RESUMO

In this study, 13 previously undescribed acorane sesquiterpenoids, namely, proliferacorins A-M, were isolated from the solid fermentation of Fusarium proliferatum. Their structures and absolute configurations were confirmed via spectroscopic analyses, quantum-chemical NMR calculations with DP4+ probability analyses, ECD calculations and comparisons, and single-crystal X-ray diffraction techniques. Proliferacorins A-E (1-5) have a 7-oxa-tricyclo[6.3.1.01,5]tridecane decorated with a rare ether bridge between C-7 and C-11, while proliferacorin F (6) possesses a 7-oxa-tricyclic[6.4.0.01,5]dodecane skeleton with an unusual ether bond between C-6 and C-11. Proliferacorins C and D (3 and 4) are a pair of isomers at the carbon bridge between C-5 and C-7, whereas proliferacorins H and I (8 and 9) are a pair of spiro carbon isomers. All isolates were tested for their cytotoxic, anti-inflammatory, and immunosuppressive activities.

RESUMO

Ten new compounds, including three pairs of diarylcyclopentenone enantiomers (±) talaromycesins A-C (1-3) and four biphenyl derivatives talaromycesins D-G (4-7), along with four known compounds (8-11), were isolated from the fungus Talaromyces adpressus. Their structures were determined by analyses of extensive NMR spectroscopic and HRESIMS data, and their absolute configurations were elucidated by the dimolybdenum tetraacetate [Mo2(AcO)4]-induced ECD spectra, X-ray crystallographic studies, and ECD calculations. These new compounds were evaluated for their immunosuppressive activities for the first time, and compound 7 probably exerted liver-protective and anti-inflammatory effects on Con A-induced AIH by decreasing the levels of inflammatory cytokines, modulating immune homeostasis, and decreasing hepatocyte apoptosis, which may become a potential drug for the treatment of autoimmune diseases.

Five pairs of undescribed enantiomeric α-pyrone derivatives (±)-adprepyrones A-E (±1-±5), together with an unreported congener adprepyrone F (6), and 6-[(E)-3-Hydroxyprop-1-enyl]-4-methoxy-5-methyl-2-pyrone (7), recently reported as synthetic compound, were isolated from the fungus Talaromyces adpressus. Their structures with absolute configurations were elucidated by HRESIMS, 1D and 2D NMR, electronic circular dichroism calculations, and single-crystal X-ray diffraction analyses. (±)-Adprepyrone A (±1) possesses an unreported carbon skeleton formed by the fusion of an α-pyrone derivative with nicotinamide. Compounds (+)-2, (±)-4, (±)-5, and 7 showed moderate inhibitory activity against concanavalin A (ConA)-induced T lymphocyte proliferation with IC50 values ranging from 8.9 to 19.8 µM.

The main objective of this study was to investigate the influence of Serendipita indica on the growth of Tartary buckwheat plants. This study highlighted that the roots of Tartary buckwheat can be colonized by S. indica and that this fungal endophyte improved plants height, fresh weight, dry weight, and grain yield. In the meantime, the colonization of S. indica in Tartary buckwheat leaves resulted in elevated levels of photosynthesis, plant hormone content, antioxidant enzyme activity, proline content, chlorophyll content, soluble sugars, and protein content. Additionally, the introduction of S. indica to Tartary buckwheat roots led to a substantial rise in the levels of flavonoids and phenols found in the leaves and seeds of Tartary buckwheat. In addition, S. indica colonization reduced the content of malondialdehyde and hydrogen peroxide when compared to non-colonized plants. Importantly, the drought tolerance of Tartary buckwheat plants is increased, which benefits from physiology and bio-chemical changes in plants after S. indica colonized. In conclusion, we have shown that S. indica can improve systematic resistance and promote the growth of Tartary buckwheat by enhancing the photosynthetic capacity of Tartary buckwheat, inducing the production of IAA, increasing the content of secondary metabolites such as total phenols and total flavonoids, and improving the antioxidant enzyme activity of the plant.

Bacterial growth, under laboratory conditions or in a natural environment, goes through different growth phases. Some gene expressions are regulated with respect to the growth phase, which allows bacteria to adapt to changing conditions. Among them, many gene transcriptions are controlled by RpoHI or RpoHII in Rhodobacter sphaeroides. In a previous study, it was proven that the alternative sigma factors, RpoE, RpoHI, and RpoHII, are the major regulators of oxidative stress. Moreover, the growth of bacteria reached a stationary phase, and following the outgrowth, rpoE, rpoHI, and rpoHII mRNAs increased with respect to the growth phase. In this study, we demonstrated the regulatory function of alternative sigma factors in the rsp_0557 gene. The gene rsp_0557 is expressed with respect to the growth phase and belongs to the RpoHI/RpoHII regulons. Reporter assays showed that the antisigma factor ChrR turns on or over the RpoE activity to regulate rsp_0557 expression across the growth phase. In the exponential phase, RpoHII and sRNA Pos19 regulate the expression of rsp_0557 to an appropriate level under RpoE control. In the stationary phase, RpoHI and Pos19 stabilize the transcription of rsp_0557 at a high level. During outgrowth, RpoHI negatively regulates the transcription of rsp_0557. Taken together, our data indicate that these regulators are recruited by cells to adapt to or survive under different conditions throughout the growth phase. However, they still did not display all of the regulators involved in growth phase-dependent regulation. More research is still needed to learn more about the interaction between the regulators and the process of adapting to changed growth conditions and environments.

