RESUMO

Goose astrovirus (GAstV) is a newly identified viral pathogen threatening waterfowl, exhibiting a high prevalence across various regions in China. Notably, the Guanghan District of Deyang City, situated in Sichuan Province, has faced a outbreak of GAstV, resulting in significant mortality among goslings due to the induction of gout-like symptoms. In our research, we successfully isolated a GAstV strain known as GAstV SCG3. This strain exhibits efficient replication capabilities, proving virulent in goslings and goose embryos. Our study delved into the characteristics of GAstV SCG3 both in vitro and in vivo. Additionally, we examined tissue phagocytosis and the distribution of GAstV SCG3 in deceased goslings using H&E staining and IHC techniques. According to the classification established by the ICTV, GAstV SCG3 falls under the category of GAstV genotype-2. Notably, it demonstrates the highest homology with the published AHAU5 sequences, reaching an impressive 98%. Furthermore, our findings revealed that GAstV SCG3 exhibits efficient proliferation exclusively in goose embryos and in LMH cells, while not manifesting in seven other types of avian and mammalian cells. Significantly, the mortality of GAstV on goslings and goose embryos are 93.1 and 80%, respectively. Moreover, the viral load in the livers of infected goslings surpasses that in the kidneys when compared with the attenuated strain GAstV SCG2. The mortality of GAstV is usually between 20% and 50%, our study marks the first report of a virulent GAstV strain with such a high mortality.

Goose astrovirus (GAstV) is a small, non-enveloped, single-stranded, positive-sense RNA virus. GAstV has rapidly spread across various regions in China since 2016. In Sichuan, out of 113 samples were collected from goose diseases between 2019 and 2022, 97 were positive for GAstV through PCR testing. Remarkably, over the past three years, GAstV outbreak in Sichuan has accounted for an astonishing 85.8% of all goose-origin viruses. Among these cases, 63.9% had single GAstV infections, 29.9% had dual infections, and 6.2% had quadruple infections. To comprehend the variations in virulence among distinct strains of GAstV. 12 representative strains of single GAstV infections were isolated. These strains exhibited distinct characteristics, such as prominent white urate depositions in organs and joints, as well as extensive tissues phagocytosis in major target organs' tissues. The conserved ORF1b genes and the variable ORF2 genes of these representative GAstV strains were sequenced, enabling the establishment of phylogenetic trees for GAstV. All GAstV strains were identified as belonging to genotype-2 with varying internal gene sequences. Experiments were conducted on GAstV genotype-2, both in vivo and in vitro, revealed significant variations in pathogenicity and virulence across susceptible cells, embryos, and goslings. This comprehensive study enhances researchers' understanding of the transmission characteristics and virulence of GAstV genotype-2, aiding in a better comprehension of their molecular epidemiology and pathogenic mechanism.

Goose astrovirus (GAstV, genus Avian Astrovirus, family Astrovirus) was first discovered in 2005, but was not considered as a pathogen of gosling gout until 2016. Since then, goose astrovirus has erupted in Chinese goslings, causing at most 50% of gosling deaths. By December 2022, the disease had become epidemic and prevailed in goose farms in Jiangsu, Shandong, Anhui, Henan, Guangdong, Liaoning, Sichuan and other places in China. The disease mainly affects goslings within 3 weeks old. The typical symptoms of goose astrovirus are large deposits of urate in the viscera, joint cavity and ureter surface of infected goslings. Goose astrovirus infection can trigger high levels of iNOS, limiting goose astrovirus replication. The ORF2 domain P2 of the goose astrovirus activates the OASL protein, limiting its replication. Goose astrovirus can also activate pattern recognition receptors (RIG-I, MDA-5, TLR-3), causing an increase in MHC-Ia, MHC-Ib and CD81 mRNA, activating humoral and cellular immunity, thereby hindering virus invasion. Goose astrovirus also regulates the activation of IFNs and other antiviral proteins (Mx1, IFITM3, and PKR) in the spleens and kidneys to inhibit viral replication. The innate immune response process in goslings also activates TGF-ß, which may be closely related to the immune escape of goose astrovirus. Gaining insight into the infection and innate immune mechanism of goose astrovirus can help researchers study and prevent the severe disease in goslings better.

