RESUMO

Haemocytes are crucial for the immune defence of mollusks. It is important to explore the immune performance of haemocytes of mollusks under the stress of heavy metals with global warming. In order to study the effects of cadmium (Cd) exposure and temperature stress on the haemocyte immune function of clam Ruditapes philippinarum, clams were exposed to different Cd concentrations (0.05, 0.10 and 0.25 mg/L) at 20 °C, 25 °C and 30 °C respectively. Haemocyte mortality, reactive oxygen species (ROS) and superoxide dismutase (SOD) activity were measured at day 1, day 3, day 5 and day 7. The results showed that the changes of the three indexes were not obvious when exposed to 0.05 mg/L of Cd at 20 °C, while significant differences were observed with the increase of temperature, Cd concentration and exposure time. Under a condition of relative high temperature coupling with high concentration of Cd, the clams were significantly influenced, showing an obvious synergistic effect. Selected indexes reflect the clam's response to the combined stress of temperature and Cd. Moreover, R. philippinarum might be an ideal biological index species to the Cd pollution.

RESUMO

Using energy plants to repair salinized soils polluted by petroleum is an efficient way to solve the problem of farmland reduction and prevent pollutants from entering the food chain simultaneously. In this study, pot experiments were conducted for the purposes of preliminarily discussing the potential of using an energy plant, sweet sorghum (Sorghum bicolor (L.) Moench), to repair petroleum-polluted salinized soils and obtain associated varieties with excellent remediation performance. The emergence rate, plant height and biomass of different varieties were measured to explore the performance of plants under petroleum pollution, and the removal of petroleum hydrocarbons in soil with candidate varieties was also studied. The results showed that the emergence rate of 24 of the 28 varieties were not reduced by the addition of 1.0 × 104 mg/kg petroleum in soils with a salinity of 0.31%. After a 40-day treatment in salinized soil with petroleum additions of 1.0 × 104 mg/kg, 4 potential well-performed varieties including Zhong Ketian No. 438, Ke Tian No. 24, Ke Tian No. 21 (KT21) and Ke Tian No. 6 with a plant height of >40 cm and dry weight of >4 g were screened. Obvious removal of petroleum hydrocarbons in the salinized soils planted with the four varieties were observed. Compared with the treatment without plants, the residual petroleum hydrocarbon concentrations in soils planted with KT21 decreased by 69.3%, 46.3%, 56.5%, 50.9% and 41.4%, for the additions of 0, 0.5 × 104, 1.0 × 104, 1.5 × 104 and 2.0 × 104 mg/kg, respectively. In general, KT21 had the best performance and application potential to remediate petroleum-polluted salinized soil.

RESUMO

With global warming, high-temperature stress has become an essential abiotic factor affecting plant growth and survival. However, little knowledge was available of the molecular mechanism that aquatic plants respond to this stress. In the present study, we explore the adaptation mechanism of Spirodela polyrhiza, a surface-water-grown duckweed species broadly distributed worldwide to high temperatures, and analyze its gene expression pattern of S. polyrhiza under heat stress. Three temperature stress treatments, including room temperature group (CK), middle high-temperature group (MTS), and high-temperature group (45 °C, HTS) were set. The results showed that the contents of SOD first increased and then decreased, and those of MDA showed an upward trend under elevated high-temperature stress. According to the transcriptome data, 3145, 3487, and 3089 differently expressed genes (DEGs) were identified between MTS and CK, HTS and CK, and HTS and MTS, respectively. The transcription factors (TFs) analysis showed that 14 deferentially expressed TFs, including HSF, ERF, WRKY, and GRAS family, were responsive to heat stress, suggesting they might play vital roles in improving resistance to heat stress. In conclusion, S. polyrhiza could resist high temperatures by increasing SOD activity and MDA at the physiological level. Several transcription factors, energy accumulation processes, and cell membranes were involved in high-temperature stress at the molecular level. Our findings are helpful in better grasping the adaptation rules of some aquatic plants to high temperatures.

