RESUMO

Nowadays, the problem of inland water pollution is acute. It is caused by vast industrial growth and agricultural intensification. Concentrations of Cd, Pb, Zn, Cu, Mn, Fe, Mg, and Kwere determined in the muscles, liver, and gonads sabrefish from Rybinsk Reservoir areas with different anthropogenic loads. The tissue samples were analyzed by atomic absorption spectrometry. Heavy metals accumulated more intensively in the body of fish from more polluted areas of the reservoir. Among the analyzed elements, the maximum accumulation levels were found for K, Zn, and Fe and the minimum levels were observed for Cd and Pb. The gonads contained the largest concentration of Cd and Mn, the muscles contained the highest concentrations of Mg, and the other elements mainly accumulated in the liver of sabrefish. The THQ and HI values for all elements did not exceed 1, which suggests that there is no potential non-carcinogenic risk to human health. The target values of carcinogenic risk (TR) for cadmium ranged from 8.32 × 10-6 to 1.22 × 10-4 in the muscles. The increased content of cadmium in the gonads of sabrefish not only poses a risk to human health, but also to the reproduction of this species in the Rybinsk Reservoir.

RESUMO

Arabinogalactan depolymerization over solid Zr-containing SBA-15-based catalyst was studied via HPLC, GPC, and theoretical modeling. Arabinogalactans (AG) are hemicelluloses mainly present in larch wood species, which can be extracted on an industrial scale. The application of solid acid catalysts in the processes of hemicellulose conversion can exclude serious drawbacks such as equipment corrosion, etc. Characterization of 5%Zr-SBA-15 confirmed the successful formation of the mesoporous structure inherent to SBA-15 with fine Zr distribution and strong acidic properties (XRD, XPS, FTIR, pHpzc). Carrying out the process at 130 °C allowed us to achieve total products yield of up to 59 wt%, which is represented mainly by galactose (51 wt%) and minor (less than 9 wt%) presence of arabinose, furfural, 5-HMF, and levulinic acid. The temperature increases up to 150 °C resulted in a total product yield drop down to 37 wt%, making temperature elevation above 130 °C obsolete. According to the theoretical investigations, arabinogalactan depolymerization follows the primary cleavage of the ß(1→3) bonds between the D-galactose units of the main chain, which is also confirmed by GPC.

RESUMO

The bacteria Serratia proteamaculans 94 have a LuxI/LuxR type QS system consisting of AHL synthase SprI and the regulatory receptor SprR. We have previously shown that inactivation of the AHL synthase sprI gene resulted in an increase in the invasive activity of S. proteamaculans correlated with an increased bacterial adhesion. In the present work, the effects of inactivation of the S. proteamaculans receptor SprR are studied. Our results show that inactivation of the receptor sprR gene leads to an increase in bacterial invasion without any increase in their adhesion. On the other hand, inactivation of the sprR gene increases the activity of the extracellular protease serralysin. Inactivation of the QS system does not affect the activity of the pore-forming toxin ShlA and prevents the ShlA activation under conditions of a limited concentration of iron ions typical of the human body. While the wild type strain shows increased invasion in the iron-depleted medium, deletion of its QS system leads to a decrease in host cell invasion, which is nevertheless similar to the level of the wild type S. proteamaculans grown in the iron-rich medium. Thus, inactivation of either of the two component of the S. proteamaculans LuxI/LuxR-type QS system leads to an increase in the invasive activity of these bacteria through different mechanisms and prevents invasion under the iron-limited conditions.

RESUMO

Quorum Sensing (QS) system regulates gene expression in response to a change in the density of the bacterial population. Facultative pathogen Serratia proteamaculans 94 has a LuxI/LuxR type QS system consisting of regulatory protein SprR and AHL synthase SprI. Invasive activity of these bacteria appears at the stationary growth phase corresponding to a maximal density of the bacterial population in vitro. To evaluate the contribution of QS system of S. proteamaculans 94 to the regulation of invasive activity, in this work, S. proteamaculans SprI(-) mutant carrying the inactivated AHL synthase gene was used. Inactivation of the AHL synthase sprI gene resulted in a more than fourfold increase in the invasive activity of S. proteamaculans preceded by the increased adhesion of bacteria to the cell surface. This effect correlated with the increased expression of the outer membrane protein ompX gene and the decrease in the activity of intrabacterial protease protealysin, whose substrate is OmpX. The inverse correlation between activity of protealysin and bacterial invasion was also observed in the model experiments under the iron-limiting culture conditions. These results show that QS system regulates the S. proteamaculans invasion. This regulation can involve changes both in the protealysin activity and in the level of the ompX gene transcription.

