpartum. A total of 103 proteins were isolated from LM; 52 gene products were identified. Abundance of specific proteins in the LM was not influenced (P > 0.11) by MR. Variation in MR of cows will make it possible to improve feed efficiency by selection. Identification of biomarkers for MR will allow selection of more efficient cows, which consume less feed and produce calves with similar weaning weights. Productive cows that require less feed for maintenance will improve efficiency of production and enhance sustainability of the environment.

The effect of acute nutritional restriction on metabolic status, gonadotropin secretion, and ovarian function of heifers was determined in 2 experiments. In Exp. 1, 14-mo-old heifers were fed a diet supplying 1.2 × maintenance energy requirements (1.2M). After 10 d, heifers were fed 1.2M or were restricted to 0.4 × maintenance requirements (0.4M; d 0). Heifers received PGF2α (25 mg, intramuscularly) on d -10, 0, and 10 to synchronize ovulation. After 30 d, 1.2M and 0.4M heifers were realimented to 1.2 M for 100 d. Blood samples were collected every other day from d 0 to 14 then 3 times weekly thereafter. Heifers in Exp. 2 were managed as in Exp. 1 except that animals were fitted with an indwelling jugular catheter and blood samples were collected at 10-min intervals for 8 h on d 9, 10, and 11. Concentrations of progesterone in plasma were used to quantify ovarian luteal function. All 1.2M heifers ovulated, whereas only 30% of 0.4M heifers ovulated in Exp. 1. Concentrations of NEFA were greater and concentrations of thyroxine and IGF-I were less (P < 0.05) in plasma of 0.4M heifers compared with 1.2M heifers. The size of dominant follicles in Exp. 1 was reduced (P < 0.05) in 0.4M compared with 1.2M heifers. Concentrations of IGF-I were increased and anovulatory heifers resumed ovarian cycles an average of 35 d after realimentation. Concentrations of insulin were greater (P < 0.05) in plasma of 1.2M compared with 0.4M heifers in Exp. 2. The frequency of LH pulses was reduced (P < 0.05) in 0.4M heifers on d 9, and FSH in plasma on d 11 was not influenced by treatment. Reduced concentrations of IGF-I in plasma of nutrient-restricted heifers were associated with the reduced size of dominant follicles and indicated a local effect of growth factors on follicles. The decreased LH pulse frequency of 0.4M heifers before luteolysis indicates that restriction of nutrients decreased LH support of follicle growth. A preovulatory increase in estradiol in plasma and an ovulatory surge of LH were not detected in nutrient restricted heifers that did not ovulate. It is concluded that restricting beef heifers to 0.4 × maintenance energy requirements reduced the availability of metabolic fuel and decreased metabolic hormones, resulting in changes within the reproductive neuroendocrine-ovarian axis that compromised the ability of the dominant follicle to secrete sufficient concentrations of estrogen to stimulate an ovulatory surge of LH.

Three experiments were conducted to evaluate methods of immunization against GnRH on antibody titer, luteal activity, and pregnancy in beef heifers. Experiment 1 evaluated the efficacy of adjuvants with 30 heifers. Control heifers were immunized against human serum albumin (HSA) emulsified in Freund's complete adjuvant (FCA). The other 4 treatments contained GnRH conjugated to HSA (HSA-GnRH) emulsified in FCA, Freund's incomplete adjuvant (FIA), DEAE dextran (DD) + mineral oil (MO), or DD+FIA. Treatment was in the mammary gland for all experiments. Titers against GnRH for heifers immunized against HSA-GnRH with FCA, DD+MO, or DD+FIA were greater than titers for HSA-GnRH with FIA or control heifers (P < 0.01). Body weight was reduced (P < 0.05) in control and FCA heifers compared with FIA, DD+MO, and DD+FIA heifers. Heifers immunized with DD+MO and DD+FIA had fewer granulomas in mammary glands than heifers treated with FCA (P < 0.01). In Exp. 2, 36 heifers were used to determine the effect of the protein conjugated to GnRH on titers against GnRH. Heifers (6/treatment) received a primary immunization against GnRH conjugated to HSA (HSA-GnRH), ovalbumin (OA-GnRH), or keyhole limpet hemocyanin (KL-GnRH), or heifers were immunized against each carrier protein. Antigens were emulsified in DD+FIA. Immunization of heifers against OA-GnRH, KL-GnRH, or HSA-GnRH suppressed luteal activity (P < 0.01) for 23, 16, and 12 wk, respectively, and antibody titers against GnRH were greater (P < 0.01) for 19, 5, and 7 wk, respectively, compared with heifers immunized against the carrier proteins. In Exp. 3, 90 heifers were used to determine the effect of immunization against GnRH on ovarian activity and pregnancy rate. Heifers (30/treatment) received a primary and 2 or 3 booster immunizations against GnRH conjugated to OA, and controls received a primary and 2 booster immunizations against OA. All antigens were emulsified in DD+FIA. At 8 wk after primary immunization, heifers were exposed to fertile bulls for 24 wk. Pregnancy rate was less (P < 0.01) for 3-booster heifers (13%) compared with control (83%) and 2-booster (62%) heifers. We conclude that immunization against GnRH, conjugated to OA and emulsified in DD+FIA, does not influence ADG and produces sufficient titers against GnRH to prevent estrous cycles with few mammary granulomas. Immunization against GnRH with 3 booster immunizations prevented luteal activity and pregnancy in most beef heifers for more than 4 mo.

