RESUMO

The limited self-healing ability of cartilage necessitates the application of alternative tissue engineering strategies for repairing the damaged tissue and restoring its normal function. Compared to conventional tissue engineering strategies, three-dimensional (3D) printing offers a greater potential for developing tissue-engineered scaffolds. Herein, we prepared a novel photocrosslinked printable cartilage ink comprising of polyethylene glycol diacrylate (PEGDA), gelatin methacryloyl (GelMA), and chondroitin sulfate methacrylate (CSMA). The PEGDA-GelMA-CSMA scaffolds possessed favorable compressive elastic modulus and degradation rate. In vitro experiments showed good adhesion, proliferation, and F-actin and chondrogenic differentiation of bone marrow mesenchymal stem cells (BMSCs) on the scaffolds. When the CSMA concentration was increased, the compressive elastic modulus, GAG production, and expression of F-actin and cartilage-specific genes (COL2, ACAN, SOX9, PRG4) were significantly improved while the osteogenic marker genes of COL1 and ALP were decreased. The findings of the study indicate that the 3D-printed PEGDA-GelMA-CSMA scaffolds possessed not only adequate mechanical strength but also maintained a suitable 3D microenvironment for differentiation, proliferation, and extracellular matrix production of BMSCs, which suggested this customizable 3D-printed PEGDA-GelMA-CSMA scaffold may have great potential for cartilage repair and regeneration in vivo.

Vortioxetine is a potent antagonist of the 5-hydroxytryptamine receptor and serotonin transporter and has been reported to function as an antidepressant in the treatment of major depressive disorder. However, its antitumor effects remain unclear. Here, we examined whether vortioxetine affects the characteristics of GC cells. Cell viability was measured by a colony formation assay and, in addition, cell invasion, migration and apoptosis assays were performed with a transwell assay and a flow cytometry assay. Protein levels were measured by western blotting. We found that vortioxetine inhibited the proliferation, invasion and migration abilities of AGS cells. Additionally, vortioxetine could induce apoptosis and autophagy by increasing the levels of Bax, active caspase-3/-9, Beclin-1 and light chain 3, as well as by downregulating Bcl-2 and P62. Further investigations indicated that vortioxetine regulated apoptosis and autophagy via activation of the phosphoinositide 3-kinase/AKT pathway. Taken together, our data suggest that vortioxetine has cytotoxic effects against GC AGS cells as a result of inhibiting proliferation, invasion and migration, as well as by inducing apoptosis and autophagy through the phosphoinositide 3-kinase/AKT pathway.

The status of treatment equipment, the emission characteristics, and the ozone formation potential (OFP) of volatile organic compounds (VOCs) for 11 typical enterprises, which were categorized into the 8 major VOC emission industries identified by the emission inventory of a typical city in the Yangtze River Delta, are discussed in this paper. There was a large difference in the removal efficiency of non-methane hydrocarbon (NMHC) between different treatment techniques, and even an increase in concentration occurred after some of the treatments. The current treatment equipment for VOCs needs further optimization. The emissions of NMHC, benzene, toluene, and xylene in most of the surveyed enterprises exceeded their corresponding standards, with toluene the worst offender. The most abundant compounds in the eight emission industries were aromatic hydrocarbons and oxygenated VOCs, whereas aromatic hydrocarbons contributed the most to ozone formation potential. There were large differences in emission characteristics of VOCs from different industries. Priority should be placed on the industries that have large OFP when control strategies of VOCs are considered.

