RESUMO

As a metal-free photocatalyst, graphitic carbon nitride (g-CN) shows great potential for photocatalytic water splitting, although its performance is significantly limited by structural defects due to incomplete polymerization. In the present work, we successfully synthesize highly conjugated g-CN nanofoam through an iodide substitution technique. The product possesses a high polymerization degree, low defect density, and large specific surface area; as a result, it achieves a hydrogen evolution rate of 9.06 mmol h-1 g-1 under visible light irradiation, with an apparent quantum efficiency (AQE) of 18.9% at 420 nm. Experimental analysis and theoretical calculations demonstrate that the recombination of photogenerated carriers at C-NHx defects was effectively depressed in the nanofoam, giving rise to the high photocatalytic activity.

Gas-liquid two-phase flow behavior in horizontal channel under heaving motion showed unique dynamic characteristics due to the complex nonlinear interaction. To further establish a description model and investigate the effects of heaving motion on horizontal gas-liquid flow, experiments in a wide range of vibration parameters and working conditions were carried out by combining vibration platform with two-phase flow loop. It was found that the flow regimes under heaving motion showed significant differences compared to the ones expected in steady state flow under the same working conditions. Increasing vibration parameters showed an obvious impact on fluctuation degree of gas-liquid interface by visualizing high-speed photographs. A method based on multi-scale entropy was applied to identify flow regimes and reveal the underlying dynamic characteristics by collecting signals of pressure-difference. The results indicated that the proposed method was effective to analyze gas-liquid two-phase flow transition in horizontal channel under heaving motion by incorporating information of flow condition and change rate of multi-scale entropy, which provided a reliable guide for flow pattern control design and safe operation of equipment. However, for slug-wave and boiling wave flow, an innovative method based on multi-scale marginal spectrum entropy showed more feasible for identification of transition boundary.

The model of high solid anaerobic digestion was used by improving the degree of homogeneity of the reaction system and biogas slurry reflux to gradually increase the material load. The vinegar residue-efficient anaerobic digestion system was successfully constructed without pretreatment.The optimum anaerobic digestibility was observed when the material loading of the reaction system reached 6.15 g·(L·d)-1, when the amount of biogas produced per unit of dry material was 396 mL·g-1, and the amount of methane produced per unit of dry material was 211 mL·g-1. The degradation rate of hemicellulose reached 63.66%, which was the main reason for the improvement of anaerobic digestion performance. The degradation rates of cellulose and lignin were 21.46% and 24.43%, respectively. The lower degradation efficiency was mainly due to the complicated degradation of the benzene ring structure in lignin and hindered hydrolysis of cellulose, which had a shielding effect on cellulose degradation.

OBJECTIVE: To investigate the effect of the spinal cord extracts (SCE) after spinal cord injuries (SCIs) on the proliferation of rat embryonic neural stem cells (NSCs) and the expressions of mRNA of Notch1 as well as of Hes1 in this process in vitro. METHODS: The experiment was conducted in 4 different mediums: NSCs+PBS (Group A-blank control group), NSCs+SCE with healthy SD rats (Group B-normal control group), NSCs+SCE with SD rats receiving sham-operation treatment (Group C-sham-operation group) and NSCs+ SCE with SCIs rats (Group D-paraplegic group). Proliferative abilities of 4 different groups were analyzed by MTT chromatometry after co-culture for 1, 2, 3, 4 and 5 d, respectively. The expressions of Notch1 and Hes1 mRNA were also detected with RT-PCR after co-culture for 24 and 48 h, respectively. RESULTS: After co-culture for 1, 2, 3, 4 and 5 d respectively, the MTT values of group D were significantly higher than those of group A, group B and group C (P<0.05). However, there were no significantly differences regarding MTT values between group A, group B and group C after co-culture for 1, 2, 3, 4 and 5 d, respectively (P>0.05). Both the expressions of Notch1 and Hes1 mRNA of group D were significantly higher than those of other 3 groups after co-culture for 24 h and 48 h as well (P<0.05). But there was no difference oin expressions of Notch1 and Hes1 mRNA among group A, group B and group C after co-culture for 24 h and 48 h (P>0.05). There was no difference in expressions of Notch1 and Hes1 mRNA between 24 h and 48 h treatment in group D. CONCLUSIONS: SCE could promote the proliferation of NSCs. It is demonstrated that the microenvironment of SCI may promote the proliferation of NSCs. Besides, SCE could increase the expression of Notch1 and Hes1 mRNA of NSC. It can be concluded that the Notch signaling pathway activation is one of the mechanisms that locally injured microenvironment contributes to the proliferation of ENSC after SCIs. This process may be performed by up-regulating the expressions of Notch1 and Hes1 gene.

AIM: Gambogic acid (GA) is the major active ingredient of gamboge, which is secreted from a Chinese traditional medicine, Garcinia hanburyi, which possesses potent antitumor activity. GA3, a new GA derivative, has been shown to possess better water solubility than GA. The aim of the present study was to examine the antitumor activity of GA3 and the mechanism underlying it. METHODS: The growth inhibition of cancer cell lines induced by GA3 was assessed using the SRB assay. DAPI staining, flow cytometry, a DNA fragment assay, and Western blot analysis were used to study the apoptotic mechanisms of GA3. RESULTS: GA3 displayed wide cytotoxicity in diversified human cancer cell lines with a mean IC(50) value of 2.15 micromol/L. GA3 was also effective against multidrug resistant cells, with an average resistance factor (RF) that was much lower than that of the reference drug, doxorubicin. Mechanistic studies revealed that GA3-induced apoptosis in HL-60 cells proceeded via both extrinsic and intrinsic pathways, with caspase-8 functioning upstream of caspase-9. In addition, GA3-driven apoptotic events were associated with up-regulation of Bax, down-regulation of Bcl-2 and cleavage of Bid. Moreover, GA3 triggered cytochrome c release from the mitochondria, in particular bypassing the involvement of the mitochondrial membrane potential. CONCLUSION: Better solubility and a potential anti-MDR activity, combined with a comparable antitumor efficacy, make GA3 a potential drug candidate in cancer therapy that deserves further investigation.
