Recent studies that used adult faces as the baseline have revealed that attentional bias toward infant faces is the strongest for neutral expressions than for happy and sad expressions. However, the time course of the strongest attentional bias toward infant neutral expressions is unclear. To clarify this time course, we combined a behavioral dot-probe task with electrophysiological event-related potentials (ERPs) to measure adults' responses to infant and adult faces with happy, neutral, and sad expressions derived from the same face. The results indicated that compared with the corresponding expressions in adult faces, attentional bias toward infant faces with various expressions resulted in different patterns during rapid and prolonged attention stages. In particular, first, neutral expressions in infant faces elicited greater behavioral attentional bias and P1 responses than happy and sad ones did. Second, sad expressions in infant faces elicited greater N170 responses than neutral and happy ones did; notably, sad expressions elicited greater N170 responses in the left hemisphere in women than in men. Third, late positive potential (LPP) responses were greater for infant faces than for adult faces under each expression condition. Thus, we propose a three-stage model of attentional allocation patterns that reveals the time course of attentional bias toward infant faces with various expressions. This model highlights the prominent role of neutral facial expressions in the attentional bias toward infant faces.

The effect of the babyface schema includes three typical responses, namely, the preference response, viewing motivation, and attention bias towards infant faces. It has been theorised that these responses are primarily influenced by infants' facial structures. However, recent studies have revealed the moderating role of facial expression, suggesting that the strongest effect of the babyface schema may be related to the neutral facial expression; this hypothesis remains to be tested. In this study, the moderating role of facial expression was assessed in three successive experiments (total N = 402). We used a series of images of the same face with multiple expression-standardised images of infants and adults to control for facial structure. The results indicated that the effect sizes of the babyface schema (i.e., response differences between infants and adults) were different for multiple expressions of the same face. Specifically, the effect sizes of neutral faces were significantly greater than those of happy and sad faces according to the preference response (experiment 1, N = 90), viewing motivation (experiment 2, N = 214), and attentional bias (experiment 3, N = 98). These results empirically confirm that neutral infant facial expressions elicit the strongest effect of the babyface schema under the condition of using adult faces as a comparison baseline and matching multiple expressions of the same face.

Orf virus (ORFV) infects sheep and goat tissues, resulting in severe proliferative lesions. To analyze cellular protein expression in ORFV-infected goat skin fibroblast (GSF) cells, we used two-dimensional liquid chromatography-tandem mass spectrometry coupled with isobaric tags for relative and absolute quantification (iTRAQ). The proteomics approach was used along with quantitative reverse transcription polymerase chain reaction (RT-qPCR) to detect differentially expressed proteins in ORFV-infected GSF cells and mock-infected GSF cells. A total of 282 differentially expressed proteins were identified. It was found that 222 host proteins were upregulated and 60 were downregulated following viral infection. We confirmed that these proteins were differentially expressed and found that heat shock 70-kDa protein 1B (HSPA1B) was differentially expressed and localized in the cytoplasm. It was also noted that HSPA1B caused inhibition of viral proliferation, in the middle and late stages of viral infection. The differentially expressed proteins were associated with the biological processes of viral binding, cell structure, signal transduction, cell adhesion, and cell proliferation.

As a zoonotic disease, ovine contagious pustular dermatitis (Orf) is a serious threat to sheep as well as humans. Orf virus (ORFV) interferon resistance protein (VIR) is the principal virulence protein that encodes a dsRNA-binding protein to inhibit host antiviral response. p53 is one of the key proteins of the host antiviral innate immunity. It not only enhances type I interferon secretion but also induces apoptosis in infected cells, and plays a crucial role in the immune response against various viral infections. However, it remains to be elucidated what role p53 plays in ORFV replication and whether ORFV's own protein VIR regulates p53 expression to promote self-replication. In this study, we showed that p53 has an antiviral effect on ORFV and can inhibit ORFV replication. In addition, ORFV nonstructural protein VIR interacts with p53 and degrades p53, which inhibits p53-mediated positive regulation of downstream antiviral genes. This study provides new insight into the immune evasion mediated by ORFV and identifies VIR as an antagonistic factor for ORFV to evade the antiviral response.

