Abstract We aimed to compare the effects of oxycodone hydrochloride and dezocine on hemodynamics and inflammatory factors in patients receiving gynecological laparoscopic surgery under general anesthesia. A total of 246 patients were divided into group A and B (n=123). Hemorheology indices were recorded 5 min after anesthesia (T0), 1 min after pneumoperitoneum (T1), when position was changed 5 min after pneumoperitoneum (T2), 15 min after pneumoperitoneum (T3), 1 min (T4) and 5 min (T5) after position was restored. Visual analogue scale scores 1, 2, 6, 12, 24 and 48 h after operation were recorded. Postoperative adverse reactions and visceral pain were observed. The expression levels of inflammatory factors were detected by enzyme-linked immunosorbent assay 12 h after operation. Compared with group A, group B had higher heart rate and mean arterial pressure at T2, lower central venous pressure and cardiac output at T1-T3, and higher systemic vascular resistance at T1-T5 (P<0.05). The incidence rate of pain syndrome in group A was lower (P<0.05). Group A had lower tumor necrosis factor-alpha and interleukin-6 expression levels and higher interleukin-10 level than those of group B (P<0.05). For gynecological laparoscopic surgery, oxycodone preemptive analgesia has superior outcomes to those of dezocine

Deregulation of microRNAs (miRNAs) is essential to tumor development, and serum miRNA profiles have been reported in several cancers. However, the serum miRNA profile in oral squamous cell carcinoma (OSCC) remained unclear. The present study aimed to explore abnormal miRNA profile in sera samples from OSCC patients and the association of miR-483-5p with patient prognosis. Microarray analysis was performed in sera from OSCC patients versus healthy controls. miR-483-5p expression was measured by reverse transcription (RT)-PCR and correlated to clinicopathological characteristics of OSCC patients. The prognostic significance was then evaluated with a Kaplan-Meier curve and log-rank tests, using a Cox proportional hazard model. According to microRNA array, 16 miRNAs were upregulated and 10 were downregulated in OSCC patient sera. miR-483-5p expression was significantly increased in OSCC patients (3.23-fold, p < 0.01), and this was significantly correlated with tumor nodal metastasis (TNM) stage and lymph nodal metastases (p < 0.01, p < 0.01). For predicting OSCC, receiver operating characteristic (ROC)/area under the curve (AUC) analysis confirmed a AUC of 0.85 (sensitivity of 0.853 and specificity of 0.746). OSCC patients with high serum miR-483-5p had lower survival than those with low expression, and multivariate analyses for overall survival revealed that high serum miR-483-5p expression was an independent prognostic factor for OSCC (HR = 2.32, 95 %CI 1.20-4.48). miR-483-5p expression increased in OSCC patient sera, and this may be a novel diagnostic and prognostic biomarker for OSCC.

PURPOSE: Several studies have investigated the association of the interleukin (IL) 17A and IL-17F polymorphisms and cancer of various organs. However, the role of the IL-17A and IL-17F polymorphisms in oral squamous cell carcinoma (OSCC) remains unclear. Thus we sought to clarify the association of the rs2275913, rs763780, and rs2397084 polymorphisms with OSCC in a Chinese population. MATERIALS AND METHODS: A TaqMan single-nucleotide polymorphism Genotyping Assay (ABI, Foster, CA) was used to measure the distributions of the IL-17A (rs2275913) and IL-17F (rs763780, rs2397084) polymorphisms in 121 OSCC patients and 103 healthy controls. The association of those polymorphisms and clinical OSCC patient characteristic also was evaluated. RESULTS: Individuals carrying the rs2275913 A allele and AA genotype had an increased risk of OSCC (odds ratio [OR], 1.463; 95% confidence interval [CI], 0.807 to 2.652; and OR, 2.713; 95% CI, 1.250 to 5.889, respectively). The frequency of the rs2397084 T allele was significantly associated with a higher risk of OSCC than the G allele (OR, 1.501; 95% CI, 1.026 to 2.196). No difference in rs763780 frequencies was observed. The rs2275913 AA and rs2397084 TT genotypes also were associated with late clinical stages and poor tumor differentiation. In addition, stratification analysis indicated that the rs2275913 AA genotype increased OSCC risk among smoking and drinking populations (OR, 4.000; 95% CI, 1.404 to 11.394; and OR, 3.500; 95% CI, 1.018 to 12.030, respectively). In a smoking population, an rs9382084 T-allele carrier has a greater potential risk of OSCC than the overall population (OR, 2.200; 95% CI, 1.009 to 4.797). CONCLUSIONS: The results of this study suggest a significant association of rs2275913 and rs2397084 but not rs763780 with OSCC risk, and this was related to tumor stage and differentiation. In addition, the IL-17A and IL-17F polymorphisms can interact with smoking and drinking to enhance the risk of OSCC developing.

OBJECTIVE: To synthesize a biodegradable non-viral gene carrier with a high transfection efficiency and a low cytotoxicity. METHODS: Poly(ethylene glycol)-block-(poly(L-glutamic acid)-graft-polyethylenimine) was prepared via ammonolysis of poly(ethylene glycol)-block-poly (γ-benzyl L-glutamate) with the low-molecular-mass polyethylenimine (600 Da). The synthesized copolymer was characterized by 1H nuclear magnetic resonance spectroscopy and gel permeation chromatography. The polyplex micelle from PEG-b-(PG-g-PEI) and plasmid DNA (pDNA) was studied using dynamic light scattering, zeta-potential measurements, and gel retardation assay. The in vitro cytotoxicity and transfection efficiency of PEG-b-(PG-g-PEI) were tested by MTT assay and luciferase assay in HEK 293T cells using PEI (25 kDa) as the control. RESULTS: PEG-b-(PG-g-PEI) could efficiently condense DNA into nanosized particles with positive surface charges when the N/P ratio of polymer and DNA was above 5:1. The zeta potential of the polyplexes was about 25 mV, and the particle size was 120 nm at a N/P ratio of 10. The cell toxicity and gene transfection evaluations showed a lower cytotoxicity and a higher gene transfection efficiency of the copolymer than PEI 25000 in HEK 293T cells. CONCLUSIONS: The polymer can be used as a potential non-viral gene carrier for gene therapy.
