Hawthorns are important medicinal and edible plants with a long history of health protection in China. Besides cultivated hawthorn, other wild hawthorns may also have excellent medicinal and edible value, such as Crataeguschungtienensis, an endemic species distributed in the Southwest of China. In this study, by integrating the flavor-related metabolome and transcriptome data of the ripening fruit of C. chungtienensis, we have developed an understanding of the formation of hawthorn fruit quality. The results show that a total of 849 metabolites were detected in the young and mature fruit of C. chungtienensis, of which flavonoids were the most detected metabolites. Among the differentially accumulated metabolites, stachyose, maltotetraose and cis-aconitic acid were significantly increased during fruit ripening, and these may be important metabolites affecting fruit flavor change. Moreover, several flavonoids and terpenoids were reduced after fruit ripening compared with young fruit. Therefore, using the unripe fruit of C. chungtienensis may allow us to obtain more medicinal active ingredients such as flavonoids and terpenoids. Furthermore, we screened out some differentially expressed genes (DEGs) related to fruit quality formation, which had important relationships with differentially accumulated sugars, acids, flavonoids and terpenoids. Our study provides new insights into flavor formation in wild hawthorn during fruit development and ripening, and at the same time this study lays the foundation for the improvement of hawthorn fruit flavor.

Gentiana rhodantha is a medicinally important perennial herb used as traditional Chinese and ethnic medicines. Secoiridoids are one of the major bioactive compounds in G. rhodantha. To better understand the secoiridoid biosynthesis pathway, we generated transcriptome sequences from four organs (root, leaf, stem and flower), followed by the de novo sequence assembly. We verified 8-HGO (8-hydroxygeraniol oxidoreductase), which may encode key enzymes of the secoiridoid biosynthesis by qRT-PCR. The mangiferin, swertiamarin and loganic acid contents in root, stem, leaf, and flower were determined by HPLC. The results showed that there were 47,871 unigenes with an average length of 1,107.38 bp. Among them, 1,422 unigenes were involved in 25 standard secondary metabolism-related pathways in the KEGG database. Furthermore, we found that 1,005 unigenes can be divided into 66 transcription factor (TF) families, with no family members exhibiting significant organ-specificity. There were 54 unigenes in G. rhodantha that encoded 17 key enzymes of the secoiridoid biosynthetic pathway. The qRT-PCR of the 8-HGO and HPLC results showed that the relative expression and the mangiferin, swertiamarin, and loganic acid contents of the aerial parts were higher than in the root. Six types of SSR were identified by SSR analysis of unigenes: mono-nucleoside repeat SSR, di-nucleoside repeat SSR, tri-nucleoside repeat SSR, tetra-nucleoside repeat SSR, penta-nucleoside repeat SSR, and hexa-nucleoside repeat SSR. This report not only enriches the Gentiana transcriptome database but helps further study the function and regulation of active component biosynthesis of G. rhodantha.

The increasing microplastics (MPs) pollution in freshwater wetlands has received global concerns. To investigate the spatiotemporal dynamics of MPs in the wetlands of Poyang Lake, surface water and sediment samples were collected from five rivers entering the lake as well as the confluence of Poyang Lake into the Yangtze River, in both dry and wet seasons. The MPs in water and sediment were extracted by the digestion-filtration method and flotation-separation-digestion-filtration method, respectively. Light microscope, Fourier transform infrared spectroscopy, and scanning electron microscope were used for microplastic characterization. The results showed that the abundance of MPs ranged from 32.1 to 127.3 n·L-1 in water samples, and from 533.3 to 1286.6 n·kg-1 in sediment samples during the wet season. In the dry season, the abundance of MPs ranged from 87.1 to 295.5 n·L-1 in water and from 460.0 to 1368.0 n·kg-1 in sediment. Compared with other freshwater wetlands, Poyang Lake had higher abundance of MPs. There were temporal and spatial differences among regions. The main forms of MPs included beads, fragment, film and fiber, and the corresponding polymer components were mainly polystyrene, polypropy-lene and polyethylene. Beads (35.7% in wet season and 52.0% in dry season) were the main form of MPs in water, while fragment (45.8% in wet season and 69.7% in dry season) was the main form of MPs in sediment. Small size (<0.1 mm) MPs were dominant (>50%) in water and sediment in both seasons. The abundance of MPs with different sizes decreased with the increases of size. The potential main sources of MPs in the wetlands of Poyang Lake included the discharge of industrial wastewater, discharge from urban and rural domestic sewage treatment plants, agricultural and fishing activities, and improper disposal of domestic wastes.