RESUMO

In this study, we investigated the quorum sensing (QS) regulatory system of the psychrotrophic strain Serratia proteamaculans 94 isolated from spoiled refrigerated meat. The strain produced several N-acyl-L-homoserine-lactone (AHL) QS signal molecules, with N-(3-oxo-hexanoyl)-L-homoserine lactone and N-(3-hydroxy-hexanoyl)-L-homoserine lactone as two main types. The sprI and sprR genes encoding an AHL synthase and a receptor regulatory protein, respectively, were cloned and sequenced. Analysis of their nucleotide sequence showed that these genes were transcribed convergently and that their reading frames partly overlapped by 23 bp in the terminal regions. The genes were highly similar to the luxI/luxR-type QS genes of other Gram-negative bacteria. An spr-box (analog of the lux-box) was identified upstream of the sprR gene and found to be overlapped with the sequence of -10 sequence site in the promoter region of this gene. Inactivation of the sprI gene led to the absence of AHL synthesis, chitinolytic activity, and swimming motility; decrease of extracellular proteolytic activity; affected the cellular fatty acid composition; and reduced suppression of the fungal plant pathogen mycelium growth by volatile compounds emitted by strain S. proteamaculans 94. The data obtained demonstrated the important role of the QS system in the regulation of cellular processes in S. proteamaculans 94.

RESUMO

Air quality, like weather, can affect everyone, but responses differ depending on the sensitivity and health condition of a given individual. To help protect exposed populations, many countries have put in place real-time air quality nowcasting and forecasting capabilities. We present in this paper an optimal combination of air quality measurements and model outputs and show that it leads to significant improvements in the spatial representativeness of air quality. The product is referred to as multi-pollutant surface objective analyses (MPSOAs). Moreover, based on MPSOA, a geographical mapping of the Canadian Air Quality Health Index (AQHI) is also presented which provides users (policy makers, public, air quality forecasters, and epidemiologists) with a more accurate picture of the health risk anytime and anywhere in Canada and the USA. Since pollutants can also behave as passive atmospheric tracers, they provide information about transport and dispersion and, hence, reveal synoptic and regional meteorological phenomena. MPSOA could also be used to build air pollution climatology, compute local and national trends in air quality, and detect systematic biases in numerical air quality (AQ) models. Finally, initializing AQ models at regular time intervals with MPSOA can produce more accurate air quality forecasts. It is for these reasons that the Canadian Meteorological Centre (CMC) in collaboration with the Air Quality Research Division (AQRD) of Environment Canada has recently implemented MPSOA in their daily operations.

RESUMO

Phase formation in the NaF-KF-AlF3 system, in the vicinity of the K2NaAl3F12 composition, has been studied. The samples have been prepared by melting the starting components at 650 °C. A new phase has been revealed, which appeared to be a low-temperature form of the well-known K2NaAl3F12 ternary fluoride obtained by the hydrothermal synthesis method. The high-temperature form melts at 598 °C and is stable in a narrow temperature region of about 15 deg below the melting point. Thermal analysis, high temperature X-ray diffraction, IR-spectroscopy, X-ray fluorescence, and X-ray powder diffraction crystal structure analysis have been applied to study the composition, crystal structure, and thermal properties of the low-temperature phase. The crystal structure consists of trigonal-hexagonal two-dimensional (2D) grids built from the [AlF6] octahedra connected via vertices. The 2D grids have a specific wave-like conformation with a wavelength of 11.88 Å and an amplitude of 0.46 Å. There is a shift of the adjacent grids relative to each other. Because of this shift, the space between the grids changes. The shift leads to the formation of pores adapted to potassium and sodium ions. The reasons for the wave-like structure of layers are discussed. It is shown that the two polymorphic forms differ in the order of cation occupations.