Two experiments were conducted to determine the effects of sunflower seed supplements with varying fatty acid profiles on performance, reproduction, intake, and digestion in beef cattle. In Exp. 1, 127 multiparous spring-calving beef cows with free-choice access to bermudagrass hay were individually fed 1 of 3 supplements for an average of 83 d during mid to late gestation. Supplements (DM basis) included 1) 1.23 kg/d of a soybean hull-based supplement (control treatment); 2) 0.68 kg/d of linoleic sunflower seed plus 0.23 kg/d of the control supplement (linoleic treatment); and 3) 0.64 kg/d of mid-oleic sunflower seed plus 0.23 kg/d of the control supplement (oleic treatment). During the first 62 d of supplementation, the BW change was 11, 3, and -3 kg for cows fed the control, linoleic, and oleic supplements, respectively (P < 0.001). No difference in BW change was observed during the subsequent period (-65 kg, P = 0.83) or during the entire 303-d experiment (-31 kg, P = 0.49). During the first 62 d of supplementation, cows fed sunflower supplements tended (P = 0.08) to lose more body condition than cows fed the control diet, but BCS was not different (P > 0.22) for any subsequent measurement. At the beginning of the breeding season, the percentage of cows exhibiting luteal activity was greater for cows fed the control diet (43%; P = 0.02) than for cows fed either linoleic (20%) or oleic (16%) supplementation; however, first-service conception rate (67%; P = 0.22) and pregnancy rate at weaning (92%; P = 0.18) were not different among supplements. No differences were detected in calf birth (P = 0.46) or weaning BW (P = 0.74). In Exp. 2, 8 ruminally cannulated steers were used to determine the effects of sunflower seed supplementation on forage intake and digestion. Treatments (DM basis) included 1) no supplement; 2) a soybean hull-based supplement fed at 0.29% of BW/d; 3) whole linoleic sunflower seed fed at 0.16% of BW/d; and 4) whole high-oleic sunflower seed fed at 0.16% of BW/d. Hay intake was not influenced (P = 0.25) by supplement (1.51% of BW/d); however, DMI was greatest (P < 0.01) for steers fed the soybean hull-based supplement (1.93% of BW/d). Sunflower seed supplementation reduced (P < 0.01) NDF and ADF digestibility while increasing (P < 0.01) apparent CP and apparent lipid digestibility. In conclusion, whole sunflower seed supplementation resulted in reduced cow BW gain during mid to late gestation, but this reduction did not influence subsequent cow BW change, pregnancy rate, or calf performance.

Spring-calving Angus cows (n = 30) were used to evaluate changes in ruminal temperature (RuT) related to parturition and estrus. Cows were synchronized and artificially inseminated with semen from a single sire. Temperature boluses were placed in the rumen at 7.0 ± 0.2 mo of gestation. Boluses were programmed to transmit RuT every 15 min. Cows (BW = 623 ± 44 kg, BCS = 4.9 ± 0.4) calved during 3 wk, and estrus was synchronized at 77 ± 7 d after calving with PGF(2α). Cows were observed every 12 h to detect estrus. Daily average ambient temperatures ranged from 2 to 22 °C during parturition (February to March) and 17 to 25 °C during estrus (May to June). Ruminal temperature from 7 d before to 3 d after parturition and 2 d before to 2 d after visual detection of estrus was analyzed using the MIXED procedure. Ruminal temperatures <37.72 °C were attributed to water consumption and excluded from analyses. Day did not influence (P = 0.36) RuT from d -2 to -7 before parturition (38.94 ± 0.05 °C). Ruminal temperature decreased (P < 0.001) from d -2 to d -1 before parturition (38.88 ± 0.05 to 38.55 ± 0.05 °C, respectively). Ruminal temperature was not influenced (P = 0.23) by day from 1 d before to 3 d after parturition (38.49 ± 0.05 °C). Ruminal temperature at 0 to 8 h after detection of estrus (38.98 ± 0.09 °C) was greater (P < 0.001) compared with RuT at the same daily hour of the day before (38.37 ± 0.11 °C) or the day after estrus (38.30 ± 0.09 °C). Ambient temperature did not influence (P > 0.30) RuT at parturition or estrus. Ruminal temperature decreased the day before parturition and increased at estrus in spring-calving beef cows and has potential use as a predictor of parturition and estrus.