RESUMO

Palmitoleic acid, a monounsaturated fatty acid which could affect glucose and lipid metabolism and reduce insulin resistance has two isomers, i.e. cis-palmitoleic acid (cPOA) and trans-palmitoleic acid (tPOA). However, the pharmacokinetic, metabolic transformation and structure-activity relationship of the two isomers have not been reported. A precise and accurate ultra performance liquid chromatography-tandem mass spectroscopy (UPLC-MS/MS) method was developed to determine cPOA and tPOA simultaneously. Both the cPOA and tPOA were administered i.g. (intragastric gavage) to rats at 75 mg/kg. Serum samples were collected and analyzed for the two isomers by UPLC-MS/MS on a reverse-phase BDS C18 column equilibrated and eluted with water (A) and acetonitrile (B) at a flow rate of 0.3 mL/min. The calibration curves for cPOA and tPOA were linear over the range 0.1-12 µg/mL. Analytes were monitored by selected-reaction monitoring in negative electrospray ionization mode. The Tmax of cPOA was 0.94 ± 0.44 h and the Cmax 8.17 ± 1.97 µg/L, and the Tmax and Cmax of tPOA were 1.50 ± 0.98 h and 14.77 ± 11.91 µg/L, respectively. AUC0-24 h of cPOA and tPOA were 59.45 ± 29.83 and 113.88 ± 72.25 mg/L·h. The method was applied in pharmacokinetic study of cPOA and tPOA in rat serum successfully. Besides, the concentrations of cPOA and tPOA in rat serums were observed fluctuating with a consistent trend, which may be due to reciprocal bio-convert in the body.

RESUMO

Considering the phasing-out of polybrominated diphenyl ethers (PBDEs), environmental concerns of PBDE alternatives and dechlorane plus (DP) are rising. Accordingly, this study investigates occurrence and ecological risks of PBDEs, PBDE alternatives and DPs in sediments of two littoral regions, the Pearl River Estuary (PRE) and Daya Bay (DYB), in southern China. Total PBDEs concentrations in surface sediments of the PRE and DYB were in the range (mean) of 0.30-28.7 (8.71) and 0.29-43.4 (6.05) ng/g dw, respectively. DP levels in surface sediments of the PRE (0.004-0.27 ng/g dw) were significantly higher than those in the DYB (0.005-0.24 ng/g dw) (p < 0.05). BDE 209 was the predominant component, followed by DBDPE, exhibiting regional variations in BFRs usage. Vertical profiles of BFRs and DP in the PRE and DYB sediment cores exhibited clear anthropogenic influences. Risk quotients suggest critical ecological risks of tetra-, penta- and deca-BDE congeners in all the surface sediments.

RESUMO

As a typical invasive species, Spartina alterniflora is widely recognized as one of the primary threats to biodiversity in various habitats, including wetlands. Although the invasion by S. alterniflora has been managed in multiple ways, it may reappear after treatment. How S. alterniflora affects the soil microbial community in coastal wetlands during its regeneration process has not yet been clarified. Here, rhizosphere soil samples (RSPs) and bulk soil samples (SSPs) were collected in the S. alterniflora community and a high-throughput sequencing method was conducted to analyze the composition and diversity of soil microorganisms. Meanwhile, we also obtain the soil physicochemical properties. In the present study, there was no significant difference in the alpha diversity of both bacterial and fungal communities in the SSP and RSP groups. The PCoA (principal coordinate analysis) also showed that the microbial community structure did not differ significantly between the SSP and RSP groups. The results showed that except for pH, the total sulfur (TS) content, total nitrogen (TN) content, and electrical conductivity (EC) did not differ significantly (p > 0.05) between the SSP and RSP groups. The composition of the bacterial and fungal community in the rhizosphere of S. alterniflora was similar to that found in the surrounding soils. The top two dominant bacterial phyla were Proteobacteria and Desulfobacterota in the present study. Venn diagram results also support this view; most OTUs belong to the common OTUs of the two groups, and the proportion of unique OTUs is relatively small. The LEfSe (LDA effect size) analysis showed that Campylobacterota (at the phylum level) and Sulfurimonas (at the genus level) significantly increased in the RSP group, implying that the increased Sulfurimonas might play an essential role in the invasion by S. alterniflora during the under-water period. Overall, these results suggest that the bacterial and fungal communities were not significantly affected by the S. alterniflora invasion due to the short invasion time.

