RESUMO

This study compared artificial insemination pregnancy rate (AI-PR) between 14-day CIDR-GnRH-PGF2α-GnRH and CIDR-PGF2α-GnRH synchronization protocol with two fixed AI times (56 or 72 hr after PGF2α). On day 0, heifers (n = 1311) from nine locations assigned body condition score (BCS: 1, emaciated; 9, obese), reproductive tract score (RTS: 1, immature, acyclic; 5, mature, cyclic) and temperament score (0, calm; and 1, excited) and fitted with a controlled internal drug release (CIDR, 1.38 g of progesterone) insert for 14 days. Within herd, heifers were randomly assigned either to no-GnRH group (n = 635) or to GnRH group (n = 676), and heifers in GnRH group received 100 µg of GnRH (gonadorelin hydrochloride, IM) on day 23. All heifers received 25 mg of PGF2α (dinoprost, IM) on day 30 and oestrous detection aids at the same time. Heifers were observed for oestrus thrice daily until AI. Within GnRH groups, heifers were randomly assigned to either AI-56 or AI-72 groups. Heifers in AI-56 group (n = 667) were inseminated at 56 hr (day 32 PM), and heifers in AI-72 group (n = 644) were inseminated at 72 hr (day 33 AM) after PGF2α administration. All heifers were given 100 µg of GnRH concurrently at the time AI. Controlling for BCS (p < .05), RTS (p < .05), oestrous expression (p < .001), temperament (p < .001) and GnRH treatment by time of insemination (p < .001), the AI-PR differed between GnRH treatment [GnRH (Yes - 60.9% (412/676) vs. No - 55.1% (350/635); p < .05)] and insemination time [AI-56 - 54.6% (364/667) vs. AI-72 - 61.8% (398/644); (p < .01)] groups. The GnRH treatment by AI time interaction influenced AI-PR (GnRH56 - 61.0% (208/341); GnRH72 - 60.9% (204/335); No-GnRH56 - 47.9% (156/326); No-GnRH72 - 62.8% (194/309); p < .001). In conclusion, 14-day CIDR synchronization protocol for FTAI required inclusion of GnRH on day 23 if inseminations were to be performed at 56 hr after PGF2α in order to achieve greater AI-PR.

A multilocation study examined pregnancy risk (PR) after delaying AI in suckled beef cows from 60 to 75 h when estrus had not been detected by 60 h in response to a 7-d CO-Synch + progesterone insert (CIDR) timed AI (TAI) program (d -7: CIDR insert concurrent with an injection of GnRH; d 0: PGF injection and removal of CIDR insert; and GnRH injection at TAI [60 or 75 h after CIDR removal]). A total of 1,611 suckled beef cows at 15 locations in 9 states (CO, IL, KS, MN, MS, MT, ND, SD, and VA) were enrolled. Before applying the fixed-time AI program, BCS was assessed, and blood samples were collected. Estrus was defined to have occurred when an estrus detection patch was >50% colored (activated). Pregnancy was determined 35 d after AI via transrectal ultrasound. Cows ( = 746) detected in estrus by 60 h (46.3%) after CIDR removal were inseminated and treated with GnRH at AI (Control). Remaining nonestrous cows were allocated within location to 3 treatments on the basis of parity and days postpartum: 1) GnRH injection and AI at 60 h (early-early = EE; = 292), 2) GnRH injection at 60 h and AI at 75 h (early-delayed = ED; = 282), or 3) GnRH injection and AI at 75 h (delayed-delayed = DD; = 291). Control cows had a greater ( < 0.01) PR (64.2%) than other treatments (EE = 41.7%, ED = 52.8%, DD = 50.0%). Use of estrus detection patches to delay AI in cows not in estrus by 60 h after CIDR insert removal (ED and DD treatments) increased ( < 0.05) PR to TAI when compared with cows in the EE treatment. More ( < 0.001) cows that showed estrus by 60 h conceived to AI at 60 h than those not showing estrus (64.2% vs. 48.1%). Approximately half (49.2%) of the cows not in estrus by 60 h had activated patches by 75 h, resulting in a greater ( < 0.05) PR than their nonestrous herd mates in the EE (46.1% vs. 34.5%), ED (64.2% vs. 39.2%), and DD (64.8% vs. 31.5%) treatments, respectively. Overall, cows showing estrus by 75 h (72.7%) had greater ( < 0.001) PR to AI (61.3% vs. 37.9%) than cows not showing estrus. Use of estrus detection patches to allow for a delayed AI in cows not in estrus by 60 h after removal of the CIDR insert improved PR to TAI by optimizing the timing of the AI in those cows.