Nine new ergosteroids (1-9) and seven known ones (10-16) were isolated from Talaromyces adpressus. Their structures and absolute configurations were determined by the interpretation of NMR spectroscopic data, HRESIMS, ECD calculations, and single-crystal X-ray diffraction analyses. Structurally, compound 1 was an ergosteroid with two epoxy and a 3α-OH group at ring A, while compounds 8 and 9 had a contracted ring A with a peroxy bridge between C-3 and C-9, which were reported for the first time. Compounds 2-6, 9, 11, and 15 displayed cytotoxic activities with IC50 values ranging from 0.4 to 32 µM, and compound 7 exhibited an immunosuppressive effect against LPS-induced B lymphocyte proliferation with an IC50 value of 8.6 µM. The structure-activity relationships of these compounds are briefly discussed.

Talarolactones A-G (1-7), seven novel α-pyrone adducts with unprecedented scaffolds, together with two pairs of α-pyrone monomers [(±)-8 and (±)-9)] were isolated from Talaromyces adpressus. Compounds 1-7 are highly modified α-pyrone dimers with a 4,7,7,8-tetrasubstitued 5,6,7,8-tetrahydro-2H-chromen-2-one. Compounds 5 and 6 possessed a significant NO production inhibitory effect, with IC50 values of 2.3 ± 0.1 and 3.7 ± 0.3 µM, respectively. Plausible biosynthetic pathways were proposed and supported by the results of heterologous expression experiments.

Six undescribed caryophyllene sesquiterpenoids named pestalotiopsins O-T, along with eight known analogues, were obtained from the fungus Pestalotiopsis chamaeropis. Their structures and absolute configurations were assigned by NMR spectroscopic analyses, HRESIMS, single-crystal X-ray diffraction, electronic circular dichroism (ECD) calculations, Mo2(OAc)4-induced ECD, and chemical derivatization. Pestalotiopsin P represents the first example of a caryophyllene sesquiterpenoid possessing an oxatricyclo [7.2.2.03.6]tridecane decorated with a rare bridgehead double bond, while pestalotiopsin Q has an oxatricyclic [6.3.1.01,4]dodecane skeleton with an unusual ether bridge between C-1 and C-5. These undescribed caryophyllene sesquiterpenoids were screened for their cytotoxic and anti-inflammatory activities.

Five undescribed sterol derivatives, (22E,24R)-7α-methoxy-5α,6α-epoxyergosta-8(14),22-diene-3ß,15ß-diol, (22E,24R)-5α,6α-epoxyergosta-8(14),22-diene-3ß,7ß,15α-triol, (22E,24R)-3ß,5α-dihydroxy-14ß,15ß-epoxyergosta-7,22-diene-6-one, (22E,24R)-6α-methoxy-7α,15ß-dihydroxyergosta-4,8(14),22-triene-3-one, and (25S)-ergosta-7,24(28)-diene-3ß,4α,6α,26-tetraol were isolated from the extract of Talaromyces stipitatus, along with eight known congeners. This is the first example of a class of ergosterols isolated from T. stipitatus. Their structures with absolute configurations were elucidated based on NMR spectroscopic data, ECD calculations, and X-ray crystallographic analyses. All these compounds were tested for their effects on three hepatoma cell lines including Hep3B, HepG2, and Huh-7. Moreover, (22E,24R)-5α,6α-epoxyergosta-8(14),22-diene-3ß,7ß,15α-triol and (22E,24R)-9α,15α-dihydroxyergosta-4,6,8(14),22-tetraen-3-one were further evaluated for their impacts on cell cycle progression and apoptosis due to their pronounced cytotoxicity, to uncover their underlying mechanisms. Our results suggested that their antiproliferative activities were mainly mediated by inducing cell apoptosis.

Seven undescribed isopimarane diterpene glycosides hypoxylonoids A-G, along with five known analogues were obtained from the fungus Xylaria hypoxylon. The structures and absolute configurations of hypoxylonoids A-G were confirmed by extensive spectroscopic and single-crystal X-ray diffraction analyses. Among these compounds, the γ-lactone moiety formed between C-19 and C-6 of hypoxylonoid A; the 1,2-methyl shift of Me-18 of hypoxylonoids B and E; and the decarboxylation of C-19 of hypoxylonoid E, make them outstanding from the isopimarane family. Single crystal X-ray diffraction analyses of hypoxylonoids A, C, F, and 15-hydroxy-16-α-D-mannopyranosyloxyisopimar-7-en-19-oic acid was performed to determine their absolute structural configuration.
