RESUMO

This study aimed to detect micrometastatic tumor cells in the lymph nodes of patients with pN0 lung adenocarcinoma using a combination of thyroid transcription factor-1 (TTF-1) expression and cytokeratin 7 (CK7) expression and to investigate the association of lymph node micrometastasis with the clinicopathological characteristics of patients with lung adenocarcinoma. A total of 54 patients with pN0 lung adenocarcinoma and whose primary tumors were positive for both TTF-1 and CK7 expression were included in this study. In total, 893 lymph nodes were obtained from these 54 patients and were analyzed for micrometastasis by immunohistochemical staining with anti-CK7 and anti-TTF-1 antibodies. CK7- and TTF-1-positive cells were found in the lymph nodes of 9 (16.7%) of 54 patients, and 21 (2.4%) of 893 lymph nodes exhibited positivity for these factors. No cells positive for both CK7 and TTF-1 were detected in the 5 lymph nodes obtained from patients with benign lung tumors. Lymph node micrometastasis was found to be associated with the differentiation grade and primary tumor position (P < 0.05). The detection of lymph node micrometastasis by a combination of CK7 and TTF-1 immunohistochemical staining provides a more accurate assessment of tumor staging for pN0 lung adenocarcinoma.

RESUMO

Sequence-known short-stranded hepatitis B virus (HBV) DNA fragment (181 bps) was obtained by PCR method. The strategy for its electrochemical detection was designed by covalently immobilizing single-stranded HBV DNA on gold electrode surface via carboxylate ester as a linkage between 3'-hydroxy end of DNA and carboxyl group of thioglycolic acid (TGA) self-assembled monolayer. The hybridization reaction on surface was evidenced by electrochemical methods using ferrocenium hexafluorophosphate (FcPF6) as an electroactive indicator. The interactions of Fc+ with single-stranded (ss) and double-stranded (ds) HBV DNA immobilized on TGA monolayer were studied. The difference between the responses of Fc+ at ss- and ds-DNA/Au electrodes suggested that this hybridization biosensor could be conveniently used to monitor DNA hybridization with a high sensitivity. AC impedance and XPS techniques have been employed to characterize the immobilization of ss-DNA on the gold surface.

The use of acidic mobile phase additives allows the chiral separation of underivatized phenylalanine analogs on a common amylosic column. In addition to decreasing retention and band-broadening arising from non-ideal interactions, acidic additives may also increase selectivity. This appears to be due to the minimization of non-selective binding in the recognition site. Effects of the additives are related to additive pKa and size. Ethanesulfonic acid was typically the most effective additive with trifluoroacetic acid being one of the least effective.

The use of basic mobile phase additives enhances the chiral separation of underivatized phenylalanine analogs on a common amylosic column. These additives appear to exert their effect through differential disruption of hydrogen binding involved in the recognition process. Several examples of amine increasing retention of the second eluting enantiomer while decreasing retention of the other enantiomer were observed. This gave dramatically increased selectivity and was most commonly observed with cyclopropylamine and cyclobutylamine. The effect was attributed to steric factors involved in the elution process.

RESUMO

Allele and genotype frequencies of four short tandem repeat loci were determined in a population sample from east China by using PCR and fluorescence techniques. After denaturing PAGE, 9 alleles were identified for CSF1PO, 8 alleles for TPOX, and 7 alleles for TH01. All loci met Hardy-Weinberg expectations. In addition, the theoretical values of these loci for paternity and forensic casework were evaluated.

RESUMO

We invented a stereoscopic localization apparatus for early diagnosis of lung cancer, facilitating fine needle aspiration biopsy especially for the deep-situated small lesion of the lung. Under X-ray, the stereoscopic localization of the three dimensions, posteroanterior sagittal and frontal plane, and transverse section of the body with small lesions can be soon established accurately with the apparatus. 227 percutaneous aspiration biopsies of lung lesions were performed by using the apparatus from 1985 to 1989. Of the 206 patients, 180 had lung lesions less than 3cm in size, and in 89.6% of the patients positive diagnosis was obtained by the first puncture biopsy. Satisfactory specimens were obtained in 96.6% of the biopsies. 26 malignant lesions about 1cm in diameter were localized, punctured and diagnosed accurately. They were subsequently confirmed surgically and pathologically. Mild pneumothorax occurred in 19 patients, and it subsided spontaneously.

RESUMO

A synthetic 55-residue peptide consisting of the carboxyl portion of the predicted genomic DNA sequence of Shope fibroma virus growth factor (SFGF residue 26-80) was found to exhibit epidermal growth factor-transforming growth factor activities in newborn mice. The synthetic SFGF accelerated precocious incisor eruption and eyelid opening in newborn mice and also retarded the overall growth rates of hair, body weight and body length when administered in dosages of 4 to 6 micrograms per gram of body weight. The results of whole animal studies indicate that SFGF belongs to the EGF-TGF alpha family and exerts similar biologic effects in newborn animals.