Energy reserve, estimated as body condition score (BCS), is the major determinant of the re-initiation of ovarian activity in postpartum cows. Leptin, IGF-I and insulin are positively related to BCS and are putative mediators between BCS and reproductive function. However, when BCS and body composition dissociates, concentrations of these metabolic hormones are altered. We hypothesized that increasing lean muscle tissue, but not fat tissue, would diminish the reproductive response to oestrus induction treatments. Thirty lactating beef cows with BCS of 3.10±1.21 and 75.94±12 days postpartum were divided in two groups. Control cows (n=15) were supplemented with 10.20 kg of concentrate daily for 60 days. Treated cows (n=15) were supplemented equally, and received a ß-adrenergic receptor agonist (ß-AA; 0.15 mg/kg BW) to achieve accretion of lean tissue mass and not fat tissue mass. Twelve days after ending concentrate supplementation/ß-AA treatment, cows received a progestin implant to induce oestrus. Cows displaying oestrus were inseminated during the following 60 days, and maintained with a fertile bull for a further 21 days. Cows in both groups gained weight during the supplementation period (Daily weight gain: Control=0.75 kg v. ß-AA=0.89 kg). Cows treated with ß-AA had a larger increase in BCS (i.e. change in BCS: control=1 point (score 4.13) v. ß-AA=2 points (score 5.06; P0.05) did not differ between groups. However, the number of cows displaying oestrus (control 13/15 v. ß-AA 8/15; P<0.05) and the percentage cycling (control 6/8 v. ß-AA 3/10; P=0.07) after progestin treatment and the pregnancy percentage at the end of the breeding period (control 13/15 v. ß-AA 8/15; P<0.05) were lower in ß-AA than control cows. In summary, the increase BCS through muscle tissue accretion, but not through fat tissue accretion, resulted in a lower response to oestrus induction, lower percentage of cycling animals and lower pregnancy percentage after progestin treatment; which was associated with a decrease in serum concentrations of leptin and IGF-I.

Two experiments were conducted to evaluate the effect of long-term (LT; a 14-day controlled internal drug release insert [CIDR]-PGF2α [PGF]-GnRH) and short-term (ST; 5-day CO-Synch + CIDR) progesterone-based protocols on pregnancy rate to fixed-time artificial insemination (FTAI) in beef heifers. In experiment 1, Angus cross beef heifers (N = 1887) at nine locations received a body condition score and a reproductive tract score (RTS). Within the herd, heifers were randomly assigned to LT-72 and ST-56 protocol groups. Heifers in the LT-72 group received a CIDR from Days 0 to 14, followed by 25 mg of PGF 16 days later (Day 30). Heifers in the ST-56 group received a CIDR and 100 µg of gonadorelin hydrochloride (GnRH) on Day 25 followed by 25 mg of PGF at CIDR removal on Day 30 and a second dose of PGF 6 hours later (Day 30). Artificial insemination was performed at 56 hours (Day 32) after CIDR removal for the ST-56 group and at 72 hours (Day 33) after CIDR removal for the LT-72 group, and all heifers were given GnRH (100 µg, intramuscular) at the time of AI. In experiment 2, Angus cross beef heifers (N = 718) at four locations received a body condition score and an RTS. Within the herd, heifers were randomly assigned to LT-72 and ST-72 protocol groups. The protocol was similar to experiment 1 except that AI was performed at 72 hours after CIDR removal for both LT-72 and ST-72 groups. In experiment 1, no difference in AI pregnancy rates between the LT-72 and ST-56 groups was observed (54.5% [489 of 897] and 55.5% [549 of 990], respectively; P = 0.92) after accounting for the RTS. The AI pregnancy rates for heifers with RTS 3 or less, 4, and 5 were 52.6%, 53.6%, and 59.9%, respectively (P < 0.05). In experiment 2, controlling for the RTS, no difference in AI pregnancy rates was observed between the LT-72 and ST-72 groups, 56.9% (198 of 347) and 57.8% (214 of 371), respectively (P = 0.87). The AI pregnancy rates for heifers with RTS 3 or less, 4, and 5 were 49.3%, 58.4%, and 62.1%, respectively (P < 0.05). In conclusion, heifers synchronized for fixed-time AI with LT and ST protocols resulted in a similar AI pregnancy rate. Approximately, 55% of the herd was pregnant to one insemination in 33 days with the LT protocol compared with just 8 days with the ST protocol.