The hawthorns (Crataegus spp.) are widely distributed and famous for their edible and medicinal values. There are ∼18 species and seven varieties of hawthorn in China distributed throughout the country. We now report the chloroplast genome sequences from C. scabrifolia, C. chungtienensis and C. oresbia, from the southwest of China and compare them with the previously released six species in Crataegus and four species in Rosaceae. The chloroplast genome structure of Crataegus is typical and can be divided into four parts. The genome sizes are between 159,654 and 159,898bp. The three newly sequenced chloroplast genomes encode 132 genes, including 85 protein-coding genes, 37 tRNA genes, and eight rRNA genes. Comparative analysis of the chloroplast genomes revealed six divergent hotspot regions, including ndhA, rps16-trnQ-UUG, ndhF-rpl32, rps16-psbK, trnR-UCU-atpA and rpl32-trnL-UAG. According to the correlation and co-occurrence analysis of repeats with indels and SNPs, the relationship between them cannot be ignored. The phylogenetic tree constructed based on the complete chloroplast genome and intergenic region sequences indicated that C. scabrifolia has a different origin from C. chungtienensis and C. oresbia. We support the placement of C. hupehensis, C. cuneata, C. scabrifolia in C. subg. Crataegus and C. kansuensis, C. oresbia, C. kansuensis in C. subg. Sanguineae. In addition, based on the morphology, geographic distribution and phylogenetic relationships of C. chungtienensis and C. oresbia, we speculate that these two species may be the same species. In conclusion, this study has enriched the chloroplast genome resources of Crataegus and provided valuable information for the phylogeny and species identification of this genus.

Microplastic pollution (plastics with particle sizes<5 mm) has become a serious problem. In this study, we sampled the surface water, sediment, and bird excrement from the shore, center of the lake, and active areas for birds in Baisha Lake, which is key habitat for migratory birds in the Poyang Lake wetlands. The microplastics were separated by flotation separation, and then, we analyzed the pollution characteristics of this area. The main results were as follows. ① There were significant differences in the abundance of microplastics in water and sediment at different sampling points in the study area. The average abundance of microplastics in water and sediment were 263.28 per·m-3 and 215.9 per·kg-1, respectively. The average abundance of microplastics in migratory bird feces was 4.93 per·g-1. ② There were four kinds of microplastics in the surface water in terms of the different morphologies of particles, namely, pellets, films, fragments, and fibers, among which pellets were the major type. The main colors of microplastics were red, yellow, green, blue, purple, black, and transparent. The rank in terms of the size of microplastics was follows: < 1 mm > 1-2 mm > 2-3 mm > 3-5 mm, and the abundance decreased with the increase of particle size. ③ There were also four kinds of microplastics with different morphologies in the sediment, namely, pellets, films, fragments, and fibers. The main colors of these microplastics were red, yellow, blue, purple, black, and transparent. Microplastic sizes on the shore of the lake and bird habitat were mainly < 1 mm, and 1-2 mm particles were mainly detected in the center of the lake. ④ There were four types of microplastics in the excrement of migratory birds, namely, pellets, films, fragments, and fibers. The colors of these microplastics were red, yellow, green, purple, and transparent, and the particle size was mainly < 1 mm. ⑤ There was no consistency in the types of microplastics in the water body, sediment, and bird droppings. However, the size distribution for water and bird excrement was consistent. The results indicate that Poyang Lake bird habitat is polluted by microplastics, and the birds are inevitably exposed to microplastics. This study warns that microplastics will be an emerging threat to migratory birds.
