RESUMO

PURPOSE: Melanoma is widely utilized as a prominent model for the development of immunotherapy, thought an inadequate immune response can occur. Moreover, the development of apoptosis-related therapies and combinations with other therapeutic strategies is impeded by the limited understanding of apoptosis's role within diverse tumor immune microenvironments (TMEs). METHODS: Here, we constructed an apoptosis-related tumor microenvironment signature (ATM) and employ multi-dimensional analysis to understand the roles of apoptosis in tumor microenvironment. We further assessed the clinical applications of ATM in nine independent cohorts, and anticipated the impact of ATM on cellular drug response in cultured cells. RESULTS: Our ATM model exhibits robust performance in survival prediction in multiple melanoma cohorts. Different ATM groups exhibited distinct molecular signatures and biological processes. The low ATM group exhibited significant enrichment in B cell activation-related pathways. What's more, plasma cells showed the lowest ATM score, highlighting their role as pivotal contributors in the ATM model. Mechanistically, the analysis of the interplay between plasma cells and other immune cells elucidated their crucial role in orchestrating an effective anti-tumor immune response. Significantly, the ATM signature exhibited associations with therapeutic efficacy of immune checkpoint blockade and the drug sensitivity of various agents, including FDA-approved and clinically utilized drugs targeting the VEGF signaling pathway. Finally, ATM was associated with tertiary lymphoid structures (TLS), exhibiting stronger patient stratification ability compared to classical "hot tumors". CONCLUSION: Our findings indicate that ATM is a prognostic factor and is associated with the immune response and drug sensitivity in melanoma.

Keloids are benign fibroproliferative tumors that severely diminish the quality of life due to discomfort, dysfunction, and disfigurement. Recently, ultrasound technology as a noninvasive adjuvant therapy is developed to optimize treatment protocols. However, the biophysical mechanisms have not yet been fully elucidated. Here, it is proposed that piezo-type mechanosensitive ion channel component 1 (Piezo1) plays an important role in low-frequency sonophoresis (LFS) induced mechanical transduction pathways that trigger downstream cellular signaling processes. It is demonstrated that patient-derived primary keloid fibroblasts (PKF), NIH 3T3, and HFF-1 cell migration are inhibited, and PKF apoptosis is significantly increased by LFS stimulation. And the effects of LFS is diminished by the application of GsMTx-4, the selective inhibitor of Piezo1, and the knockdown of Piezo1. More importantly, the effects of LFS can be imitated by Yoda1, an agonist of Piezo1 channels. Establishing a patient-derived xenograft keloid implantation mouse model further verified these results, as LFS significantly decreased the volume and weight of the keloids. Moreover, blocking the Piezo1 channel impaired the effectiveness of LFS treatment. These results suggest that LFS inhibits the malignant characteristics of keloids by activating the Piezo1 channel, thus providing a theoretical basis for improving the clinical treatment of keloids.

BACKGROUND: Plantar warts are common infectious cutaneous growths causing severe physiological and psychological discomforts in patients and heaving global financial burdens. However, paucity of clear-cut guidelines for plantar warts, selecting appropriate treatments for plantar warts remains challenging. The objective of the study is to evaluate the efficacy and safety of common treatments for plantar warts. METHODS: PubMed, EMbase, and The Cochrane Library were searched from inception to March 1, 2023 for randomized controlled trials (RCTs) of plantar warts. The primary outcome (complete response) and secondary outcome (recurrence and pain) were extracted and combined using Bayesian network meta-analysis (NMA) with random-effect and fixed-effect models. RESULTS: Totally, 33 RCTs were included in the systematic review and quantitative NMA. In NMA of complete response, topical application of 1% cantharidin, 20% podophylotoxin, 30% salicylic acid (CPS), microneedles plus bleomycin (MNB), and intralesional bleomycin injection (INB) were the only three treatments significantly superior to no treatment (NT) and CPS was of the highest possibility to be the top-ranked treatment (SUCRA = 0.9363). However, traditional warts treatments, salicylic acid (SA) and cryotherapy were not superior to NT. CONCLUSIONS: The NMA has produced evidence for using CPS, MNB, and INB, which are all topical antimitotic treatments, to improve the management of plantar warts. The classic treatment modalities for plantar warts, including SA and cryotherapy, may play a less important role in the clinical practice of plantar warts.