Profitability of a beef operation is determined by the proportion of cows attaining pregnancy early in the breeding season and those that are pregnant at the end of breeding season. Many factors, including temperament, contribute to those reproductive parameters. The objective of this study was to evaluate effects of temperament on reproductive performance of beef cows. In Experiment 1, Angus and Angus-cross beef cows (n = 1546) from eight locations were assigned a body condition score (BCS; 1 = emaciated; 9 = obese) and chute exit and gait score (1 = slow exit, walk; calm temperament; 2 = jump, trot or run; excitable temperament). Cows were grouped with bulls (1 : 25 to 1 : 30; with satisfactory breeding potential and free of venereal disease) for an 85-day breeding season. Pregnancy status and stage of gestation were determined (transrectal palpation) 35 days after the end of the breeding season. Controlling for BCS (p < 0.01) and handling facility (p < 0.0001) and handling facility by temperament score interaction (p < 0.001), breeding season pregnancy rate was lower in excited versus calm cows [88.6% (798/901) vs 94.1% (607/645); p < 0.001]. Cows with an excitable temperament took 24 more days to become pregnant compared to calm cows (median days to pregnancy, 35 vs 59 days; p < 0.0001). In Experiment 2, Angus and Angus-cross beef cows (n = 1407) from 8 locations were assigned scores for body condition and chute exit and gait (as described in Experiment 1) and assigned to bulls (breeding sound and free of venereal disease; 1 : 25 to 1 : 30) for 85 days. Pregnancy status was determined by transrectal palpation at 2 and 6 months after the onset of the breeding season. Controlling for BCS (p < 0.05), pregnancy loss was higher in excited versus calm cows [5.5% (36/651) vs 3.2% (20/623), p < 0.0001]. In conclusion, beef cows with an excitable temperament had significantly lower reproductive performance than calmer cows. The modified two-point chute exit-gait scoring method was repeatable and identified cattle with an excitable temperament.

The objectives were (i) to evaluate the effect of temperament, determined by modified 2-point chute exit and gait score, on artificial insemination (AI) pregnancy rates in beef heifers following fixed time AI and (ii) to determine the effect of temperament on cortisol, substance-P, prolactin and progesterone at initiation of synchronization and at the time of AI. Angus beef heifers (n = 967) at eight locations were included in this study. At the initiation of synchronization (Day 0 = initiation of synchronization), all heifers received a body condition score (BCS), and temperament score (0 = calm; slow exit and walk or 1 = excitable; fast exit or jump or trot or run). Blood samples were collected from a sub-population of heifers (n = 86) at both synchronization initiation and the time of AI to determine the differences in serum progesterone, cortisol, prolactin and substance-P concentrations between temperament groups. Heifers were synchronized with 5-day CO-Synch+ controlled internal drug release (CIDR) protocol and were inseminated at 56 h after CIDR removal. Heifers were examined for pregnancy by ultrasound 70 days after AI to determine AI pregnancy. Controlling for synchronization treatment (p = 0.03), facility design (p = 0.05), and cattle handling facility design by temperament score interaction (p = 0.02), the AI pregnancy differed between heifers with excitable and calm temperament (51.9% vs 60.3%; p = 0.01). The alley-way with acute bends and turns, and long straight alley-way had lower AI pregnancy rate than did the semicircular alley-way (53.5%, 56.3% and 67.0% respectively; p = 0.05). The serum hormone concentrations differed significantly between different types of cattle handling facility (p < 0.05). The cattle handling facility design by temperament group interactions significantly influenced progesterone (p = 0.01), cortisol (p = 0.01), prolactin (p = 0.02) and substance-P (p = 0.04) both at the initiation of synchronization and at the time of AI. Inter- and intra-rater agreement for temperament scoring were moderate and good (Kappa = 0.596 ± 0.07 and 0.797 ± 0.11) respectively. The predictive value for calm and pregnant to AI was 0.87, and excited and non-pregnant to AI was 0.76. In conclusion, the modified 2-point temperament scoring method can be used to identify heifers with excitable temperament. Heifers with excitable temperament had lower AI pregnancy. Further, cattle handling facility design influenced the temperament and AI pregnancy.

The objective was to determine the effects of reproductive tract score (RTS) on reproductive performance in beef heifers bred by timed artificial insemination followed by natural service (AI-NS) or by natural service only (NSO). Angus cross beef heifers (n = 2660) in the AI-NS group were artificially inseminated at a fixed time (5- or 7-day CO-Synch + controlled internal drug release protocol) once, then exposed to bulls 2 weeks later (bull-to-heifer ratio = 1:40-1:50) for the reminder of the 85-day breeding season. Angus cross beef heifers (n = 1381) in NSO group were submitted to bulls (bull-to-heifer ratio = 1:20-1:25) for the entire 85-day breeding season. Heifers were reproductive tract scored from 1 (prepubertal) to 5 (cyclic) 4 weeks before, and were body condition scored (BCS) from 1 (emaciated) to 9 (obese) at the beginning of breeding season. Pregnancy diagnosis was performed 70 days after AI for AI-NS group and 2 months after the end of breeding season for both groups. Heifers in both groups were well managed and of similar age (14.9 ± 0.4 [AI-NS] and 14.7 ± 0.8 [NSO] months). Pregnancy rates (PRs) and number of days to become pregnant were calculated using PROC GLIMMIX and PROC LIFETEST procedures of SAS. Adjusting for BCS (P = 0.07), expressed estrus (P < 0.05), year (P < 0.05), and BCS by year interaction (P < 0.05), the AI-PR was greater for heifers in AI-NS group with higher RTS (P < 0.0001; 40.7%, 48.3%, 57.6%, and 64.6% for RTS of 2 or less, 3, 4, and 5, respectively). Controlling for BCS (P < 0.05), year (P < 0.05) and the breeding season pregnancy rates (BS-PRs) were greater for heifers in the AI-NS group with higher RTS (P < 0.01; 81.2%, 86.5%, 90.4%, and 95.2% for RTS of 2 or less, 3, 4, and 5, respectively). Similarly, adjusting for BCS, year (P < 0.05), the BS-PR was greater for heifers in NSO group with higher RTS (P < 0.01; 79.7%, 84.3%, 88.4%, and 90.2% for RTS of 2 or less, 3, 4, and 5, respectively). Heifers with higher RTS in both groups became pregnant earlier in the breeding season compared with heifers with lower RTS (log-rank statistics: P < 0.0001). Heifers in the AI-NS group become pregnant at a faster rate compared with those in the NSO group (P < 0.01). The BS-PR for heifers with RTS 5 was different between AI-NS and NSO groups (P < 0.0001). In conclusion, the RTS influenced both the number of beef heifers that became pregnant during the breeding season and the time at which they become pregnant. Furthermore, irrespective of RTS, heifers bred by NSO required more time to become pregnant than their counterparts in herds that used timed AI. The application of RTS system is reliant on the use of synchronization protocol. The application of RTS for selection may plausibly remove precocious females with lower RTS. On the contrary, application of RTS would help select heifers that will become pregnant earlier in breeding season.