Background: There issome research on school climate impact on adolescent alcohol use in the Western social context; however, there is no research on school climate impact on adolescent alcohol use in China. This study aimed to explore the effect of school climate on Chinese adolescents' alcohol use, and the moderating role of psychological suzhi between them. Methods: A total of 801 adolescents (45.8% boys, 14.96±1.66 years) completed self-reports on school climate, psychological suzhi, and alcohol use. Results: Moderation analyses revealed that both school climate and psychological suzhi significantly negatively predicted adolescents' alcohol use, and the interaction of school climate and psychological suzhi significantly positively predicted adolescents' alcohol use. Moreover, the effect of school climate on adolescents' alcohol use was stronger for low psychological suzhi adolescents than high psychological suzhi adolescents. Conclusions: We can build good school climate by formulating of national level legal and regulations and good social norms, and use mature interventions or cultivation strategies to improve adolescents' psychological suzhi in order to better play its role in protection.

RESUMO

Tumor progression locus 2 (TPL2) is a serine/threonine kinase that belongs to the MAP3K family. The activated TPL2 regulates the innate immune-relevant signaling pathways, such as ERK, JNK, and NF-κB, and the differentiation of immune cells, for example, CD4+ T and NK cells. Therefore, TPL2 plays a critical role in regulating the innate immune response. The present review summarizes the recent advancements in the TPL2-regulated innate immune response.

BACKGROUND/AIMS: This study aimed to investigate the anti-inflammatory activity of Berbamine (BER), a bisbenzylisoquinoline alkaloid extracted from Berberis amurensis (Xiao Bo An), and the underlying mechanisms. METHODS: Macrophages and neutrophils were treated with BER in vitro and stimulated with LPS and fMLP. The effects of BER on the expression of pro-inflammatory mediators in macrophages were evaluated with quantitative RT-PCR and ELISA. The effects of BER on the activation and superoxide release of neutrophils were determined with flow cytometry and WST-1 reduction test. The inhibitory effects of BER on the activation of signaling pathways related to inflammatory response in macrophages were evaluated by western blot analysis. In addition, a mouse peritonitis model was made by peritoneal injection of thioglycollate medium and anti-inflammatory effects of BER were investigated in vivo by quantitative analysis of pro-inflammatory factor production and leukocyte exudation. RESULTS: BER significantly inhibited inflammatory factor expression by LPS-stimulated macrophages and suppressed activation and superoxide release of fMLP-stimulated neutrophils. In the mouse peritonitis model, BER significantly inhibited the activation of macrophages and exudation of neutrophils. According to analysis, BER significantly suppressed phosphorylation of NF-κB and MAPK (JNK and ERK1/2) signaling pathways in LPS-stimulated macrophages. CONCLUSIONS: Collectively, data from this study suggest that BER has anti-inflammatory potential, which is effected via inhibition of NF-κB and MAPK signaling pathways, and thus holds promise for treatment of inflammatory disease.

Over the past 20 years, resilience theory has attracted great attention from both researchers and mental health practitioners. Resilience is defined as a process of overcoming the negative effects of risk exposure, coping successfully with traumatic experiences, or avoiding the negative trajectories associated with risks. Three basic models of resilience have been proposed to account for the mechanism whereby promotive factors operate to alter the trajectory from risk exposure to negative consequences: compensatory model, protective model, and inoculation model. Assets and resources are two types of promotive factors found to be effective in decreasing internalizing and externalizing problems. Considering the protective or compensatory role of assets and resources in helping youth be resilient against negative effects of adversity, resilience could be applied to Chinese migrant and left-behind children who are at risk for internalizing (e.g., depression, anxiety) and externalizing problems (e.g., delinquent behaviors, cigarette and alcohol use). Additionally, psychological suzhi-based interventions, a mental health construct for individuals that focuses on a strengths-based approach, can be integrated with resilience-based approach to develop more balanced programs for positive youth development.

A series of novel unsaturated five-membered benzo-heterocyclic amine derivatives were synthesized and assayed to determine their in vitro broad-spectrum antiviral activities. The biological results showed that most of our synthesized compounds exhibited potent broad-spectrum antiviral activity. Notably, compounds 3f (IC50=3.21-5.06 µM) and 3g (IC50=0.71-34.87 µM) showed potent activity towards both RNA viruses (influenza A, HCV and Cox B3 virus) and a DNA virus (HBV) at low micromolar concentrations. An SAR study showed that electron-withdrawing substituents located on the aromatic or heteroaromatic ring favored antiviral activity towards RNA viruses.