Given the increasingly prominent contradiction between the supply of and demand for wood, the abundant resource of bamboo can be a good substitute. Bamboo scrimber can effectively improve the utilization rate of bamboo and has good mechanical properties. However, bamboo scrimber has the problem of poor mildew resistance, and does not meet the requirements for outdoor applications. In this study, in order to further improve the mildew resistance and mechanical properties of bamboo scrimber, alkali treatment was used to remove some nutrients from the bamboo bundles and change the pH of the bamboo scrimber. The results showed that nutrients such as hemicellulose, lignin, starch, and sugar were notably removed from bamboo bundles, and the pH of bamboo was slightly alkaline. The anti-mildew effect was significantly enhanced, which could allow use in outdoor environments, and the mechanical properties and dimensional stability were also improved. Among them, TB6 bamboo scrimber showed comprehensively excellent properties. The infection time in the laboratory mildew test increased from 3 days to more than 30 days, and the infection time in the outdoor mildew resistance test increased from 1 week to more than 8 weeks; the static bending intensity of TB6 increased by 62.6% to 150 MPa, and the bending modulus increased by 71.7% to 14.2 GPa; the change rate of water absorption thickness was reduced to 0.58%. This modification method effectively improved the mildew resistance of bamboo scrimber, while maintaining high mechanical strength, and provides a new method for the outdoor application of bamboo scrimber.

Mid-infrared fiber combiners have great potential in power and spectral combination. However, studies on mid-infrared transmission optical field distributions using these combiners are limited. In this study, we designed and fabricated a 7 × 1 multimode fiber combiner based on sulfur-based glass fibers and observed approximately 80% per-port transmission efficiency at 4.778 µm wavelength. We investigated the propagation properties of the prepared combiners and explored the effects of transmission wavelength, output fiber length, and fusion deviation on the transmitted optical field and beam quality factor M2. Additionally, we assessed the effect of coupling on the excitation mode and spectral combination of the mid-infrared fiber combiner for multiple light sources. Our results provide an in-depth understanding of the propagation properties of the mid-infrared multimode fiber combiners, which may find applications in high-beam-quality laser devices.

Chalcogenide hollow-core anti-resonant fibers (HC-ARFs) are a promising propagation medium for high-power mid-infrared (3-5 µm) laser delivery, while their properties have not been well understood and their fabrications remain challenging. In this paper, we design a seven-hole chalcogenide HC-ARF with touching cladding capillaries, which was then fabricated from purified As40S60 glass by combining the "stack-and-draw" method with a dual gas path pressure control technique. In particular, we predict theoretically and confirm experimentally that such medium exhibits higher-order mode suppression properties and several low-loss transmission bands in the mid-infrared spectrum, with the measured fiber loss being as low as 1.29 dB/m at 4.79 µm. Our results pave the way for the fabrication and implication of various chalcogenide HC-ARFs in mid-infrared laser delivery systems.

