RESUMO

Cryptobiosis describes the state of an organism whose body water is completely dehydrated and metabolic activity has become undetectable. Our study aimed to elucidate the physiological mechanism of cryptobiosis in the highest cryptobiotic invertebrate, Polypedilum vanderplanki. Larvae of this insect rapidly accumulated a large amount of the carbohydrate, trehalose, (18% of dry body mass) during desiccation for 2 days, suggesting that a high level of trehalose accumulation contributed to the successful induction of cryptobiosis in P. vanderplanki as well as in other lower cryptobiotic organisms. When larvae deprived of the brain, suboesophageal ganglion (SG) and thoracic ganglia (TG) were completely dehydrated and then rehydrated, they were able to recover and move actively. During desiccation, such larvae also accumulated trehalose, although only about half as much as the intact larvae. It is concluded that the brain, SG and TG do not affect the induction and termination of cryptobiosis, and hence in this higher multicellular animal cryptobiosis is independent of brain, SG and TG regulation, just as in plants or in unicellular organisms.

We investigated the effects of two ecdysteroids, ecdysone (E) and 20-hydroxyecdysone (20E), on silkworm larval development. Silkworm larvae, Bombyx mori, were fed an artificial diet supplemented with 20E during the fourth instar to promote premature molting. At the onset of the fifth instar, these precocious fifth-instar larvae were fed diets supplemented with either E or 20E to determine the effects of the two ecdysteroids on the morphology of midgut epithelial cells. Regeneration of midgut epithelial cells normally occurs only during the molting period. However, in larvae fed E, complete replacement of midgut epithelial cells was observed 24 h before the larvae entered apolysis. In larvae fed 20E, the morphology of midgut epithelial cells was disrupted, leading to death of the larvae during the fifth instar. We also observed similar differences in the effects of the two ecdysteroids in an in vitro experiment. These results suggest that E has a specific effect on the morphological change of midgut epithelial cells in precocious fifth-instar larvae that is distinct from 20E.

1-(4-Phenoxyphenoxypropyl)imidazole (KS-175), which has two types of characteristic moieties of insect growth regulators (IGRs), the phenoxyphenoxyalkyl group of juvenile hormone analogs (JHAs) and imidazole of 1,5-disubstituted imidazole such as KK-42, was tested for its biological activity on the silkworm, Bombyx mori. Penultimate (4th) instar larvae topically treated with KS-175 did not molt for more than 20 days. This activity was different from that reported for any IGRs. After the treatment, ecdysteroid levels in the hemolymph did not increase and the cells of the prothoracic gland had shrunk. When the treated penultimate larvae were fed an artificial diet supplemented with 20 ppm of 20-hydroxyecdysone, the larvae molted to the ultimate (5th) instar with a timing similar to that of control larvae fed a diet with or without 20-hydroxyecdysone. These results suggest that topical application of KS-175 irreversibly damages ecdysone biosynthesis in the prothoracic glands.

RESUMO

A new family member of insect defensin, an antibacterial peptide, has been isolated from larvae of a beetle, Allomyrina dichotoma. The peptide consisted of 43 amino acids and 6 cystein residues were conserved in the same position as that of other insect defensins. The new defensin was found to be inducible by bacterial injection. Analysis of the antibacterial spectrum of A. dichotoma defensin indicated that this peptide showed antibacterial activity against Gram-positive bacteria like Staphylococcus aureus and Bacillus subtilis but not against Gram-negative bacteria like Escherichia coli and Pseudomonas aeruginosa, indicating a typical spectrum of the insect defensin family. In addition, A. dichotoma defensin also exhibited antibacterial activity against methicillin-resistant S. aureus (MRSA) isolated from a patient.
