RESUMO

Long non-coding RNAs (lncRNAs) have been shown to be involved in the regulation of skeletal muscle development through multiple mechanisms. The present study revealed that the lncRNA SOX6 AU (SRY-box transcription factor 6 antisense upstream) is reverse transcribed from upstream of the bovine sex-determining region Y (SRY)-related high-mobility-group box 6 (SOX6) gene. SOX6 AU was significantly differentially expressed in muscle tissue among different developmental stages in Xianan cattle. Subsequently, knockdown and overexpression experiments discovered that SOX6 AU promoted primary skeletal muscle cells proliferation, apoptosis, and differentiation in bovine. The overexpression of SOX6 AU in bovine primary skeletal muscle cells resulted in 483 differentially expressed genes (DEGs), including 224 upregulated DEGs and 259 downregulated DEGs. GO functional annotation analysis showed that muscle development-related biological processes such as muscle structure development and muscle cell proliferation were significantly enriched. KEGG pathway analysis revealed that the PI3K/AKT and MAPK signaling pathways were important pathways for DEG enrichment. Notably, we found that SOX6 AU inhibited the mRNA and protein expression levels of the SOX6 gene. Moreover, knockdown of the SOX6 gene promoted the proliferation and apoptosis of bovine primary skeletal muscle cells. Finally, we showed that SOX6 AU promoted the proliferation and apoptosis of bovine primary skeletal muscle cells by cis-modulation of SOX6 in cattle. This work illustrates our discovery of the molecular mechanisms underlying the regulation of SOX6 AU in the development of beef.

RESUMO

OBJECTIVE: To observe the whole process of cranio-maxillofacial development of an animal model with bilateral premaxillary clefts, after being exerted with premaxillary retro-pressing forces. METHODS: Experimental rabbits with premaxillary clefts and without clefts were exerted with retro-pressing forces respectively. Roentgenographic cephalometric analysis and different investigations of premaxillary suture were performed during the whole craniofacial development. RESULTS: The longitudinal cephalometric analysis showed that a temporary retarding effect of mid-facial growth existed. The hindrance of maxillary growth potentiality did not occur in spite of being exerted retro-pressing force as to this experimental extent. CONCLUSIONS: (1) Infant rabbit combined with the designed retro-pressing appliance is an ideal experimental model for quantitative study of active orthopedic treatment of protruding premaxilla. (2) Using active preoperative orthopedics to reposit the protruding premaxilla of bilateral cleft palate is a reasonable and effective approach.

RESUMO

OBJECTIVE: To gain a clear idea of the function of middle ear and hearing in cleft palate with secretory otitis media after pressure equalization tube insertion. METHODS: 53 cleft palate cases with secretory otitis media including 62 ears were divided into treatment and control groups. They were examined by tympanogram and hearing threshold. RESULTS: Hearing threshold in treatment group after PE tube insertion respectively was 23.3 dB and 23.4 dB in the near future and at a specified future date. It was normal. But there were still 64% tympanogram B and C. CONCLUSIONS: PE tube insertion could solve hearing problem, but could not improve the function of middle ear.

OBJECTIVE: To compare results of the growth and development of the upper dental arch and the velopharyngeal closure of the cleft patients treated by two methods. METHODS: The dental cast of patient and X-ray films were measured and the statistical medical records were analyzed. RESULTS: The transverse distance of upper dental arch was found to be wider in group A than in group B. The anterior-posterior distance of the dental arch in bilateral cleft group was shorter in group A than in group B. The difference of the two groups were gradually lessened as age increases. Bony bridge in alveolar gap was 63% and 83.3% in unilateral and bilateral cleft group respectively. 15% of cases in group A and 35.2% in group B needed pharyngeal flap. CONCLUSIONS: The stable upper dental arch in group A can opposes the pressure from the lip muscles, this maintains the width of the arch. But A-P distance of upper dental arch in BCLP in group A should be followed up after the age of 9 years. Pharyngeal flap is needed less in group A than in group B.