Background: Transient receptor potential vanilloid (TRPV) is one of the transient receptor potential protein groups; cardiovascular system disease is a crucial cause of mortality among people globally. Objective: This article is intended to accomplish a bibliometric analysis of the trends and public interest since TRPV was reported for the first time. Methods: The article summarized the Web of Science (WOS) Core Collection on the relationship between TRPV and cardiovascular system disease each year from 2000 to 2021. Data extraction and visualization were completed by R package bibliometrix. Keyword citation burst and co-citation networks were generated and produced by CiteSpace. The map evaluating the distribution of country and region was painted in GunnMap 2 (lert.co.nz). The ranking was performed using the Standard Competition Ranking method. Co-authorship and co-occurrence were analyzed with VOSviewer. Results: After removing duplicated data, books, conference proceedings, and articles of uncertain age, 493 were included, and 17 were excluded. The pattern of publication years showed that the number of publications increased rapidly from 2008 to 2021 with no peak in the number of publications until 2021. The geographical distribution pattern revealed a considerable gap in the number of publications between the United States, China, and other countries, with East Asian institutions leading the world in this area. The pattern of co-authorship showed that 77 institutions were divided into 19 clusters, each covering one country or region.These results suggest that intercontinental cooperation among institutions should be strengthened. The core authors section displayed the change in the most published authors. Keyword analysis listed six burst keywords. Co-citation analysis of references from 2011 to 2021 showed the number and centrality of citations to leading articles. Conclusion: Our findings reveal trends and public interest in transient receptor potential vanilloid for cardiovascular disease. These findings suggest that the field has experienced significant growth since 2008, with the United States and China in dominant positions. Our findings also suggest that intercontinental cooperation should be strengthened, and that future research hotspots may focus on pharmacological mechanisms and in-depth exploration of drug clinical trials and new clinical disease application areas such as hypertension, diabetes, and cardiac arrhythmias, which could serve as a foundation for further research.

BACKGROUND: Photodynamic therapy (PDT) has been widely accepted in keratocyte carcinoma and an increasing number of literatures concerning PDT in skin cancer is published. But a detailed examination of publication patterns of PDT in skin cancer has not yet been carried out. METHODS: Bibliographies were retrieved from Web of Science Core Collection restricted the publication date from January 1, 1985 to December 31, 2021. The search terms were photodynamic therapy and skin cancer. Visualization analysis and statistical analysis were performed by VOSviewer (Version 1.6.13), R software (Version 4.1.2) and Scimago Graphica (Version 1.0.15). RESULTS: 3248 documents were selected for analysis. The results showed that the number of annual publications related to PDT in skin cancer was gradually increased and would continue to increase in the future. The results illustrated that "melanoma", "nanoparticles", "drug-delivery", "mechanism", "delivery" and "in-vitro" are newly occurred topics. The most prolific country was the United States and the most productive institution was the University of Sao Paulo in Brazil. German researcher Szeimies RM published the most papers related to PDT in skin cancer. British Journal of Dermatology was the most popular journal in this field. CONCLUSION: The topic that PDT in skin cancer is a heated issue. Our study revealed the bibliometric result of the field, which might provide the prospects for further research. We recommend future investigations focusing on PDT in treating melanoma, innovation of photosensitizer, improvement of drug delivery and the mechanism of PDT in skin cancer.

BACKGROUND: Melanoma is one of the most life-threatening skin cancers; immune checkpoint blockade is widely used in the treatment of melanoma because of its remarkable efficacy. OBJECTIVE: This study aimed to conduct a comprehensive bibliometric analysis of research conducted in recent decades on immune checkpoint blockade for melanoma, while exploring research trends and public interest in this topic. METHODS: We summarized the articles in the Web of Science Core Collection on immune checkpoint blockade for melanoma in each year from 1999 to 2020. The R package bibliometrix was used for data extraction and visualization of the distribution of publication year and the top 10 core authors. Keyword citation burst analysis and cocitation networks were calculated with CiteSpace. A Gunn online world map was used to evaluate distribution by country and region. Ranking was performed using the Standard Competition Ranking method. Coauthorship analysis and co-occurrence were analyzed and visualized with VOSviewer. RESULTS: After removing duplicates, a total of 9169 publications were included. The distribution of publications by year showed that the number of publications rose sharply from 2015 onwards and either reached a peak in 2020 or has yet to reach a peak. The geographical distribution indicated that there was a large gap between the number of publications in the United States and other countries. The coauthorship analysis showed that the 149 top institutions were grouped into 8 clusters, each covering approximately a single country, suggesting that international cooperation among institutions should be strengthened. The core author extraction revealed changes in the most prolific authors. The keyword analysis revealed clustering and top citation bursts. The cocitation analysis of references from 2010 to 2020 revealed the number of citations and the centrality of the top articles. CONCLUSIONS: This study revealed trends in research and public interest in immune checkpoint blockade for melanoma. Our findings suggest that the field is growing rapidly, has several core authors, and that the United States is taking the lead position. Moreover, cooperation between countries should be strengthened, and future research hot spots might focus on deeper exploration of drug mechanisms, prediction of treatment efficacy, prediction of adverse events, and new modes of administration, such as combination therapy, which may pave the way for further research.

