BACKGROUND: To determine whether items of the Chinese version of the Montreal Cognitive Assessment Basic (MoCA-BC) could discriminate among cognitively normal controls (NC), and those with mild cognitive impairment (MCI), mild Alzheimer's disease (AD), and moderate-severe (AD), as well as their sensitivity and specificity. METHODS: MCI (n = 456), mild AD (n = 502) and moderate-severe AD (n = 102) patients were recruited from the memory clinic, Huashan Hospital, Shanghai, China. NC (n = 329) were recruited from health checkup outpatients. Five MoCA-BC item scores were collected in interviews. RESULTS: The MoCA-BC orientation test had high sensitivity and specificity for discrimination among MCI, mild AD and moderate-severe AD. The delayed recall memory test had high sensitivity and specificity for MCI screening. The verbal fluency test was efficient for detecting MCI and differentiating AD severity. CONCLUSIONS: Various items of the MoCA-BC can identify MCI patients early and identify the severity of dementia.

RATIONALE: Absence or hypoplasia of the internal carotid artery (ICA) are rare developmental anomalies. Usually, patients with ICA agenesis are asymptomatic due to collateral circulation, but they may present with seizures, headache, or transient ischemic attack. We report a patient with right ICA absence in whom "paroxysmal right eye amaurosis" was the main symptom. PATIENT CONCERNS: A 76-year-old male patient suffered from "paroxysmal right eye amaurosis for 3 years". Three years prior, the patient had suffered sudden one-minute right eye amaurosis without any obvious cause. The attack reoccurred 1-2 times/year until one week before admission when he experienced two sudden right eye amaurosis. DIAGNOSIS: Congenital absence of the right ICA was diagnosed. In this patient with congenital absence of the right ICA, the ipsilateral anterior cerebral artery (ACA) was compensated by the anterior communicating artery (ACOM), and the ipsilateral middle cerebral artery (MCA) emerged from the carotid siphon of the contralateral ICA. INTERVENTIONS: The patient was given antiplatelet treatment consisting of aspirin and atorvastatin after admission and instructed to maintain the treatment after discharge. OUTCOMES: No symptom onset was observed during follow-up. LESSONS: Here, we report the patient's clinical manifestations and imaging findings and analyze the cause of the condition to provide a clinical reference for the study of congenital absence of the ICA.

Early diagnosis and therapy of cancer metastasis are of great importance for disease outcome. Circulating tumor cells (CTCs) offer the ability for noninvasive tumor profiling in real time. However, simply capturing and counting tumor cells are inadequate to provide valuable information about tumor. Efficiently releasing the captured cells is necessary for the downstream characterization. Herein, we describe a mild electrochemical strategy to effectively isolate CTCs from the bloodstream and rapidly release the captured cells in 2 min for downstream molecular characterization, as realized on a conductive poly(aminophenylboronic acid) derivatized electrode. The boronic ester linkage between dopamine (DA) and boronic acids-functionalized electrode is stable, and only upon the application of a weak potential perturbation does the boronic ester dissociate and release cells without compromising cell viability. This platform is reusable after acid treatment and has the potential to be the next-generation platform for cell capture and release, realizing the clinical value of CTCs as biomarkers.

We demonstrated a sensitive method for detection of glutathione (GSH) based on LSPR scattering spectrum using gold core-satellite nanostructure linked by T-Hg2+-T base pair. The core-satellite assembly caused coupling between plasmonic nanoparticles, which inducing distinct change of LSPR peak wavelength. As the interaction between Hg2+ and GSH, the core-satellite nanostructure would be disassembled, which accompanied with spectral blue-shift of the scattering spectrum. By using this method, GSH could be quantitatively detected, and the detection limits can reach to 0.1 µM.

We demonstrate a core-satellite plasmonic nanoprobe assembled via metal-ion-dependent DNA-cleaving DNAzyme linker for imaging intercellular metal ion based on plasmon coupling effect at a single-particle level. As metal ions are present in the system, the DNAzyme linker will be cleaved, and thus, disassembly of the core-satellite nanoprobes occurs, which results in distinct blue shift of the scattering spectra of Au core-satellite probes and naked color change of the scattering light. This change in scattering spectra has been supported by theoretical simulations. As a proof of concept, sensitive detection of Cu2+ with a limit of detection down to 67.2 pM has been demonstrated. The nanoprobes have been further utilized for intracellular Cu2+ imaging in living cells. The results demonstrate that the present strategy provides a promising platform for detection and imaging of metal ions in living cells and could be potentially applied to imaging other interesting target molecules simply by substituting the oligonucleotide sequence.