An efficient and practical method to construct benzimidazoles via Rh(III)-catalyzed sequential C-H amination and annulation cascade reaction has been developed. The cascade reaction displays high step, atom, and redox economy, is compatible with the air, and has good functional group tolerance and high efficiency. The titled products can be easily further converted into imidazo[4,5-c]acridines, which were observed unique fluorescent properties.

An efficient protocol to synthesize substituted benzo[4,5]imidazo[1,2- c]quinazolines starting from N-LG-2-phenylbenzoimidazole and dioxazolones catalyzed by Rh(III) or Ir(III) has been developed. Various substituted benzo[4,5]imidazo[1,2- c]quinazolines could be easily provided in up to 99% yield. A large range of substrates and functional groups are compatible for this transformation. This method features low catalyst loading, acid-free conditions, and low solvent consumption.

An efficient tandem reaction of imidamides and N-hydroxycarbamates has been developed. Valuable 2-alkylbenzimidazoles could be easily obtained in up to 97% yield for more than 20 examples. The products would further streamline the synthesis of molecules, which are essential building blocks in organic synthesis and drug discovery. This protocol features high regioselectivity, efficiency, good tolerance of functional groups, and mild reaction conditions.

RESUMO

A polarization-insensitive and angle-independent graphene absorber (GA) with periodically patterned grating is demonstrated. A periodic nanocavity composed of multilayer subwavelength grating and metal substrate supports a strongly localized mode inside the cavity, where the mode helps to absorb more electromagnetic waves. The proposed GA exhibits polarization-insensitive behavior and maintains the high absorption above 90% within a wide range of incident angle (more than 80°). We attribute the high absorption to the excitation of the cavity mode resonance and magnetic resonance for the transverse electric and transverse magnetic polarizations, respectively. The proposed GA has potential applications in the design of various devices, such as optical modulators or tunable absorption filters because of its remarkable angle-insensitive absorption performance.

A multiline absorber based on the excitation of guided-mode resonance of one-dimensional photonic crystals (1D-PhCs), including a surface graphene monolayer under the attenuated-total-reflection configuration, is proposed and demonstrated. By carefully designing the structure parameters of the 1D-PhCs, the guided mode can be modulated by the periodic distribution of the refractive index. Our results reveal that the critical coupling of the guided resonance in periodical PhCs to graphene produces the perfect absorption. The number of absorption peaks within the photonic band corresponds to the number of unit cells. An ultrahigh Q-factor value of 4.75×106 is obtained at resonance with unity absorption, which could serve as a promising replacement of metallic thin film as a sensor probe for future biosensing applications.

In this work, we show that the application of a sol-gel coating renders a microbubble whispering gallery resonator into an active device. During the fabrication of the resonator, a thin layer of erbium-doped sol-gel is applied to a tapered microcapillary, then a microbubble with a wall thickness of 1.3 µm is formed with the rare earth ions diffused into its wall. The doped microbubble is pumped at 980 nm and lases in the emission band of the Er3+ ions at 1535 nm. The laser wavelength can be shifted by aerostatic pressure tuning of the whispering gallery modes of the microbubble. Up to 240 pm tuning is observed with 2 bar of applied pressure. We also show that the doped microbubble could be used as a compact, tunable laser source.

We report Raman lasing and the optical analog of electromagnetically-induced-transparency (EIT) in a whispering-gallery-mode (WGM) microtoroid resonator embedded in a low refractive index polymer matrix together with a tapered fiber coupler. The microtoroid resonator supports both single mode and multimode Raman lasing with low power thresholds. Observations of Fano and EIT-like phenomena in a packaged microresonator will enable high resolution sensors and can be used in networks where slow-light effect is needed. These results will open up new possibilities for portable, robust, and stable WGM microlasers and resonator-based sensors for applications in various environments.

Frequency comb generation in microresonators at visible wavelengths has found applications in a variety of areas such as metrology, sensing, and imaging. To achieve Kerr combs based on four-wave mixing in a microresonator, dispersion must be in the anomalous regime. In this Letter, we demonstrate dispersion engineering in a microbubble resonator (MBR) fabricated by a two-CO2 laser beam technique. By decreasing the wall thickness of the MBR to 1.4 µm, the zero dispersion wavelength shifts to values shorter than 764 nm, making phase matching possible around 765 nm. With the optical Q-factor of the MBR modes being greater than 107, four-wave mixing is observed at 765 nm for a pump power of 3 mW. By increasing the pump power, parametric oscillation is achieved, and a frequency comb with 14 comb lines is generated at visible wavelengths.