Polydopamine (PDA)-modified NaEr0.8Yb0.2 F4nanoparticles were synthesized, with strong NIR-II emission, quantum yield of 29.63%, and excellent photothermal performance. Crystal phases and microstructures are characterized. Optical properties such as absorption, NIR-II emission, and light stability are studied, and the luminescence mechanism is discussed in detail. Key factors in NIR-II imaging were evaluated in fresh pork tissue, including penetration depth, spatial resolution, and signal-to-noise ratio (SNR). A high penetration depth of 5 mm and a high spatial resolution of 1 mm were detected. Mice are imaged in vivo afterintravenousinjection. Due to the accumulation of nanoparticles in the liver, high image quality with an SNR of 5.2 was detected in the abdomen of KM mice with hair. The photothermal conversion effect of PDA-modified NPs was twice that of the reported material. These NIR-II nanoparticles have superior optical properties, high photothermal efficiency and low cytotoxicity, and are potential fluorescent probes for further disease diagnosis and treatment.

Background: Macrolides have been widely used to treat moderate-to-severe acne for more than 50 years. However, the prevalent antibiotic resistance of Propionibacterium acnes, along with the absence of clinically available resistance tests, has made macrolide misuse a frequent occurrence around the globe, with serious consequences. Objective: We developed Cutibacterium acnes quantitative PCR (qPCR)-based antibiotics resistance assay (ACQUIRE) to enable fast and accurate detection of C. acnes macrolide resistance in clinical settings, representing an opportunity to administer antibiotics more wisely and improve the quality of care. Methods: A cross-sectional observational study (n = 915) was conducted to probe into the macrolide resistance of C. acnes in patients with acne. Results: The high sensitivity of ACQUIRE enabled us to reveal a much higher C. acnes 23S recombinant DNA (rDNA) point mutation rate (52%) and thus a higher macrolide resistance (75.5%) compared to previous reports. Carriage of ermX gene was discovered on 472 (53%) subjects, which concurs with previous studies. Conclusion: The macrolide resistance of C. acnes is much higher than previously reported. Integrating ACQUIRE into acne treatment modalities may eliminate macrolide misuse and achieve better clinical improvements.

Cymbidium iridioides D. Don 1852 is a Class I endangered species in China having important ornamental and breeding values. This study used Illumina high-throughput sequencing technology to sequence the complete chloroplast genome of C. iridioides. The genome features of C. iridioides and its phylogenetic relationships were determined. The complete chloroplast genome is 156,599 bp, containing a pair of 26,736 bp inverse duplication regions, a large 85,242 bp single-copy region, and a small 17,885 bp single-copy region. The entire genome contains 76 protein-coding genes, 37 tRNA genes, and 4 rRNA genes. A phylogenetic tree of 25 Orchidaceae species revealed that C. iridioides was grouped within Sect. Iridorchis and comprised a clade with Cymbidium tracyanum.

Since the 1950s, gradual changes in the gut microbiota of patients with hepatic encephalopathy have been observed. Previous research has indicated potential associations between the gut and brain, and the gut microbiota is becoming a hot topic in research on diseases of the nervous system. However, for the past few decades, studies of hepatic encephalopathy have been restricted to controlling the gut microbiota during macroscopic manipulation, such as probiotic intervention, while its clinical use remains controversial, and the cellular mechanisms underlying this condition are still poorly understood. This thesis seeks to comprehensively understand and explain the role of gut microbiota in hepatic encephalopathy as well as analyze the effects of intervention by regulating the gut microbiota. Evidence is presented that shows that dysbiosis of the gut microbiota is the primary pathological driver of hepatic encephalopathy and impacts pathologic progression via complex regulatory networks. As a result, suggestions were identified for future mechanistic research and improvements in therapeutic strategies for hepatic encephalopathy.

