RESUMO

Cationic amphiphiles derived from trehalose have been synthesized; trehalose analogues substituted with n-pentyl or n-hexyl ethers exhibited membrane disrupting activities against clinically important Gram positive and Gram negative bacteria and fungi. Our results demonstrate that trehalose is a useful disaccharide scaffold for the development of broad-spectrum antimicrobial and antifungal agents.

In this work, the changes in expression of the adhesion molecules ICAM-1/LFA-1 on inflammatory cells of the liver were studied by immunohistochemistry. Mice sensitized with SEA and infected with S. mansoni and S. mansoni-infected controls were examined from day 35 to day 56 postinfection. A significant upregulation of ICAM-1 and LFA-1 in both the SEA group and the infected control group started shortly after egg deposition at day 35 and persisted up to day 56 p.i. Notably, both ICAM-1 and LFA-1 expression peaks were shifted earlier to day 38 p.i. in the SEA group compared to day 40 in the infected control group. The distribution of ICAM-1 and LFA-1 in both groups was comparable. At the early phase of infection before granuloma formation, both ICAM-1 and LFA-1 were detected along the sinusoidal wall of small blood vessels. At the acute cellular granuloma phase, they were homogeneously distributed all over the inflammatory cells, while at the chronic fibrocellular stage a non-homogeneous staining of granuloma cells at the periphery of the granuloma was apparent. The present data suggest that adhesion molecules play a role in the initiation and maintenance of granuloma formation. Thus, the granulomatous hyporesponsiveness induced by sensitization with SEA was associated with reduced expression of adhesion molecules.

This work studied the histopathological changes and the changes in the expression of macrophage adhesion molecule-1 (Mac-1) and macrophage inflammatory protein-1alpha (MIP-1alpha) in a murine model of soluble egg antigen (SEA) - induced granulomatous hyporesponsiveness. Histopathological results of hepatic sections in an SEA group showed early acceleration of ova destruction and markedly diminished granuloma cellularity with eosinophils and macrophages still being the predominant cells. Later, giant cells and pigmented macrophages that were scattered among granuloma cells and in intimate contact with the deposited eggs were more predominant in the SEA group than in the infected control group. Concurrently, the counts of Mac-1 positive cells were significantly increased in liver sections of the SEA group than the infected control group during the course of infection. MIP-1alpha showed early higher counts followed by lower counts in the later stages of infection on granuloma cells in the SEA group than the infected control group. During the course of infection, similar distribution of Mac-1 and MIP-1alpha was present in both groups. This study suggests that sensitization with SEA probably leads to enhancement of phagocytic activity of macrophages via increasing expression of Mac-1 and hence engulfment of ic3b coated schistosomal products such as ova. It leads to rapid destruction of ova and hence decreases the host inflammatory response to infection and amelioration of hepatic pathology which would be a promising approach in reduction of host morbidity and mortality.

RESUMO

The anti-miracidial potential of recombinant Schistosoma mansoni glutathione S-transferase 26 (rSmGST26) or native crude soluble egg antigens (SEA) was assessed. The associated dynamics of granuloma formation and immune responses were evaluated. Naive C57BL/6 mice were injected intravenously with multiple doses of either SEA (SEA-group) or rSmGST26 (GST-group) 7 days before cercarial infection. The immunized groups and the respective controls were sacrificed 6, 8 and 16 weeks postinfection (p.i.). Acceleration of ova destruction and reduction of granuloma diameter were greater in the GST-group than the SEA-group, mainly at 8 weeks p.i. However, the amelioration of hepatic pathology and function was more evident in the SEA-group. Concurrently, serum-specific IgG1 levels were elevated throughout the course of infection in the immunized groups compared to the infected controls. Initial rise of all splenic cytokines and serum anti-SEA IgE levels at 6 weeks p.i. was observed, followed by a dramatic drop in the levels of the proinflammatory cytokines IL-2, IFNgamma, IL-4 and TNF-alpha and IgE at 8 weeks of infection. IL-10 level was lower at 8 weeks p.i. than at 6 weeks, but was higher in immunized groups than in infected controls. Several responses may be implicated as an outcome of the present immunization protocol, such as increased levels of blocking antibody (IgG1) and IL-10 with decreased levels of proinflammatory cytokines and IgE.

A mouse monoclonal antibody (mAb) raised against serum immunoglobulins (Ig) of the snake. Psammophis sibilans stained in indirect immunofluorescence a proportion of snake splenic and peripheral blood lymphocytes, whereas it did not react with thymocytes, erythrocytes, brain, heart, lung, liver or kidney cells. The mAb, designated SR-2, combined in enzyme-linked immunosorbent assay (ELISA) with serum proteins of each of 20 individual P. sibilans tested. On Western blots of P. sibilans reduced whole serum proteins, purified Ig, or anti-rat erythrocyte (RRBC) antibodies eluted from glutaraldehyde-fixed RRBC, mAb SR-2 identified two bands of apparent molecular weight (m.w.) of 60,000 and 51,000 daltons. These bands were due to distinct polypeptides and not resulting from heterogeneous glycosylation of a single polypeptide, as they both were readily detected after periodate oxidation or endoglycosidase-F treatment of serum proteins and isolated Ig. MAb SR-2 bound to CNBr-activated Sepharose 4B precipitated from P. sibilans 125I-labeled serum proteins under non-reducing conditions a band that did not enter 7.5 or 9-16% gel and one of about 150,000 daltons. Under reducing conditions, two heavy bands of approximately 63,000 and 50,000 daltons and two light chains of apparent mass 23,000 and 20,000 bands were precipitated. The data presented provide, for the first time, substantial information on the molecular characteristics of snake Ig.

