We designed a cascaded all-soft-glass fiber structure and simulate midinfrared 2-20 µm ultrawideband supercontinuum (SC) generation numerically. The cascaded fiber structure consists of a 1.5 m I n F 3 fiber, a 0.2 m chalcogenide photonic crystal fiber, and a 0.2 m tellurium-based chalcogenide photonic crystal fiber. Using a 2 µm pulse pumping this cascaded structure, the generated SC covering the wavelengths longer than 20 µm has been demonstrated theoretically. The 30 dB bandwidth reaches 20.87 µm from 1.44 to 22.31 µm. The effect of different pulse widths on SC generation is considered. With the increase of peak power and the decrease of pulse width, the energy of SC in the 15-20 µm waveband increases gradually. The mechanism of SC broadening process has also been analyzed. The SC generation of more than 20 µm in this cascade structure is caused by the self-phase modulation, soliton effects, four-wave mixing, and redshifted dispersive wave. This method demonstrates the possibility of generating ultrawide bandwidth SCs up to a 20 µm waveband by a commercial 2 µm pump source and all-fiber structure.

Leaf angle is a major trait of ideal architecture, which is considered to influence rice (Oryza sativa) cultivation and grain yield. Although a few mutants with altered rice leaf inclination angle have been reported, the underlying molecular mechanism remains unclear. In this study, we showed that a WRKY transcription factor gene, OsWRKY72, was highly expressed in the leaf sheath and lamina joint. Phenotypic analyses showed that oswrky72 mutants have smaller leaf angles than the wild type (WT), while OsWRKY72 overexpression lines exhibited increased leaf angle. This observation suggests that OsWRKY72 functions as a positive regulator, promoting the enlargement of the leaf angle. Our bioinformatics analysis identified LAZY1 as the downstream gene of OsWRKY72. Electrophoretic mobility shift assays (EMSA) and dual luciferase analysis revealed that OsWRKY72 directly inhibits LAZY1 by binding to its promoter. Moreover, knocking out OsWRKY72 enhanced shoot gravitropism, which is in contrast to the phenotype of lazy1 plants. These results imply that OsWRKY72 regulates leaf angle through gravitropism by reducing the expression of LAZY1. In addition, OsWRKY72 could directly regulate the expression of other leaf angle-related genes, such as FLOWERING LOCUS T-LIKE 12 (OsFTL12) and WALL-ASSOCIATED KINASE 11 (OsWAK11). Our study indicates that OsWRKY72 contributes positively to the expansion of the leaf angle by interfering with shoot gravitropism in rice.

Detailed and accurate urban landscape mapping, especially for urban blue-green-gray (UBGG) continuum, is the fundamental first step to understanding human-nature coupled urban systems. Nevertheless, the intricate spatial heterogeneity of urban landscapes within cities and across urban agglomerations presents challenges for large-scale and fine-grained mapping. In this study, we generated a 3 m high-resolution UBGG landscape dataset (UBGG-3m) for 36 Chinese metropolises using a transferable multi-scale high-resolution convolutional neural network and 336 Planet images. To train the network for generalization, we also created a large-volume UBGG landscape sample dataset (UBGGset) covering 2,272 km2 of urban landscape samples at 3 m resolution. The classification results for five cities across diverse geographic regions substantiate the superior accuracy of UBGG-3m in both visual interpretation and quantitative evaluation (with an overall accuracy of 91.2% and FWIoU of 83.9%). Comparative analyses with existing datasets underscore the UBGG-3m's great capability to depict urban landscape heterogeneity, providing a wealth of new data and valuable insights into the complex and dynamic urban environments in Chinese metropolises.

