Aim To investigate the effect of PI3K/AKT/NF-κB signaling pathway in the treatment of physicion-8-O-β-D-monoglu-coside(PMG) on acute liver injury induced by carbon tetracloride(CCl 4) in mice . Methods Mice were randomly assigned into control group, model group, PMG low-dose, medium-dose and high-dose groups and Bifendate groups. After the continuous intervention with PMG for three days, CCl 4oil solution was intraperitoneally injected to establish acute liver injury mouse models, and samples were collected sixteen hours later. HE staining was used to observe the pathological changes of liver tissue. Hoechst 33342 staining was used to detect the number of apoptotic hepatocytes. Western blot was employed to detect the expression levels of caspase-3, PI3K, p-PI3K, AKT, p-Akt, IκB, p-IκB total protein and the nuclear protein NF-κB p65 in mouse liver tissue. The proportion of Th17 cells in mouse liver tissue was detected by FACS. Results After three days of PMG treatment, the pathological injury of liver tissue was relieved, the apoptosis of liver cells and the protein levels of caspase-3(P<0.01) were induced compared with model group.PMG could significantly decrease the nuclear expression of NF-κB p65 in the liver of mice with acute liver injury(P<0.05 or P<0.01). The phosphorylation levels of PI3K, AKT and IκB significantly decreased by PMG(P<0.05 or P <0.01). Otherwise, the proportion of Th17 cells in liver tissue was significantly reduced after PMG treatment(P<0.01). Conclusion PMG can alleviate CCl4 - induced acute liver injury through PI3K/AKT/NF-κB signaling pathway.

Transcriptome sequencing was used to obtain the differentially expressed genes of conjugated linoleic acid (CLA) on the basal diet of Royal chickens on intramuscular fat metabolism in related signal pathways and candidate genes that may play important roles by bioinformatics analysis, which could provide a further understanding of the molecular mechanism of CLA on intramuscular fat deposition. In this study, 55-day-old healthy Royal chickens were selected as experimental animals, and 0%, 1% and 2% of CLA were added to the basic diet. The pre-feeding period was 1 week and the normal feeding period was 6 weeks. The breast muscle tissues were collected for transcriptome sequencing, and the sequencing data were subjected for differential expression analysis, such as GO function and KEGG pathway enrichment analysis of differentially expressed genes. Differentially expressed genes related to breast muscle lipid metabolism were screened out, and qRT-PCR was used to verify differentially expressed genes. The results showed that a total of 1 065 differentially expressed genes were obtained, of which 703 were up-regulated genes and 362 were down-regulated genes. GO enrichment results show that differentially expressed genes are concentrated in cellular processes, single-biological processes, biological regulation and metabolic processes in biological processes. Enrichment analysis of KEGG signaling pathway shows that differentially expressed genes were significantly enriched in focal adhesion, unsaturated fatty acid biosynthesis, fatty acid biosynthesis, and steroid biosynthesis. Totally 11 candidate genes were found mainly related to intramuscular fat metabolism, including FADS1, FADS2, ELOVL5, ACOX2, SLC27A1, FABP5, LPL, LOC107050163, ENSGALG00000030996, ENSGALG00000005043 and ENSGALG00000048882. Six genes were randomly selected for qRT-PCR verification, and their relative expression changes were consistent with the sequencing results. This study screened the differentially expressed genes related to CLA affecting breast muscle lipid metabolism in Royal chickens, and analyzed 11 genes related to fat metabolism, laying the foundation for revealing the molecular mechanism of CLA regulating intramuscular fat deposition.

Objective:To investigate the effects of Huber 360 neuromuscular control training and evaluation system (Huber 360) on balance dysfunction for patients with stroke. Methods:From January to November, 2020, 16 patients with stroke were randomly assigned to control group (n = 8) and observation group (n = 8). Both groups received routine motor function training, while the observation group accepted Huber 360 training in addition, for three weeks. They were assessed with Berg Balance Scale, Huber 360, 10-meter Walking Test and Timed 'Up & Go' Test before and after training. Results:All the indexes improved after training in both groups (t > 1.368, P < 0.05), and improved more in the observation group than in the control group (t > 2.230, P < 0.05). Conclusion:Huber 360 can promote the recovery of balance for stroke patients.

