RESUMO

OBJECTIVE: To investigate the correlation between blood concentrations of tacrolimus (FK506) and cystatin C (Cys C) and the effect of FK506 on glycolipid metabolism in renal transplant recipients. METHODS: A total of 325 patients receiving renal transplantation between August, 2014 and September, 2015 in Nanfang Hospital were divided into 4 groups according to the postoperative time (1 month group, 1-3 months group, 4-6 months group, and 7-12 months group). FK506 blood trough concentration was measured at the time of postoperative follow-up, and creatinine (Scr) and Cys C levels were also detected. Results Plasma FK506 concentration decreased with age in the recipients and showed a positive correlation with Cys C (r=0.985, P=0.015) but no obvious correlation with Scr (r=0.259, P=0.741). FK506 had no effect on blood glucose (5.53-5.59 mmol

OBJECTIVE: To optimize the formulation of an emollient for treatment of atopic dermatitis prepared using ceramide, sodium hyaluronate, paeonol, and camellia-seed oil. METHODS: The emollients with different ratios of the 4 components were designed according to the L9(34)orthogonal table with 4 factors and 3 levels. The efficacy of the prepared emollients was tested in 4-6 week-old BALB/c mouse models of atopic dermatitis to determine the optimal formulation of the emollient by evaluating skin water content, transepidermal water loss (TEWL), pharmacodynamics and skin irritation. RESULTS: Range analysis of the orthogonal table and analysis of variance showed that ceramide and camellia seed oil contents had the greatest impact on the skin water content and TEWL, respectively, and the optimal composition of the emollient contained the 4 components at the ratios of D1E1F1G1. Pharmacodynamic experiments showed that at high, medium and low doses, the emollient with the optimal formulation significantly improved the skin water content, pH and TEWL in the mice (P<0.05) with similar effects in the positive control group (P>0.05) and a skin irritation test score of 0. CONCLUSION: The emollient we prepared can significantly improve skin water content, pH and TEWL in the mouse model of atopic dermatitis without skin irritations.

OBJECTIVE: To study the effects of Qing-Xuan tablets (QXT) on behavior pattern and striatal TNF-alpha in mice model of Parkinson's disease (PD). METHODS: The PD models were established by intraperitoneal injection of MPTP (30 mg/kg). 30 C57BL/6J mice were randomly divided into six groups: control group, PD model group, QXT high dosage group, QXT middle dosage group, QXT low dosage group and trihexyphenidyl hydrochloride group. After 7 days of treatment, the behavior pattern of mice were observed, and striatum were seperated to detect the content of TNF-alpha by ELISA. RESULTS: QXT increased the behavior of mice in behavioral tests (open field, pole test, grid test) (P<0.05 or P<0.01) but depressed TNF-alpha activity in striatum (P<0.05). CONCLUSION: QXT can significantly enhance the behavioral activity of mice,and depress TNF-alpha content in striatum,which suggest QXT can effectively relieve the symptom of PD.

OBJECTIVE: To evaluate the anti-proliferative and apoptosis-inducing effect of podophyllotoxin solid lipid nanoparticles (PDP-SLN) in human cervical carcinoma cells in vitro. METHODS: Hela cells were treated with PDP and PDP-SLN at different concentrations (0.0005-5 micromol/L), and the proliferation of the cells was assessed using MTT assay and the apoptotic index was determined by flow cytometry. RESULTS: Both PDP and PDP-SLN showed obvious inhibitory effect on the cell proliferation in a dose- and time-dependent manner. At the same concentration, PDP-SLN produced stronger inhibitory effect on the cells than PDP, with IC50 24, 48, and 72 h after the cell exposure to PDP-SLN and PDP of 4.10, 0.65, 0.20 micromol/L and 9.2, 4.0, 1.3 micromol/L, respectively. Both PDP and PDP-SLN significantly induced the apoptosis of the Hela cell, and the apoptosis rates of the cells incubated in the presence of 0.5 micromol/L PDP-SLN reached 90.8% at 24 h and 94.2% at 72 h, significantly higher than the rate of cells incubated with PDP (64.1% at 24 h and 68.4% at 72 h, P<0.01). CONCLUSION: PDP-SLN can effectively suppress the proliferation and induce apoptosis of Hela cells in vitro.

OBJECTIVE: To determine arsenic content in Changtong oral liquid, inductively coupled plasma-mass spectrometry (ICP-MS) was employed, which generated linear calibration curves in the range of 5-25 ng/ml for As (r=0.9998). The average recovery of As was 98.94% (n=5, RSD=2.58%).

