RESUMO

With the 14-year enclosed grassland and the grazed grassland as control, the impacts of anthropogenic shrublands (Caragana korshinskii) with the different planting years (3, 12, 22 a) and planting spaces (2, 8, 40 m) on soil organic carbon (SOC) contents were examined in the desert steppe of Eastern Ningxia, China. We further analyzed the spatial pattern and heterogeneity of SOC in 0-40 cm soil layer of the grassland area with introduced shrubs. The results showed that SOC in C. korshinskii shrublands had an increase trend with increased planting years and decreased spaces. The mean SOC with different planting years and spaces was 42.7% and 32.8% more than that in grazing land, respectively. There was no significant difference of SOC between shrublands and the 14-year enclosed grassland. The increase trend of SOC decreased by 27.0% in 22-year planting shrubland. The SOC content of 0-40 cm soil layer varied from 0.21 g·kg-1 to 26.04 g·kg-1 (with a mean of 3.75 g·kg-1), and the coefficient of SOC variation ranged from 90.9% to 114.7%. The SOC in 0-5 cm and 15-40 cm soil layers fitted the optimal theory formulation of Gaussian model, while that in 5-15 cm soil layer fitted a spherical model. The ranges (A0) of spatial autocorrelation in the 0-5 and 5-15 cm soil layers were smaller (3.11, 3.00 km) than that in 15-40 cm soil layer (10.10 km). The nugget/sill C0/(C0+C) of SOC in 0-5, 5-15 cm soil layer was 0.2% and 16.3%, indicating a strong spatial correlation, while that in 15-40 cm soil layer was 36.9%, with a moderate correlation. The shrub introdution could significantly accelerate the accumulation and fixation of SOC in top 40 cm soil layer in degraded desert steppe, but also intensified the spatial heterogeneity and SOC fragmentation. The SOC content in the anthropogenic shrublands had no significant difference from that in the enclosed desert steppe (14 years). The SOC spatial heterogeneity and the degree of fragmentation were weakened and decreased with the increasing soil layer depth.

In performing our experiment, impaired autophagy increased hepatocellular damage during the reperfusion period. It was demonstrated by the effect of blocking autophagy using bafilomycin A1 or knocking Atg5 gene out reduces the anti-apoptotic effect of Stat3. Here we focus on the role of signal transducer and activator of transcription 3 (Stat3) in regulating autophagy to alleviate hepatic IRI. We found that Stat3 was up-regulated during hepatic IRI and was associated with an activation of the autophagic signaling pathway. This increased Stat3 expression, which was allied with high autophagic activity, alleviated liver damage to IR, an effect which was abrogated by Stat3 epletion as demonstrated in both in vivo and in vitro methods. The levels of Atg5 protein were decreased when Stat3 was inhibited by HO 3867 or siStat3. We conclude that Stat3 appeared to exert a pivotal role in hepatic IRI, by activating autophagy to alleviate hepatic IRI, and Atg5 was required for this process. The identification of this novel pathway, that links expression levels of Stat3 with Atg5-mediated autophagy, may provide new insights for the generation of novel protective therapies directed against hepatic IRI.

In this study, the thrombospondin-related adhesive protein (TRAP) gene and the heat shock protein 70 (Hsp70) gene of Babesia gibsoni isolated from the naturally infected dog in the Nanjing area were cloned and sequenced. Twenty blood samples were collected from the suspected cases of babesiosis at the animal hospital of Nanjing Agriculture University. Genomic DNA was extracted from the blood samples, and the 18S rDNAs were amplified by PCR to confirm Babesia infection. As a result, 10 cases of Babesia 18S rDNA gene amplification were positive in the 20 blood samples, confirming that the 10 cases were infected with canine Babesia. The TRAP and the Hsp70 gene fragments were amplified from all 10 positive cases. The four isolates, named NJN1, NJN2, NJN3 and NJN4, were sequenced and compared with other isolates in Asian. The similarity of TRAP and Hsp70 gene sequences among four isolates in Nanjing were above 99%. The Nanjing isolates were closely related to isolates from Taiwan and Japan. Indian isolates were different form Chinese and Japanese isolates, despite the very high similarity of the 18s rRNA genes sequence. These results suggest that the TRAP and Hsp70 genes have a reference value for the genetic diversity analysis of Babesia gibsoni.

RESUMO

Treatment of bacterial infections due to the fast emergence of drug-resistant bacteria is a significant challenge faced in modern medicine. Here the authors report a drug-induced self-assembly nanoantibiotic for treating bacterial infection, with temperature/pH-sensitivity, synergistic antibacterial effect of silver and antibiotics, and immunoregulatory effect. In this nanoantibiotic, smart polymer p(N-isopropylacrylamide-b-acrylic acid) triblock polymer (PNA) utilized to encapsulate the drugs provides convenience in preparing this structure simply through drug-induced self-assembly and controllable release profile by changing the sequence of addition of different drugs. The polymer also allows the nanoantibiotic to be responsive to multiple external stimuli such as pH, temperature, and ionic strength. The silver and antibiotics codelivered in this nanoantibiotic can exert a synergistic antibacterial effect due to the different antibacterial mechanisms. More importantly, macrophages can be activated into an M2 phenotype to promote tissue repair by this nanoantibiotic for the negative surface charge and the antibiotics contained. The self-assembly nanoantibiotic exhibited great promise to be applied in the treatment of bacterial infection and provide favorable utility for inflammation treatment, tissue engineering, and targeted therapy.