A family of glycoconjugates has recently been shown to share a common carbohydrate epitope recognized by the mouse monoclonal antibody HNK-1. The specificity of HNK-1 was found to be similar to that of another monoclonal antibody, NC-1. These two IgM monoclonal antibodies were raised after immunization of mice with a human T-cell line and avian neural crest-derived ganglia, respectively. The antigens recognized by these antibodies include the myelin-associated glycoprotein, MAG, a glycolipid of defined structure, and a set of molecules involved in cell adhesion. The timing and pattern of appearance of these antigens are distinct. Moreover, the epitope may be absent on an antigen at a given stage or in a given tissue. Therefore, although the molecules able to carry the NC-1/HNK-1 epitope are numerous and expressed in various tissues, the use of the monoclonal antibodies on tissue sections has proven adequate for following the migration of avian neural crest cells, the major cell lineage recognized by NC-1 and HNK-1 during early embryogenesis. Analogies in several other species have been found on the basis of HNK-1 reactivity. In this study we show that NC-1 and HNK-1 can be used successfully to label migrating neural crest cells in dog, pig and human. On the other hand, the NC-1/HNK-1 epitope was not present on migrating crest cells in amphibians or mice and was found only transiently on the neural crest of rats.

To study the ontogeny of lymphocytes in the lizard, Chalcides ocellatus antisera were raised in rabbits against thymocytes collected from adult lizards (AATS) or from embryos of stages 40 and 41 (AETS), and against serum immunoglobulins (AGGS). AATS recognized in adult lizards a surface membrane antigenic system (TA) specific to thymocytes and thymus-derived (T-) cells in spleen, and a distinct antigenic entity (Tt) found exclusively on intrathymic lymphocytes. After exhaustive absorption with adult thymocytes, AETS defined a further antigen (TE). On the other hand, AGGS combined with the immunoglobulin (Ig) determinants present in the cytoplasm and surface membrane of lizard presumptive B lymphocytes. Immunofluorescence assays during the development of the T and B lineage showed that TA+ and Tt+ thymocytes increased from 35% at stage 37 to 96% at stage 41, and were exported precociously to the embryonic splenic environment. Embryonic thymocytes were shown to totally lack surface Ig, whereas 40-50% of splenocytes carried cytoplasmic and/or surface membrane Ig determinants. The data revealed further the presence of a subset of splenocytes bearing the specific markers of both T and B cells.

Lymphocytes of thymus, spleen, peripheral blood (PB) and bone marrow (BM) collected from adult lizards, Chalcides ocellatus were cultured for 24 hr in the presence of 10(-3)M hydrocortisone acetate (HC) in order to assess the effect of in vitro HC on lizard T and B cell viability. The results indicated that HC induced stepwise, time-dependent mortality of the majority of thymocytes carrying T cell specific antigen(s) (TSA), 30-50% of T cells of spleen, PB and BM, and of a proportion of splenic B lymphocytes. Administration of 1 mg/g body weight HC to adult Ch. ocellatus lead to depletion of all TSA+ thymocytes. In contrast, T lymphocytes in the peripheral lymphoid compartments revealed both sensitivity and resistance to HC; similarly, B lymphocytes constituted susceptible and resistant subpopulations.

A single dose of 1 mg/g body weight of hydrocortisone acetate (HC) administered intraperitoneally to adult lizards, Chalcides ocellatus induced rapidly a reduction of about 85% of thymic lymphocytes. Histological evidence indicated that cortical, as well as, medullary thymocytes are sensitive to HC exposure. Around 40-50% of lymphocytes in peripheral blood (PB) and spleen were depleted at 3-7 days post-HC injection; such depletion durated about 4 weeks for PB but was rather temporary in spleen. Increase in number of bone marrow (BM) lymphocytes was negligible and transient and could by no way account for the dramatic cell losses in the different lymphoid tissues. The findings thus suggested that HC-mediated lymphocyte depletion in lizards is not attributable to redistribution between the different lymphoid compartments but rather to destruction. In direct conformation, lymphocytes were readily lysed in vitro by 10(-3)M HC, thymocytes being more vulnerable greater than PB greater than spleen greater than BM lymphocytes.

RESUMO

Major features of interest in the mature chondrocranium of Agama pallida are striking curvature of the nasal region, lack of the paranasal cartilage and concha nasalis, and presence of a cartilaginous roof over Jacobson's organ. In addition, the course of the ethmoid nerve deviates from the normal lacertilian pattern; there is no foramen epiphaniale, and the temporal region is reduced. The prefacial commissure and facial foramen lie in front of the cochlear portion of the auditory capsule, whereas the prominentia semicircularis anterior is separated from the rest of the otic capsule. Several chondrocranial characters are suggested to be unique to the agamids.