RESUMO

In the present study, winter jujube organs including fruit, fruiting leaf and foliage leaf, and associated soils in 14 typical orchards in Binzhou City, Shandong Province, China were collected and determined for the mass fractions of Co, Ni, Cu, Zn, and Cd. The mass fractions of Co, Ni, Cu, Zn, and Cd in plant tissues generally showed an order of Cu > Zn > Ni > Co > Cd as well as those in the soils decreased as Zn > Cu > Ni > Co > Cd. The values of single factor index and Nemerow pollution index suggested the jujube fruits were not polluted by heavy metals. Values of estimated daily intake for all the elements were far below their associated acceptable reference values, indicating no health risks would be caused by a single trace element. The results of targeted hazard quotient (THQ) of the metals in the fruits decreased as Cu > Ni > Zn > Cd accompanying total THQ (TTHQ) lower than 1 showing no hazard would be caused by those metals. Correlation analysis showed soil might not be the main source of heavy metals in winter jujube organs. Bioaccumulation factors (BAFs) for Co, Ni, Zn and Cd in fruits and leaves were far below 1 suggesting their low bioavailablities. The relatively great BAFs of Cu in the leaves might be due to the application of fertilizers and pesticides containing great amounts of Cu through soil and foliar spraying. To sum up, heavy metals tended not to be a major threat to winter jujube cultivation, and winter jujube had great edible safety.

RESUMO

Heavy metals are major pollutants that pose threats to wetland environments. In the present study, surface sediments from wetlands vegetated by invasive species Spartina alterniflora in the Yellow River Delta were collected and determined for the mass fractions of Co, Ni, As, Cd and Pb. Results showed mass fractions of Co, Ni, As, Cd and Pb in the sediments of the S. alterniflora communities ranged from 8.5 to 16.0, 13.9−27.9, 3.2−13.8, 0.08−0.24, and 17.6−37.5 mg/kg dw, respectively, generally presenting an order of Pb > Ni > Co > As > Cd. The levels of heavy metals in sediments in the S. alterniflora communities were higher than those in the wetland vegetated by the native plant species Suaeda heteroptera. Correlations among metal elements were highly significant, suggesting that they might have the same sources. Clay and TOC were important factors affecting the spatial distribution of metals. The Igeo values of the investigated elements in the sediments were frequently lower than 0, revealing the slight pollution status of these metals. Relatively slight values of Eri and RI suggested that the potential ecological risks caused by the 5 metals were low. Our findings could provide a better understanding of the correlation between metal pollution and bio-invasion in wetland ecosystems.

RESUMO

Spartina alterniflora invasion has negative effects on the structure and functioning of coastal wetland ecosystems. Therefore, many methods for controlling S. alterniflora invasion have been developed. S. alterniflora control methods can affect plant community, which results in changes in microbial communities and subsequent changes in soil ecological processes. However, the effects of controlling S. alterniflora on soil microbial communities remain poorly understood. We aimed to examine the responses of bacterial and fungal communities to invasion control methods (cutting plus tilling treatment: CT; mechanical rolling treatment: MR). Soil bacterial and fungal community diversity and composition structure were assessed using high-throughput sequencing technology. The findings of the study showed that bacterial diversity and richness in the CT treatment reduced substantially, but fungal diversity and richness did not show any remarkable change. Bacterial and fungal diversity and richness in the MR treatment were not affected considerably. In addition, the two control methods significantly changed the soil microbial community structure. The relative abundance of bacteria negatively associated with nutrient cycling increased considerably in the CT treatment. The considerable increases in the relative abundance of certain bacterial taxa in the MR treatment may promote soil nutrient cycling. Compared with mechanical rolling, soil bacterial community diversity and structure were more sensitive to cutting plus tilling.

