RESUMO

Double perovskites (DPs) have attracted attention in the field of luminescence due to their inherent broadband emission of self-trapping excitons. In this work, we choose [(CH3)3NCH2CHCH2]+ and [CH3CHOHCH2NH2]+ as organic cations to synthesize two new organic-inorganic hybrid DPs, [(CH3)3NCH2CHCH2]2KInCl6 (1) and [CH3CHOHCH2NH2]2KInCl6 (2). The [KCl6]3- and [InCl6]3- octahedra are interchangeably connected by sharing two opposite faces, forming a one-dimensional coordination chain. Each K atom coordinates with six chlorine atoms in 1, while it coordinates with two oxygen atoms in addition to the six chlorine atoms in 2. The coordination between ions K and O in compound 2 may have significantly reduced its luminescence. As a result, compound 1 shows bright-yellow light with a quantum yield of more than 90%, while 2 shows weak blue light with a quantum yield of only 0.98%. In addition, different from no phase transition found in 2, 1 undergoes a reversible phase transition at 324/307 K in the heating-cooling cycle. Through structural and spectral analysis and density functional theory calculation, we conclude that the larger degree of [InCl6]3- octahedral distortion and the larger anion distance (In···In) also cause the PLQY of compound 1 to be higher than that of compound 2.

Osteoarthritis (OA), a joint disease that is associated with inflammatory processes is involved in joint destruction. Scutellarein (Scu), a component of the medicinal herbs Scutellaria barbata D. Don and Erigeron breviscapus (vant) Hand Mass, has anti-inflammatory effects. We explored the role of Scu in the development of OA and the underlying mechanisms. CCK-8 assays, Calcein-AM/PI and EdU staining were used to determine chondrocyte viability after Scu exposure. Western blot, qPCR, as well as ELISA were utilized to measure extracellular matrix (ECM) degradation and inflammation. Immunofluorescence (IF), western blot and luciferase assays were used to examine the NF-kappaB (NF-κB) pathway. Scu interacting proteins were predicted using network pharmacology analysis and molecular docking. X-ray, H&E, Safranin O-Fast Green(S-O), toluidine blue, and immunohistochemistry analysis were used to examine the therapeutic effects of Scu in OA using destabilization of medial meniscus (DMM) models. Scu demonstrated inhibitory effects on ECM degradation and pro-inflammatory factor levels in chondrocytes treated with IL-1ß. Mechanistically, Scu inhibited the IL-1ß-induced activation of the PI3K/Akt/ NF-κB signaling pathway cascades. Furthermore, Scu has been shown to have significant binding capacities to PI3K. Additionally, Scu ameliorated the OA progression in DMM models. Our findings suggest that Scu may contribute to the amelioration of OA progression by targeting the PI3K/Akt/NF-κB signaling pathway, implying Scu possesses promising therapeutic potential for the treatment of OA.

The cannabinoid receptor 1 (CB1) is famous as the target of Δ9-tetrahydrocannabinol (THC), which is the active ingredient of marijuana. Suppression of CB1 is frequently suggested as a drug target or gene therapy for many conditions (e.g., obesity, Parkinson's disease). However, brain networks affected by CB1 remain elusive, and unanticipated psychological effects in a clinical trial had dire consequences. To better understand the whole brain effects of CB1 suppression we performed in vivo imaging on mice under complete knockout of the gene for CB1 (cnr1-/-) and also under the CB1 inverse agonist rimonabant. We examined white matter structural changes and brain function (network activity and directional uniformity) in cnr1-/- mice. In cnr1-/- mice, white matter (in both sexes) and functional directional uniformity (in male mice) were altered across the brain but network activity was largely unaltered. Conversely, under rimonabant, functional directional uniformity was not altered but network activity was altered in cortical regions, primarily in networks known to be altered by THC (e.g., neocortex, hippocampal formation). However, rimonabant did not alter many brain regions found in both our cnr1-/- results and previous behavioral studies of cnr1-/- mice (e.g., thalamus, infralimbic area). This suggests that chronic loss of cnr1 is substantially different from short-term suppression, subtly rewiring the brain but largely maintaining the network activity. Our results help explain why pathological mutations in CB1 (e.g., chronic pain) do not always provide insight into the side effects of CB1 suppression (e.g., clinical depression), and thus urge more preclinical studies for any drugs that suppress CB1.