This study aims to explore the prevalence and influence factors of depressive symptoms in Chinese older adults from 1987 to 2012. The study investigated 81 published papers on depressive symptoms in Chinese older adults using the 30-item Geriatric Depression Scale (GDS-30) or the Center for Epidemiologic Studies Depression Scale (CES-D) as a measuring tool through meta-analysis. A total of 88,417 Chinese older adults were included in this review. The pooled prevalence of depressive symptoms in Chinese older adults was 23.6% (95% CI: 20.3-27.2%). The pooled prevalence of depressive symptoms was greater in women. In addition, the prevalence of depressive symptoms in married older adults was lower than in single adults (i.e., divorced, unmarried or widowed). Depressive morbidity with GDS-30 was significantly higher than with CES-D. The prevalence of depressive symptoms in Chinese older adults presented a downward tendency with increasing educational level. Moreover, the prevalence of depressive symptoms gradually increased with the year of data collection. Significant differences were found in the measurement tools, gender, degree of education and marital status, but none in age.

A series of novel (5-oxazolyl)phenyl amine derivatives were synthesized and their antiviral activities against the hepatitis C virus (HCV) and the coxsackie virus B3 (CVB3) and B6 (CVB6) were evaluated in vitro. Bioassays showed that the synthesized compounds 17a1, 17a4, 17a6, 17b1, 17d1, 17e2 and 17g3 exhibited potent antiviral activity against HCV (IC50 = 0.28-0.92 µM) and most synthesized compounds exhibited low cytotoxicity in Huh7.5 cells, compared to telaprevir. The compounds 17a1, 17a4, 17a5, 17a6, 17b1, 17b2, 17g1 and 17g3 showed strong activity against the CVB3 and/or CVB6 at low concentrations (IC50 < 2.0 µM). The (5-oxazolyl)phenyl amines 17a1, 17a4, 17a8, 17b1, 17d1, 17e2, 17f3 and 17g3 were identified as the most active on the biological assays, and will be studied further.

Event-related potentials were used to investigate the neural correlates of two-digit exact and approximate addition calculations. The results showed that the dissociation between exact and approximate calculations began at approximately 150 ms after stimulus. In the left hemisphere, N170 amplitude elicited by exact calculation was larger than that elicited by approximate calculation; however, in the right hemisphere, N170 amplitudes elicited by the two calculation strategies were not significantly different. Moreover, a larger P3 amplitude was elicited by exact calculation than by approximate calculation. In the 130-210 ms windows, dipole source analysis of the difference wave (exact calculation minus approximate calculation) indicated that the generator of N170 was localized in the left fusiform gyrus.

OBJECTIVE: To explore the status and influencing factors on anxiety sensitivity among middle school students in Chongqing. METHODS: 58 classes from 12 schools were randomly selected in four administrative districts of Chongqing city. A total number of 2700 students was included for final analysis including 48.5% from junior high school and 51.5% from senior high school students with 49.2% boys and 50.8% girls. The Chinese version of the Anxiety Sensitivity Index-Revision, Adolescent Self-Rating Life Events Check List (ASLEC) and State-Trait Anxiety Inventory (STAI) were used. RESULTS: (1) There was no significant difference between grade groups (P = 0.49). (2) The level of girl's anxiety sensitivity was always higher than boy's (P < 0.001). (3) Data from multiple linear regression showed that the influential factors to the degree of anxiety sensitivity were: state of anxiety, trait anxiety, life events, sex, stress from learning, etc (standard coefficients of regression were 0.258, 0.163, 0.112, 0.093, 0.124, -0.096, 0.096). CONCLUSION: The major influential factors of anxiety sensitivity would include: sex, stress from learning, life events, interpersonal relationship, state of anxiety and trait anxiety.