RESUMO

Parthenium hysterophorus L., as an invasive plant, has negatively impacted the ecosystem functioning and stability of the terrestrial ecosystem in China. However, little information was available for its effects on microorganisms in the Yellow River Delta (YRD), the biggest newly-formed wetland in China. In the present study, high-throughput sequencing technology was used to obtain the bacterial community in soils and roots of different plant species, including P. hysterophorus and some native ones in the YRD. Our results showed that the Proteobacteria, Acidobacteriota, Gemmatimonadota, and Actinobacteriota were dominant in the rhizosphere soils of P. hysterophorus (84.2%) and Setaria viridis (86.47%), and the bulk soils (80.7%). The Proteobacteria and Actinobacteriota were dominant within the root of P. hysterophorus. A total of 2468 bacterial OTUs were obtained from different groups among which 140 were observed in all the groups; 1019 OTUs were shared by P. hysterophorus non-rhizosphere soil bacteria (YNR) P. hysterophorus rhizosphere soil bacteria (YRR) groups. The indexes of the ACE (823.1), Chao1 (823.19), Simpson (0.9971), and Shannon (9.068) were the highest in the YRR groups, showing the greatest bacterial community diversity. Random forest analysis showed that the Methylomirabilota and Dadabacteria (at the phylum level) and the Sphingomonas, and Woeseia (at the genus level) were identified as the main predictors among different groups. The LEfSe results also showed the essential role of the Acidobacteriota in the YRR group. The SourceTracker analysis of the bacterial community of the YRR group was mainly from GBS groups (average 53.14%) and a small part was from YNR groups (average 6.56%), indicating that the P. hysterophorus invasion had a more significant effect on native plants' rhizosphere microorganisms than soil microorganisms. Our observations could provide valuable information for understanding the bacterial diversity and structure of the soil to the invasion of P. hysterophorus.

RESUMO

BACKGROUND: Anterior serratus plane block for analgesia in many procedures, but there have been no reports of analgesia undergoing laparoscopic cholecystectomy (LC). We investigated the effects of ultrasonic-guided low serratory anterior block on patients undergoing LC. METHODS: One hundred patients who undergo LC were selected and randomly divided into 2 groups: Patients in group C with routine general anesthesia and patients in group S treat low anterior serratus block combined with general anesthesia. The serratus anterior block was performed at the T8 to T9 point after anesthesia induction, before cutting leather (T0), stamp card placement (T1), 2 groups of patients' heart rate (HR), blood pressure were recorded, at the same time dosage of analgesic drugs and postoperative 0.5, 6, 12, 24 hours when resting visual analogue scale (VAS) scores were recorded in 2 groups of patients. RESULTS: The T0 point, patients' HR, blood pressure had significant difference (P > .05), the T1 point, patients' HR, mean arterial pressure in group S lower than group C (P < .05), the amount of intra-operative propofol and remifentanil, patients in group S were less than in group C (P < .05), and resting VAS pain score at the 0.5, 6, 12 hours after operation patients in group S were lower than group C also (P < .05), resting VAS pain had no significant difference at postoperative 24 hours between 2 groups (P > .05). CONCLUSION: Low serratus anterior plane block in LC can provide safe and effective analgesia for patients.

RESUMO

BACKGROUND: Sevoflurane (Sev) has been reported to inhibit cancer development, and sevoflurane treatment in cancers is implicated with the deregulation of specific non-coding RNAs (ncRNAs). This study aimed to investigate the relationship between sevoflurane and circular RNA reelin (circRELN) in glioma. METHODS: The expression of circRELN, microRNA-1290 (miR-1290) and RAR-related orphan receptor A (RORA) was measured by quantitative real-time PCR (qPCR). Cell proliferative capacity was assessed by cell counting kit-8 (CCK-8) and colony formation assays. Cell apoptosis and cell cycle distribution were monitored by flow cytometry assay. Cell migration was assessed by wound healing assay and transwell assay, and cell invasion was assessed by transwell assay. The protein levels of matrix metalloproteinase-2 (MMP2), MMP9 and RORA were quantified by western blot. Tumor growth in vivo was assessed by Xenograft models. The binding relationship between miR-1290 and circRELN or RORA was verified by dual-luciferase reporter assay and RNA immunoprecipitation (RIP) assay. RESULTS: We found that circRELN expression was declined in glioma tissues and cells, while Sev treatment enhanced circRELN expression. In function, Sev notably inhibited glioma cell proliferation, migration and invasion and promoted apoptosis and cell cycle arrest, while circRELN knockdown reversed these effects. MiR-1290 served as a target of circRELN, and glioma cell malignant phenotypes recovered by circRELN knockdown were partly repressed by miR-1290 deficiency. In addition, RORA was a target of miR-1290, and glioma cell malignant phenotypes promoted by miR-1290 restoration were partly blocked by RORA overexpression. CircRELN regulated RORA expression by targeting miR-1290. In Xenograft models, Sev inhibited tumor growth by upregulating circRELN. CONCLUSION: Sev blocked the progression of glioma by increasing circRELN expression, and circRELN played roles in glioma partly by regulating the miR-1290/RORA network.