OBJECTIVE: To investigate the effects of mitomycin magnetic polybutylcyanoacrylate nanoparticles (MMC-MPBCA- NP), magnetic polybutylcyanoacrylate nanoparticles (MPBCA-NP) and magnetic fluids (MF) and MMC on the proliferation and apoptosis of normal human liver cell strain (L-02 cells) in vitro. METHODS: The effect of MMC, MMC-MPBCA-NP, MPBCA-NP, and MF on the apoptosis and cell cycle of L-02 cells was studied by flow cytometry. The changes in the cell proliferation in response to treatments of the preparations and agents were examined by MTT colorimetric assay. RESULTS: MMC, MMC-MPBCA-NP, MPBCA-NP produced inhibition of the cell proliferation to different degrees but MF failed to result in cell proliferation inhibition. The results of MTT assay showed that MM, MPBCA-NP and MMC-MPBCA-NP all possessed concentration-dependent cytotoxicity, which increased with the concentration increment in comparison with the blank control. CONCLUSION: The descending order of the 4 agents in their effect of L-02 cell strain growth inhibition is MF >MPBCA-NP>MMC-MPBCA-NP>MMC. MMC and MMC-MPBCA-NP can inhibit the proliferation and cause cell cycle changes of L-02 cells, while MPBCA-NP inhibits the cell proliferation without inducing cell cycle alterations.

OBJECTIVE: To evaluate the biocompatibility of polybutylcyanoacrylate nanoparticles (PBCA-NP) manufactured by emulsion polymerization technique. METHODS: MTT assay was performed for evaluation of the cytotoxicity of PBCA-NP (e.g. cell proliferation and hemolytic test) experiments of long-term subcutaneous implantation and implantation in the muscular tissues of PBCA-NP was conducted in rabbits to observe the inflammatory reactions due to the implantation. RESULTS: No obvious cytotoxicity or hemolytic reactions were observed (with the hemolytic rate at different concentrations of PBCA-NP(5%). Three months after PBCA-NP implantation, obvious lymphocyte infiltration was not observed in the tissues around the implants which underwent gradual degradation. CONCLUSION: PBCA-NP possesses good biocompatibility.

AIM: The aim of this study was to observe the effect of a Chinese medicine compound Changtong oral liquid (CT) on tissue plasminogen activity (t-PA), plasminogen activator inhibitor (PAI), TGF-beta1 and hydroxyproline (OHP). METHODS: Two sets of animal experiments were performed in the present study. Forty New Zealand rabbits and 48 Sprague-Dawley (SD) rats were assigned randomly to one of the five groups: sham adhesion, adhesion with saline, adhesion with low dosage of the CT, adhesion with middle dosage of the CT and adhesion with high dosage of the CT. t-PA and PAI activity in plasma, OHP and TGF-beta1 expression in adhesion were investigated. Analysis of variance was used to test differences among groups. RESULTS: CT treatment increased plasma t-PA activity in rabbits but decreased TGF-beta1 activity in rats. The data were expressed from low to high dose respectively as follows: t-PA, 46.1+/-8.6 microkat/L, 59.6+/-10.1 microkat/L, 64.0+/-11.5 microkat/L; TGF-beta1 28+/-7.23%, 31+/-3.05%, 30+/-4.04%. There were significant differences compared with saline-treated animals (t-PA 26.4+/-5.1 microkat/L, TGF-beta1 54+/-5.51%). OHP content in cecum of rabbits from middle and high but not low dose of CT lowered significantly as compared with saline-treated rabbits, 0.3641+/-0.1373, 0.3348+/-0.0321, 0.2757+/-0.0497 mg/g vs 0.4183+/-0.0883 mg/g of protein, P>0.05, P<0.05, P<0.05 respectively. The rabbit plasma PAI activity and OHP content in abdominal wall had no difference in all groups. CONCLUSION: CT treatment significantly enhanced t-PA activity in rabbits, but decreased TGF-beta1 content in rats, OHP content in cecum of rabbits, and failed to affect the activity of PAI and OHP content in abdominal wall in rabbits, compared with saline group. The result suggests that CT could effectively prevent adhesions without interfering wound healing.

OBJECTIVE: To determine the content of dexamethasone sodium phosphate in Fuyankang cream, a preparation for treatment of various skin conditions. METHODS: Dexamethasone sodium phosphate content in Fuyankang cream was determined using high-performance capillary electrophoresis (HPCE) at the detection wavelength of 254 nm, under the optimal condition achieved with a fused-silica capillary tube (60 cm x 75 microm) and 100 mmol/L sodium tetraborate buffer (pH 9.2) at a constant voltage of 25 kV with a sampling time of 10 s at 25 degrees Celsius;. RESULTS: The calibration curve displayed good linear relationship within dexamethasone sodium phosphate concentration range of 20 to 100 microg/ ml r=0.999 8 . The average recovery of dexamethasone sodium phosphate was 99.75% n=5, RSD=1.03% . CONCLUSION: HPCE is simple, quick and sensitive in determination of dexamethasone sodium phosphate content in Fuyankang cream.