It has been well established that cancer cells can evade immune surveillance by mutating themselves. Understanding genetic alterations in cancer cells that contribute to immune regulation could lead to better immunotherapy patient stratification and identification of novel immune-oncology (IO) targets. In this report, we describe our effort of genome-wide association analyses across 22 TCGA cancer types to explore the associations between genetic alterations in cancer cells and 74 immune traits. Results showed that the tumor microenvironment (TME) is shaped by different gene mutations in different cancer types. Out of the key genes that drive multiple immune traits, top hit KEAP1 in lung adenocarcinoma (LUAD) was selected for validation. It was found that KEAP1 mutations can explain more than 10% of the variance for multiple immune traits in LUAD. Using public scRNA-seq data, further analysis confirmed that KEAP1 mutations activate the NRF2 pathway and promote a suppressive TME. The activation of the NRF2 pathway is negatively correlated with lower T cell infiltration and higher T cell exhaustion. Meanwhile, several immune check point genes, such as CD274 (PD-L1), are highly expressed in NRF2-activated cancer cells. By integrating multiple RNA-seq data, a NRF2 gene signature was curated, which predicts anti-PD1 therapy response better than CD274 gene alone in a mixed cohort of different subtypes of non-small cell lung cancer (NSCLC) including LUAD, highlighting the important role of KEAP1-NRF2 axis in shaping the TME in NSCLC. Finally, a list of overexpressed ligands in NRF2 pathway activated cancer cells were identified and could potentially be targeted for TME remodeling in LUAD.

The energy exchange between orthogonal polarization components is crucial for the build-up of vector solitons (VSs). Unlike previous observations of energy exchange in the frequency domain, our experiments analyzed pulse energy flows in the time domain. We provide evidence to demonstrate the influence of the four-wave mixing (FWM) and cross-phase modulation (XPM) effect on VSs build-up in passive mode-locked fiber lasers through a perspective of pulse energy exchange. The results indicate that the energy exchange of PRVS caused by FWM and XPM is stronger than that of PLVS. The liner energy exchange caused by the birefringence of fiber and PC influences the period of energy exchange. After stabilization, the intra-cavity energy evolution period is one roundtrip for PLVS while serval roundtrips for PRVS. In the future, we can achieve PLVS by adjusting the linear energy exchange through cavity birefringence, thereby meeting the industrial demand for stable and uniform pulse trains.

Effective treatment of sewage requires accurate measurement of important water quality parameters, such as chemical oxygen demand (COD), pH value, total nitrogen (TN), total phosphorus (TP), and ammonia nitrogen (NH3-N). Traditional detection techniques can result in secondary contamination and are time- and labor-intensive. Near infrared spectroscopy was used in this study to create a model of these parameters of pig manure anaerobic fermentation sewage. The models' viability for quickly estimating the aforementioned water quality characteristics was reviewed, and the models' performance in predicting the results of several samples (biogas slurry, supernatant, and biogas residue) was contrasted. By analyzing the near infrared spectrograms with a spectral range of 4000 cm-1 and 12 500 cm-1 and using second derivative (SD), Savitzky-Golay smoothing (SG) and standard normal variable (SNV) to preprocess the spectra, partial least squares (PLS) was selected to establish the prediction model. The results showed that the effect of the NIR model constructed from the supernatant was better than that of biogas slurry and biogas residue. The determination coefficients for COD, pH value, NH3-N and TN were 0.69, 0.87, 0.81, and 0.94, respectively. This study could provide reference for on-line monitoring of wastewater in the future.

Urban Green Space (UGS), providing environmental, social and economic benefits simultaneously, has been regarded as a cost-effective Nature-based Solution (NbS) to combat the effects of urban heat island (UHI). Under the dual pressure of increasing demand for limited land resources and mitigating UHI, how to scientifically and effectively use the limited space to obtain the maximum cooling efficiency (scaling of cooling intensity and UGS size) is an important component of strategic urban green planning. However, the scale dependence of UGS cooling effect has not yet been sufficiently quantified, particularly with respect to involving small and medium size UGS. Here, we explored the size-dependent UGS cooling efficiency in Beijing using 10,003 UGS patches extracted from high-resolution remote sensing images. We found that 5922 UGS (59.20 %) exhibited a "cooling island effect", the cooling service of UGS could reduce land surface temperature by 0.06 ± 0.05 °C to 3.81 ± 1.01 °C, and the cooling intensity enhanced nonlinearly with increasing size and closely related to the complexity of UGS shape and vegetation quality. We further showed that the cooling efficiency of small, medium and large UGS was -0.004 ± 0.03 (n = 2201), 0.79 ± 0.01 (n = 3570), 0.19 ± 0.03 (n = 151), respectively, suggesting that strategic urban greening to combat urban heat should target on increasing medium-sized UGS and managing the layout of green space. These findings emphasize the significance of considering and further exploring the scale dependence of UGS cooling effect in mitigating urban heat.