The objectives were (1) to determine the effects of gonadorelin hydrochloride (GnRH) injection at controlled internal drug release (CIDR) insertion on Day 0 and the number of PGF2α doses at CIDR removal on Day 5 in a 5-day CO-Synch + CIDR program on pregnancy rate (PR) to artificial insemination (AI) in heifers; (2) to examine how the effect of systemic concentration of progesterone and size of follicles influenced treatment outcome. Angus cross beef heifers (n = 1018) at eight locations and Holstein dairy heifers (n = 1137) at 15 locations were included in this study. On Day 0, heifers were body condition scored (BCS), and received a CIDR. Within farms, heifers were randomly divided into two groups: at the time of CIDR insertion, the GnRH group received 100 µg of GnRH and No-GnRH group received none. On Day 5, all heifers received 25 mg of PGF2α at the time of CIDR insert removal. The GnRH and No-GnRH groups were further divided into 1PGF and 2PGF groups. The heifers in 2PGF group received a second dose of PGF2α 6 hours after the administration of the first dose. Beef heifers underwent AI at 56 hours and dairy heifers at 72 hours after CIDR removal and received 100 µg of GnRH at the time of AI. Pregnancy was determined approximately at 35 and/or 70 days after AI. Controlling for herd effect (P < 0.06), the treatments had significant effect on AI pregnancy in beef heifers (P = 0.03). The AI-PRs were 50.3%, 50.2%, 59.7%, and 58.3% for No-GnRH + PGF + GnRH, No-GnRH + 2PGF + GnRH, GnRH + PGF + GnRH, and GnRH + 2PGF + GnRH groups, respectively. The AI-PRs were ranged from 50% to 62.4% between herds. Controlling for herd effects (P < 0.01) and for BCS (P < 0.05), the AI pregnancy was not different among the treatment groups in dairy heifers (P > 0.05). The AI-PRs were 51.2%, 51.9%, 53.9%, and 54.5% for No-GnRH + PGF + GnRH, No-GnRH + 2PGF + GnRH, GnRH + PGF + GnRH, and GnRH + 2PGF + GnRH groups, respectively. The AI-PR varied among locations from 48.3% to 75.0%. The AI-PR was 43.5%, 50.4%, and 64.2% for 2.5 or less, 2.75 to 3.5, and greater than 3.5 BCS categories. Numerically higher AI-PRs were observed in beef and dairy heifers that exhibited high progesterone concentrations at the time of CIDR insertion (>1 ng/mL, with a CL). In addition, numerically higher AI-PRs were also observed in heifers receiving CIDR + GnRH with both high and low progesterone concentration (<1 ng/mL) initially compared with heifers receiving a CIDR only with low progesterone. In dairy heifers, there were no differences in the pregnancy loss between 35 and 70 days post-AI among the treatment groups (P > 0.1). In conclusion, GnRH administration at the time of CIDR insertion is advantageous in beef heifers, but not in dairy heifers, to improve AI-PR in the 5-day CIDR + CO-Synch protocol. In addition, in this study, both dairy heifers that received either one or two PGF2α doses at CIDR removal resulted in similar AI-PR in this study regardless of whether they received GnRH at CIDR insertion.