Angus x Hereford heifers (15 mo and artificially inseminated to a single sire) were used to evaluate the effect of prenatal nutritional restriction on postnatal growth and development. At d 32 of gestation, dams were stratified by BW and BCS and allotted to a low-nutrition [55% of NRC (1996) requirements, n = 10] or moderate-nutrition [100% of NRC (1996) requirements, n = 10] diet. After 83 d of feeding, dams were commingled and received a diet in excess of requirements. Dams were allowed to calve naturally, and birth weights and growth of calves were recorded. Bulls were castrated at birth. Steers (16 mo of age, 5 per treatment) received a high-concentrate diet ad libitum to a constant age (88 ± 1 wk). Steers were slaughtered and weights of the empty body and organs were recorded. Samples of organs, muscle (complexus), and perirenal and subcutaneous adipose tissue were stored at -80 degrees C, and then DNA and protein concentrations were quantified and expression of genes associated with fatty acid metabolism and glucose uptake were measured in adipose and muscle tissue. Dams had similar (P > 0.33) BW and BCS at the beginning of the experiment. At the end of restriction, dams on the low-nutrition diet weighed less (P ≤ 0.01) and had less BCS (P < 0.001) than those on the moderate-nutrition diet. Length of gestation was 274 ± 2 d for dams in the low-nutrition treatment and 278 ± 2 d (P = 0.05) for dams in the moderate-nutrition treatment. Nutrient restriction during gestation did not influence birth weight or postnatal growth of calves. Lungs and trachea of steers whose dams were fed the low-nutrition diet weighed less (P = 0.05) at slaughter than those of steers whose dams were fed the moderate-nutrition diet; weights of other organs were not influenced by treatment. Complexus muscle from steers whose dams were fed the low-nutrition diet had a greater (P = 0.04) concentration of DNA and larger muscle fiber area compared with steers whose dams were fed the moderate-nutrition diet. Abundance of mRNA for fatty acid binding protein 4, fatty acid translocase, and glucose transporter 4 was less in perirenal adipose tissue of steers whose dams were fed the low-nutrition diet compared with those whose dams were fed the moderate-nutrition diet. Nutritional restriction of dams during early gestation did not alter postnatal calf growth. However, concentrations of DNA in muscle tissue and muscle fiber area were greater in steers from dams exposed to restricted nutrient intake during early gestation.

Angus x Hereford heifers (15 mo and AI to a single sire) were used to evaluate the effect of prenatal nutritional restriction on postnatal growth and regulation of glucose in plasma. Dams (d 32 of gestation) were stratified by BW and BCS and allotted to low [LN, 55% of NRC (1996) requirements, n = 7] or moderate nutrition [MN, 100% of NRC (1996) requirements, n = 7]. After 83 d of feeding, dams were commingled and received a diet in excess of requirements. Dams were allowed to calve naturally, and bull calves were castrated at birth. Dams and calves were maintained as a group until weaning, and calves were maintained as a group after weaning. Calves (15 mo of age) were adapted to a similar diet during 2 wk; catheters were placed in both jugular veins; and calves were confined in stalls. Two days later, calves were subjected to an intravenous glucose challenge and the next day to an insulin challenge. Dams had similar (P = 0.31) BW at the beginning of the experiment. At the end of restriction, LN dams weighed less (P ≤ 0.01) and had less BCS (P < 0.001) compared with MN dams. Length of gestation was not affected by prenatal nutritional treatment. Nutrient restriction during gestation did not influence birth weight or postnatal growth. Concentrations of glucose (P = 0.49) and insulin (P = 0.29) were not different in plasma of LN and MN calves before glucose infusion. Plasma concentrations of glucose, after intravenous administration of glucose, decreased more rapidly (P = 0.05) in LN compared with MN calves. Concentrations of glucose (P = 0.68) and insulin (P = 0.55) in plasma of LN and MN calves were similar after infusion of insulin. Nutritional restriction of dams during early gestation did not influence postnatal growth, but altered clearance of glucose after a bolus infusion of glucose.

