RESUMO

Utilization of female livestock can be optimized by application of Fixed-Time Artificial Insemination (FTAI), which plays an important role in large-scale livestock production. However, molecular mechanism of FTAI affecting reproductive performance remains unclear. To investigate the effects of FTAI on corpus luteum in 16 and 25 days of pregnancy gilts, 12 pregnancy gilts were selected from Altrenogest + PMSG + GnRH (APG) group and control group. The number of left and right CL in APG group were significantly higher than control (p < 0.05). Furthermore, result showed the number of differentially expressed genes between 16APG group and 16 C group was obviously larger than that between 25APG and 25 C group (2394 vs. 1476). Up regulated genes in APG were mainly associated with cytokine production and secretion, cell communication and transport (day 16) and angiogenesis, cell cycle and cell-cell signaling (day 25). The expression of differentially expressed genes (RPL10, CYP17A1, DCN, IL18, LDLR and PRLR) in luteal tissue were verified by real-time PCR. In summary, APG program significantly improve reproductive efficiency of gilts through up regulation of cytokine production/secretion, cell communication and transport in Day 16 pregnancy and angiogenesis, cell cycle and cell-cell signaling at Day 25 pregnancy in porcine.

RESUMO

Intrauterine cigarette smoke exposure(ICSE)refers to a condition under which pregnant women actively inhale and/or passively inhale cigarettes and smogs mostly consisting of nicotine, tar and carbon monoxide during the first, second and third trimesters of pregnancies.As an adverse environmental factor in early life, ICSE is associated with increased risks of various diseases at childhood and adulthood.ICSE is closely related with children′s cognitive and behavioral developmental disorders.It has been reported that ICSE led to elevated risks of cognitive impairments including disorders in fine motor skills, language and vocabulary, reading comprehension, matrix reasoning, working memory, learning skills, orientation and other capabilities.Additionally, it is associated with increased risks of attention deficit hyperactivity disorder and sleep disorder, but the association with autism spectrum disorder is still controversial.The review focuses on the effects of ICSE on cognitive and behavioral development, and summarizes the underlying mechanisms, providing more clinical ideas for etiological studies of cognitive and behavioral developmental disorders in children.

Objective:To study the effects of hydrogen molecule on the formation of pelvic and abdominal adhesions in dogs, and to explore its possible mechanism.Methods:The Beagle pelvic and abdominal adhesion model was constructed. 15 Beagle dogs were randomly divided into 3 groups: model control group, hydrogen treatment group and blank control group. The Phillips adhesion score was used to evalute the morphological adhesion degree. The morphology of intestinal, uterus and ovarian tissues was evaluated by hematoxylin-eosin (HE) staining. The inflammation biomarkers and oxygen free radical index were detected before modeling and on the 1st, 4th, 7th and 14th day after modeling. C-reactive protein (CRP), tumor necrosis factors-α (TNF-α) and interleukin(IL)-6 content in serum were detected by enzyme-linked immunosorbent assay (ELISA) kit; hydroxylamine method was used to detect the serum superoxide dismutase (SOD); thiobarbituric acid (TBA) colorimetric method was used to measure malondialdehyde concentration.Results:The Phillips adhesion score of the model control group was significantly higher than that of the blank control group ( P<0.05). The Phillips adhesion score of the hydrogen treatment group was significantly lower than that of the model control group ( P<0.05). In the hydrogen treatment group, the thickening of serous layer on the surface of intestine, uterus and ovary and the degree of inflammatory infiltration were lower than those in the model control group.The inflammatory indexes (leukocyte count, CRP, IL-6, TNF-α) and malondialdehyde level in the model control group were higher than those in the blank control group on the first day after modeling, and the superoxide dismutase (SOD) level was lower than that in the blank control group (all P<0.05); the inflammatory indexes and malondialdehyde level in the hy-drogen treatment group were lower than those in the model control group on the first day after modeling ( P<0.05), and the SOD level was higher than that in the model control group ( P<0.05). Conclusions:Hydrogen molecule can effectively inhibit the formation of pelvic adhesion in Beagle dogs, and its possible mechanism is to neutralize oxygen free radicals and reduce inflammatory reaction.

This study investigated the effects of a CoCl2-simulated hypoxic environment on the muscle fiber switching signaling pathways calcineurin A/nuclear factor of activated T cells cytoplasmic 1 (CnA/NFATc1) and myostatin. In this study, C2C12 muscle cells were cultured in vitro under CoCl2-simulated chemical hypoxic conditions, the expression levels of CnA and myostatin were detected through qRT-PCR and Western blot analyses, and a positioning study of NFATc1 was carried out by immunofluorescence labeling. Results showed that CoCl2 treatment significantly increased the expression levels of CnA and myostatin. Moreover, the position of NFATc1 expression changed; actually, its expression in the nucleus considerably increased. Furthermore, CoCl2-induced hypoxia inhibited the differentiation of C2C12 cells and reduced the expression levels of many slow- and fast-twitch muscles marker genes, but immunofluorescence staining results showed that the proportion of MyHC I type muscle fiber increased after CoCl2 treatment. The hypoxic environment simulated by CoCl2 can activate the signaling pathways CnA/NFATc1 and myostatin and increases the proportion of MyHC I type muscle fibers.