The objective of this experiment was to determine if 2 doses of prostaglandin F(2α) (PGF) administered concurrently at controlled intravaginal drug release (CIDR) removal was an efficacious method for delivery of PGF in the 5-d CO-Synch + CIDR protocol. Postpartum beef cows (n = 2,465) from 13 herds in 8 states were enrolled in the 5-d CO-Synch + CIDR protocol and assigned to receive 2 doses of PGF (25 mg/dose) 8 h apart with the initial injection given at CIDR insert removal (8h-PGF), 2 doses (25 mg/dose) of PGF delivered in 2 injection sites, both administered at CIDR insert removal (Co-PGF), or a single 25-mg dose of PGF at CIDR insert removal (1x-PGF). Cows were fixed timed-artificially inseminated (FTAI) 72 h after CIDR removal concurrent with GnRH administration. Estrus-cycling status (54% cyclic) was determined by evaluation of progesterone in 2 blood samples collected before CIDR insertion. Determination of pregnancy was performed by transrectal ultrasonography 39 ± 0.1 d after FTAI and at least 35 d after the conclusion of the breeding season. Fixed timed-AI pregnancy rates were greater (P < 0.05) for the 8h-PGF (55%) than the 1x-PGF (48%) treatment, with the Co-PGF (51%) treatment intermediate and not different (P > 0.10) from the other treatments. Contrast analysis demonstrated that cows receiving 50 mg of PGF (8h-PGF and Co-PGF) had greater (P < 0.05) FTAI pregnancy rates than those receiving 25 mg (1x-PGF). Pregnancy rates to FTAI were greater (P < 0.05) in cyclic (55%) than noncyclic (47%) and greater (P < 0.05) in multiparous (≥3 yr of age; 54%; n = 1,940) than primiparous cows (40%; n = 525). Luteolysis after PGF treatment was assessed in a subset of cows (n = 277) and treatment tended (P = 0.09) to affect the proportion of cows having luteolysis. The percentage of cows that had luteolysis was least in the 1x-PGF treatment (89%) and greatest in the 8h-PGF treatment (97%), with the Co-PGF treatment (94%) being intermediate. Breeding season pregnancy rate (88%) did not differ (P > 0.10) among treatments but was greater (P < 0.01) in multiparous (90%) than primiparous (78%) cows. In summary, 50 mg of PGF was required in the 5-d CO-Synch + CIDR protocol to maximize pregnancy rates; however, pregnancy rate did not differ when 50 mg of PGF was administered simultaneously with CIDR removal or split with 25 mg administered at 0 and 8 h after CIDR removal.

The objective was to determine whether timed artificial insemination (TAI) 56 h after removal of a Controlled Internal Drug Release (CIDR, 1.38 g of progesterone) insert would improve AI pregnancy rate in beef heifers compared to TAI 72 h after CIDR insert removal in a 5-days CO-Synch + CIDR protocol. Angus cross beef heifers (n = 1098) at nine locations [WA (5 locations; n = 634), ID (2 locations; n = 211), VA (one location; n = 193) and WY (one location; n = 60)] were included in this study. All heifers were given a body condition score (BCS; 1-emaciated; 9-obese), and received a CIDR insert and 100 µg of gonadorelin hydrochloride (GnRH) on Day 0. The CIDR insert was removed and two doses of 25 mg of dinoprost (PGF(2α)) were given, first dose at CIDR insert removal and second dose 6 h later, on Day 5. A subset of heifers (n = 629) received an estrus detector aid at CIDR removal. After CIDR removal, heifers were observed thrice daily for estrus and estrus detector aid status until they were inseminated. Within farm, heifers were randomly allocated to two groups and were inseminated either at 56 h (n = 554) or at 72 h (n = 544) after CIDR removal. All heifers were given 100 µg of GnRH at AI. Insemination 56 h after CIDR insert removal improved AI pregnancy rate compared to insemination 72 h (66.2 vs. 55.9%; P < 0.001; 1 - ß = 0.94). Locations, BCS categories (≤ 6 vs. > 6) and location by treatment and BCS by treatment interactions did not influence AI pregnancy rate (P > 0.1). The AI pregnancy rates for heifers with BCS ≤ 6 and > 6 were 61.8 and 60.1%, respectively (P > 0.1). The AI pregnancy rates among locations varied from 54.9 to 69.2% (P > 0.1). The AI pregnancy rate for heifers observed in estrus at or before AI was not different compared to heifers not observed in estrus [(65.4% (302/462) vs. 52.7% (88/167); P > 0.05)]. In conclusion, heifers inseminated 56 h after CIDR insert removal in a 5-days CO-Synch + CIDR protocol had, on average, 10.3% higher AI pregnancy rate compared to heifers inseminated 72 h after CIDR insert removal.

