RESUMO

Cordyceps cicadae, a medicinal fungus that is abundant in bioactive compounds such as N6-(2-hydroxyethyl)-adenosine (HEA) and polysaccharides, possesses remarkable anti-inflammatory, antioxidant, and nerve damage recovery properties. Deep ocean water (DOW) contains minerals that can be absorbed and transformed into organic forms by fungi fermentation. Recent studies have shown that culturing C. cicadae in DOW can enhance its therapeutic benefits by increasing the levels of bioactive compounds and minerals' bioavailibility. In this study, we investigated the effects of DOW-cultured C. cicadae (DCC) on brain damage and memory impairment induced by D-galactose in rats. Our results indicate that DCC and its metabolite HEA can improve memory ability and exhibit potent antioxidant activity and free radical scavenging in D-galactose-induced aging rats (p < 0.05). Additionally, DCC can mitigate the expression of inflammatory factors, such as tumor necrosis factor-alpha (TNF-α), interleukin-6 (IL-6), interleukin-1ß (IL-1ß), cyclooxygenase-2 (COX-2), and inducible nitric oxide synthase (iNOS), thereby preventing brain aging. Furthermore, DCC showed a significant decrease in the expression of the aging-related proteins glial fibrillary acidic protein (GFAP) and presenilin 1 (PS1). By reducing brain oxidation and aging-related factors, DOW-cultured C. cicadae demonstrate enhanced anti-inflammatory, antioxidant, and neuroprotective effects, making it a promising therapeutic agent for preventing and treating age-related brain damage and cognitive impairment.

During the last century, Grotta Romanelli (Southern Italy) has been a reference site for the European Late Pleistocene stratigraphy, due to its geomorphological setting and archaeological and palaeontological content. The beginning of the sedimentation inside the cave was attributed to the Last Interglacial (MISs 5e) and the oldest unearthed evidence of human occupation, including remains of hearths, was therefore referred to the Middle Palaeolithic. Recent surveys and excavations produced new U/Th dates, palaeoenvironmental interpretation and a litho-, morpho- and chrono-stratigraphical reassessment, placing the oldest human frequentation of the cave between MIS 9 and MIS 7, therefore embracing Glacial and Interglacial cycles. These new data provide evidence that the sea reached the cave during the Middle Pleistocene and human occupation occurred long before MISs 5e and persisted beyond the Pleistocene- Holocene boundary.

A new analytical method has been developed to determine atomic 236U/238U ratios in samples with only femtograms of 236U using a secondary electron multiplier (SEM) on a multicollector high-resolution inductively coupled plasma mass spectrometer (MC-ICPMS). The abundance sensitivity of the 238U tail at 236 atomic mass unit is reduced from 10-6 to 10-10 with the deployment of a retarding potential quadrupole lens. This method features the reduction of polyatomic interferences from hydride, nitride, lead, and plutonium and the evaluation of nonlinear SEM behavior. The instrument sensitivity is 1-2%, and the estimated methodological detection limit of the 236U/238U atomic ratio is as low as 2 × 10-10. Measurements on reference materials with 236U/238U ratios of 10-7-10-9, including the IRMM-075 series and the ETH Zurich in-house standard ZUTRI, demonstrate the accuracy of our MC-ICPMS technique. The analytical precisions (2σ) are ±4% for 5 fg of 236U at a 236U/238U of 1 × 10-8 and ±8% for 2 fg of 236U at a 236U/238U of 4 × 10-9 level. Compared to state-of-the-art accelerator mass spectrometry techniques and triple quadrupole-based ICPMS, our detection limit is not as low, but the required sample size is 3-40 times lower, and the throughput is as high as 3-4 samples per hour. The new MC-ICPMS-SEM technique is sensitive enough for determining 236U/238U in various small natural samples, such as marine carbonates and seawater.

Uranium (U) geochemistry and its isotopic compositions of reservoir sediments in U mine area were poorly understood. Herein, U and Th isotopic compositions were employed to investigate source apportionment and geochemical behavior of U in 41 reservoir sediments from a U mining area, Guangdong, China. The remarkably high contents of both total U (207.3-1117.7 mg/kg) and acid-leachable U (90.3-638.5 mg/kg) in the sediments exhibit a severe U contamination and mobilization-release risk. The U/Th activity ratios (ARs) indicate that all sediments have been contaminated apparently by U as a result of discharge of U containing wastewater, especially uranium mill tailings (UMT) leachate, while the variations of U/Th ARs are dominated by U geochemical behaviors (mainly redox process and adsorption). The U isotopic compositions (δ238U) showed a large variance through the sediment profile, varying from - 0.62 to - 0.04‰. The relation between δ238U and acid-leachable U fraction demonstrates that the U isotopic fractionation in sediments can be controlled by bedrock weathering (natural activity), UMT leachate (anthropogenic activity) and subsequent biogeochemical processes. The findings suggest that U-Th isotopes are a powerful tool to better understand U geochemical processes and enrichment mechanism in sediments that were affected by combined sources and driving forces.