We have experimentally demonstrated an on-chip all-silk fibroin whispering gallery mode microresonator by using a simple molding and solution-casting technique. The quality factors of the fabricated silk protein microresonators are on the order of 105. A high-sensitivity thermal sensor was realized in this silk fibroin microtoroid with a sensitivity of -1.17 nm/K, that is 8 times higher than previous WGM resonator-based thermal sensors. This opens the way to fabricate biodegradable and biocompatible protein based microresonators on a flexible chip for biophotonics applications.

Ir-catalyzed direct C-H sulfamidation of benzaldehydes has been achieved. A series of ortho-amided benzaldehydes were obtained in up to 95% yields for 21 examples with excellent regioselectivity and broad functional group tolerance. This transformation could proceed smoothly with low catalyst loading under external-oxidant-, acid-, or base-free conditions. Molecular nitrogen was released as the sole byproduct, providing an environmentally benign sulfamidation process.

One method for enhancement and manipulation of light absorption with monolayer graphene covered on a single-layer guided mode resonant Brewster filter surface is demonstrated. By means of the rigorous coupled-wave analysis method, the effect of geometrical parameters on the optical response of the structure is investigated. It is possible to achieve a maximum absorption of 60% at the Brewster angle; dual-band optical absorption can also be realized when the depth of the grating is increased. The situation of oblique incidence for TM polarization is studied as well; the absorption property can be controlled by adjusting the incident angle without changing the structural parameters. The proposed structure has the advantage of more free geometry parameters compared to the graphene disk and ribbon, so the absorption could be tuned more flexibly.

A waveguide-coupled surface phonon resonance (SPhR) sensor with super-resolution based on Fano resonance (FR) by using a multilayer system within the Kretschmann configuration in the mid-infrared wavelength region is proposed. Due to the coherent interference of the waveguide and the surface phonon polariton modes, the calculated reflectivity spectrum possesses sharp asymmetric FR dips. An ultra-small linewidth is formed because of the Fano coupling, and the physical features contribute to a highly efficient nano-sensor for refractive index sensing. The bulk and surface sensitivity by intensities are greatly enhanced relative to those of conventional SPhR sensors.

RESUMO

A new C3-symmetric Schiff-base fluorescent probe (L) based on 8-hydroxy-2-methylquinoline has been developed. As expected, the probe L can display high fluorescent selectivity for Cd(2+) over Zn(2+) and most other common ions in neutral ethanol aqueous medium. Moreover, the mechanism of the L-Cd(2+) complex has been confirmed by X-ray crystallography and density functional theory calculation results. More importantly, L could be used to image Cd(2+) within living cells.

We demonstrate the design of an efficient angle-insensitive guided mode resonance filter (GMRF), with narrow bandwidth and low sideband reflection, for TE-polarized waves. The reflection properties of the multilayer structure have been studied, and the results verify that the thin film design of the gradient-index layer is important for the realization of an angle-insensitive filter. Various gradient coefficients of the thin film have distinct effects on the reflection spectrum. For an increasing incident angle, although the line-shape symmetry becomes less perfect, the positions of the resonant peak remain the same. The GMRF proposed here has many desirable attributes that lends itself to being an excellent platform, for devices such as lasers, detectors, filters, and sensors.

The insulin receptor (IR) recruits adaptor proteins, so-called insulin receptor substrates (IRS), to connect with downstream signalling pathways. A family of IRS proteins was defined based on three major common structural elements: Amino-terminal PH and PTB domains that mediate protein-lipid or protein-protein interactions, mostly carboxy-terminal multiple tyrosine residues that serve as binding sites for proteins that contain one or more SH2 domains and serine/threonine-rich regions which may be recognized by negative regulators of insulin action. The current model for the role of IRS proteins therefore combines an adaptor function with the integration of mostly negative input from other signal transduction cascades allowing for modulation of signalling amplitude. In this review we propose an extended version of the adaptor model that can explain how signalling specificity could be implemented at the level of IRS proteins.