For the last 65 years, the industry standard for time of artificial insemination (AI) relative to onset of estrus has been established as 12 h after observed estrus. More recent experiments indicated the optimum time for AI was 4-12 h after onset of estrus in dairy cattle, but little recent information is available on the effect of time of AI relative to the onset of estrus in beef cattle. Therefore, we conducted a retrospective study on the impact of time from onset of estrus to artificial insemination using records from estrous synchronized crossbred Angus heifers (n=1205). Data were compiled from experiments on estrous synchronization systems and AI where estrous detection was performed electronically with HeatWatch(®). Only data from heifers detected in estrus was used. Heifers averaged 440 days of age, and weighed 368 kg at insemination. Heifers were assigned to one of the following 4h blocks of time based on time from onset of estrus (hour 0) to insemination: 0-4 (n=79), 4-8 (n=163), 8-12 (n=211), 12-16 (n=163), 16-20 (n=193), 20-24 (n=149), >24 h (n=247). Pregnancy rates to AI were not affected by AI technician or season of breeding (P>0.80). However, time of insemination relative to onset of estrus affected (P<0.01) pregnancy rates to AI. Heifers inseminated between 4 and 24h after onset of estrus had greater AI pregnancy rates (P<0.05; 63.7%) compared to the 0-4h (48.1%) and >24h (55.9%) groups. The time period for achieving acceptable pregnancy rates to AI in beef heifers appears to be approximately 20 h in length. Estrous synchronization programs resulting in a majority of heifers in estrus between 4 and 24 h before AI should yield the greatest pregnancy rates.

The objective was to compare the timed AI pregnancy rate of Angus-cross beef cows synchronized with a 5-d CO-Synch + CIDR (a progesterone-releasing intravaginal insert) protocol and given two doses of PGF(2 alpha) (PGF), with the first dose in conjunction with CIDR withdrawal on Day 5, and the second dose given either early or late relative to the first dose. All cows (N = 1782) at 16 locations received 100 microg of GnRH + CIDR on Day 0. Cows received 25 mg of PGF concurrent with removal of the CIDR on Day 5, and were randomly allocated within locations to receive a second PGF either early (N = 881; from 0.5 to 3.9 h) or late (N = 901; from 4.5 to 8.15 h) relative to the first PGF treatment. On Day 8 (72 h after CIDR removal), all cows were inseminated and concurrently given 100 microg of GnRH. Cows were fitted with a pressure-sensitive mount detection device (Kamar) at CIDR removal. Cows were observed twice daily through Day 7 and at the time of AI on Day 8 for estrus and Kamar status (estrus - red, partial and lost Kamar versus no estrus - white Kamar) was recorded. Accounting for location, season, AI sire, cow observed in estrus or not at or before timed AI, and treatment by cows observed in estrus interaction, timed AI pregnancy rates were greater for the late (6.45 +/- 0.03 h) than the early (2.25 +/- 0.05 h) interval, 57.2 vs. 52.7%, respectively (P < 0.05). In conclusion, cows that received the second PGF late after the first PGF on the day of CIDR removal in a 5 d CO-Synch + CIDR synchronization protocol had significantly higher timed AI pregnancy rates than those receiving the second PGF early after the first PGF.

Crossbred cows (n = 1073) from five locations had oestrous cycles synchronized with 100 microg of GnRH IM and insertion of controlled internal drug release device (CIDR) on Day 0 followed by 25 mg of PGF(2alpha) IM and CIDR removal on Day 7. Kamar patches were placed on all cows at CIDR removal. Cows were observed three times daily for oestrus after PGF(2alpha) administration. In the Ovsynch-CIDR group, cows detected in oestrus (n = 193) within 48 h after PGF(2alpha) were inseminated using the AM-PM rule. Among these cows, 80 received and 113 did not receive a second GnRH at 48 h after PGF(2alpha). Cows (n = 345) not detected in oestrus received a second GnRH at 48 h after PGF(2alpha) on Day 9, and fixed-time AI 16 h after the GnRH on Day 10. In the CO-Synch-CIDR group, cows detected in oestrus (n = 224) within 48 h after PGF(2alpha) were inseminated using the AM-PM rule. Among these cows, 79 received and 145 did not receive a second GnRH at 64 h after PGF(2alpha). Cows (n = 311) not detected in oestrus received a second GnRH on Day 10 at the time of AI, 64 h after PGF(2alpha). The AI pregnancy rates were not different between the Ovsynch-CIDR and CO-Synch-CIDR groups (p = 0.48). There were no differences in the AI pregnancy rates for cows inseminated at a fixed time (p = 0.26) or at detected oestrus (p = 0.79) between the treatment groups. Among cows inseminated in oestrus, there were no differences in the AI pregnancy rates between cows that received or did not receive the second GnRH (p = 0.47). In conclusion, acceptable AI pregnancy rates can be achieved with or without inclusion of oestrus detection in the Ovsynch-CIDR and CO-Synch-CIDR protocols. Among cows detected in oestrus, cows that received a second GnRH yielded similar pregnancy rates when compared with cows that did not receive the second GnRH.