Mature Angus and Angus x Hereford cows were used to determine the effects of confinement area and number of cows in estrus on estrous behavior. During each of 2 replicates, 32 nonpregnant, nonlactating cows were maintained in a drylot (60 x 100 m) or in a pasture (12 ha). Estrous cycles were synchronized with 2 injections of PGF(2alpha) 10 to 14 d apart at the initiation of the experiment. Thereafter, PGF(2alpha) was administered between d 6 and 18 of the estrous cycle so that 1, 2 to 3, 4 to 6, or > or = 7 cows were in estrus at the same time. Concentrations of progesterone were quantified in plasma at treatment to ensure cows had a normal response to PGF(2alpha). Duration of estrus and the number of mounts received during estrus were recorded by HeatWatch. If any part of estrus for a cow occurred at the same time as estrus for another cow, the cow was considered to have concurrent estrus with the other cow. Cows in the drylot had a shorter (P < 0.02; 61.8 +/- 3.1 h) interval to estrus after PGF(2alpha) compared with cows on pasture (72.8 +/- 3.3 h). The interval to estrus was longer (P < 0.07) when cows were treated with PGF(2alpha) on d 10 to 13 (76.7 +/- 3.3 h) of the estrous cycle than when treated on d 6 to 9 (62.3 +/- 4.7 h) or d 14 to 18 (62.9 +/- 3.6 h). Increasing the number of cows concurrently in estrus increased the number of mounts each cow received per estrus (P < 0.001) and the duration of estrus (P < 0.01). When only 1 cow was in estrus, she received 11.0 +/- 6.2 mounts during 11.6 +/- 1.5 h. When > or = 7 cows were in estrus at the same time, each cow received 50.4 +/- 3.2 mounts during 17.3 +/- 0.8 h. Cows in drylot were in estrus longer (P < 0.04; 16.4 +/- 0.8 h) than cows on pasture (14.2 +/- 0.7 h). Duration of the longest interval between mounts received decreased (P < 0.002) as the number of cows in estrus at one time increased (5.3 +/- 0.7 h for 1 estrous cow; 2.6 +/- 0.3 h when > or = 7 cows were in estrus). We conclude that increasing the number of beef cows in estrus at the same time will increase the number of times a cow is mounted and the duration of estrus. The increase in estrous behavior associated with more cows in estrus could increase the number of estrous cows detected with infrequent visual observation.

The objective of this study was to determine the effects of BCS at calving and the amount of postpartum protein supplementation on the dominant follicle (DF) and behavioral characteristics at the first postpartum estrus of mature beef cows. Multiparous Angus x Hereford cows (n = 45) were fed to calve in thin (T; < 5) or moderate (M; >or=5) BCS. Cows were stratified by BCS and calving date, and randomly assigned to receive lesser (L; 1.2 kg/d) or greater (G; 2.5 kg/d) amounts of a 42% CP supplement. All cows grazed the same native grass pasture and were fed in individual stalls for 49 +/- 2 d. Beginning 20 d after calving, blood samples were collected from each cow thrice weekly, and estrous behavior was monitored continuously with a radiotelemetry system. At 4 to 16 h after the onset of estrus, size of the DF was determined by ultrasonography. Body condition score of T cows was less (P < 0.01) at calving than M cows; L and G cows had similar BCS at calving and at the end of the feeding period. Body weight gains during treatment did not differ for L or G cows. Duration from calving to first estrus was greater (P < 0.01) for T than M cows. The incidence of a short luteal phase before first estrus was not influenced by BCS or protein supplement. Concentrations of IGF-I in plasma tended (P < 0.07) to be greater and size of the DF was greater (P < 0.01) for M than T cows. Size of the DF tended (P < 0.06) to be greater for G than L cows. Duration and number of mounts received at the first estrus were not influenced by BCS or supplement. Pregnancy rate of M cows during the breeding season was greater (P < 0.05) than T cows. Postpartum protein intake and BCS at calving influenced the size of the DF at the first postpartum estrus in mature suckled beef cows. Cows should be managed to calve in moderate BCS and maintain BW after parturition to decrease the interval to first estrus, increase follicular development, and maximize pregnancy rate.