Objective: To observe the clinical efficacy of acupuncture plus subtle adjustment manipulation of the cervical spine for peripheral facial paralysis.Methods: A total of 60 patients with peripheral facial paralysis who met the screening criteria were selected and divided into two groups according to the method of random number table, with 30 cases in each group. The patients in the acupuncture plus tuina group received routine treatment of acupuncture and subtle adjustment manipulation of the cervical spine, while the patients in the acupuncture group only received the same acupuncture treatment. Assessment of peripheral facial paralysis with integrated traditional Chinese and Western medicine was adopted for rehabilitation assessment. Results: The total effective rate of the acupuncture plus tuina group was 93.3%, while that of the acupuncture group was 70.0%, and there was a significant between-group difference (P<0.05). After treatment, the total scores of rehabilitation assessment in both groups were significantly higher than those before treatment (both P<0.05), and the score of the acupuncture plus tuina group was significantly higher than that of the acupuncture group (P<0.05). Conclusion: The treatment of acupuncture plus subtle adjustment manipulation of the cervical spine can improve the function of facial muscles in patients with peripheral facial paralysis, and its clinical efficacy is better than that of the acupuncture treatment alone.

Study the suitability of organic film for salvianolic acid in the ultrafiltration process of Danshen Dizhuye. UPLC was used to analyze the migration of nine phenolic active ingredients in Danshen Dizhuye during ultrafiltration of PES hollow fiber membrane and PS hollow fiber membrane. The structural composition of multi-components was analyzed by three different batches of Danshen Dizhuye before and after ultrafiltration of the two membranes. The results showed that 9 phenolic active ingredients in Danshen Dizhuye did not change significantly after ultrafiltration through PES membrane. However, after ultrafiltration through PS membrane, the content of sodium danshensu, protocatechualdehyde, caffeic acid, 3-hydroxy-4-methoxycinnamic acid and rosmarinic acid in Danshen Dizhuye did not change significantly, while salvianolic acid D, salvianolic acid B and lithospermic acid decreased by about 20%, and the content of salvianolic acid A decreased significantly. The final content in equilibrium was only about 20% of the original solution. Therefore, an in-depth study on the migration particularity of salvianolic acid A in ultrafiltration membrane was the focuse. The results showed that the loss of salvianolic acid A was caused by both membranes during ultrafiltration, and salvianolic acid A was lost more in PS membrane. When the membrane was washed and regenerated, it was found that salvianolic acid A was detected in the ethanol washing solution, but not in the washing liquid, indicating that the loss of salvianolic acid A during the ultrafiltration was mainly adsorptive action. The results suggested that the migration of phenolic active ingredients in Danshen Dizhuye during the membrane ultrafiltration process did not completely follow the molecular weight passing rule of the membrane pore size. At the same time, it may be affected by factors, such as the structure of the membrane material, and the interaction between the membrane structure and the structure of components, and exhibit different migration behaviors during the ultrafiltration of the membrane.

Traditional Chinese medicine believes that the occurrence and development of tumors is related to the body's Qi deficiency. " Invigorating Qi for consolidation of exterior" has became an effective way to treat tumors by traditional Chinese medicine. This study is based on the " invigorating Qi for consolidation of exterior" to explore the effect of flavonoid components in Qi-invigorating herbs Astragali Radix( AR) on the growth and immune function of mouse Lewis lung cancer xenografts,and further explore its mechanism of action. In the present study,high performance liquid chromatography was performed to analyze the flavonoid components in AR.The Lewis lung cancer model of C57 BL/6 mice was constructed,and the tumor volume of mice was determined by Visual Sonics Vevo2100 high frequency color ultrasound. The levels of IL~(-1)7 and RORγt in serum and tumor tissues were detected by ELISA and immunohistochemistry. The expression of IRE~(-1)/XBP~(-1) pathway-related proteins in tumor tissues was detected by Western blot. The results revealed that treatment of 5 and 10 g·kg~(-1)·d~(-1) of flavonoid components in AR significantly inhibited tumor growth of C57 BL/6 tumorbearing mice. The inhibition rates at the dose of 5 and 10 g·kg~(-1)·d~(-1) of flavonoid components in AR were( 29. 5±4. 4) % and( 43. 4±5. 2) %,respectively. The expression of IL~(-1)7 and RORγt in serum and tumor tissues of Lewis lung cancer mice were decreased,and the spleen index and thymus index were significantly enhanced by the flavonoid components in AR. Flavonoid components in AR could decrease the expression of X-box binding protein 1( XBP1),inositol-requiring enzyme( IRE1) and glucose regulated protein 78 k D( GRP78),and increase the expression of C/EBP homologous protein( CHOP),and the high-dose group is better,suggesting that the anti-lung cancer effect of flavonoid components in AR is related to the regulation of XBP1 mediated ERs. This study provides new evidence that the flavonoid components in AR could inhibit the tumor growth of C57 BL/6 tumor-bearing mice by regulating the body's immune function through " invigorating Qi for consolidation of exterior".