The purpose of this study is to investigate the serum inflammatory factors in patients with high-altitude polycythemia (HAPC) and their correlation with cognitive function. The subjects were recruited and placed into a HAPC group and control group. Serum samples were collected, and inflammatory factors (interleukin-1beta [IL-1ß], monocyte chemoattractant protein-1 [MCP-1], and tumor necrosis factor-alpha [TNF-α]) were measured using ELISA kits. The mini-mental State Examination (MMSE) was used to assess cognitive function. According to the MMSE scores, HAPC group was further divided into normal cognitive function group (HNCF) and cognitive dysfunction group (HCDF). In comparison with the control group, the MMSE scores in the HAPC group were significantly low (P < .05), whereas the serum levels of IL-1ß, MCP-1, and TNF-α were significantly high (P < .01). Among the HAPC group (n = 60), 21 belonged to the HCDF and 39 belonged to the HNCF. Compared with the HNCF, the IL-1ß, MCP-1, and TNF-α in the HCDF were significantly increased (P < .01). The Pearson correlation analysis showed that inflammatory factors were positively correlated with hemoglobin, and negatively correlated with MMSE. Serum inflammatory cytokines IL-1, MCP-1, and TNF-α were increased in HAPC, and HAPC exhibited cognitive dysfunction. Considering chronic hypoxia environment influences the change of the red blood cell metabolic and inflammatory factor, red blood cells and inflammatory factor in plateau is likely to be affected by patients with vascular lesions, increase cognitive impairment.

Hepatocellular carcinoma (HCC), ranking as the seventh most prevalent cancer worldwide, poses a significant health challenge. Actinidia chinensis Planch Root extracts (acRoots), a traditional Chinese medicine, has exhibited promising inhibitory effects on the proliferation, invasion, and migration of various cancer cell types. Nevertheless, its specific impact and underlying mechanisms concerning HCC remain unclear. This research aimed to elucidate the anticancer properties and potential molecular mechanisms of acRoots in the HepG2 and LM3 cell lines. Our findings demonstrate that acRoots effectively hampers the in vitro proliferation, migration, and invasion of HCC cells. Furthermore, acRoots induces apoptosis and autophagy by impeding the AKT/mTOR signaling pathway, with its inhibitory effects on cells being restored under AKT activator induction. This study, for the first time, elucidates that acRoots can suppress HepG2 and LM3 cell proliferation by blocking the Akt/mTOR pathway, thereby activating apoptosis and autophagy. These results underscore the potential of acRoots as a promising antitumor agent for HCC.

[(CH3)3P(CH2)2OH]2Cd3(SCN)8 (1) and [(CH3)3P(CH2)2OH]Cd(SCN)3 (2) were obtained with completely different structures and properties under the same synthesis conditions. Compound 1, showing green fluorescence, has a rare three-dimensional 4,6-connected fsh topology having (43.63)2(46.66.83) Schläfli notation, while compound 2 with blue-violet phosphorescence displays a one-dimensional perovskite structure with an infinite {[Cd(SCN)3]-}∞ chain and exhibits both ferroelastic and dielectric switching characteristics.

The study of neural circuits, which underlies perception, cognition, emotion, and behavior, is essential for understanding the mammalian brain, a complex organ consisting of billions of neurons. To study the structure and function of the brain, in vivo neuronal labeling and imaging techniques are crucial as they provide true physiological information that ex vivo methods cannot offer. In this paper, we present a new strategy for in vivo neuronal labeling and quantification using MRI. We demonstrate the efficacy of this method by delivering the oatp1a1 gene to the target neurons using rAAV2-retro virus. OATP1A1 protein expression on the neuronal membrane increased the uptake of a specific MRI contrast agent (Gd-EOB-DTPA), leading to hyperintense signals on T1W images of labeled neuronal populations. We also used dynamic contrast enhancement-based methods to obtain quantitative information on labeled neuronal populations in vivo.

