RESUMO

Time-of-flight mass spectrometry-most notably matrix-assisted laser-desorption-ionization time-of-flight (MALDI-TOF) spectrometry-is an important class of techniques for the study of proteins and other biomolecules. Although these techniques provide excellent performance for masses up to about 20,000 daltons, there has been limited success in achieving good mass resolution at higher masses. This is because the sensitivity of the microchannel plate (MCP) detectors used in most systems decreases rapidly with increasing particle mass, limiting the utility of MCP detectors for very large masses. It has recently been proposed that cryogenic particle detectors may provide a solution to these difficulties. Cryogenic detectors measure the thermal energy deposited by the particle impact, and thus have a sensitivity that is largely independent of particle mass. Recent experiments have demonstrated the sensitivity of cryogenic particle detectors to single biomolecules, a quantum efficiency several orders of magnitude larger than the MCP detectors, and sensitivity to masses as large as 750,000 daltons. Here we present results demonstrating an order of magnitude better energy resolution than previous measurements, allowing direct determination of particle charge state during acceleration. Although application of these detectors to practical mass spectrometry will require further development of the detectors and cryogenics, these detectors can be used to elucidate the performance-limiting processes that occur in such systems.

The proliferation of low-cost microcomputer systems has led to the use of these systems as alternatives to expensive display devices for visual physiology and psychophysics experiments. The video displays of these systems often lack the flexibility of achieving wide linear luminance ranges and high vertical refresh rates--two parameters which may influence data acquisition. We have examined the responses of neurons and pairs of neurons in cat LGN and striate cortex to bar and sinusoidal grating stimuli generated by a conventional PC-based VGA graphics card and displayed on a NEC Multisync + color monitor with a 60 Hz vertical (display) refresh rate. Responses to these stimuli were autocorrelated and power spectral densities (PSD) were calculated, revealing that the majority of simple and complex cortical cells and nearly all LGN cells exhibited significant peaks in their autocorrelations at 16.7 ms and in the PSD at 60 Hz. Responses to identical stimuli generated with an optical bench using an incandescent light source contained no power at 60 Hz. Furthermore, cross-correlations between the spike trains of neuron-pairs were severely contaminated by peaks directly attributable to the entrainment of the two elements of the pair to the vertical refresh signal. Thus, we suggest that the use of conventional computer displays introduces a temporal artifact into neuronal spike trains in both single and multiple spike train analysis.

RESUMO

In an effort to help determine the current status and needs of hearing impaired children in Western Samoa, the Commonwealth Society for the Deaf sent a team of three professionals to this Pacific island nation in 1991. Initial information on known children with severe and profound hearing loss was gathered and a small prevalence survey of urban school children's ear health was conducted. No previously published information was available for this population. Results are presented and related to data from other areas of the Pacific. Hearing health care implications of the survey team's findings are discussed.