Electrochromic materials are widely used in smart windows. An ideal future electrochromic window would be able to control visible light transmission, tune building's heat conversion of near-infrared (NIR) solar radiation, and reduce attacks by microorganisms. To date, most of the reports have primarily focused on visible-light transmission modulation using electrochromic materials. Herein, we report the fabrication of an electrochromic-photothermal film by integrating electrochromic WO3 with plasmonic Au nanostructures and demonstrate its adjustability during optical transmission and photothermal conversion of visible and NIR lights. The localized surface plasmon resonance (LSPR) of Au nanostructures and the broadband nonradiative plasmon decay are proposed to be tunable using both the electric field and the WO3 substrate. Further enhanced photothermal conversion is achieved in colored state, which is attributed to coupling of traditional visible-band optical switching with NIR-LSPR extinction. The resulted electrochromic-photothermal film can also effectively reduce the numbers of attacking microorganisms, thus promising for use as a sterile smart window for advanced applications.

Silver nanoparticles (AgNPs) have been widely used as photocatalysts and nanosensors. Observation of the spectroscopy of a single AgNP greatly helps us understand the catalytic characteristics and morphology change of the AgNP during reactions. In the present study, AgNPs physically adsorbed on indium tin oxide (ITO) conductive glass were electrochemically reduced and oxidized, and the plasmonic resonance Rayleigh scattering (PRRS) spectrum of an individual AgNP was observed under a dark-field microscopy (DFM) equipped with a spectrometer. The electrochemical oxidization of the AgNP under constant potential caused a redshift of the PRRS peak for 30±5 nm. However, electrochemical reduction of the AgNP could not make the PRRS peak completely shift back to the initial position. In situ AFM and SEM characterization confirmed that very small Ag fragments (<10 nm) formed around the AgNP core during electrochemical oxidization. Results showed that dark-field microspectroscopy could be used as a sensitive tool for estimating the morphology/structural changes of nanoparticles that can hardly be observed through the cyclic voltammograms of multiple AgNPs.

RESUMO

The effective capture and release of circulating tumor cells (CTCs) is of significant importance in cancer prognose and treatment. Here we report a highly efficient method to capture and release human leukemic lymphoblasts (CCRF-CEM) using aptamers modified gold nanowire arrays (AuNWs). The gold nanowires, showing tunable morphologies from relatively random pillar deposit to relatively uniform arrays, were fabricated by electrochemical deposition using anodic aluminum oxide (AAO) as template. Upon simply being modified with aptamers by Au-S chemistry, the AuNWs exhibit higher specificity to target cells. Also compared to flat gold substrate, the AuNWs with nanostructure can capture target cells with much higher capture yield. Moreover, the captured CCRF-CEM cells can be released from AuNWs efficiently with little damage through an electrochemical desorption process. We predict that our strategy has great potential in providing a simple and economical platform for CTCs isolation, cancer diagnosis, and therapy.

OBJECTIVE: To compare the advantages and disadvantages between exercise challenge test (ECT) and methacholine challenge test (MCT) in the measurement of airway hyperresponsiveness (AHR), in order to identify a better and safer method to measure AHR. METHODS: Forty-seven children with controlled asthma after regular treatment were enrolled. ECT and MCT were performed for each child successively, and sensitivity was obtained through comparison with the golden standard (PD20). The occurrence of bronchospasm symptoms during the two tests was recorded. RESULTS: Taking PD20 as the gold standard, in children with moderate or severe AHR, the sensitivity of MCT (61%) for the measurement of AHR was significantly higher than that of ECT (9%) (P<0.05). The consistency between MCT results and PD20 was relatively high (κ=0.614), while the consistency between ECT results and PD20 was relatively low (κ=0.006). However, in the MCT, the incidence of bronchospasm symptoms was high and positively correlated with the incidence of cough and chest distress (P<0.05). CONCLUSIONS: MCT has a higher sensitivity for the measurement of AHR, but has a higher incidence of adverse events, compared with ECT in children with controlled asthma after regular treatment.

Efficient water splitting through electrocatalysis holds great promise for producing hydrogen fuel in modern energy devices. Its real application however suffers from sluggish reaction kinetics due to the lack of high-performance catalysts except noble metals such as platinum. Herein, we report an active system of plasmonic-metal Au nanorods/molybdenum disulfide (MoS2) nanosheets hybrids for the hydrogen evolution reaction (HER). The plasmonic Au-MoS2 hybrids dramatically improve the HER, leading to a ∼3-fold increase of current under excitation of Au localized surface plasmon resonance (LSPR). A turnover of 8.76 s(-1) at 300 mV overpotential is measured under LSPR excitation, which by far exceeds the activity of MoS2 catalysts reported recently. The HER enhancement can be largely attributed to the increase of carrier density in MoS2 induced by the injection of hot electrons of Au nanorods. We demonstrate that the synergistic effect of the hole scavengers can further facilitate electron-hole separation, resulting in a decrease of the overpotential of HER at MoS2 to ∼120 mV. This study highlights how metal LSPR activates the HER and promises novel opportunities for enhancing intrinsic activities of semiconducting materials.