Hybrid organic-inorganic perovskites (HOIPs) have been widely studied for their interesting functions and potential applications. Here, we report a novel sulfur-containing hybrid organic-inorganic perovskite based on a one-dimensional ABX3-type compound: [C3H7N2S]PbI3 ([C3H7N2S]+ is 2-amino-2-thiazolinium) (1). Compound 1 undergoes two high-temperature phase transitions at 363 K and 401 K, respectively, showing a band gap of 2.33 eV, and has a narrower band gap compared to other one-dimensional materials. Moreover, by introducing thioether groups into the organic component, 1 has the ability to uptake Pd(II) ions. Compared with previously reported low-temperature isostructural phase transition sulfur-containing hybrids, the molecular motion of 1 becomes more intense under the stimulation of high temperature, leading to changes in the space group during the two phase transitions (Pbca → Pmcn → Cmcm), which are no longer the previous isostructural phase transitions. Significant changes in the phase transition behavior and semiconductor properties before and after metal absorption make it possible to monitor the absorption process of metal ions. The study of the effect of Pd(II) uptake on phase transitions may be helpful to reveal the mechanism of phase transitions more deeply. This work will broaden the hybrid organic-inorganic ABX3-type semiconductor family and pave the way for the development of organic-inorganic hybrid-based multifunctional phase transition materials.

Molecular ferroelectrics have gradually aroused great interest in both fundamental scientific research and technological applications because of their easy processing, light weight and mechanical flexibility. Hybrid organic-inorganic perovskite ferroelectrics (HOIPFs), as a class of molecule-based ferroelectrics, have diverse functionalities owing to their unique structure and have become a hot spot in molecular ferroelectrics research. Therefore, they are extremely attractive in the field of ferroelectrics. However, there seems to be a lack of systematic review of their design, performance and potential applications. Herein, we review the recent development of HOIPFs from lead-based, lead-free and metal-free perovskites, and outline the versatility of these ferroelectrics, including piezoelectricity for mechanical energy-harvesting and optoelectronic properties for photovoltaics and light detection. Furthermore, a perspective view of the challenges and future directions of HOIPFs is also highlighted.

The remarkable material stability and structural diversity of two-dimensional (2D) organic-inorganic hybrid perovskites (OIHPs) constitute a vast available library of versatile materials. In particular, ferroelastic property, for which the spontaneous strain can be transformed by applying mechanical stress, is very promising for extensive nanotechnological applications. However, integrating ferroelastic property into 2D OIHPs is still in its infancy. Herein, we designed two new 2D OIHPs (C3 H5 CH2 NH3 )2 [MCl4 ] (M=Mn for 1 and Cd for 2), which undergo reversible ferroelastic phase transitions with an Aizu expression 4/mmmFmmm. The templating influence of the more distorted inorganic framework on the disordering of organic cations and the stronger hydrogen bonds has a key role in the striking improvement of Curie temperature from 246 K in 1 to 273 K in 2. Meanwhile, the minimized alteration of structural motif ensures the well maintaining of the ferroelastic performance in the forms of crystals and thin films, as demonstrated by the identifiable evolution of domain structures. This work will provide a fertile new ground for enlarging the limited number of 2D ferroelastic OIHPs with better practical utility.

Dynamic contrast-enhanced MRI (DCE-MRI) is routinely included in the prostate MRI protocol for a long time; its role has been questioned. It provides rich spatial and temporal information. However, the contained information cannot be fully extracted in radiologists' visual evaluation. More sophisticated computer algorithms are needed to extract the higher-order information. The purpose of this study was to apply a new deep learning algorithm, the bi-directional convolutional long short-term memory (CLSTM) network, and the radiomics analysis for differential diagnosis of PCa and benign prostatic hyperplasia (BPH). To systematically investigate the optimal amount of peritumoral tissue for improving diagnosis, a total of 9 ROIs were delineated by using 3 different methods. The results showed that bi-directional CLSTM with ± 20% region growing peritumoral ROI achieved the mean AUC of 0.89, better than the mean AUC of 0.84 by using the tumor alone without any peritumoral tissue (p = 0.25, not significant). For all 9 ROIs, deep learning had higher AUC than radiomics, but only reaching the significant difference for ± 20% region growing peritumoral ROI (0.89 vs. 0.79, p = 0.04). In conclusion, the kinetic information extracted from DCE-MRI using bi-directional CLSTM may provide helpful supplementary information for diagnosis of PCa.

Ferroelastic materials have received special attention because of their great promise for mechanical switches, piezoelectric sensors, and data storage applications. Here, we report a novel ferroelastic semiconducting hybrid organic-inorganic perovskite (C3H7N2S)PbBr3 (1) [(C3H7N2S)+ is 2-amino-2-thiazolinium] with a ferroelastic phase transition at 395 K and an optical band gap of 3.43 eV. 1 has a one-dimensional BaNiO3-type structure and undergoes a high-temperature ferroelastic phase transition with an Aizu notation of mmmF2/m. Meanwhile, 1 exhibits dielectric switch near the phase transition temperature. By introducing the thioether group, the motion of the molecules or ions of 1 is hindered after the sorption of Pd(II) metal ions, which leads to the disappearance of the high-temperature ferroelastic phase transition and dielectric switch. This is the first reported ferroelastic semiconductor material with Pd(II) adsorption property, by studying the influence of Pd(II) adsorption on high-temperature ferroelastic phase transition, it may be enlightening to further uncover the mechanism of phase transition or the origin of ferroelasticity, which represents an important step toward multifunctional applications of lead-hybrid perovskite-based ferroelastic materials.