OBJECTIVE: To investigate the self-esteem level and social adaptation ability of hospitalized burn patients in our burn ward. METHODS: One hundred and twenty hospitalized burn patients in our burn ward were enrolled in the study and evaluated according to their sex, severity of burn injury and education level. Their self-esteem level and social adaptation ability were scored with the Felling of Inadequacy Scale and Abbreviated Burn Specific Health Scale. RESULTS: The general score of self-esteem of the patients with mild burns( 183+/-23) was obviously lower than that with moderate and severe burns (167+/-21 and 154 +/-24) , ( P <0.01). The self-esteem level of burn patients was different in different sex and education level. Among the self-esteem scores, male burn patients presented evidently higher scores of self evaluation, social ability, appearance, as well as the general score than those in the female ( P < 0.05). Moreover, the self evaluation score and study ability was higher in those with higher education level than those with lower education. Furthermore, the score of social adaptation ability was higher in the patients with mild burns than that in patients with moderate and severe burns ( P < 0. 01). The social adaptation ability and psychological function were much higher in male patients than those in female patients, but the former were weaker than the latter in regard to the body function. The psychological function, social relationship and general condition of the patients with lower education were better than those with higher education ( P <0. 05 ). CONCLUSION: There existed difference in the self-esteem and social adaptation ability in different burn patients during different periods.

A new quartz crystal microbalance immunoassay method based on a novel transparent immunoaffinity reactor was developed for clinical immunoassay. To construct such an affinity reactor, resonators with a frequency of 10 MHz were fabricated by affinity binding of functionalized gold nanoparticles (nanogold) to quartz crystal with immobilized specific ligand for the label-free analysis of the affinity reaction between a ligand and its receptor. [Recombinant human tumor markers, carcinoembryonic antigen (CEA) was chosen as a model ligand.] The binding of target molecules onto the immobilized antibodies decreased the sensor's resonant frequency, and the frequency shift was proportional to the CEA concentration in the range of 3.0-50 ng/ml with a detection limit of 1.5 ng/ml at a signal/noise ration of 3. A glycine-HCl solution (pH 2.3) was used to release antigen-antibody complexes from the biorecognition surface. Good reusability was exhibited. Moreover, spiking various levels of CEA into normal human sera was diagnosed using the proposed immunoassay. Analytical results show the precision of the developed immunoassay is acceptable, implying a promising alternative approach for detecting CEA in clinical immunoassay. Compared with the conventional enzyme-linked immunosorbent assay, the proposed immunoassay system was simple and rapid without multiple labeling and separation steps. Importantly, the proposed immunoassay system could be further developed for the immobilization of other antigens or biocompounds.

Mammary tumor virus (Mtv29)-encoded superantigen expressed by SJL/J mouse B cell lymphomas stimulates CD4+V16+ T cells and thereby acquires T cell help necessary for lymphoma growth. Mtv29 mouse mammary tumor virus env transcriptional activator (META) env-controlled Mtv29 superantigen (vSAg29) mRNA transcripts (1.8 kb) are not expressed in normal B or other somatic cells. Real-time PCR-based assays with DNA from normal SJL liver and vSAg29- lymphoma (cNJ101), digested with methylation-sensitive enzymes, showed hypermethylation at AvaI, FspI, HpaII, ThaI, and the distal HgaI sites of the META env, but vSAg29+ lymphoma cells showed significant demethylation at AvaI, HpaII, and the distal HgaI sites. The distal HgaI site that is adjacent to an Ikaros binding site is significantly demethylated in the META env DNA from primary lymphomas. Gel shift assays showed binding of Ikaros to a sequence representing this region in the META env. SJL lymphomas expressed the Ikaros isoform Ik6 that was absent in normal B cells. vSAg29+ cells exhibited increased DNaseI accessibility to chromatin at the vSAg29 initiation site. Treatment of cNJ101 cells with a demethylating agent, 5-azacytidine, and a histone deacetylase inhibitor, trichostatin A, caused hypomethylation at AvaI, HpaII, and distal HgaI sites and led to chromatin structural change at the vSAg29 initiation site, accompanied by the expression of vSAg29 transcripts. This enabled cNJ101 cells to stimulate SJL lymphoma-responsive CD4+V16+ T hybridoma cells. Thus, demethylation at the distal HgaI site of the Mtv29 META env permits vSAg29 expression, which may have an impact on the development of germinal center-derived B cell lymphomas of SJL/J mice.