Objective To evaluate the year-round vaccine persistency of trivalent inactivated influenza vaccine (TIV) among elderly aged 65 years and above. Methods Searching "National Center for Biotechnology Information" , "Cochrane Library, CL", "China Biology Medicine disc", "China National Knowledge Infrastructure", "Wanfang Database", the studies of the year-round vaccine persistency of trivalent inactivated influenza vaccine among the elderly aged 65 years and above were included, and meta-analysis were made by Stata 11.0 software. Results A total of 17 studies were included. Compared with the antibody level before immunization, the standard mean differences (SMDs) of geometric mean titer (GMT) was 0.10 (95%CI: -0.12-0.32), 0.57 (95%CI: 0.23-0.91), 0.47 (95%CI:-0.20-1.15) and -0.27 (95%CI: -0.51--0.04) for the total haemagglutination inhibition (HI) antibody, the HI antibody of A/H1N1, the HI antibody of A/H3N2 and the HI antibody of B, respectively, by 360 day. The risk differences (RD) of sero-protection rate (SP) was 0.06 (95%CI: -0.04-0.16), 0.09 (95%CI: -0.15-0.33), -0.08 (95%CI: -0.28-0.13) and 0.00 (95%CI: -0.13 - -0.13) for the total haemagglutination inhibition (HI) antibody, the HI antibody of A/H1N1, the HI antibody of A/H3N2 and the HI antibody of B, respectively, by 360 day. Conclusion The HI antibody responses following TIV vaccination do not reliably persist year-round in the elderly aged 65 years and above . Clinic trials and health economic evaluation studies are needed to assess the feasibility and cost-effectiveness of multiple doses schedule of TIV in elderly.

Objective To evaluate vaccine efficacy(VE)of Haemophilus influenza type b vaccine (Hib)among children under five years of age .Methods Searching “National Center for Biotechnology Information,NCBI”,“Cochrane Library, CL”,“China Biology Medicine disc,CBMd”,“China National Knowledge Infrastructure,CNKI”,“Wanfang Database, the studies of VE for Hib among children under five years of age were included,and meta -analysis were made by RevMan 5.3 software.Results A total of 9 studies were included,seven were randomised clinical trial(RCT)design and two were semi RCT design.The VEs of one dose,two doses and three doses of Hib were 65%(95%CI:23% -84%),79%(95%CI:54% -90%),and 85%(95%CI:77% -90%),respectively.Conclusion We found that Hib is highly effective and that the two dose regime is as good as the three dose regime.

Objective To evaluate immunogenicity of one dose or two dose salk strain inactivated poliovirus vaccine(Salk -IPV )among children under seven years .Methods Searching "National Center for Biotechnology Information ,NCBI ","Cochrane Library ""China Biology Medicine disc ,CBMd ","China National Knowledge Infrastructure ,CNKI ","Wanfang Database,WF",studies for assessing immunogenicity of Salk -IPV were included.The seroconversion rate (SR)of neutralization antibody (NA)of specific type of polio virus with an interval of one month after vaccination was used as our endpoints.Pooled SR with 95% Confidence Interval (CI)was calculated by RevMan5.1 software.The association between seroconversion and age at administration was described by R 3.2.2 program.Results A total of 1 1 studies were included.One dose of Salk -IPV seroconvert 29% (95%CI:1 9% -39%),41 %(95%CI:31 % -51 %)and 38% (95%CI:28% -48%)of infants aged 7 years against serotypes I,Ⅱ,and Ⅲ on average,while 2 doses seroconvert 82% (95% CI:74% -90%),83% (95% CI:77% -88%) and 91 % (95% CI:87% -95%),respectively. Seroconversion increased with the starting age for both one dose and two dose schedules.Conclusion Routine immunization with at least 2 doses of Salk -IPV was recommended as a sequential schedule with two -valent oral polio virus vaccine and Salk -IPV was applied.

Objective To evaluate the status and equity of resource allocation for community vaccination services in Zhejiang Province.Methods A descriptive analysis was conducted,and Gini coefficient and Theil index were used to analyze the reasonableness and the demographic equity of resource allocation for community vaccination services in Zhejiang Province in 2013.Results The Gini coefficient of vaccination clinic and clinic staff based on population distribution in Zhejiang Province were 0.403 6 and 0.355 4,respectively.The total Theil index of vaccination clinic and clinic staff were 0.204 4 and 0.207 1,respectively.Both vaccination clinic and clinic staff,Theil index within the region were far higher than inter -regional Theil index.Conclusion The demographic equity of resource allocation for community vaccination services in Zhejiang Province should be improved.The disparity within the region is the main reason of unequal in resource allocation for community vaccination service.