RESUMO

Twenty biota species were collected from the Northern Beibu Gulf to understand the heavy metal pollution status and biomagnification characteristics. Mean concentrations (µg/g) of Mn, Zn, Pb, Cr, Ni, As, Cu, and Cd in the biota species were 0.99, 38.33, 0.14, 0.079, 0.085, 51.10, 7.92, and 0.21, respectively. As levels in most biota species exceeded the corresponding guidelines. Crabs were detected with high metal accumulation ability for each heavy metal except Pb. The ranges of δ15N and δ13C in organisms were from 8.0‰ to 15.6‰ and from -21.4‰ to -15.6‰, respectively. Cr, Mn, Ni, and As showed potential biomagnification trends in the food webs. The EDI values of these elements decreased by the sequence of As > Zn > Cu > Mn > Cd > Pb > Ni > Cr. High THQ and CR values for As indicated a potential health risk by consumption of these aquatic products.

RESUMO

Due to the inefficient reproduction of microorganisms in oxygen-deprived environments of the reservoir, the applications of microbial enhanced oil recovery (MEOR) are restricted. To overcome this problem, a new type of air-assisted MEOR process was investigated. Three compounding oil degradation strains were screened using biochemical experiments. Their performances in bacterial suspensions with different amounts of dissolved oxygen were evaluated. Water flooding, microbial flooding and air-assisted microbial flooding core flow experiments were carried out. Carbon distribution curve of biodegraded oil with different oxygen concentration was determined by chromatographic analysis. The long-chain alkanes are degraded by microorganisms. A simulation model was established to take into account the change in oxygen concentration in the reservoir. The results showed that the optimal dissolved oxygen concentration for microbial growth was 4.5~5.5mg/L. The main oxygen consumption in the reservoir happened in the stationary and declining phases of the microbial growth systems. In order to reduce the oxygen concentration to a safe level, the minimum radius of oxygen consumption was found to be about 145m. These results demonstrate that the air-assisted MEOR process can overcome the shortcomings of traditional microbial flooding techniques. The findings of this study can help for better understanding of microbial enhanced oil recovery and improving the efficiency of microbial oil displacement.

RESUMO

Seven heavy metals including Hg, Cu, Pb, Cd, Zn, Cr, and As were examined in seventeen marine nekton species from the outer Pearl River Estuary (PRE), South China Sea. On the wet weight basis, the metal concentration ranges were 0.016-0.157 µg/g for Hg, 0.18-14.3 µg/g for Cu, 0.26-1.48 µg/g for Pb, 0.021-0.873 µg/g for Cd, 1.35-57.15 µg/g for Zn, 0.15-0.53 µg/g for Cr, and 0.42-7.83 µg/g for As, respectively. The levels of tested metals except for Pb in crustaceans were found to be higher than those in fish and cephalopods, suggesting that the diet and habitat played important roles on heavy metal accumulation ability of marine organism. Except for Cd in Champsodon capensis, Calappa lophos, and Portunus argentatus, all the left metal concentrations of investigated nekton species were below their permissible upper limits, indicating that consumption of examined marine nekton should be considered as safe for human health. The values of single target hazard quotient (THQ) and total THQ were all less than 1 and also suggested that there was no health risk for consumption. Even so, the local people should control their daily intake of crustacean foods from the outer PRE, since there might be potential As and Cd cumulative risks.

