RESUMO

Objective: Low-grade glioma (LGG) mainly threatens the elderly population, with undesirable prognoses. This study uncovered the immune cell infiltration (ICI) landscape in LGG. Methods: RNA-seq profiles of LGG were retrieved from TCGA and CGGA databases. CIBERSORTx and ESTIMATE algorithms were employed to characterize the ICI landscape in LGG tissues. Through unsupervised clustering analysis, ICI subtypes were clustered. ICI scores were computed via principal component analysis (PCA). The differences in survival, tumor-infiltrating immune cells, stromal scores, immune scores, immune checkpoint genes, immune activity genes, and tumor mutation burden (TMB) were assessed between high and low ICI score groups. Results: Three ICI subtypes were constructed in LGG, with distinct survival outcomes, PD-L1 expression, and infiltration levels of immune cells. Furthermore, ICI scores were developed. Both in TCGA and CGGA datasets, low ICI scores were indicative of undesirable outcomes. High ICI scores were significantly correlated to increased infiltration levels of memory B cells, CD8 T cells, CD4 naïve T cells, T follicular helper cells, macrophages M0, and eosinophils, while low ICI scores were characterized by increased infiltration levels of naïve B cells, plasma cells, CD4 memory resting T cells, Tregs, resting NK cells, macrophages M2, and activated dendritic cells. High ICI scores exhibited correlations with lower immune activity genes and immune checkpoint genes. Furthermore, TMB was distinctly reduced in the high ICI score group. Conclusion: The ICI scores may serve as a promising prognostic index and predictive indicator for immunotherapies, extending our understanding of immune microenvironment in LGG.

Neovascularization is required in high-grade glioma (HGG). The objective of this study was to explore neovascularization-related genes and their corresponding MRI biomarkers during the early-growth stage of HGG. Tumor tissues from 30 HGG patients underwent perfusion MRI scanning prior to surgery were used to establish orthotopic xenograft models, pathologically analyze the tumor vasculature and perform transcriptome sequencing. The cases were divided into two groups based on whether the xenograft was successfully established. Microvascular density and BMPER, CXCL10, and HOXA9 expression of surgical specimens in the xenograft-forming group was significantly elevated and the microvascular diameter was significantly reduced, in vitro inhibition of BMPER, CXCL10, or HOXA9 in the glioma stem cell significantly suppressed its tube formation abilities. The in vivo experiment showed that BMPER was highly expressed in the early tumor growth phase (20 days), CXCL10 and HOXA9 expression was elevated with tumor progress, and spatially associated with tumor vasculature. Perfusion weighted MRI (PWI-MRI) derived parameters, rCBV, rCBF, Ktrans, and Vp, were also increased in the xenograft-forming group. In conclusion BMPER, CXCL10, and HOXA9 promote early tumor growth and progression by stimulating neovascularization of primary HGG. The rCBV, rCBF, Ktrans, and Vp can be used as imaging biomarkers to predict the expression statuses of these genes.

Developing efficient, sustainable, and biocompatible high-tech nanoplatforms derived from naturally existing components in living organisms is highly beneficial for diverse advanced biomedical applications. Melanins are nontoxic natural biopolymers owning widespread distribution in various biosystems, possessing fascinating physicochemical properties and playing significant physiological roles. The multifunctionality together with intrinsic biocompatibility renders bioinspired melanin-like nanomaterials considerably promising as a versatile and powerful nanoplatform with broad bioapplication prospects. This panoramic Review starts with an overview of the fundamental physicochemical properties, preparation methods, and polymerization mechanisms of melanins. A systematical and well-bedded description of recent advancements of melanin-like nanomaterials regarding diverse biomedical applications is then given, mainly focusing on biological imaging, photothermal therapy, drug delivery for tumor treatment, and other emerging biomedicine-related implementations. Finally, current challenges toward clinical translation with an emphasis on innovative design strategies and future striving directions are rationally discussed. This comprehensive and detailed Review provides a deep understanding of the current research status of melanin-like nanomaterials and is expected to motivate further optimization of the design of novel tailorable and marketable multifunctional nanoplatforms in biomedicine.

In this work, we report on biodegradable eumelanin-Fe3O4 hybrid nanoparticles (euMel-Fe3O4 NPs) for multiple imaging-assisted local photothermolysis. The euMel-Fe3O4 NPs are rationally prepared via a facile one-step co-precipitation method in the aqueous phase in the presence of eumelanin. The obtained NPs exhibited high stability and aqueous dispersity. euMel-Fe3O4 NPs could provide T2-weighted magnetic resonance (MR) and photoacoustic (PA) imaging contrast enhancement prior to the treatment, due to their high r2 relaxivity that was determined to be 245.88 mM-1 s-1 (a value higher than those for other commercially available iron oxide NPs) and good PA signal intensity. Besides, with effective NIR absorbance, the euMel-Fe3O4 NPs promoted an effective photothermolysis treatment in an experimental mouse model of cancer. Cytotoxicity and in vivo histological examination demonstrated no appreciable toxicity. Our work suggests that euMel-Fe3O4 NPs could be proposed as a potential class of theranostic nanoplatform with multiple imaging modalities for cancer therapy.

OBJECTIVE: To develop a brief version scale with good validity and reliability to evaluate self-management knowledge, attitude and behavior (KAB) of diabetes patients in a shorter time. METHODS: 20 authority experts who specialized in the field of diabetes clinical and non-communicable disease self-management, and familiar with the relevant content of diabetes self-management were selected. Face to face and Email consultation methods were applied in two rounds delphi expert consultations to choose items from the Diabetes Self-management Knowledge, Attitude, and Behavior Assessment Scale (DSKAB, total scale, 98 items). In the first round of expert consultation, experts were asked to select the core items from every dimension and score the extent of familiarity and basis of judgment. In the second round of expert consultation, the significance of the core items which were selected by experts in first round, were graded. The items which entered into the brief scale were determined by harmonious condition of expert opinions. RESULTS: Two rounds of consultation reclaimed 16 and 15 responses respectively, Positive coefficients of experts were 80% and 94% respectively, and both authoritative coefficients were 0.90. In the first round of consultation, experts selected 44.3 ± 6.2 items on average, 42 items were selected according to the integrity of the scale for measuring dimension and content at last. In the second round of consultation, experts thought that the 42 items could measure the important contents of each dimension comprehensive and representative, the item importance scores of knowledge, attitude, and behavior subscale were 8.42 ± 1.17, 8.61 ± 1.24, 8.61 ± 1.02 respectively, and the coordination coefficients of the three subscale were 0.36, 0.20, 0.49 (χ(2) were 89.74, 11.13, 96.03, P<0.05). The total coordination coefficient was 0.38 (χ(2) =199.36, P<0.001), which indicated the concentration of experts(')opinion was acceptable, showed the brief scale with good face validity and content validity. CONCLUSION: The brief scale could evaluate Diabetes Self-management KAB Scale rapidly and relatively comprehensive in groups or individuals diabetes with good face validity and content validity.