[(CH3)3PCH2CH2CH3]2(n-C4H9NH3)Bi2Br9 (1) was obtained by a mixed organic cation strategy. The introduction of phosphonium cations increases the potential energy barrier for the motion of cations, which raises the ferroelastic phase transition temperature of 1 above room temperature, unlike the low-temperature ferroelasticity of [n-C4H9NH3]2[BiBr5]. This provides a new idea for exploring and designing molecular ferroelastic materials with excellent performance.

BACKGROUND: Falls are one of the main reasons for mortality and morbidity in older adults. However, despite adoption of prevention strategies, the number of falls in older adults has not declined. This study aimed to develop a questionnaire to assess fall prevention knowledge and explore associated factors among Chinese community-dwelling older adults. METHODS: The Home-Based Fall Prevention Knowledge (HFPK) questionnaire was developed by Delphi expert consultation. We tested the internal consistency, reliability, and content validity of the HFPK. A total of 374 community-dwelling older adults participated in this study. The HFPK was used to assess their fall prevention knowledge. RESULTS: After being evaluated by 15 experts, the item content validity index ranged from 0.867 to 1, and the scale content validity index was 0.985, which met the criterion for content validity. Cronbach's α coefficient was 0.933, which satisfied the reliability criterion. Stepwise linear regression analysis showed that fall prevention knowledge was significantly associated with having higher education, being female, having a higher monthly income, people who were public officials before retirement, and having fewer children (p < 0.05). CONCLUSION: Fall prevention knowledge should be improved among older males and those with lower education, lower monthly income, people who were not public officials before retirement, and more children.

Direct detection of circularly polarized light (CPL) is a challenging task due to limited materials and ambiguous structure-property relationships that lead to low distinguishability of the light helicities. Perovskite ferroelectric semiconductors incorporating chirality provide new opportunities in dealing with this issue. Herein, a pair of 2D chiral perovskite ferroelectrics is reported, which have enhanced CPL detection performance due to interplays among lattice, photon, charge, spin, and orbit. The chirality-transfer-induced chiral&polar ferroelectric phase enhances the asymmetric nature of the photoactive sublattice and achieves a switchable self-powered detection via the bulk photovoltaic effect. The single-crystal-based device exhibits a CPL-sensitive detection performance under 430 nm with an asymmetric factor of 0.20 for left- and right-CPL differentiation, about two times that of the pure chiral counterparts. The enhanced CPL detection performance is ascribed to the Rashba-Dresselhaus effect that originates from the bulk inversion asymmetry and strong spin-orbit coupling, shown with a large Rashba coefficient, which is demonstrated by density functional theory calculation and circularly polarized light excited photoluminescence measurement. These results provide new perspectives on chiral Rashba ferroelectric semiconductors for direct CPL detection and ferroelectrics-based chiroptics and spintronics.

Layered hybrid perovskites, due to their broad application potential in optical, electrical, and luminescence fields, are attracting increasing attention. Herein, we report two novel two-dimensional hybrid organic-inorganic perovskites [C5H10N2S]PbBr4 (1) and [C5H10N2S]PbI4 (2) ([C5H10N2S]2+ is 3-ethylaminothiazolium), which possess phase transition and semiconductor properties. Intriguingly, 1 has semiconductor properties with an optical band gap of 2.70 eV and a photocurrent to dark current ratio of 50 for the photoresponse. By varying the halogen, the band gap of 2 is significantly reduced to 2.06 eV and the photocurrent to dark current ratio of the photoresponse reaches 104. Moreover, compounds 1 and 2 are able to adsorb Pd(II) resulting from the presence of -SH groups in the cation. The adsorption of Pd(II) can turn a semiconductor into an insulator, weaken the fluorescence intensity, and cause the phase transition behavior to disappear. This work has remarkable implications for the continuous development of hybrid perovskites with phase transitions, metal adsorption, and semiconductor properties.