The Indian Ocean Dipole (IOD) affects climate and rainfall across the world, and most severely in nations surrounding the Indian Ocean1-4. The frequency and intensity of positive IOD events increased during the twentieth century5 and may continue to intensify in a warming world6. However, confidence in predictions of future IOD change is limited by known biases in IOD models7 and the lack of information on natural IOD variability before anthropogenic climate change. Here we use precisely dated and highly resolved coral records from the eastern equatorial Indian Ocean, where the signature of IOD variability is strong and unambiguous, to produce a semi-continuous reconstruction of IOD variability that covers five centuries of the last millennium. Our reconstruction demonstrates that extreme positive IOD events were rare before 1960. However, the most extreme event on record (1997) is not unprecedented, because at least one event that was approximately 27 to 42 per cent larger occurred naturally during the seventeenth century. We further show that a persistent, tight coupling existed between the variability of the IOD and the El Niño/Southern Oscillation during the last millennium. Indo-Pacific coupling was characterized by weak interannual variability before approximately 1590, which probably altered teleconnection patterns, and by anomalously strong variability during the seventeenth century, which was associated with societal upheaval in tropical Asia. A tendency towards clustering of positive IOD events is evident in our reconstruction, which-together with the identification of extreme IOD variability and persistent tropical Indo-Pacific climate coupling-may have implications for improving seasonal and decadal predictions and managing the climate risks of future IOD variability.

Qualitative and quantitative assessments of fluid cycling are essential to address the role and transport of deeply sourced fluids in subduction systems. In this study, sediment cores distributed across a submarine mud volcano (SMV) offshore southwestern Taiwan were investigated to determine the characteristics of fluids generated through the convergence between the Eurasian and Phillippine Sea Plates. The low dissolved chloride concentration combined with the enrichment of 18O, and depletion of 2H of pore fluids suggest the discharge of deep freshwater formed by smectite dehydration at an equilibrium temperature of 100 to 150 °C. The upward fluid velocities, decreasing from 2.0 to 5.0 cm yr-1 at the center to a negligible value at margin sites, varied with the rate and efficiency of anaerobic methanotrophy, demonstrating the impact of fluid migration on biogeochemical processes and carbon cycling. By extrapolating the velocity pattern, the flux of fluids exported from 13 SMVs into seawater amounted up to 1.3-2.5 × 107 kg yr-1, a quantity accounting for 1.1-28.6% of the smectite-bound water originally stored in the incoming sediments. Our results imply that SMVs could act as a conduit to channel the fluids produced from great depth/temperature into seafloor environments in a subduction system of the western Pacific Ocean.

Denisovans are members of a hominin group who are currently only known directly from fragmentary fossils, the genomes of which have been studied from a single site, Denisova Cave1-3 in Siberia. They are also known indirectly from their genetic legacy through gene flow into several low-altitude East Asian populations4,5 and high-altitude modern Tibetans6. The lack of morphologically informative Denisovan fossils hinders our ability to connect geographically and temporally dispersed fossil hominins from Asia and to understand in a coherent manner their relation to recent Asian populations. This includes understanding the genetic adaptation of humans to the high-altitude Tibetan Plateau7,8, which was inherited from the Denisovans. Here we report a Denisovan mandible, identified by ancient protein analysis9,10, found on the Tibetan Plateau in Baishiya Karst Cave, Xiahe, Gansu, China. We determine the mandible to be at least 160 thousand years old through U-series dating of an adhering carbonate matrix. The Xiahe specimen provides direct evidence of the Denisovans outside the Altai Mountains and its analysis unique insights into Denisovan mandibular and dental morphology. Our results indicate that archaic hominins occupied the Tibetan Plateau in the Middle Pleistocene epoch and successfully adapted to high-altitude hypoxic environments long before the regional arrival of modern Homo sapiens.

We propose an improved method for precise sulfur isotopic measurements by multi-collector inductively coupled plasma mass spectrometry (MC-ICP-MS) in conjunction with a membrane desolvation nebulization system. The problems of sulfur loss through the membrane desolvation apparatus are carefully quantified and resolved. The method overcomes low intrinsic sulfur transmission through the instrument, which was initially 1% when operating at a desolvation temperature of 160 °C. Sulfur loss through the membrane desolvation apparatus was resolved by doping with sodium. A Na/S ratio of 2 mol mol-1 produced sulfur transmissions with 98% recovery. Samples of 3 nmol (100 ng) sulfur achieved an external precision of ±0.18‰ (2 SD) for δ34S and ±0.10‰ (2 SD) for Δ33S (uppercase delta expresses the extent of mass-independent isotopic fractionation). Measurements made on certified reference materials and in-house standards demonstrate analytical accuracy and reproducibility. We applied the method to examine microbial-induced sulfur transformation in marine sediment pore waters from the sulfate-methane transition zone. The technique is quite versatile, and can be applied to a range of materials, including natural waters and minerals.

Extensive use of pharmaceuticals and growth hormone in farm animal and live stocks has resulted in their frequent detection in soils, groundwater, and wastewater. The fate and transport of these compounds are strongly affected by their sorptive behavior to the soil minerals and humic materials. In this research, we conducted the sorption of tetracycline (TC), a common antibiotic, on palygorskite (PFL-1), a fibrous clay mineral of high surface area and high sorptivity towards organic compounds. The results showed that the sorption capacity of TC on PFL-1 was as high as 210 mmol/kg at pH 8.7. The sorption was relatively fast and reached equilibrium in 2h. Solution pH and ionic strength had significant effects on TC sorption. The sorption of TC by palygorskite is endothermic and the free energy of sorption is in the range of -10 to -30 kJ/mol, suggesting a strong physical sorption. The X-ray diffraction patterns before and after TC sorption revealed no changes in d-spacing and intensity under different pH and initial TC concentrations, indicating that the sorbed TC molecules are on the external surface of the mineral in contrast to intercalation of TC into swelling clays, such as montmorillonite. The small positive value of entropy change suggested that TC molecules are in disordered arrangement on palygorskite surfaces. Surface sorption of TC on PFL-1 is further supported by the derivative of gravimetric analysis and by the calculation of the amount of TC sorption normalized to the surface area. The results suggest that palygorskite could be a good candidate to remove TC from wastewater containing higher amounts of TC.