Two experiments were conducted to determine the effects of whole soybean supplementation on intake, digestion, and performance of beef cows of varying age. Treatments were arranged in a 2 x 3 factorial with 2 supplements and 3 age classes of cows (2-yr-old, 3-yr-old, and mature cows). Supplements (DM basis) included 1) 1.36 kg/d of whole raw soybeans, and 2) 1.56 kg/d of a soybean meal/hulls supplement. Supplements were formulated to provide similar amounts of protein and energy, but a greater fat content with the whole soybeans. Supplements were individually fed on Monday, Tuesday, Thursday, and Saturday mornings. During the treatment period, cows had free choice access to bermudagrass hay [Cynodon dactylon (L.) Pers.; 8.4% CP; 72% NDF; DM basis]. In Exp. 1, 166 spring-calving Angus and Angus x Hereford crossbred beef cows were individually fed supplements for an average of 80 d during mid to late gestation. During the first 50 d of supplementation, cows fed soybean meal/hulls gained more BW (10 kg; P < 0.001) and body condition (0.18 BCS units; P = 0.004) than cows fed whole soybeans. However, BW change (P = 0.87) and BCS change (P = 0.25) during the 296-d experiment were not different between supplements. Although calves from cows fed soybean meal/hulls were 2 kg heavier at birth, there was no difference in calf BW at weaning between supplements. Additionally, first service conception rate (68%; P = 0.24) and pregnancy rate (73%; P = 0.21) were not different between supplements. In Exp. 2, 24 cows from Exp. 1 were used to determine the effect of supplement composition on forage intake and digestion; cows remained on the same supplements, hay, and feeding schedule as Exp. 1. Crude fat digestibility was the only intake or digestibility measurement influenced by supplement composition; fat digestibility was higher for cows fed whole soybeans compared with cows fed the soybean meal/hulls supplement (58.1 vs. 48.8%). Hay intake and DMI averaged 1.63 and 1.92% of BW daily, respectively. Dry matter, NDF, and CP digestibility averaged 54.1, 55.1, and 63.2%, respectively. Compared with supplementation with soybean meal/ hulls, whole soybean supplementation during mid to late gestation resulted in reduced BW weight gain during supplementation, inconsistent effects on reproduction, no effect on calf weaning weight, and no effect on forage intake or digestion.

Spring calving Angus and Angus x Hereford multiparous cows were utilized to determine the effects of intramammary treatment with penicillin G procaine (200,000 IU) and novobiocin (400 mg) at the time of weaning on udder health and calf growth after the subsequent calving. Cows were stratified by age and breed and assigned randomly to receive intramammary treatment (n = 99) at weaning or as untreated controls (n = 97). Quarter milk samples were collected at weaning and at 8 to 14 d after calving. Milk samples were analyzed for somatic cell counts (SCC) and mastitis-causing bacteria. Dry cow treatment decreased (P = 0.005) the number of cows infected after calving. Treatment decreased (P = 0.04) the number of cows that developed new infections and reduced (P = 0.03) the number of quarters with mastitis-causing bacteria after calving that were infected at weaning. Somatic cell counts after calving were greatest (P = 0.008) for cows infected with Staphylococcus aureus. Treatment did not alter (P = 0.19) SCC of quarters after calving that were infected with S. aureus at weaning but reduced (P = 0.002) SCC after calving of quarters that were infected with coagulase-negative staphylococci at weaning. Body weight of calves during early lactation was increased (P = 0.006) if cows with intramammary infection were treated at weaning. Treatment of noninfected cows at weaning increased (P = 0.008) adjusted 205-d weaning weights of calves after the subsequent lactation when compared with untreated noninfected cows. We conclude that treatment of beef cows at weaning with intramammary antibiotics decreased intramammary infections after calving, improved udder health during the subsequent lactation, and increased BW gain of the calves.