Erectile dysfunction (ED) is closely associated with smooth muscle dysfunction, but its underlying mechanisms remains incompletely understood. We here reported that the reduced expression of myosin phosphatase target subunit 1 (MYPT1), the main regulatory unit of myosin light chain phosphatase, was critical for the development of vasculogenic ED. Male MYPT1 knockout mice had reduced fertility and the penises displayed impaired erections as evidenced by reduced intracavernous pressure (ICP). The penile smooth muscles of the knockout mice displayed enhanced response to G-Protein Couple Receptor agonism and depolarization contractility and resistant relaxation. We further identified a natural compound lotusine that increased the MYPT1 expression by inhibiting SIAH1/2 E3 ligases-mediated protein degradation. This compound sufficiently restored the ICP and improved histological characters of the penile artery of Mypt1 haploinsufficiency mice. In diabetic ED mice (db/db), the decreased expression of MYPT1 was measured, and ICP was improved by lotusine treatment. We conclude that the reduction of MYPT1 is the major pathogenic factor of vasculogenic ED. The restoration of MYPT1 by lotusine improved the function of injured penile smooth muscles, and could be a novel strategy for ED therapy.

BACKGROUND: Falls are common among adults aged 60 years and older because of physiological changes. Most falls in older adults occur most often at home. Coupled with the lack of awareness and knowledge of preventing falls, the proportion of injuries and deaths among older adults due to falls is increasing yearly. Our study developed a WeChat mini-program for urban elderly to implement teach-back health education (TBHE) that a repeated cycle process of health education, assessment, and re-education in preventing falls at home. OBJECTIVES: This study aimed to evaluate the application effect of the TBHE-based WeChat mini-program on health education knowledge for fall prevention at home for urban older adults. DESIGN: A single-blinded, two-arm parallel-group, randomized controlled trial was conducted. SETTING: Three residential communities, named Hot Spring Apartment, Hualinyuan, and Dongtang Community in Gulou District, Fuzhou, China. PARTICIPANTS: Participants were older adults recruited from communities in Fuzhou from January to March 2021. METHODS: Fifty-nine participants agreed to participate and were assigned randomly to the intervention group (n = 29) or the control group receiving traditional health education (n = 30). Each participant in the intervention group received twice a week for a total of 8 weeks of health education interventions performed by the first author that she is intervenor according to specific themes. The trial statistician, recruiters, and participants were blinded to group allocation. The intervenor (first author) was blinded to the study hypotheses. To evaluate the effects of the intervention, we assessed participants' knowledge total score and scores of physiology and disease; drug application; mental, cognitive, and spiritual well-being; lifestyle; and house environment at baseline and 1-week post-intervention and compared scores between two groups. A two-way repeated-measures analysis of variance was conducted to examine the effects of time, group, and their interaction. RESULTS: There was a significant difference in knowledge of house environment (p = 0.003) between the two groups. Within groups, total and five dimensions knowledge scores had a significant difference (p < 0.001). Moreover, interaction effects were significant on drug application (p = 0.012) and mental, cognitive, and spiritual well-being (p = 0.015). CONCLUSIONS: The TBHE can improve knowledge on fall prevention at home among urban older adults. The TBHE based on the WeChat mini-program could enhance the efficiency and effectiveness of being educated among urban older adults. RESUMO

A long-established strategy for transcription regulation is the tethering of transcription factors to cellular membranes. By contrast, the principal effectors of Hedgehog signalling, the GLI transcription factors, are regulated by microtubules in the primary cilium and the cytoplasm. How GLI is tethered to microtubules remains unclear. Here, we uncover DNA mimicry by the ciliary kinesin KIF7 as a mechanism for the recruitment of GLI to microtubules, wherein the coiled-coil dimerization domain of KIF7, characterized by its striking shape, size and charge similarity to DNA, forms a complex with the DNA-binding zinc fingers in GLI, thus revealing a mode of tethering a DNA-binding protein to the cytoskeleton. GLI increases KIF7 microtubule affinity and consequently modulates the localization of both proteins to microtubules and the cilium tip. Thus, the kinesin-microtubule system is not a passive GLI tether but a regulatable platform tuned by the kinesin-transcription factor interaction. We retooled this coiled-coil-based GLI-KIF7 interaction to inhibit the nuclear and cilium localization of GLI. This strategy can potentially be exploited to downregulate erroneously activated GLI in human cancers.