A 3 month study was conducted on the ruggedness of a multiresidue method for accuracy and stability. The results indicate that in terms of Youden pair ratios of 201 pesticide aged tea samples falling approximately within 1.00-1.20 of the ratio of theoretical spraying concentrations, the differences do not exceed 5% for percentages made up by ratios of the fixed values obtained by two kinds of instruments for two teas and those made up by 18 circular determinations. However, regarding two kinds of SPE cartridges, the Cleanert TPT cartridge is higher than the ENVI-CARB+primary secondary amine (PSA) cartridge by 10%. Pertaining to RSD values of "parallel samples" and whether it is green tea or Woolong tea, the percentages of RSD≤15% values of the parallel samples all exceed 88%. Whether it is the first or circular determination for two teas and analytical results from two kinds of instruments, the percentages of RSD≤15% values have a difference of less than 6%, while the TPT cartridge is better than ENVI-CARB+PSA by above 6% for the two cartridges. Concerning RSDs of Youden pair ratios, RSD≤15% values have a proportion exceeding 85% for both green tea and Woolong tea, and the percentage is greater than 87% whether it is for two kinds of SPE cartridges or two kinds of instruments. In terms of Youden pair ratios and the classified statistical analysis of the ruggedness data of parallel samples, the proportion of RSD≤15% values of Youden pair ratios is 8% higher for the TPT cartridge than the ENVI-CARB+PSA cartridge; the proportion of RSD≤15% values of parallel samples is 6.2% higher for the TPT cartridge than the ENVI-CARB+PSA cartridge. Data show no marked differences for two teas and two kinds of instruments. A comparison of the aforementioned aspects finds that good ruggedness was obtained with both SPE cleanup methods, and the results from the TPT cartridge are better than those from the ENVI-CARB+PSA cartridge.

BACKGROUND: To provide a point of reference to study the epidemiology and clinical expression of canine babesiosis in China. METHODS: A total of 30 dogs infected with canine babesiosis were evaluated by mean of clinical history, physical examination, hematological, restriction fragment length polymorphism of PCR products (PCR-RFLP) and sequencing analysis. RESULT: The most prevalent clinical abnormalities were lethargy (100%), anorexia (100%), pale or icteric mucous membranes (80%), fever (70%) and dark urine (70%). Hematology parameters revealed that anemia and thrombocytopenia were the major abnormalities in blood of dogs infected with canine babesia. The results of PCR-RFLP and sequencing analysis indicated that B. gibsoni was the main species responsible for canine babesiosis cases at the time of the study in Nanjing, China. CONCLUSIONS: The results provide valuable information for better understanding of the epidemiology of canine babesiosis in China.

A comparative study was conducted over three stages on the cleanup efficiency of SPE cartridge Cleanert TPT, newly developed for multigroups of pesticide residues in tea. In Stage I, different SPE cartridges C18, graphite carbon black (GCB), primary secondary amine (PSA), and amino (NH2) were purchased and combined into 12 different sequences. Through the comparative test on cleanup efficiency of 84 representative pesticides in tea, Envi-Carb GCB + PSA with a good cleanup effect was selected. In Stage II, GC/MS test results from the comparative study of the extraction efficiency of 201 pesticides spiked into green tea and Woolong tea with Cleanert TPT and Envi-Carb + PSA SPE showed that average recoveries fell within 70-110% and RSD <20% for 193 and 184 pesticides, respectively, for green tea, accounting for 96.0 and 91.0% of the total number, respectively. GC/MS/MS test results also found 193 and 184 pesticides, respectively, meeting the recovery and RSD conditions, accounting for 96.0 and 91.5%, respectively, of the total number. For Woolong tea samples, GC/MS results showed that with Cleanert TPT and Envi-Carb + PSA SPE for cleanup, there were 192 and 177 pesticides, respectively, meeting the conditions, accounting for 95.5 and 88.1% of the total number, respectively. GC/MS/MS results demonstrated that there were 195 and 184 pesticides, respectively, meeting the conditions, accounting for 97.0 and 91.5% of the total number, respectively. It was seen that Cleanert TPT was superior to Envi-Carb + PSA in cleanup efficiency, whether for green or Woolong tea samples, or GC/MS or GC/MS/MS determination. In Stage III, 61104 results of the average content value of pesticides and RSD (two teas xtwo Youden pair concentrations x two kinds of SPE cartridges x two instruments x 19 tests x 201 pesticides) were derived from the 19 times stability tests over 3 months by paralleling three samples every 5 days via two instruments with two kinds of SPE cartridges for cleanup, respectively, against Youden Pair samples of the 201 incurred pesticides from green and Woolong teas. The statistical analysis found that detected values from the target pesticides of the incurred Youden pair samples showed no marked differences with cleanup by either Cleanert TPT or Envi-Carb + PSA, whether for green or Woolong tea, or G/IMS or G/IM/IMS. The test results using the two aforementioned kinds of SPE cleanup for above 93% pesticides had a tolerance less than 15%, which testifies that both cartridge cleanups met the requirement for pesticide residue analysis.

RESUMO

A canine influenza virus (CIV) strain of avian origin designated A/Canine/Jiangsu/06/2010 (H3N2) was isolated from dogs exhibiting severe respiratory disease in Jiangsu, China. We announce the complete genome sequence of this viral strain and report major findings from the genomic analysis. This sequence will help us understand the molecular characteristics and evolutionary of H3N2 CIV in China.

RESUMO

Sodium hypochlorite digestion (NaClo) method and enzyme method were compared in the hatching of Taenia solium eggs. Both methods are effective, but the sodium hypochlorite digestion shows higher hatching rate (96.4%) and viability (89.0%) in a shorter reaction time (< 5 min) without needing expensive reagents.