The objective of this study was to compare fixed-time AI pregnancy rate in Angus crossbred beef cows inseminated with frozen-thawed or fresh-extended semen. Two ejaculates from each of two Angus bulls were collected by artificial vagina and pooled for each bull. The pooled semen from each bull was divided into two aliquots; Aliquot 1 was extended using Caprogen (LIC, Hamilton, New Zealand) to a concentration of 3 x 10(6)sperm/straw and Aliquot 2 was extended using egg-yolk-glycerol extender to a concentration of 20 x 10(6)sperm/straw. Semen extended with Caprogen was maintained at ambient temperature and semen extended with egg-yolk-glycerol extender was frozen and maintained at -196 degrees C until insemination. In each of two breeding seasons (Fall 2007 and Spring 2008), Angus-crossbeef cows (N=1455) at 12 locations were randomly assigned within location to semen type [Fresh (N=736) vs. Frozen (N=719)] and sire [1 (N=731) vs. 2 (N=724)]. All cows were synchronized with 100 microg of GnRH im and a progesterone Controlled Internal Drug Release insert (CIDR) on Day 0, and on Day 7, 25mg of PGF2(alpha) im and CIDR removal. All cows received 100 microg of GnRH im and were inseminated at a fixed-time on Day 10, 66 h after CIDR removal. Timed-AI pregnancy rates were influenced by season (P<0.05), cows detected in estrus prior to and at AI (P<0.001), and dam age (P<0.01). Pregnancy rates were not affected by semen type (Fresh=51.5% vs. Frozen=50.4%; P=0.66) and there were no significant interactions of semen type by estrus expression, semen type by sire, or semen type by season (P>0.1). In conclusion, commercial beef cows inseminated with fresh-extended semen (3 x 10(6)sperm/straw) yielded comparable pregnancy rates to conventional frozen-thawed semen in a progesterone supplemented, CO-Synch fixed-time AI synchronization protocol and may provide an alternate to frozen semen for more efficient utilization of superior genetics.

The objective was to compare reproductive performance of Angus-cross beef cows synchronized with GnRH, a progesterone-based intravaginal insert (Controlled Internal Drug Release, CIDR) for 5-d, and one dose of either dinoprost (PGF) or cloprostenol (CLP, a PGF analogue) or two doses of PGF on the day of CIDR withdrawal. All cows (N=830) at six locations received 100microg of GnRH and a CIDR on Day 0. Within farm, cows were randomly allocated to receive 25mg of PGF at the time of CIDR insert removal on Day 5 (1xPGF; N=277), two 25mg doses of PGF, the first given on Day 5 at the time of CIDR removal and the second 7h later (2xPGF; N=282), or 500microg of CLP at the time of CIDR removal on Day 5 (1xCLP; N=271). All cows were given 100microg of GnRH on Day 8 (72h after CIDR removal) and concurrently inseminated (5-d CO-Synch+CIDR). Cows were fitted with a pressure-sensitive estrus detection device at the time of CIDR withdrawal. Timed-AI pregnancy rates were greater (P<0.0001) in the 2xPGF (69.0%) than the 1xPGF (52.0%) and 1xCLP (54.3%) treatments. However, breeding-season pregnancy rates were not different among treatments (87.0% for 1xPGF, 92.9% for 2xPGF and 87.5% for 1xCLP; P>0.1). In conclusion, cows that received two doses of PGF on the day of CIDR removal in a 5-d CO-Synch+CIDR synchronization protocol had excellent timed-AI pregnancy rates that were greater than in cows receiving a single treatment with either PGF or CLP.

The objective was to determine the sire effect on the pregnancy outcome in beef cows in which stage of estrus was synchronized with progesterone based fixed-time artificial insemination (AI) protocols. Three Angus sires with more than 300 breedings were evaluated for differences in pregnancy outcome from 1868 inseminations. Angus cross beef cows (N=1868) were synchronized with Ovsynch-CIDR or CO-Synch-CIDR protocols for fixed-time AI. Cows in both groups that showed estrus on day 9 before 1500 h were designated to Selectsynch-CIDR group and were inseminated according to AM-PM rule. Results indicated that Sire 2 had lower fixed-time AI pregnancy rate compared to Sire 3 (48.1% versus 58.7%; P=0.01). Significant sirexsynchronization program and sirexlocation interactions were observed for fixed-time AI (P<0.05). Sire 2 had a lesser fixed-time AI pregnancy in both Ovsynch-CIDR and CO-Synch-CIDR groups compared to Sire 3. In two of four locations, Sire 2 had a lesser fixed-time pregnancy rate compared to Sire 3. No sire differences were observed in AI pregnancy for cows in Selectsynch-CIDR group. In conclusion, evidence in this study suggest that there are differences in sire fertility when they were used in fixed-time AI protocols, possibly due to the sire differences in sperm capacitation process. Further studies are needed to investigate association of the sire differences in fixed-time AI protocols with sire differences in the sperm capacitation process.