<p><b>OBJECTIVE</b>To analyze the correlations between clinical features in paediatric patients with acute respiratory tract infection (ARTI) and viral load of human bocavirus.</p><p><b>METHODS</b>A prospective study was conducted on 956 children < 5 years admitted with an acute respiratory tract infection from November 2009 to December 2010, and 251 healthy children conclused as control group in the corresponding period. Human bocavirus was investigated in nasopharyngeal aspirates (NPA) and throat swab by PCR, and viral load was detected by real-time polymerase chain reaction (RT-PCR) in HBoV positive sample. Clinical data were also prospectively recorded.</p><p><b>RESULTS</b>A significant difference was found in HBoV positive rate between children with ARTI and control group at enrollment. There was a significant difference in HBoV viral load between children with upper respiratory tract infection and lower respiratory tract infection. HBoV viral load did not differ significantly between children with upper respiratory tract infection and control group. Among children with lower respiratory tract infection, no significant difference were detected between common and severe cases in HBoV viral load. HBoV viral load did not differ significantly whether the children were with or without co-infection.</p><p><b>CONCLUSIONS</b>HBoV could be detected perennial and considered as a major pathogen associated with acute respiratory tract infection in children. However, HBoV may not be a independent factor in children with ARTI and the HBoV viral load was not associated with the severity of respiratory illness.</p>

<p><b>BACKGROUND</b>Proprioception plays an important role in knee movements. Since there are controversies surrounding the overall recovery time of proprioception following surgery, it is necessary to define the factors affecting proprioceptive recovery after anterior cruciate ligament (ACL) reconstruction and to investigate the relationship between proprioception and muscle strength.</p><p><b>METHODS</b>A total of 36 patients who had their ACL reconstructed with a semitendinosus/gracilis graft (reconstructed group: 6 months post-surgery) and 13 healthy adults without any knee injury (control group) were included in the study. Knee proprioception was evaluated with a passive reproduction test. Isokinetic strength was measured using the Biodex System. Statistical analysis was used to compare proprioception of the reconstructed group versus the control group, and to define causal factors, including sex, hamstring/quadriceps ratio, and the course of injury before reconstruction. We also investigated the correlation between the passive reproduction error and quadriceps index.</p><p><b>RESULTS</b>There was a significant difference in proprioception between the reconstructed and control groups (P < 0.05). When the course of injury before reconstruction was less than 4 months, there was a linear correlation with proprioception 6 months after the operation (r = 0.713, P < 0.05). There was a positive correlation between post-surgery proprioception and the quadriceps index at 6 months post-surgery.</p><p><b>CONCLUSIONS</b>Impaired knee proprioception is observed 6 months after ACL reconstruction. Within 4 months of injury, early undertaking of reconstruction is associated with better proprioception outcome. Patients with enhanced proprioception have a better quadriceps index.</p>

RESUMO

With the development of structural and functional genomics, nowadays specific plant genome and transcriptome sequences can be cloned much easier and faster. Next step is to identify the functions of different genes and regulating elements to unravel the genetic mechanisms behind plant growth and development. Expression and its regulation are the language and dynamic property of genetic material, so expression and regulation analysis of target genes and sequences in plant cell is the basis for function study. Besides stable genetic transformation, plant transient expression system gains broad application in recent years, and its combination with other new technologies as gene shuffling, VIGS and RNAi plays a more and more important role in plant functional genomics.

<p><b>OBJECTIVE</b>To understand the incidence of falls in different residents of four communities in Zhejiang Province to provide a basis for their prevention and control.</p><p><b>METHODS</b>A household questionnaire survey on falls was conducted in 16 899 residents of two urban and two rural communities during November 2000 to October 2001.</p><p><b>RESULTS</b>Standardized incidence rate of falls was 5.07%, higher in rural areas (5.23%) than that in urban areas (4.95%) and showing no significant differences by gender. Falls occurred in September and October accounted for 11.68% and 19.22% and those occurred in pre-school children and the elderly accounted for 22.94% and 19.46% of the total episodes, respectively. Leading causes of falls varied in different population, from falls during their play (7.65%) and sports (4.06%) in young people and pre-school children (19.12%), slipping down when walking in adult (1.21%) and elderly (4.28%) men, and slipping down when up and down stairs in adult and elderly women. Burden and severity of injury caused by falls were increased with age.</p><p><b>CONCLUSIONS</b>Incidence of falls in pre-school children and the elderly was higher in local residents of communities in Zhejiang Province, causing heavy burden to the society and their families. Varied relevant measures should be taken to prevent and control for falls in different subgroups of population.</p>