Objective To investigate the effects of general anesthesia and hard epidural anesthesia on the incidence of postoperative cognitive dysfunction (POCD),serum S-100β and Aβ1-42 protein in elderly patients with transurethral resection of the prostate (TURP).Methods 120 cases of elderly male patients who wanted to implement TURP were enrolled in this study.From March 2014 to August 2016,60 patients underwent general anesthesia (general anesthesia group) and 60 patients underwent epidural anesthesia (hard epidural Group).The effects of two anesthesia methods on the cognitive function,serum S-100 β and Aβ1-42 protein were compared.Results There was no significant difference in mini-mental state examination (MMSE) score in preoperative,postoperative 12 h,postoperative 24 h,postoperative 72 h,and postoperative 1 week between hard epidural group and general anesthesia group (P > 0.05).The MMSE scores at 12 h,24 h and 72 h after operation in both groups were significantly lower than those before operation in both groups (P <0.05).There was no significant difference in clock drawing task (CDT) score in preoperative,24 h after operation,72 h after operation and one week after operation (P > 0.05).The CDT scores of both groups at 12 h,24 h,72 h after operation were significantly lower than those before operation (P <0.05).There was no significant difference in serum S-100β levels between the two groups at preoperative and 12 h,72 h after operation (P >0.05).Serum S-100β levels at 12 h and 72 h after surgery in both groups were significantly higher than those before surgery (P < 0.05).There was no significant difference in preoperative and postoperative 12 h,postoperative between hard epidural group and general anesthesia group (P > 0.05).Serum Aβ1-42 levels at preoperative,12 h and 72 h after operation in both groups were significantly lower than those before operation (P < 0.05).There was not statistically significant in the incidence of POCD between hard epidural group [28.33 % (17/60)] and general anesthesia group [35% (21/ 60)] (P > 0.05).Conclusions There was no significant difference in the incidence of POCD between general anesthesia and hard epidural anesthesia group in elderly patients with TURP.The incidence of POCD in elderly patients was related to the decrease of serum S-100β and the decrease of Aβ1-42.

A microwave digestion system was preparation for the digestion of the sediment samples of 16 stations in the Southe mid_Atlantic ridge by using HNO3_H2 O2_HF as the digestion reagent. The rare earth elements ( RE ) in sediments were determined by inductively coupled plasma mass spectrometry, and distribution characteristics of rare earth elements were studied. The microwave digestion_ICP_MS method was used for the determination of rare earth elements with a good linear relationship ( r=0 . 9997-1 . 0000 ) for each element. The detection limit reached ng/L level, the relative standard deviation ( RSD, n=3 ) was less than 3% and the relative error was 6%. The total amount of rare earth elements (ΣRE) in sediment samples from 16 stations varied in the range of 37. 25-134. 77 μg/g, the ratio range of light RE/heavy RE ( LRE/HRE) was 0 . 61-1 . 70 , the average value was 1 . 27 , and the enrichment of light rare earth elements in sediments was slightly obvious. The RE distribution patterns were basically the same in each station with obvious fractionation between LRE and HRE. The RE distribution patterns were also similar in sediments from different sources with slightly difference between terrestrial and marine sediments. The δEu and δCe in the sediments had negative anomaly which showed that the rare earth elements in sediments came from the seawater. This study first analyzed the content and distribution of rare earth elements in the southern Atlantic, providing data and technical support for further study of the distribution of rare earth elements in the Atlantic.

Objective To evaluate the usefulness of anti -HCV signal-to-cutoff (S/CO)ratio, AST, ALT and the combined examination of anti-HCV S/CO, AST, ALT for predicting HCV RNA results by a model of logistic regression and receiver -operating characteristic (ROC) curve.Methods Five hundred and eighty -eight anti-HCV positive samples were tested by ELISA , followed by RT-PCR to detect HCV-RNA and enzyme rate method to detect AST, ALT.Patients were divided into viremia and non -viremia groups according to HCV-RNA results.Logistic regression and ROC curve analysis was performed to evaluate the diagnostic accuracy of each index for a diagnosis of viremia.Results The serum anti-HCV S/CO ratio, AST, ALT of HCV-RNA positive group were higher than HCV-RNA negative group, showing significant statistical difference ( P AST >ALT.The Area Under Curve(AUC) of the combined examina-tion of anti-HCV S/CO ratio, AST and ALT was 0.949(95% confidence interval,0.932 to 0.966), higher than the AUC of anti-HCV S/CO ratio, AST and ALT single index examinations , which was 0.894(95 % confidence interval, 0.862 to 0.926), 0.823(95%confidence interval, 0.789 to 0.856) and 0.788(95% confidence interval, 0.750 to0.826 ) respectively.C onclusions The diag-nostic relevance of the three biochemical markers for predicting the presence of viremia was anti -HCV S/CO ratio >AST >ALT.The combined examination of anti-HCV S /CO ratio, AST and ALT in predicting hepatitis C viremia is superior to any single index examina -tion and it can increase the detecting ability of HCV -RNA greatly.

