RESUMO

In recent years, the application of numerical simulation in the research and development(R&D) as well as the pharmaceutical processes of new drugs has expanded considerably. The discrete element method(DEM), an important approach among numerical simulation methods, offers an effective tool for the simulation of discontinuous media. Referring to the research progress of DEM and the formulation of solid traditional Chinese medicine(TCM) preparations in recent years, this paper summarizes and analyzes the application of DEM in the pharmaceutical processes of solid TCM preparations, and discusses the challenges of its application in these processes, in order to provide new methods and ideas for promoting the high-quality production of TCM preparations.

RESUMO

The 21st century is a highly information-driven era, and traditional Chinese medicine(TCM) pharmacy is also moving towards digitization and informatization. New technologies such as artificial intelligence and big data with information technology as the core are being integrated into various aspects of drug research, manufacturing, evaluation, and application, promoting interaction between these stages and improving the quality and efficiency of TCM preparations. This, in turn, provides better healthcare services to the general population. The deep integration of emerging technologies such as artificial intelligence, big data, and cloud computing with the TCM pharmaceutical industry will innovate TCM pharmaceutical technology, accelerate the research and industrialization process of TCM pharmacy, provide cutting-edge technological support to the global scientific community, boost the efficiency of the TCM industry, and promote economic and social development. Drawing from recent developments in TCM pharmacy in China, this paper discussed the current research status and future trends in digital TCM pharmacy, aiming to provide a reference for future research in this field.

In recent years, the use of active substances as excipients or as substitutes for other excipients in the design of modern drug delivery systems has received widespread attention, which has promoted the development of the theory of unification of medicines and excipients in the design of traditional Chinese medicine(TCM) preparations. Adopting the theory of unification of medicines and excipients to design drug delivery systems can reduce the use of excipients and thus the cost of preparations, reduce drug toxicity, increase drug solubility and biocompatibility, enhance synergistic effect, and realize targeted delivery and simultaneous delivery of multiple components. However, the research on the application of this theory in the modern drug delivery system of TCM preparations is still insufficient, with few relevant articles. In addition, the TCM active substances that can be used as the excipients remain to be catalogued. In this paper, we review the types and applications of the drug delivery systems with TCM active substances as excipients and describe their common construction methods and mechanisms, aiming to provide references for the in-depth research on the modern drug delivery systems for TCM preparations.

Introduction: Lactation mastitis seriously severely affects the health of lactating females and their infants, yet the underlying causes of clinical lactation mastitis remain unclear. Methods: In this study, we used microbiota-humanized mice as a model to investigate the role of gut microbiota in lactation mastitis. We compared the fecal microbiota of lactation mastitis patients and healthy individuals and conducted fecal microbiota transplantation (FMT) experiments in an antibiotic-pretreated mouse model to test whether gut microbes contribute to human lactation mastitis. Results: Our results showed that gut microbiota diversity was reduced and dysbiosis was present in lactating mastitis patients. FMT from lactation mastitis patients (M-FMT), but not from healthy individuals (H-FMT), to antibiotic-treated mice resulted in lactation mastitis. The inflammation in mice caused by gut microbiota from lactating mastitis patients appears to be pervasive, as hepatocytes from mice that received feces from lactating mastitis patients showed marked swelling. In addition, serum pro-inflammatory factors, including IL-4, IL-17, MPO, IL-6, IL-1ß, and TNF-α, were significantly increased in the M-FMT group. The Firmicutes/Bacteroidetes ratio (F/B), a biomarker of gut dysbiosis, was significantly increased in the M-FMT group. At the phylum level, Actinobacteria were significantly increased, and Verrucomicrobia were significantly decreased in the M-FMT group. At the genus level, Ruminococcus and Faecalibacterium were significantly reduced, while Parabacteroides were significantly increased in the feces of both patients with lactation mastitis and M-FMT mice. Moreover, our study revealed an "amplification effect" on microbiota differences and mastitis disease following human-to-mouse FMT. Conclusion: Collectively, our findings demonstrate that the gut microbiota in lactating mastitis patients is dysbiotic and contributes to the pathogenesis of mastitis.

Ionic liquids(ILs) are salts composed entirely of anions and cations in a liquid state at or near room temperature, which have a variety of good physicochemical properties such as low volatility and high stability. This paper mainly reviewed the research overview of ILs in the application of traditional Chinese medicine(TCM) volatile oil preparation technology. Firstly, it briefly introduced the application of TCM volatile oil preparation technology and composition classification and physicochemical properties of ILs, and then summarized the application of ILs in the extraction, separation, analysis, and preparation of TCM volatile oil. Finally, the problems and challenges of ILs in the application of TCM volatile oil were explained, and the application of ILs in TCM volatile oil in the future was prospected.

Novel polyvinyl alcohol (PVA) blend membranes containing cellulose nanocrystals (CNs) and silver nanoparticles (AgNPs) were prepared via a simple method. CNs were prepared by sulfuric acid treatment of microcrystalline cellulose. AgNO3 aqueous solution mixed with the CNs aqueous suspension and was reduced by NaBH4 at room temperature. Purified CNs/AgNPs nanocomposites as functional fillers mixed with polyvinyl alcohol to prepare blend membrane. The morphology, mechanical properties, and antibacterial activities of PVA/CNs/AgNPs composite films were investigated. The PVA/CNs/AgNPs composite films were stable and homogeneous. The tensile strength of PVA was increased from 57.02 MPa to 81.21 MPa when filled with CNs/AgNPs. Antibacterial ratio of PVA/CNs/AgNPs composite against Gram-negative Escherichia coli and Gram-positive Staphylococcus aureus was 96.9% and 88.2%, respectively. The CNs/AgNPs nanocomposites could be applied as bi-functional nanofillers within PVA to improve the mechanical properties and antibacterial activities.

The goal of the study was to examine the differences in trajectories of change in the volume of white matter hyperintensities (WMH) in healthy adults within a relatively short period. We measured volumes of periventricular and deep WMH in frontal, temporal, parietal, and occipital lobes of healthy volunteers (age 49-83) on 3 occasions, approximately 15 months apart. At baseline, 40 participants underwent magnetic resonance imaging (MRI), 37 returned for the first and 30 for the second follow-up. Latent growth curve models estimated the variance and mean change in WMH volume and examined their associations with age, sex, education, and hypertension. In both regions and for both WMH types, the positive association between volume and age was stronger among the middle-aged adults and became weaker in older ages, as a logarithmic function of age. Individual variations were present in initial WMH volume but not in WMH volume progression. Frontal deep WMH volume was greater in hypertensive participants, whereas lower education was associated with greater posterior deep WMH volume. Thus, white matter of healthy middle-aged and older adults undergoes significant regional deterioration in a relatively short period, and is negatively affected by vascular risk and lower educational level.