The effect of days after calving on IGF-I, IGFBP, progesterone, androstenedione, estradiol, and aromatase mRNA in dominant ovarian follicles (DF) was evaluated in Angus x Hereford cows. Growth of DF (>9 mm) was monitored daily by ultrasonography and fluid from DF was collected in vivo at either 30+/-2 d or 47+/-2 d postpartum. Follicular fluid (FF) was also aspirated from DF of contemporary ovulatory cows at proestrus. Estrous behavior was monitored continuously using the HeatWatch system, and progesterone in plasma collected twice weekly was used to assess luteal activity. Anovulatory DF aspirated 30 and 47 d postpartum had similar concentrations of IGF-I, IGFBP, progesterone, estradiol and androstenedione in FF and IGF-I and IGFBP in plasma. The intervals from aspiration to estrus were similar for cows aspirated 30 and 47 d postpartum. Proestrous follicles had greater (P<0.01) estradiol (435+/-79 ng/mL) than DF at 30 d (107+/-63 ng/mL) or 47 d (68+/-53 ng/mL) after calving. Concentrations of androstenedione in FF were also greater (P<0.01) in proestrous follicles than in DF aspirated at 30 or 47 d after calving. Concentrations of insulin-like growth factor-1 (IGF-I) and insulin-like growth factor binding proteins (IGFBP) in FF and plasma, and aromatase mRNA in granulosa cells were similar for anovulatory and proestrous cows. In conclusion, estradiol production by DF of postpartum anovulatory cows may be limited by inadequate production of androstenedione during the postpartum anovulatory interval and this may influence follicular maturation. Concentrations of IGF-I and IGFBP were similar in anovulatory and proestrous cows, an indication that alterations in the IGF-I system in the DF at 30-47 d after calving are not associated with delayed follicular development in postpartum beef cows.

The effect of plasma from cyclic versus nutritionally induced anovulatory beef heifers was evaluated on proliferation of bovine granulosa cells in vitro. Granulosa cells were obtained from small (1-5mm) follicles of cattle and cultured for 4 days. During the last 2 days of culture, cells were exposed to medium containing 0, 1 or 10% plasma from cyclic or anovulatory heifers in the presence or absence of IGF-I (100ng/ml). Cell numbers were determined. Regardless of source, increasing percentage of plasma to culture medium increased cell numbers. However, the plasma-induced increase was greater in granulosa cells exposed to cyclic heifer plasma versus anovulatory heifer plasma. In addition, concomitant treatment with IGF-I dramatically improved cell proliferation induced by anovulatory heifer plasma. These results indicate that plasma from cyclic heifers contain factors that are a greater stimulus to granulosa cell proliferation than plasma from anovulatory heifers. Systemic factors such as IGF-I may play a role in directly regulating granulosa cell proliferation in cattle.

The objective of this experiment was to evaluate the effects of active immunization against 2 GnRH isoforms on gonadotropin secretion and testicular function in pigs. Synthetic chicken (c) GnRH-II and lamprey (l) GnRH-III peptides, with the common pGlu-His-Trp-Ser sequence at the N-terminal omitted, were conjugated to BSA. Forty-eight male piglets were randomly assigned to 1 of 4 treatments. Pigs on treatment 1 were actively immunized against cGnRH-II, whereas pigs on treatment 2 were actively immunized against lGnRH-III. Control pigs on treatment 3 were actively immunized against the carrier protein (BSA), and pigs on treatment 4 were castrated and actively immunized against BSA. The BSA conjugate was emulsified in Freund's Incomplete Adjuvant and diethylaminoethyldextran. Primary immunization was given at 13 wk of age (WOA) with booster immunizations given at 16 and 19 WOA. Body weight and plasma samples were collected weekly beginning at 11 WOA. Treatments did not affect BW during the experimental period. Antibody titers were increased in animals immunized against cGnRH-II and lGnRH-III (P < 0.001). Cross-reactivity of the antibodies to mammalian GnRH or between cGnRH-II and lGnRH-III was minimal. Concentrations of testosterone were maximal in control boars (treatment 3) and minimal in control barrows (treatment 4) and immunized pigs (treatment x week; P < 0.01). Immunized animals had concentrations of LH (P < 0.001) and FSH (treatment x week; P < 0.03) that were less than control barrows and similar to control boars. At the end of the experiment, intact (noncastrated) pigs were exsanguinated. Testes were removed immediately; Leydig cells were isolated and treated with 0, 1, or 10 ng/mL of LH. There was an LH x GnRH treatment effect on testosterone concentrations (P < 0.03), indicating that Leydig cells were sensitive to the immunization protocol and doses of LH. Taken together, these data suggest that immunization against GnRH isoforms decreased gonadotropin secretion compared with control barrows. Additionally, immunization against cGnRH-II and lGnRH-III reduced the ability of Leydig cells to respond to LH challenges.