Recently, rapidly increased demands of integration and miniaturization continuously challenge energy densities of dielectric capacitors. New materials with high recoverable energy storage densities become highly desirable. Here, by structure evolution between fluorite HfO2 and perovskite hafnate, we create an amorphous hafnium-based oxide that exhibits the energy density of ~155 J/cm3 with an efficiency of 87%, which is state-of-the-art in emergingly capacitive energy-storage materials. The amorphous structure is owing to oxygen instability in between the two energetically-favorable crystalline forms, in which not only the long-range periodicities of fluorite and perovskite are collapsed but also more than one symmetry, i.e., the monoclinic and orthorhombic, coexist in short range, giving rise to a strong structure disordering. As a result, the carrier avalanche is impeded and an ultrahigh breakdown strength up to 12 MV/cm is achieved, which, accompanying with a large permittivity, remarkably enhances the energy storage density. Our study provides a new and widely applicable platform for designing high-performance dielectric energy storage with the strategy exploring the boundary among different categories of materials.

Mid-infrared (MIR) pulsed lasers near a 3 µm waveband show great potential for the high absorption of water molecules and many important gas molecules. A passively Q-switched mode-locked (QSML) E r 3+-doped fluoride fiber laser with a low laser threshold and high slope efficiency around a 2.8 µm waveband is reported. The improvement is achieved by depositing bismuth sulfide (B i 2 S 3) particles onto the cavity mirror directly as a saturable absorber and using the cleaved end of the fluoride fiber as output directly. -QSML pulses begin to appear with the pump power of 280 mW. The repetition rate of the QSML pulses reaches a maximum of 33.59 kHz with the pump power of 540 mW. When the pump power is further increased, the output of the fiber laser switches from the QSML to the continuous-wave mode-locked operation with the repetition rate of 28.64 MHz and the slope efficiency of 12.2%. The results indicate that B i 2 S 3 is a promising modulator for the pulsed lasers near a 3 µm waveband, which paves the way for further development of various applications in MIR wavebands, including material processing, MIR frequency combs, and modern healthcare.

Voice user interface (VUI) is widely used in developing intelligent products due to its low learning cost. However, most of such products do not consider the cognitive and language ability of elderly people, which leads to low interaction efficiency, poor user experience, and unfriendliness to them. Firstly, the paper analyzes the factors which influence the voice interaction behavior of elderly people: speech rate of elderly people, dialog task type, and feedback word count. And then, the voice interaction simulation experiment was designed based on the wizard of Oz testing method. Thirty subjects (M = 61.86 years old, SD = 7.16; 15 males and 15 females) were invited to interact with the prototype of a voice robot through three kinds of dialog tasks and six configurations of the feedback speech rate. Elderly people's speech rates at which they speak to a person and to a voice robot, the feedback speech rates they expected for three dialog tasks were collected. The correlation between subjects' speech rate and the expected feedback speech rate, the influence of dialog task type, and feedback word count on elderly people's expected feedback speech rate were analyzed. The results show that elderly people speak to a voice robot with a lower speech rate than they speak to a person, and they expected the robot feedback speech rate to be lower than the rate they speak to the robot. There is a positive correlation between subjects' speech rate and the expected speech rate, which implies that elderly people with faster speech rates expected a faster feedback speech rate. There is no significant difference between the elderly people's expected speech rate for non-goal-oriented and goal-oriented dialog tasks. Meanwhile, a negative correlation between the feedback word count and the expected feedback speech rate is found. This study extends the knowledge boundaries of VUI design by investigating the influencing factors of voice interaction between elderly people and VUI. These results also provide practical implications for developing suitable VUI for elderly people, especially for regulating the feedback speech rate of VUI.

Enhancing soil organic carbon (SOC) sequestration and food supply are vital for human survival when facing climate change. Site-specific best management practices (BMPs) are being promoted for adoption globally as solutions. However, how SOC and crop yield are related to each other in responding to BMPs remains unknown. Here, path analysis based on meta-analysis and machine learning was conducted to identify the effects and potential mechanisms of how the relationship between SOC and crop yield responds to site-specific BMPs in China. The results showed that BMPs could significantly enhance SOC and maintain or increase crop yield. The maximum benefits in SOC (30.6%) and crop yield (79.8%) occurred in mineral fertilizer combined with organic inputs (MOF). Specifically, the optimal SOC and crop yield would be achieved when the areas were arid, soil pH was ≥7.3, initial SOC content was ≤10 g kg-1 , duration was >10 years, and the nitrogen (N) input level was 100-200 kg ha-1 . Further analysis revealed that the original SOC level and crop yield change showed an inverted V-shaped structure. The association between the changes in SOC and crop yield might be linked to the positive role of the nutrient-mediated effect. The results generally suggested that improving the SOC can strongly support better crop performance. Limitations in increasing crop yield still exist due to low original SOC level, and in regions where the excessive N inputs, inappropriate tillage or organic input is inadequate and could be diminished by optimizing BMPs in harmony with site-specific conditions.