RESUMO

Tetrabromobisphenol A (TBBPA) and hexabromocyclododecanes (HBCDs) were examined in sediments and biota species from two mangrove wetlands of the Pearl River Estuary (PRE) and the Jiulong River Estuary (JRE), South China, to investigate their distribution, bioaccumulation and biomagnification in mangrove food webs. Levels of TBBPA and ΣHBCD (sum of α-, ß- and γ-HBCDs) ranged from 0.003 to 0.31 and not detected (nd) to 1.11 ng/g dry weight in the sediments, and from 0.56 to 22.1 and nd to 56.3 ng/g lipid weight in the biota species, respectively. γ-HBCD was the major diastereoisomer in the PRE sediments, while α- and γ-HBCDs predominated in the JRE sediments. In contrast, α-HBCD was dominant in the biota. Mean enantiomeric fractions (EFs) of α-, ß- and γ-HBCDs in the sediments all followed the trend of JRE > racemic standard > PRE. A significant enrichment of (-)-α-HBCD was found in the biota (p = 0.04), with EFs in the range of 0.297-0.485. Bioaccumulations were seen for TBBPA and α-HBCD as their biota-sediment accumulation factors (BSAFs) were greater than 1. (-)-α-HBCD had significantly greater BSAFs than (+)-α-HBCD (p = 0.04), indicating the preferential bioaccumulation of (-)-α-HBCD. Biomagnification factors (BMFs) of TBBPA ranged from 0.83 to 1.51, which varied among feeding relationships and mangroves. Positive relationships were found between TBBPA concentrations and trophic levels of the biota species with trophic magnification factors of 2.17 for the PRE and 1.22 for the JRE, suggesting that TBBPA biomagnifies in the mangrove food webs. No biomagnifications were observed for ΣHBCD, α-HBCD and its enantiomers.

RESUMO

Accumulating evidence has elucidated that human mesenchymal stem cells (hMSCs) exert profound analgesic effects on numerous animal models of neuropathic pain, including drug-induced peripheral nerves, diabetes-induced neuropathy, and chronic constriction injury. We aimed to address whether forcing expression of sirtuin 1 (SIRT1) can enhance the efficacy of hMSCs on alleviation of pain sensation. A rat model of chronic constriction injury (CCI) mimicking peripheral nerve injury was incorporated in the study. Paw withdrawal threshold (PWT) and paw withdrawal latency (PWL) testing were used to measure pain-related behavior. Our results demonstrated that SIRT1 was decreased post-CCI surgery. Compared to hMSCs-control implantation, the hMSCs-SIRT1 (hMSCs overexpressing SIRT1) implantation exhibited superior effects on reducing pro-inflammatory cytokine levels in serum and spinal dorsal horn, while ameliorating neuropathic pain in CCI rat. Engineering hMSCs such as overexpressing SIRT1 may serve as a promising strategy for the treatment of patients with neuropathic pain.

RESUMO

Aquatic biota including fish, shrimp and bivalves were collected from the Yellow River Delta (YRD), China to investigate the levels, composition profile and dietary exposure of polybrominated diphenyl ethers (PBDEs), decabromodiphenyl ethane (DBDPE) and dechlorane plus (DP). The concentrations of PBDEs, DBDPE and DP in the organisms ranged from 5.3 to 149, not detected (nd) - 49, and 0.5-29 ng/g lipid weight, respectively. Higher levels of PBDEs and DP were found in mullet (Liza haematocheila).PBDEs were the major pollutants and BDE 209 was the predominant congener of PBDEs suggesting the great production and application of deca-BDE in YRD. The average fanti values for different species were similar to or a little lower than that of the commercial DP, suggesting syn-DP might be selectively accumulated by the organisms. The estimated daily intake values of HFRs suggested consuming fish was the main pathway for the exposure of halogenated flame retardants.

RESUMO

Microbial fuel cell (MFC) technology is a simple way to accelerate the treatment of the oily sludge which is a major problem affecting the quality of oil fields and surrounding environment while generating electricity. To investigate the oil removal and the characteristics of changes in the composition of bacteria, sediment microbial fuel cells (SMFCs) supplemented with oily sludge was constructed. The results showed that the degradation efficiency of total petroleum hydrocarbon (TPH) of SMFC treatment was 10.1 times higher than the common anaerobic degradation. In addition, the degradation rate of n-alkanes followed the order of high carbon number > low carbon number > medium carbon number. The odd-even alkane predominance (OEP) increased, indicating that a high contribution of even alkanes whose degradation predominates. The OUT number, Shannon index, AEC index, and Chao1 index of the sludge treated with SMFC (YN2) are greater than those of the original sludge (YN1), showing that the microbial diversity of sludge increased after SMFC treatment. After SMFC treatment the relative abundance of Chloroflexi, Bacteroidia and Pseudomonadales which are essential for the degradation of the organic matter and electricity production increased significantly in YN2. These results will play a crucial role in improving the performance of oily sludge MFC.