This experiment was conducted to determine the effects of interval feeding of whole sunflower seeds on the performance of beef cows and their progeny. During mid to late gestation, 144 multiparous, spring-calving beef cows (588 kg of initial BW; 5.6 initial BCS; 4 to 13 yr old) were individually fed 1 of 3 supplements 4 d/wk for a 76-d period. Supplements (DM basis) included: 1) 0.68 kg of soybean meal/feeding (NCON); 2) 3.01 kg of a soybean hull-based supplement/feeding (PCON); and 3) 1.66 kg of whole sunflower seeds high in linoleic acid/feeding (WSUN). Supplements were formulated to provide similar amounts of CP and ruminally degraded intake protein; PCON and WSUN were also formulated to be isocaloric. During the supplementation period, cows had free-choice access to bermudagrass (Cynodon dactylon) and tall-grass prairie hay. By the end of the 76-d supplementation period, cows fed PCON (P < 0.01) and NCON (P < 0.01) had gained more BW than cows fed WSUN (33, 23, and 10 kg, respectively). However, from the end of this supplementation period to the beginning of the breeding season 84 d later, cows supplemented with PCON had lost more (P < 0.01) BW than cows supplemented with WSUN (-123 kg vs. -111 kg). Cow BW change through weaning (-50 kg, P = 0.43) and final cow BW (536 kg, P = 0.70) at weaning were not different among supplement groups. Furthermore, cow BCS was similar among supplement treatment groups at the end of the supplementation period (5.3, P = 0.09), at the beginning of the breeding season (4.8, P = 0.38), and at weaning (4.7, P = 0.08). No difference among treatments was detected for calf birth weight (36 kg, P = 0.42), calf weaning weight (235 kg, P = 0.67), percentage of cows exhibiting luteal activity at the beginning of the breeding season (57%, P = 0.29), or pregnancy rate (88%, P = 0.44). However, first service conception rate was greater (P = 0.01) for cows fed PCON (79%) and tended (P = 0.07) to be greater for cows fed WSUN (74%) than for cows fed NCON (53%). After weaning, all steer calves were placed in a feedlot and fed a high-concentrate finishing diet for an average of 188 d. Supplements fed to dams during gestation did not influence feedlot performance or carcass characteristics. Prepartum energy supplementation, regardless of energy source or prepartum energy balance, resulted in improved conception rate, but other measures of reproduction, calf and feedlot performance, and carcass characteristics were not affected.

Spring-born Hereford x Angus heifers (n = 206) were used to determine effects of energy supplementation programs and amount of starch in the diet on incidence of puberty. In Exp. 1, heifers (205 +/- 5 kg; n = 68) grazing dormant native pasture were fed 0.9 kg/d (as-fed basis) of a 42% CP supplement from November until February 14. Heifers were stratified by weaning weight and allotted randomly to treatment before breeding (May to July). Treatments were 1) 0.9 kg (as-fed basis) of a 42% CP supplement/d and pasture (control); 2) a high-starch (HS) diet (73% corn; 53% starch) fed in a drylot for 60 d (HS-60); 3) a HS diet fed in drylot for 30 d (HS-30); or 4) a low-starch (LS) diet (49% corn; 37% starch) self-fed on pasture for 30 d (LS-30). The HS-60 and HS-30 heifers were limited-fed to gain 0.9 kg/d, and the LS-30 heifers had ad libitum access to the diet. High-starch-60 and LS-30 heifers were heavier (P < 0.05) than control and HS-30 heifers at the beginning of the breeding season. Thirty-one, 25, and 26% more HS-60 heifers were pubertal (P < 0.05) on May 1 compared with LS-30, HS-30, and control heifers, respectively. At puberty, HS-60 heifers were 24 and 22 d younger (P < 0.05) than LS-30 and control heifers, and 31 kg lighter (P < 0.01) than LS-30 heifers. In Exp. 2, heifers grazed dormant pasture and were fed 0.9 kg (as-fed basis) of a 42% CP supplement/d from weaning in October to late February; then heifers were assigned randomly to treatments for 60 d before the breeding season. In two years, control heifers (n = 46) grazed pasture and received 0.9 kg of SBM supplement/d; LS (n = 46) heifers were self-fed a distiller's grain and soybean hull-based diet in drylot; and HS heifers (n = 46) were limited-fed a corn-based diet in drylot. During treatment, HS and LS heifers had greater weight gains than control heifers. Pubertal BW (313 +/- 6 kg) was not influenced by treatment, but HS and LS heifers were younger (P < 0.03) than control heifers at puberty. During a 60-d breeding period, the incidence of puberty was greater (P < 0.05) for HS and LS heifers than for control heifers and was greater (P < 0.05) in HS than in LS heifers in Year 1. Feeding a LS or a HS diet for 30 d before breeding may be inadequate to stimulate puberty in beef heifers, but feeding a diet with a greater amount of starch for 60 d before breeding may increase the incidence of puberty during breeding of heifers that have inadequate yearling weight.