Identifying traffic congestion accurately is crucial for improving the expressway service level. Because the distributions of microscopic traffic quantities are highly sensitive to slight changes, the traffic congestion measurement is affected by many factors. As an essential part of the expressway, service areas should be considered when measuring the traffic state. Although existing studies pay increasing attention to service areas, the impact caused by service areas is hard to measure for evaluating traffic congestion events. By merging ETC transaction datasets and service area entrance data, this work proposes a traffic congestion measurement with the influence of expressway service areas. In this model, the traffic congestion with the influence of service areas is corrected by three modules: 1) the pause rate prediction module; 2) the fitting module for the relationship between effect and pause rate; 3) the measurement module with correction terms. Extensive experiments were conducted on the real dataset of the Fujian Expressway, and the results show that the proposed method can be applied to measure the effect caused by service areas in the absence of service area entry data. The model can also provide references for other traffic indicator measurements under the effect of the service area.

As sessile organisms, plants must overcome various stresses. Accordingly, they have evolved several plant-specific growth and developmental processes. These plant processes may be related to the evolution of plant-specific protein families. The WRKY transcription factors originated in eukaryotes and expanded in plants, but are not present in animals. Over the past two decades, there have been many studies on WRKYs in plants, with much of the research concentrated on their roles in stress responses. Nevertheless, recent findings have revealed that WRKYs are also required for seed dormancy and germination, postembryonic morphogenesis, flowering, gametophyte development, and seed production. Thus, WRKYs may be important for plant adaptations to a sessile lifestyle because they simultaneously regulate stress resistance and plant-specific growth and development.

Straw incorporation is typically employed to enhance the nutrient content of soil and promote crop growth in intensive agricultural systems. Despite studies regarding the effects of straw incorporation on soil microbial communities, the underlying mechanisms of its effect on community co-occurrence interactions and assembly processes remain poorly understood. Herein, soil samples with or without straw incorporation were collected across a latitudinal gradient from north to central China. We found that straw incorporation considerably altered the structure of soil microbial community. The relative abundance of bacterial Latescibacterota and fungal Mortierellomycota were higher in straw-amended soils owing to their ability to decompose straw residues. The co-occurrence network in straw-amended soil exhibited greater complexity, including more network connectivity and keystone species, and higher average degrees and clustering coefficients compared with the control sample network. The network robustness and vulnerability indices suggested that straw incorporation increased the microbial network stability. Normalized stochastic ratios demonstrated that the stochastic process was the dominant mechanisms shaping the assembly of microbial communities in straw-amended soils. Concurrently, null model analysis revealed that straw increased the contribution of dispersal limitation to the assembly of bacterial and fungal communities. The migration rate of the microbial community, obtained from Sloan neutral community model, was relatively low in straw-amended soil at all the sample sites, potentially indicating the great importance of dispersal limitation. These findings would enhance our understanding of the ecological patterns and interactions of soil microbial communities in response to straw incorporation.

The understanding of how different cell types contribute to amyotrophic lateral sclerosis (ALS) pathogenesis is limited. Here we generated a single-nucleus transcriptomic and epigenomic atlas of the frontal cortex of ALS cases with C9orf72 (C9) hexanucleotide repeat expansions and sporadic ALS (sALS). Our findings reveal shared pathways in C9-ALS and sALS, characterized by synaptic dysfunction in excitatory neurons and a disease-associated state in microglia. The disease subtypes diverge with loss of astrocyte homeostasis in C9-ALS, and a more substantial disturbance of inhibitory neurons in sALS. Leveraging high depth 3'-end sequencing, we found a widespread switch towards distal polyadenylation (PA) site usage across ALS subtypes relative to controls. To explore this differential alternative PA (APA), we developed APA-Net, a deep neural network model that uses transcript sequence and expression levels of RNA-binding proteins (RBPs) to predict cell-type specific APA usage and RBP interactions likely to regulate APA across disease subtypes.