Our objectives were to examine the effects of added fat in late-gestation cow diets on neonatal response to cold. In Exp. 1, pregnant fall-calving heifers received control (n = 5), safflower seed (n = 5), or whole cottonseed (n = 5) diets. The hay-based, isonitrogenous, and isocaloric diets, fed for 47 d prepartum, contained 1.5, 4.0, and 5.0% fat for control, safflower, and whole cottonseed diets, respectively. At calving, calf BW and vigor score, as well as fat, lactose, and IgG in colostrum were not affected (P > 0.30) by diet. Heifers fed the safflower diet tended to have greater colostral solids (P < 0.10) than heifers fed the control or whole cottonseed diets. At 6.5 h of age, calves were placed in a 5 degrees C cold room for 90 min. Calf vigor, shivering, body temperature, and blood samples were taken every 15 min. During cold stress, calf body temperature decreased 0.7 degrees C (P < 0.03). Across all diets, shivering and serum glucose concentrations increased (P < 0.05), whereas calf vigor and cortisol concentrations decreased (P < 0.02) during cold exposure. In Exp. 2, pregnant spring-calving cows (n = 98) received a control (n = 47) or whole cottonseed (n = 51) supplement. Hay-based diets fed for 68 d prepartum contained 2.0 and 5.0% fat for control and whole cottonseed diets, respectively. Calf BW, vigor, shivering, dystocia score, time to stand, time to nurse, serum glucose concentrations, and serum IgG were not affected (P > 0.50) by diet. Between 30 and 180 min, body temperature of calves from dams fed the whole cottonseed supplement decreased (P < 0.05) more than calves from dams fed the control supplement. Serum glucose concentrations in calves were not affected by diet (P > 0.30). Serum cortisol concentrations tended (P < 0.09) to be greater for calves from dams fed whole cottonseed than control calves. When ambient temperature was < 6 degrees C, calves born to dams fed whole cottonseed had greater (P < 0.05) BW, tended (P < 0.1) to stand earlier, and had greater serum IgG concentrations. We conclude that calves from dams fed high-fat diets containing safflower or whole cottonseed respond similarly to cold stress, but these responses may not be consistent with greater cold resistance. In addition, high-fat dietary supplementation of late-gestation cows may only be beneficial during calving seasons with prolonged cold weather.

The objective of this study was to investigate metabolic alterations in young ruminating calves associated with the sudden introduction of readily fermentable diets in an attempt to induce laminitis. Sixteen dairy bull calves, at 17 wk of age were fed equal amounts of one of four diets that contained either 71 or 81% total digestible nutrients (TDN) and 15 or 20% crude protein in a 2 x 2 factorial with time as a factor. Jugular blood and ruminal fluid were sampled, and hoof temperature was measured postfeeding at frequent intervals over a subsequent 2-d period. Hooves were examined for abnormalities and the orientation of the pedal bone radiographed prior to the experiment, 48 to 72 h into the experiment and at 3 and 7 mo later. Calves responded acutely to the 81% TDN diets by inappetence, stiffness, and diarrhea. Ruminal pH was lower and both D- and L-lactate concentrations were greater in the rumens of calves fed the 81% TDN diets. Total ruminal volatile fatty acid concentration decreased as pH declined. Whole blood L-lactate did not differ across treatments, but blood D-lactate increased in calves fed the 81% TDN diets, peaking at 32 h (7.2 mM). Hoof temperature responses could not be explained by dietary treatments. Laminitis was not detected despite the reduction of ruminal pH and a manyfold increase in blood D-lactate.

Colostrum ingestion by neonatal calves is widely recognized to provide passive transfer of immunity. In this study immunoglobulin absorption from colostrum was evaluated in 54 IVF-produced calves. The IVF calves were delivered by Cesarean section on Days 275 to 277 of gestation, 24 h after the dams had been administered 30 mg dexamethasone. The calves suckled bottles or were force-fed 6 L of colostrum in the first 12 h of life. Colostrum was obtained from the first post-calving milking of recipient dams or from frozen storage reserves if dam secretion was not adequate. Immunoglobulin type G (IgG) content of both sources of colostrum was determined. Serum samples from the calves were collected at 0, 12 and 24 h of age and analyzed for IgG. Twenty dairy calves born vaginally served as the controls and were subjected to the same colostrum management protocol except that the colostrum was obtained only from frozen post-calving milk of dairy cows from the same farm. The control calves were also subjected to the same sampling protocol. The IVF group of calves ingested more IgG (P < 0.0001) and absorbed more IgG by 24 h of age (P < 0.0001) than their control group counterparts. Absorption of IgG was analyzed by comparing the g/kg body weight of IgG with serum IgG values at corresponding times after birth. Colostrum absorption efficiency was the same for both IVF and control groups of calves at 12 and 24 h of age. There was a maximum IgG dose above which additional increases in serum IgG were not realized. The slightly premature, Cesarean delivered IVF calves absorbed IgG from colostrum similarly to control calves delivered vaginally.