RESUMO

Fatty acid synthase (FAS) catalyses the reaction between acetyl-CoA and malonyl-CoA to produce fatty acids. It is one of the most important enzyme in lipid biosynthesis. FAS of the oleaginous yeast Rhodosporidium toruloides has two acyl carrier protein (ACP) domains and a distinct subunit composition compared with FASs of other species. As ACP is a protein cofactor crucial for fatty acid chain elongation, more ACPs in the FAS may facilitate the reaction. To study the biochemical and structural properties of this novel FAS from R. toruloides, plasmids were constructed and transformed into Escherichia coli BL21 (DE3). The strain ZWE06 harboring plasmids pET22b-FAS1 and pET24b-FAS2 could co-overexpress the two subunits. The recombinant FAS was purified by sequentially using ammonium sulphate precipitation, sucrose density gradient centrifugation and anion exchange chromatography. The specific activity of the recombinant FAS was 548 mU/mg. The purified complex would be used to study enzyme kinetics and protein structure of FAS, and heterogeneous expression and purification will facilitate revealing the mechanism of this novel FAS with double ACPs.

Tanshinones are the bioactive components of the Chinese medicinal herb Salvia miltiorrhiza, while its biosynthetic pathway remains to be characterized. Rapid identification and characterization of the genes correlated to tanshinones biosynthesis is very important. As one of the intermediates of tanshinones biosynthesis, the ferruginol content is relative low in both root and engineered bacteria. It is urgent to construct an efficient system for conversion of miltiradiene to ferruginol to obtain large amount of ferruginol as the substrates for further identifying other downstream genes involved in tanshinones biosynthesis. In this study, we constructed the whole-cell yeast biocatalysts co-expressing miltiradiene oxidase CYP76AH1 and cytochrome P450 reductases (SmCPR1) from Salvia miltiorrhiza, and then characterized it with RT-PCR. After permeabilization, the yeast whole-cell could catalyze turnover of miltiradiene to ferruginol efficiently through single-step biotransformation with a conversion efficiency up to 69.9%. The yeast whole-cell biocatalyst described here not only provide an efficient platform for producing ferruginol in recombinant yeast but also an alternative strategy for identifying other CYP genes involved in tanshinones biosynthesis.

Objective To investigate the correlation of father involvement with early social adaptation behaviors.Methods With social adaptation scale for infant-junior middle school' s student and a self-made questionnaire,1246 normal children,age from 16 to 72 months,were investigated.Regression analysis was performed to get the correlation of social adaptation with time and activities of father involved.Results After controlling for mother-,child- and family-related factors,the regression analysis showed that standard score of social adaptation was positively rehted with the time of father involved ( β =0.05,P=0.03 ),the time father invloved explain 12%of the variation of social adaptation.The father-child activities were related with social adaptation behavior,playing was related with work skills (β =0.09,P<0.01 ) and socialization ( β =0.06,P<0.05),story listening related with communication ( β =0.07,P < 0.05) and self-management ( β =0.06,P < 0.05 ),other activities related with self-dependence ( β =0.07,P<0.05) and work skills ( β =0.06,P<0.05).Conclusion The time and activities of father involved are related with children' s social adaptation.The effect of father involvement on early social adaptation is positive.

RESUMO

Objective To explore imaging fingdings of pigmented villonodular synovitis (PVS) so as to improve the diagnostic accuracy of PVS. Methods Imaging data of 12 patients with PVS confirmed by operation and pathology were analyzed retrospectively.12 cases were all examined with X-ray and MR imaging,5 cases examined with CT.Results In total 12 cases, swelling of soft tissue was seen in 8 cases on plain films.CT showed subcartilaginous bony erosion in 3 cases.MRI showed synovium nodular hyperplasia irregularly, deposition of paramagnetic hemosiderin-containing in all cases,and the invasion of ligament in 5 cases , menisci in 1 case and infrapatellar fat pad in 3 cases; subcartilaginous and cartilaginous erosion in 3 cases, joint effusion in 9 cases. Conclusion PVS has typical imaging characteristics,PVS can be diagnosed accurately with MRI.