Light can significantly influence users' physiological and behavioural performance. However, how light breathing influences users' mood regulation remains unknown. To fill this gap, this study conducted a 2-by-2 experiment (N = 20) with light breathing as the between-subject factor and light condition as the within-subject factor. Both physiological indicators and subjective indicators are measured to reflect mood regulation. The data were analysed using a generalised linear mixed model. The results showed that breathing lights are effective in regulating users' moods. More specifically, breathing lights help users lower their electrodermal values and heart rates. Users did not report any significant difference in terms of subjective measures, which suggest that the influence of a breathing light happens unconsciously. Furthermore, this effect is significant for both cold and warm colour temperatures. Designers and engineers can use the research findings to manage user emotion when necessary.

Most studies on microplastics (MPs) focused on gut, liver, and brain, and MPs toxicity was size-dependent, but less has been reported on gill. Here, zebrafish were exposed to three sizes of MPs (45-53 µm, 90-106 µm, and 250-300 µm). Next, comparative transcriptome analysis and determination of physiological indices were performed in zebrafish gills to elucidate the size-associated toxicity of MPs to fish gills. Compared with the control, 60, 344, and 802 differentially expressed genes (DEGs) were identified after exposure to 45-53 µm, 90-106 µm, and 250-300 µm MPs for 5 days, respectively. More DEGs in treatment with bigger MPs suggested that bigger MPs might induce more changes in zebrafish gills than smaller ones. These DEGs were significantly enriched in the FoxO signaling, cellular senescence, circadian rhythm and p53 signaling pathways. Besides, 90-106 µm and 250-300 µm MPs treatments inhibited the cell cycle and prevented the apoptosis. The GSH content significantly increased after MPs exposure, suggesting the induction of oxidative stress. AChE and Na+/K+-ATPase activities were significantly lowered in all MPs treatments than in the control, suggesting the inhibition of neurotransmission and ion regulation. These changes might negatively influence the normal functioning of gills, such as osmoregulation, ion regulation, and respiration.

The environmental microbiome harbors a vast repertoire of antibiotic resistance genes (ARGs) which can serve as evolutionary predecessors for ARGs found in pathogenic bacteria, or can be directly mobilized to pathogens in the presence of selection pressures. Thus, ARGs from benign environmental bacteria are an important resource for understanding clinically relevant resistance. Here, we conduct a comprehensive functional analysis of the Antibiotic_NAT family of aminoglycoside acetyltransferases. We determined a pan-family antibiogram of 21 Antibiotic_NAT enzymes, including 8 derived from clinical isolates and 13 from environmental metagenomic samples. We find that environment-derived representatives confer high-level, broad-spectrum resistance, including against the atypical aminoglycoside apramycin, and that a metagenome-derived gene likely is ancestral to an aac(3) gene found in clinical isolates. Through crystallographic analysis, we rationalize the molecular basis for diversification of substrate specificity across the family. This work provides critical data on the molecular mechanism underpinning resistance to established and emergent aminoglycoside antibiotics and broadens our understanding of ARGs in the environment.

Purpose: To explore the relationship between single-nucleotide polymorphisms (SNPs) in complement factor I (CFI), interleukin-8 (IL-8), transferrin (TF), and transferrin receptor 2 (TFR2) and age-related macular degeneration (AMD) in a northeastern Chinese population. Methods: A total of 400 AMD patients (200 wet AMD and 200 dry AMD) and 200 controls were enrolled in this study, and genetic polymorphisms in the above genes were detected by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP). The χ2 test was used to compare differences in allele frequencies in each group, and the associations of genotype frequencies with AMD were determined by multivariate logistic regression analysis. Results: Our research shows that CFI rs141853578, IL-8 rs2227543, TF rs8177178 and TFR2 rs2075674 are associated with the incidence of AMD. In wet AMD, allele T of CFI rs141853578, IL-8 rs2227543 and TFR2 rs2075674 may be a risk factor. Allele A of TF rs8177178 may increase the risk of dry AMD. Conclusions: CFI rs141853578, IL-8 rs2227543, TF rs8177178 and TFR2 rs2075674 genetic polymorphisms are associated with the occurrence of AMD in a northeastern Chinese population, especially wet AMD.