Pregnant Angus x Hereford cows (n = 73) were used to determine the effects of amount of nutrient intake and BCS on concentrations of IGF-I, insulin, leptin, and thyroxine in plasma. At 2 to 4 mo of gestation, cows were blocked by BCS and assigned to one of four nutritional treatments: high (H = a 50% concentrate diet fed ad libitum in a drylot) or adequate native grass pastures and one of three amounts of a 40% CP supplement each day (M = moderate, 1.6 kg; L = low, 1.1 kg; or VL = very low, 0.5 kg; as-fed basis). After 110 d of treatment, all cows grazed dormant native grass pasture and received 1.6 kg/d of a 40% CP supplement. At 68, 109, and 123 d of treatment, cows were gathered, and plasma samples were collected by tail venipuncture (fed sample). After 18 h without feed and water, a second plasma sample was collected (fasted sample). At 109 d of treatment, BCS was greatest (P < 0.05) for H cows, similar for M and L cows, and least for VL cows. Concentrations of insulin and leptin were greater (P < 0.05) for H cows than for M and VL cows at 68 and 109 d, but similar for all groups at 123 d. Thyroxine in plasma was greatest (P < 0.05) for H cows at 68 d and similar for cows on all treatments at 123 d. Concentrations of IGF-I, insulin, and leptin in fed and fasted cows were positively correlated with BCS at 109 d. Body condition was predictive of concentrations of IGF-I, insulin, and leptin when cows had different nutrient intakes, but BCS accounted for less than 12% of the variation in plasma concentrations of IGF-I, insulin, and leptin when nutrient intake was the same for all cows. We conclude that amount of nutrient intake has a greater influence than body energy reserves on IGF-I, insulin, and leptin concentrations in the plasma of gestating beef cows.

RESUMO

Two experiments were conducted using 48 Angus x Angus-Hereford steers in each experiment to determine the effect of previous winter grazing BW gain on jugular concentrations of metabolites and hormones during feedlot finishing. In each experiment, steers were randomly assigned to one of three treatments: 1) high rate of BW gain grazing winter wheat (HGW), 2) low rate of BW gain grazing winter wheat (LGW), or 3) grazing dormant tallgrass native range (NR) with 0.91 kg/d of a 41% CP (DM basis) supplement. Steers grazed for 120 or 144 d in Exp. 1 and 2, respectively. Plasma and serum were collected from all steers before placement into a feedlot, and six or seven times during finishing in Exp. 1 and 2, respectively. In Exp. 1, before steers entered the feedlot, concentrations of insulin, triiodothyronine (T3), and thyroxine (T4) were greater (P < 0.05) in HGW than in LGW or NR steers, and concentrations of IGF-I and plasma urea-N were greater (P < 0.05) in steers that grazed wheat pasture than in NR steers. In Exp. 2, concentrations of glucose, T3, T4, and IGF-I were greater (P < 0.05) in steers that grazed wheat pasture than NR steers. In Exp. 1 (P < 0.19) and 2 (P < 0.86), glucose concentration did not differ among treatments during finishing. In Exp. 1, insulin concentration across days on feed was greater for HGW than LGW steers, which were greater than for NR steers (treatment x day interaction, P < 0.03). In Exp. 2, insulin concentration increased (P < 0.001) as days on feed increased. Concentrations of IGF-I were greater in steers that had grazed wheat pasture, whereas the increase in IGF-I with increasing days on feed was greater for NR steers (treatment x day interaction, P < 0.003). Concentrations of T3 and T4 during finishing were greater (P < 0.001) in HGW and LGW than in NR steers in Exp. 1. In Exp. 2, T4 concentration also differed (P < 0.009) among treatments (HGW > LGW > NR). In Exp. 2, final concentration of glucose was greater (P < 0.01) in NR than in HGW and LGW steers, and serum insulin concentration was greater (P < 0.04) in NR than LGW steers. Final concentrations of T3 (P < 0.01) and T4 (P < 0.004) were greater in NR than in HGW steers. Our data show that previous BW gain can affect blood metabolites and hormones in steers entering the feedlot. However, lower concentrations of T3, T4, and IGF-I in steers when they entered the feedlot did not inhibit the growth response of previously restricted steers.