OBJECTIVES: We sought to systematically investigate the effectiveness of secukinumab in psoriatic arthritis (PsA) patients who previously received TNFs inhibitor (TNFi) treatment and those who were TNFi naïve. METHODS: Databases (PubMed, EMBase and Cochrane library) and ClinicalTrials.gov were searched from inception to 22 May 2020 for randomized control trails and observational studies of secukinumab, with or without a history of previous anti-TNFi treatment, in PsA. Effectiveness data were extracted and combined using a random-effects meta-analysis. The ACR20 and ACR50 (20% and 50% improvement in American College of Rheumatology response criteria) responses were the endpoints. RESULTS: Six randomized controlled trials that reported the effectiveness of secukinumab by previous anti-TNFi treatment were included. Among patients exposed to a prior anti-TNFi treatment (n = 738), 33.7% (249/738) of patients achieved an ACR20 response. In contrast, in the anti-TNFi-naïve group (n = 1754), 49.8% (873/1754) of patients achieved an ACR20 response. Prior treatment with anti-TNFi was significantly associated with a poorer response to secukinumab compared with the anti-TNFi-naïve group with an effect size of 2.09 (95% CI: 1.69, 2.58). CONCLUSION: Some patients benefit from switching from TNFi to secukinumab, but previous anti-TNFi treatment is associated with poorer effectiveness of secukinumab.

Rare earth-doped nanoparticles have been widely used in disease diagnosis, drug delivery, tumor therapy, and bioimaging. Among various bioimaging methods, the fluorescence imaging technology based on the rare earth-doped nanoparticles can visually display the cell activity and lesion evolution in living animals, which is a powerful tool in biological technology and has being widely applied in medical and biological fields. Especially in the band of near infrared (700-1700 nm), the emissions show the characteristics of deep penetration due to low absorption, low photon scattering, and low autofluorescence interference. Furthermore, the rare earth-doped nanoparticles can be endowed with the water solubility, biocompatibility, drug-loading ability, and the targeting ability for different tumors by surface functionalization. This confirms its potential in the cancer diagnosis and treatment. In this review, we summarized the recent progress in the application of rare earth-doped nanoparticles in the field of bioimaging and tumor treatment. The luminescent mechanism, properties, and structure design were also discussed.

Water-soluble K5 HoLi2 F10 (KHLF) nanoprobes with the excitation and emission both in the near-infrared (NIR) region were developed and first demonstrated for in vivo imaging of living mice. The PEG400 coating endows the nanoprobes with good water solubility and biocompatibility. Doping with Ho3+ ions is capable of emitting NIR fluorescence with two peaks centered, respectively, at 887 and 1,180 nm once excited by a 808 nm laser; meanwhile, it also possess good photothermal conversion performance. The KHLF matrix with specifically structure of large ion-distance and low photon energy imparts the nanoprobes low quenching effect and excellent photostability (fluorescence decrease <5% upon 120 min illumination of 808 nm continuous laser with a power density of 1 W/cm2 ). The nanoparticles (NPs) were tested for in vitro bioimaging with living mice. The results show the NPs have low biotoxicity, rapid metabolism, normal biodistribution, together with the photothermal imaging performance and a high-contrast fluorescence images (signal-to-background ratio of 14:1). The superior performances of these nanoprobes in vivo imaging of mice proclaim the great potential of this type of probe for high-contrast imaging and photothermal treatment in practical applications.

Bacillus is a common bacterial genus in Chinese traditional sun-brewed moromi. Some Bacillus species can metabolize sauce flavour and aroma substances. To address the insufficiency of flavour microbes in soy sauce fermentation, we used 16S rDNA sequence analysis to identify the CS1.03 strain, which was isolated from moromi produced through the traditional and natural sun brewing methods. A Box-Behnken design was used to optimize the addition technology of CS1.03, and ammoniacal nitrogen content and sensory evaluation were adopted as determination indices. GC-MS analysis was also performed to analyse the flavour components. Results identified CS1.03 as Bacillus subtilis. The optimal conditions yielded 10.7 g/L ammoniacal nitrogen in fermented soy sauce with rich aroma and flavour, and a sensory evaluation score of 90.5. GC-MS analysis revealed that the contents of phenol, aldehyde and ketone are all increased. These results demonstrated that CS1.03 could improve the flavour of soy sauce and shows good potential for application in soy sauce fermentation.