The present study was designed to culture and purify high proliferative potential endothelial progenitor cells (HPP-EPCs) from murine bone marrow and their progeny cells using a culture system which has been established by us recently, in order for more intensive researches on these cells. Fresh murine bone marrow cells were preplated to allow the preferential attachment of non-EPCs to tissue culture plates and then unattached cells were repreplated in the culture system containing the bone marrow endothelial cell line-conditioned medium (BMEC-CM). The colonies containing about 2 x 10(4) cells were collected respectively and single colony-derived cells were cultured for their expansion in the above-mentioned culture system. These single colonies were able to yield 8 x 10(6) progeny cells. The endothelial-lineage characteristics of expanded cells were confirmed by immunofluorescent staining for CD31 and vWF, FITC-UEA-1 binding and Dil-Ac-LDL uptake. These data suggest that murine bone marrow HPP-EPCs can proliferate exuberantly so that their progeny cells can be obtained with high yield by using the single colony-derived cell culture in the presence of BMEC-CM.

Resonance Raman spectra were obtained for 2,5-di-iodothiophene in cyclohexane solution with excitation wavelengths in resonance with its A-band absorption. These spectra indicate that the Franck-Condon region photodissociation dynamics have multidimensional character with motion mostly along the nominal symmetric C(2)C(3)C(4)C(5) stretch nu(2) (1388 cm(-1)) of the thienyl ring and this is accompanied by motions along the nominal symmetric CSC stretch nu(6) (640 cm(-1)), the nominal symmetric C[Single Bond]I stretch nu(7) (187 cm(-1)), the nominal antisymmetric CSC stretch nu(19) (728 cm(-1)), the nominal antisymmetric C(2)C(3)C(4)C(5) stretch nu(16) (1497 cm(-1)), the nominal C(3)C(4) stretch nu(3) (1286 cm(-1)) and four other vibrational modes. A preliminary resonance Raman intensity analysis was done and the results for 2,5-di-iodothiophene were compared to previous results for 2-iodothiophene and di-iodomethane to discuss the mechanism of the symmetric breaking in the initial A-band photodissociation dynamics of 2,5-di-iodothiophene and 2-iodothiophene.

This study was purpose to examine the effect of dimethyl sulfoxide (DMSO) and Tween 80 on the growth and viability of stromal cells (BMSC), colony-forming units for granulocytes and macrophages (CFU-GM) and bone marrow endothelial cell line (BMEC) from murine bone marrow in vitro, and to analyze the concentration-effect relationship. The colony yields of colony-forming units fibroblastic (CFU-F) and CFU-GM were assessed in the murine bone marrow cell cultures at various concentrations of DMSO or Tween 80 and in the control groups. The MTT assay and trypan blue exclusion were used to determine the cell viability and percentage of survival in BMSC and BMEC cultures with or without either of these organic solvents. The results showed that the colony yields of both CFU-F and CFU-GM were decreased significantly (p<0.05 or <0.01) at the concentrations (v/v final) of 2% DMSO or 0.005%-0.01% Tween 80 respectively, as compared with control. The cell viability and percentage of survival of BMSC and BMEC cultures were significantly reduced (p<0.05 or <0.01) at 0.5%-1.0% DMSO or 0.002%-0.005% Tween 80, as compared with control. With the increase of volume fractions of these solvents, the decreased percentages of corresponding measurements were increased by degrees. It is concluded that when the concentration of DMSO or Tween 80 goes to a certain level in cell culture medium, either of the organic solvents has an inhibitory action or/and cytotoxicity on the growth and viability of BMSCs, CFU-GM and BMECs. The growth inhibition and cytotoxic response are more significant at higher concentrations of these solvents.

Resonance Raman spectra were obtained for 2-iodothiophene in cyclohexane solution with excitation wavelengths in resonance with the A-band absorption spectrum. These resonance Raman spectra indicate that the Franck-Condon region photodissociation dynamics have multidimensional character with motion mainly along the nominal symmetric C=C stretch of the thienyl ring and accompanied by a moderate amount of motion along the nominal symmetric CSC stretch, the nominal antisymmetric CSC stretch, and the nominal C-I stretch vibrational modes. A preliminary resonance Raman intensity analysis was done for the A-band resonance Raman spectra of 2-iodothiophene. These results were compared to previous results for related iodobenzene and iodoalkane molecules that also contain a C-I chromophore and the similarities and differences in the short-time photodissociation dynamics were discussed.
