RESUMO

The development of new sensors for on-site food toxin monitoring that combine extraction, analytes distinction and detection is important in resource-limited environments. Surface-enhanced Raman scattering (SERS)-based signal readout features fast response and high sensitivity, making it a powerful method for detecting mycotoxins. In this work, a SERS-based assay for the detection of multiple mycotoxins is presented that combines extraction and subsequent detection, achieving an analytically relevant detection limit (∼ 1 ng/mL), which is also tested in corn samples. This sensor consists of a magnetic-core and mycotoxin-absorbing polydopamine-shell, with SERS-active Au nanoparticles on the outer surface. The assay can concentrate multiple mycotoxins, which are identified through multiclass partite least squares analysis based on their SERS spectra. We developed a strategy for the analysis of multiple mycotoxins with minimal sample pretreatment, enabling in situ analytical extraction and subsequent detection, displaying the potential to rapidly identify lethal mycotoxin contamination on site.

Copy number variation (CNV) refers to a kind of structural variation, having functional and evolutionary effects on phenotypes. Thus far, further elucidation of the CNVs in different Chinese indigenous cattle breeds by whole genome sequencing have yet not been done. In this study, a comprehensive genomic analysis was performed on 75 cattle individuals including six Chinese indigenous cattle breeds and two non-native specialized beef cattle breeds. Based on the 11,486 CNVRs discovered, population analysis was performed, showed that all the cattle breeds clustered in to three clades, consistent with their lineages Bos taurus, Bos taurus × Bos indicus and Bos indicus. Importantly, a set of CNVRs related genes were found to be associated with the traits of interest, which include meat production or quality (CAST, ACTC1, etc.), adaption (BLA-DQB, EGLN2, etc.) and coat color (KIT, MITF, etc.). These results provide valuable full genome variation resources for Chinese bovine genome research and would be helpful for cattle breeding and selection programs in the future.

Alzheimer's disease (AD) is a complex and multifactorial neurodegenerative disorder of aged people. The development of multitarget-directed ligands (MTDLs) to act as multifunctional agents to treat this disease is the mainstream of current research. As a continuation of our previous studies, a series of 4-flexible amino-2-arylethenylquinoline derivatives as multi-target agents was efficiently synthesized and evaluated for the treatment of AD. Among these synthesized derivatives, some compounds exhibited strong self-induced Aß1⁻42 aggregation inhibition and antioxidant activity. The structure-activity relationship was summarized, which confirmed that the introduction of a flexible amino group featuring a N,N-dimethylaminoalkylamino moiety at the 4-position increased the Aß1⁻42 aggregation inhibition activity, with an inhibition ratio of 95.3% at 20 µM concentration. Compound 6b1, the optimal compound, was able to selectively chelate copper (II), and inhibit Cu2+-induced Aß aggregation effectively. It also could disassemble the self-induced Aß1⁻42 aggregation fibrils with a ratio of 64.3% at 20 µM concentration. Moreover, compound 6b1 showed low toxicity and a good neuroprotective effect against Aß1⁻42-induced toxicity in SH-SY5Y cells. Furthermore, the step-down passive avoidance test indicated compound 6b1 significantly reversed scopolamine-induced memory deficit in mice. Taken together, these results suggested that compound 6b1 was a promising multi-target compound worthy of further study for AD.