In this study, bromelain was used to break soy protein molecules into polypeptide chains, and triglycidylamine (TGA) was added to develop a bio-adhesive. The viscosity, residual rate, functional groups, thermal behavior, and fracture surface of different adhesives were measured. A three-ply plywood was fabricated and evaluated. The results showed that using 0.1 wt% bromelain improved the soy protein isolate (SPI) content of the adhesive from 12 wt% to 18 wt%, with viscosity remaining constant, but reduced the residual rate by 9.6% and the wet shear strength of the resultant plywood by 69.8%. After the addition of 9 wt% TGA, the residual rate of the SPI/bromelain/TGA adhesive improved by 13.7%, and the wet shear strength of the resultant plywood increased by 681.3% relative to that of the SPI/bromelain adhesive. The wet shear strength was 30.2% higher than that of the SPI/TGA adhesive, which was attributed to the breakage of protein molecules into polypeptide chains. This occurrence led to (1) the formation of more interlocks with the wood surface during the curing process of the adhesive and (2) the exposure and reaction of more hydrophilic groups with TGA to produce a denser cross-linked network in the adhesive. This denser network exhibited enhanced thermal stability and created a ductile fracture surface after the enzymatic hydrolysis process.

The purpose of this study was to improve the performance of soy protein isolate (SPI) adhesives using a polyurethane elastomer. Triglycidylamine (TGA), SPI, thermoplastic polyurethane elastomer (TPU), and γ-(2,3-epoxypropoxy) propyltrimethoxysilane (KH-560) were used to develop a novel SPI-based adhesive. The residual rate, functional groups, thermal stability, and fracture surface micrographs of the cured adhesives were characterized. Three-ply plywood was fabricated, and the dry/wet shear strength was determined. The experimental results suggested that introducing 2% TGA improved the residual rate of the SPI/TGA adhesive by 4.1% because of the chemical cross-linking reaction between epoxy groups and protein molecules. Incorporating 7% TPU into the SPI/TGA adhesive, the residual rate of the adhesive increased by 5.2% and the dry/wet shear strength of plywood bonded by SPI/TGA/TPU adhesive increased by 10.7%/67.7%, respectively, compared with that of SPI/TGA adhesive. When using KH-560 and TPU together, the residual rate of the adhesive improved by 0.9% compared with that of SPI/TGA/TPU adhesive. The dry and wet shear strength of the plywood bonded by the SPI/TGA/TPU/KG-560 adhesive further increased by 23.2% and 23.6% respectively when compared with that of SPI/TGA/TPU adhesive. TPU physically combined with the SPI/TGA adhesive to form a interpenetration network and KH-560 acted as a bridge to connect TPU and SPI/TGA to form a joined crosslinking network, which improved the thermo stability/toughness of the adhesive and created a uniform ductile fracture section of the adhesive.

Chalcogenide glasses as kind of diamagnetic magneto-optical materials have promising applications in the field of integrated optics and optical communication systems due to their excellent properties, such as easy to be processed into waveguide and temperature independence of the Verdet constants. For clarifying the influence factors following the compositional variation on Faraday effect and finding a glass with a large Verdet constant, novel pseudo-ternary chalcogenide glass system, GeS2 - In2S3 - PbI2, was prepared and investigated. The composition, wavelength and temperature dependences on the Verdet constants were systematically investigated at the wavelengths of 635, 808, 980 and 1319 nm. PbI2 was confirmed to have positive contribution to the Verdet constant and the Becquerel rule was proved to be an effective guidance for predicting the Verdet constant in chalcogenide glasses. The 60GeS2·15In2S3·25PbI2 glass was found to possess the largest Verdet constant (V = 0.215 min·G-1·cm-1, @808nm), which is great larger than that of commercial diamagnetic glasses. These glasses also possess good glass-forming ability and VIS-IR transmittance, therefore be a good candidate for next-generation integrated optical isolator and other magneto-optical devices.
